1Changes in intestinal mucosal immune barrier in rats with endotoxemia显示文摘AIM:To investigate the dysfunction of the immunological barrier of the intestinal mucosa during endotox-emia and to elucidate the potential mechanism of this dysfunction.METHODS:Male Wistar rats were randomly distributed into two groups:control group and lipopolysaccharide(LPS)group.Endotoxemia was induced by a single caudal venous injection of LPS.Animals were sacrifi ced in batches 2,6,12 and 24 h after LPS infusion.The number of microfold(M)-cells,dendritic cells(DCs),CD4+ T cells,CD8+ T cells,regulatory T(Tr)cells and IgA+ B cells in the intestinal mucosa were counted after immunohistochemical staining.Apoptotic lympho-cytes were counted after TUNEL staining.The levels of interleukin(IL)-4,interferon(IFN)-γ and forkhead box P3(Foxp3)in mucosal homogenates were measured by ELISA.The secretory IgA(sIgA)content in the total protein of one milligram of small intestinal mucus was detected using a radioimmunological assay.RESULTS:This research demonstrated that LPS-induced endotoxemia results in small intestinal mucosa injury.The number of M-cells,DCs,CD8+ T cells,and IgA+ B cells were decreased while Tr cell and apoptotic lymphocyte numbers were increased signifi cantly.The number of CD4+ T cells increased in the early stages and then slightly decreased by 24 h.The level of IL-4 significantly increased in the early stages and then reversed by the end of the study period.The level of IFN-γ increased slightly in the early stages and then decreased markedly by the 24 h time point.Level of Foxp3 increased whereas sIgA level decreased.CONCLUSION:Mucosal immune dysfunction forms part of the intestinal barrier injury during endotoxemia.The increased number and function of Tr cells as well as lymphocyte apoptosis result in mucosal immunodefi ciency.Chong Liu Ang Li Yi-Bing Weng Mei-Li Duan Bao-En Wang Shu-Wen Zhang 2009World Journal of Gastroenterology2009,15,46:35
2Lushan M_S7.0 earthquake:A blind reserve-fault event显示文摘In the epicenter of the Lushan MS7.0 earthquake there are several imbricate active reverse faults lying from northwest to southeast,namely the Gengda-Longdong,Yanjing-Wulong,Shuangshi-Dachuan and Dayi faults.Emergency field investigations have indicated that no apparent earthquake surface rupture zones were located along these active faults or their adjacent areas.Only brittle compressive ruptures in the cement-covered pavements can be seen in Shuangshi,Taiping,Longxing and Longmen Townships,and these ruptures show that a local crustal shortening occurred in the region during the earthquake.Combining spatial distribution of the relocated aftershocks and focal mechanism solutions,it is inferred that the Lushan earthquake is classified as a typical blind reverse-fault earthquake,and it is advised that the relevant departments should pay great attention to other historically un-ruptured segments along the Longmenshan thrust belt and throughout its adjacent areas.XU XiWei WEN XueZe HAN ZhuJun CHEN GuiHua LI ChuanYou ZHENG WenJun ZHNAG ShiMin REN ZhiQun XU Chong TAN XiBin WEI ZhanYu WANG MingMing REN JunJie HE ZhongTai LIANG MingJian 2013Chinese Science Bulletin2013,58,28:35
3AXL is a candidate receptor for SARS-CoV-2 that promotes infection of pulmonary and bronchial epithelial cells显示文摘The current coronavirus disease 2019(COVID-19)pandemic presents a global public health challenge.The viral pathogen responsible,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),binds to the host receptor ACE2 through its spike(S)glycoprotein,which mediates membrane fusion and viral entry.Although the role of ACE2 as a receptor for SARS-CoV-2 is clear,studies have shown that ACE2 expression is extremely low in various human tissues,especially in the respiratory tract.Thus,other host receptors and/or co-receptors that promote the entry of SARS-CoV-2 into cells of the respiratory system may exist.In this study,we found that the tyrosine-protein kinase receptor UFO(AXL)specifically interacts with the