1Erianin,a novel dibenzyl compound in Dendrobium extract,inhibits lung cancer cell growth and migration via calcium/calmodulin-dependent ferroptosis显示文摘Ferroptosis,a novel form of programmed cell death,is characterized by iron-dependent lipid peroxidation and has been shown to be involved in multiple diseases,including cancer.Stimulating ferroptosis in cancer cells may be a potential strategy for cancer therapy.Therefore,ferroptosis-inducing drugs are attracting more attention for cancer treatment.Here,we showed that erianin,a natural product isolated from Dendrobium chrysotoxum Lindl,exerted its anticancer activity by inducing cell death and inhibiting cell migration in lung cancer cells.Subsequently,we demonstrated for the first time that erianin induced ferroptotic cell death in lung cancer cells,which was accompanied by ROS accumulation,lipid peroxidation,and GSH depletion.The ferroptosis inhibitors Fer-1 and Lip-1 but not Z-VAD-FMK,CQ,or necrostatin-1 rescued erianin-induced cell death,indicating that ferroptosis contributed to erianin-induced cell death.Furthermore,we demonstrated that Ca^(2+)/CaM signaling was a critical mediator of erianin-induced ferroptosis and that blockade of this signaling significantly rescued cell death induced by erianin treatment by suppressing ferroptosis.Taken together,our data suggest that the natural product erianin exerts its anticancer effects by inducing Ca^(2+)/CaMdependent ferroptosis and inhibiting cell migration,and erianin will hopefully serve as a prospective compound for lung cancer treatment.Peng Chen Qibiao Wu Jiao Feng Lili Yan Yitian Sun Shuiping Liu Yu Xiang Mingming Zhang Ting Pan Xiaying Chen Ting Duan Lijuan Zhai Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Xuemeng Han Yicong Li Liuxi Chen Ying Liu Xingxing Huang Ting Jin Wenzheng Zhang Hong Luo Xiaohui Chen Yongqiang Li Qiujie Li Guohua Li Qin Zhang Lvjia Zhuo Zuyi Yang Huifen Tang Tian Xie Xiaoping Ouyang Xinbing Sui 2020Signal Transduction and Targeted Therapy2020,5,1:42
2Co-delivery of TRAIL and paclitaxel by fibronectin-targeting liposomal nanodisk for effective lung melanoma metastasis treatment显示文摘Melanoma is a highly aggressive cancer which often forms metastatic tumors in the lung,leading to sharply reduced patients'survival rate.Effectively treating these tumors thus could improve late stage melanoma with lung metastasis.In this study,we fabricated a Cys-Arg-Glu-Lys-Ala with N-methylated Glu(CR(NMe)EKA)decorated disk shaped nano vehicle to co-deliver tumor necrosis factor-related apoptosis-inducing ligand(TRAIL)and paclitaxel(PTX)to lung melanoma tumor sites(TRAIL-[ND-PTX]^(CR(NMe)EKA)).These nanodisks displayed better tumor-targeting and penetration capability than spherical nanoparticles,while the fibronectin-targeting CR(NMe)EKA motif also increased the tumor accumulation of loaded drugs.The combined usage of TRAIL and PTX both killed tumor cells and reduced local nutrition supply,leading to stronger overall anti-tumor effect.This TRAIL-[ND-PTX]^(CR(NMe)EKA)system performed remarkably better than free paclitaxel and also significantly elongated survival rate of melanoma lung metastasis bearing mice,without displaying significant toxicity.Hence,this designing strategy and the fabricated nanoplatform possess potential for further development.Shiqi Huang Lang Deng Hanming Zhang Luyao Wang Yicong Zhang Qing Lin Tao Gong Xun Sun Zhirong Zhang Ling Zhang 2022Nano Research2022,15,1:1

4Effects of dynamic flow rates on degradation deposition behavior of Mg scaffold显示文摘Degradability of bone tissue engineering scaffold that matching the regeneration rate could allow a complete replacement of host tissue.However,the porous structure of biodegradable Mg scaffolds certainly generated high specific surface area,and the three-dimensional interconnected pores provided fast pervasive invasion entrance for the corrosive medium,rising concern of the structural integrity during the degradation.To clarify the structural evolution of the three-dimensional(3D)porous structure,semi-static immersion tests were carried out to evaluate the degradation performance in our previous study.Nevertheless,dynamic