N-terminal domain of SARS-CoV-2 S.Using both a SARS-CoV-2 virus pseudotype and authentic SARS-CoV-2,we found that overexpression of AXL in HEK293T cells promotes SARS-CoV-2 entry as efficiently as overexpression of ACE2,while knocking out AXL significantly reduces SARS-CoV-2 infection in HI 299 pulmonary cells and in human primary lung epithelial cells.Soluble human recombinant AXL blocks SARS-CoV-2 infection in cells expressing high levels of AXL.The AXL expression level is well correlated with SARS-CoV-2 S level in bronchoalveolar lavage fluid cells from COVID-19 patients.Taken together,our findings suggest that AXL is a novel candidate receptor for SARS-CoV-2 which may play an important role in promoting viral infection of the human respiratory system and indicate that it is a potential target for future clinical intervention strategies.Shuai Wang Zongyang Qiu Yingnan Hou Xiya Deng Wei Xu Tingting Zheng Peihan Wu Shaofang Xie Weixiang Bian Chong Zhang Zewei Sun Kunpeng Liu Chao Shan Aifu Lin Shibo Jiang Youhua Xie Qiang Zhou Lu Lu Jing Huang Xu Li 2021Cell Research2021,31,2:39

14Roles of neural stem cells in the repair of peripheral nerve injury显示文摘Currently, researchers are using neural stem cell transplantation to promote regeneration after peripheral nerve injury, as neural stem cells play an important role in peripheral nerve injury repair. This article reviews recent research progress of the role of neural stem cells in the repair of peripheral nerve injury. Neural stem cells can not only differentiate into neurons, astrocytes and oligodendrocytes, but can also differentiate into Schwann-like cells, which promote neurite outgrowth around the injury. Transplanted neural stem cells can differentiate into motor neurons that innervate muscles and promote the recovery of neurological function. To promote the repair of peripheral nerve injury, neural stem cells secrete various neurotrophic factors, including brain-derived neurotrophic factor, fibroblast growth factor, nerve growth factor, insulin-like growth factor and hepatocyte growth factor. In addition, neural stem cells also promote regeneration of the axonal myelin sheath, angiogenesis, and immune regulation. 16Insights on the solubilization products after combined alkaline and ultrasonic pre-treatment of sewage sludge显示文摘This work provides insights on the solubilization products after a simultaneous combination of alkaline and ultrasonic(ALK + ULS) pre-treatment of sewage sludge.Soluble chemical oxygen demand(SCOD) increased from 1200 to 11,000 mg/L after such treatment. Organics with molecular weight around 5.6 k Da were solubilized because of the synergistic effect of ultrasound and alkali. Organics with molecular weight larger than300 k Da increased from 7.8% to 60%, 16% and 42.3% after ULS, ALK and ALK + ULS treatment, respectively. Excitation emission matrix fluorescence spectroscopy analysis identified soluble microbial product-like and humic acid-like matters as the main solubilization products. Sludge anaerobic biodegradability was significantly enhanced with the simultaneous application of ALK + ULS pre-treatment. ALK + ULS pre-treatment resulted in 37.8% biodegradability increase compared to the untreated sludge. 17Long-term outcomes of a phase Ⅱ randomized controlled trial comparing intensity-modulated radiotherapy with or without weekly cisplatin for the treatment of locally recurrent nasopharyngeal carcinoma显示文摘Background:Salvage treatment for locally recurrent nasopharyngeal carcinoma(NPC) is complicated and relatively limited.Radiotherapy,combined with effective concomitant chemotherapy,may improve clinical treatment outcomes.We conducted a phase Ⅱ randomized controlled trial to evaluate the efficacy of intensity-modulated radiotherapy with concomitant weekly cisplatin on locally recurrent NPC.Methods:Between April 2002 and January 2008,69 patients diagnosed with non-metastatic locally recurrent NPC were randomly assigned to either concomitant chemoradiotherapy group(n = 34) or radiotherapy alone group(n = 35).All patients received intensity-modulated radiotherapy.The radiotherapy dose for both groups was 60 Gy in 27 fractions for 37 days(range 23-53 days).The concomitant chemotherapy schedule was cisplatin 30 mg/m^2 by intravenous infusion weekly during radiotherapy.Results:The median follow-up period of all patients was 35 months(range 2-112 months).Between concomitant chemoradiotherapy and radiotherapy groups,there was only significant difference in the 3-year and 5-year overall survival(OS) rates(68.7%vs.42.2%,P = 0.016 and 41.8%vs.27.5%,P = 0.049,respectively).Subgroup analysis showed that concomitant chemoradiotherapy significantly improved the 5-year OS rate especially for patients in stage rT3-4(33.0%vs.13.2%,P = 0.009),stages Ⅲ-Ⅳ(34.3%vs.13.2%,P = 0.006),recurrence interval >30 months(49.0%vs.20.6%,P = 0.017),and tumor volume >26 cm^3(37.6%vs.0%,P = 0.006).Conclusion:Compared with radiotherapy alone,concomitant chemoradiotherapy can improve OS of the patients with locally recurrent NPC,especially those with advanced T category(rT3-4) and stage(lll-IV) diseases,recurrence intervals >30 months,and tumor volume >26 cm^3.Ying Guan Shuai Liu Han-Yu Wang Ying Guo Wei-Wei Xiao Chun-Yan Chen Chong Zhao Tai-Xiang Lu Fei Han 2016Chinese Journal of Cancer2016,35,4:19

19Chemical constituents,biological functions and pharmacological effects for comprehensive utilization of Eucommia ulmoides Oliver显示文摘Eucommia ulmoides Oliver is a native plant and valuable tonic Chinese medicine in China with a long history,great economic value and comprehensive development potential.Traditionally,the comprehensive utilization rate of E.ulmoides Oliv.is still very low,only bark has been used as medicine and other parts of Eucommia ulmoides Oliv.cannot be fully utilized,even the leaves have been well utilized in food products in Japan in the past decades.In order to improve the comprehensive utilization efficiency of E.ulmoides Oliv.,in this review,we summarized the varieties and contents of main active compounds,biological functions and pharmacological effects in different parts of E.ulmoides Oliv.The findings suggest that other parts of E.ulmoides Oliv.could replace the bark of E.ulmoides Oliv.to some extent besides of their respective applications.The unique and extensive physiological functions between different parts of E.ulmoides Oliv.indicate that the comprehensive utilization of E.ulmoides Oliv.has a wide space to develop,which is also an effective way to protect E.ulmoides Oliv.resources and improve its the utilization rate.Yi-Fan Xing Dong He Yi Wang Wen Zeng Chong Zhang Yuan Lu Nan Su Yan-Hua Kong Xin-Hui Xing 2019Food Science and Human Wellness2019,8,2:18
20Applications of HPLC/MS in the analysis of traditional Chinese medicines显示文摘In China,traditional Chinese medicines(TCMs)have been used in clinical applications for thousands of years.The successful hyphenation of high-performance liquid chromatography(HPLC)and mass spectrometry(MS)has been applied widely in TCMs and biological samples analysis.Undoubtedly,HPLC/MS technique has facilitated the understanding of the treatment mechanism of TCMs.We reviewed more than 350 published papers within the last 5 years on HPLC/MS in the analysis of TCMs.The present review focused on the applications of HPLC/MS in the component analysis,metabolites analysis,and pharmacokinetics of TCMs etc.50% of the literature is related to the component analysis of TCMs,which show that this field is the most popular type of research.In the metabolites analysis,HPLC coupled with electrospray ionization quadrupole time-of-flight tandem mass spectrometry has been demonstrated to be the powerful tool for the characterization of structural features and fragmentation behavior patterns.This paper presented a brief overview of the applications of HPLC/MS in the analysis of TCMs.HPLC/MS in the fingerprint analysis is reviewed elsewhere.Miao Li Xiao- Fang Hou Jie Zhang Si-Cen Wang Qiang Fu Lang- Chong He 2011Journal of Pharmaceutical Analysis2011,1,2:17