immersion tests mimicking the in vivo circulatory fluid through the interconnected porous structure have yet been investigated.Moreover,the effects of dynamic flow rates on the degradation deposition behavior of 3D porous Mg scaffolds were rarely reported.In this study,Mg scaffolds degraded at three flow rates exhibited different degradation rates and deposition process.A flow rate of 0.5 m L/min introduced maximum drop of porosity by accumulated deposition products.The deposition products provided limited protection against the degradation process at a flow rate of 1.0 m L/min.The three-dimensional interconnected porous structure of Mg scaffold degraded at 2.0 m L/min well retained after 14 days showing the best interconnectivity resistance to the degradation deposition process.The dynamic immersion tests disclosed the reason for the different degradation rates on account of flow rates,which may bring insight into understanding of varied in vivo degradation rates related to implantation sites.Gaozhi Jia Meng Zhou Yicong Huang Chenxin Chen Liang Jin Qian Wu Jian Weng Fei Yu Ao Xiong Guangyin Yuan Frank Feyerabend Hui Zeng 2023Journal of Magnesium and Alloys2023,11,6:0
5Comparison of enhancement techniques based on neural networks for attenuated voice signal captured by flexible vibration sensors on throats显示文摘Wearable flexible sensors attached on the neck have been developed to measure the vibration of vocal cords during speech.However,highfrequency attenuation caused by the frequency response of the flexible sensors and absorption of high-frequency sound by the skin are obstacles to the practical application of these sensors in speech capture based on bone conduction.In this paper,speech enhancement techniques for enhancing the intelligibility of sensor signals are developed and compared.Four kinds of speech enhancement algorithms based on a fully connected neural network(FCNN),a long short-term memory(LSTM),a bidirectional long short-term memory(BLSTM),and a convolutional-recurrent neural network(CRNN)are adopted to enhance the sensor signals,and their performance after deployment on four kinds of edge and cloud platforms is also investigated.Experimental results show that the BLSTM performs best in improving speech quality,but is poorest with regard to hardware deployment.It improves short-time objective intelligibility(STOI)by 0.18 to nearly 0.80,which corresponds to a good intelligibility level,but it introduces latency as well as being a large model.The CRNN,which improves STOI to about 0.75,ranks second among the four neural networks.It is also the only model that is able to achieves real-time processing with all four hardware platforms,demonstrating its great potential for deployment on mobile platforms.To the best of our knowledge,this is one of the first trials to systematically and specifically develop processing techniques for bone-conduction speed signals captured by flexible sensors.The results demonstrate the possibility of realizing a wearable lightweight speech collection system based on flexible vibration sensors and real-time speech enhancement to compensate for high-frequency attenuation.Shenghan Gao Changyan Zheng Yicong Zhao Ziyue Wu Jiao Li Xian Huang 2022Nanotechnology and Precision Engineering2022,5,1:0
6Natural variation of an autophagy-family gene among rice subspecies affects grain size and weight显示文摘Elucidating the genetic basis of natural variation in grain size and weight among rice varieties can help breeders develop high-yielding varieties.We identified a novel gene,GW3a(Grain Weight 3a)(LOC_Os03g27350),that affects rice grain size and weight.gw3a mutants showed higher total starch content and dry matter accumulation than the wild type(WT),Nipponbare,suggesting that GW3a negatively regulates grain size and weight.Moreover,our study found that GW3a interacted with OsATG8 by cleaving it,suggesting that GW3a may be involved in the assembly of autophagosomes and starch degradation in plants.The haplotype analysis of GW3a showed functional differences between indica and japonica rice.Taken together,we conclude that GW3a is expressed in the autophagosome pathway regulating starch metabolism in rice,affecting yield-related traits,such as grain size,grain weight and thousand grain weight(TGW).Our findings also shed new light on autophagy-mediated yield trait regulation,proposing a possible strategy for the genetic improvement of high-yield germplasm in rice.Shiying Huang Peng Wang Caijing Li Qingwen Zhou Tao Huang Yicong Cai Qin Cheng Hui Wang Qi Zhong Zhihao Chen Liping Chen Haohua He Jianmin Bian 2024The Crop Journal2024,12,1:0
7Mechanisms of imperatorin on regulating Pglycoprotein in blood-brain barrier based on network pharmacology and in vitro experiment显示文摘OBJECTIVE To explore mecha⁃nisms of imperatorin on regulating P-glycoprotein(P-gp)in blood-brain barrier(BBB)based on net⁃work pharmacology combined with in vitro experi⁃ment.METHODS Drug targets were predicted using the Pharmapper and Swiss targets data⁃bases;disease targets were obtained through the Genecards database;intersections between drugs and disease targets were screened by Cytoscape software;the obtained core targets were used to construct protein-protein interaction(PPI)network,gene ontology(GO)functions,and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis.The effects of imperatorin(20,50,100μmol·L^(-1))on P-gp activity were monitored in hCMEC/D3 in vitro BBB model,and the effects of imperatorin on the expression of target proteins were verified using Western blot method.RESULTS 55 drug targets and 3102 disease targets were obtained from the network pharmacology screening,and 37 core targets were obtained after the combination.Enrichment analysis showed that core targets were closely related to chemical synaptic trans⁃mission regulation,neurotransmitter receptor activity,protein kinase regulation activity,G proteincoupled receptor signaling pathway,neural active ligand receptor interaction pathway,PI3K-Akt sig⁃naling pathway,VEGF signaling pathway,etc..In vitro experimental validation suggested that all tested concentration groups of imperatorin signifi⁃cantly reduced the activity and expression of P-gp,which were achieved by significantly downregu⁃lating the phosphorylation levels of PI3K and Akt,and repressing the expression of VEGFR2 pro⁃tein.CONCLUSION Network pharmacology was used to predict the core targets and signaling pathways of imperatorin on regulating P-gp in BBB and relevant validation was conducted through in vitro experiments,providing a refer⁃ence basis for further exploration of the mecha⁃nisms of imperatorin on regulating P-gp in BBB.SONG Yizhen YIN Wanxin MA Yicong AN Yufan HUANG Jiaqi YAN Zhongjie WU Xiuwen WANG Yuanyu 2023中国药理学与毒理学杂志2023,37,S01:0

9MicroRNA–disease Network Analysis Repurposes Methotrexate for the Treatment of Abdominal Aortic Aneurysm in Mice显示文摘Abdominal aortic aneurysm(AAA)is a permanent dilatation of the abdominal aorta and is highly lethal.The main purpose of the current study is to search for noninvasive medical therapies for AAA,for which there is currently no effective drug therapy.Network medicine represents a cuttingedge technology,as analysis and modeling of disease networks can provide critical clues regarding the etiology of specific diseases and therapeutics that may be effective.Here,we proposed a novel algorithm to quantify disease relations based on a large accumulated microRNA–disease association dataset and then built a disease network covering 15 disease classes and 304 diseases.Analysis revealed some patterns for these diseases.For instance,diseases tended to be clustered and coherent in the network.Surprisingly,we found that AAA showed the strongest similarity with rheumatoid arthritis and systemic lupus erythematosus,both of which are autoimmune diseases,suggesting that AAA could be one type of autoimmune diseases in etiology.Based on this observation,we further hypothesized that drugs for autoimmune diseases could be repurposed for the prevention and therapy of AAA.Finally,animal experiments confirmed that methotrexate,a drug for autoimmune diseases,was able to alleviate the formation and development of AAA.Yicong Shen Yuanxu Gao Jiangcheng Shi Zhou Huang Rongbo Dai Yi Fu Yuan Zhou Wei Kong Qinghua Cui 2023Genomics, Proteomics & Bioinformatics2023,21,5:0
