| 2 | miR-137, a new target for post-stroke depression?显示文摘Expression of miR-137 is downregulated in brain tissue from patients with depression and suicidal behavior,and is also downregulated in peripheral blood from stroke patients.However,it is not yet known if miR-137 acts as a bridge between stroke and depression.To test this,we used middle cerebral artery occlusion and chronic mild stress to establish a post-stroke depression model in rats.Compared with controls,we found significantly lower miR-137 levels in the brain and peripheral blood from post-stroke depression rats.Injection of a miR-137 antagonist into the brain ventricles upregulated miR-137 levels,and improved behavioral changes in post-stroke depression rats.Luciferase assays showed miR-137 bound to the 3′UTR of Grin2A,regulating Grin2A expression in a neuronal cell line.Grin2A gene overexpression in the brain of post-stroke depression rats,noticeably suppressed the inhibitory effect of miR-137 on post-stroke depression.Overall,our results show that miR-137 suppresses Grin2A protein expression through binding to Grin2A mRNA,thereby exerting an inhibitory effect on post-stroke depression.Our results offer a new therapeutic direction for post-stroke depression. | Lixia Zhao Huazi Li Ruiyou Guo Teng Ma Rongyao Hou Xiaowei Ma Yifeng Du | 2013 | Neural Regeneration Research2013,8,26: | 8 |

| 5 | N-doped ordered mesoporous carbon spheres derived by confined pyrolysis for high supercapacitor performance显示文摘Herein, we report a confined pyrolysis strategy to prepare mesoporous carbon nanospheres by which surface area of carbon spheres is increased, pore size is enlarged and effective N-doping is achieved. In this method, the mesoporous polymer sphere as carbon precursor and 2-methylimidazole as nitrogen precursor are encapsulated in a compact silica shell which provides a confined nano-space for the pyrolysis treatment. The in situ generated gases from mesoporous polymer sphere and 2-methylimidazole under pyrolysis diffuse into the pores of mesoporous polymer sphere in the confined compact silica shell,resulting in increased surface area, larger pore size and N-doping due to self-activation effect. | 6 | Significant effect of as-cast microstructure on texture evolution and magnetic properties of strip cast non-oriented silicon steel显示文摘In this study, two types of as-cast microstructure produced by strip casting were cold rolled and annealed to investigate the effect of initial microstructure on the textural evolution and magnetic properties of non-oriented silicon steel. The results indicated that the cold-rolled sheets of coarse-grained strip with pronounced {100} components exhibited stronger 入 fiber(<100>//ND) and weaker γ fiber(<111>//ND)texture as composed to the fine-grained strip with strong Goss({110}(001)) texture. After annealing, the former was dominated by η fiber(<001>//RD) texture with a peak at {110}<001)orientation, while the latter consisted of strong {111}(112) and relatively weak {110}(001) texture. | 7 | Effects of L-3-n-butylphthalide on caspase-3 and nuclear factor kappa-B expression in primary basal forebrain and hippocampal cultures after beta-amyloid peptide 1-42 treatment显示文摘BACKGROUND:L-3-n-butylphthalide(L-NBP) can inhibit phosphorylation of tau protein and reduce the neurotoxicity of beta-amyloid peptide 1-42(Aβ_(1-42)). OBJECTIVE:To observe the neuroprotective effects of L-NBP on caspase-3 and nuclear factor kappa-B(NF-κB) expression in a rat model of Alzheimer's disease. DESIGN,TIME AND SETTING:A cell experiment was performed at the Central Laboratory of Provincial Hospital affiliated to Shandong University between January 2008 and August 2008. MATERIALS:L-NBP(purity>98%) was provided by Shijiazhuang Pharma Group NBP Pharmaceutical Company Limited.Aβ_(1-42),3-[4,5-dimethylthiazolo-2]-2,5 iphenyltetrazolium bromide(MTT),and rabbit anti-Caspase-3 polyclonal antibody were provided by Cell Signaling, USA;goat anti-choactase and rabbit anti-NF-κB antibodies were provided by Santa Cruz,USA. METHODS:Primary cultures were generated from rat basal forebrain and hippocampal neurons at 17 or 19 days of gestation.The cells were assigned into five groups:the control group,the Aβ_(1-42) group(2μmol/ L),the Aβ_(1-42) + 0.1μmol/L L-NBP group,the Aβ_(1-42)+ 1μmol/L L-NBP group,and the Aβ_(1-42)+ 10μmol/L L-NBP group.The neurons were treated with Aβ_(1-42)(2μmol/L) alone or in combination with L-NBP(0.1,1,10μmol/L) for 48 hours.Cells in the control group were incubated in PBS. MAIN OUTCOME MEASURES:Morphologic changes were evaluated using inverted microscopy, viability using the MTT method,and the changes in caspase-3 and NF-κB expression using Western blot. RESULTS:Induction with Aβ_(1-42) for 48 hours caused cell death and soma atrophy,and increased caspase-3 and NF-κB expression(P<0.05).L-NBP blocked these changes in cell morphology, decreased caspase-3 and NF-κB expression(P<0.05),and improved cell viability,especially at the high dose(P<0.05). | 8 | Effect of olfactory ensheathing cell transplantation on myelin repair and motor function in a rat model of experimental allergic encephalomyelitis显示文摘BACKGROUND:Olfactory ensheathing cell transplantation can activate axonal regeneration and enhance myelin repair,which are beneficial for treating demyelinating diseases. OBJECTIVE:To explore the effects of olfactory ensheathing cell transplantation on myelin repair, synaptophysin expression,and motor function in a rat model of experimental allergic encephalomyelitis. DESIGN,TIME AND SETTING:A randomized,controlled experiment was performed at the Laboratory of Provincial Hospital affiliated to Shandong University between August 2006 and September 2007. MATERIALS:Dibenzylamine(Hoechst 33342),luxol fast blue,and rabbit anti-rat synaptophysin antibody were provided by Sigma,USA. METHODS:Olfactory ensheathing cells extracted from neonatal Wistar rats were cultured for 10-14 days and labeled with dibenzylamine.Spinal cord extracted from a healthy guinea pig was homogenized and equally mixed with complete Freund's adjuvant;thereafter,the mixture was intracutaneously injected into two posterior voix pedis of healthy male Wistar rats to establish models of experimental allergic encephalomyelitis.Rats were randomly divided into a control encephalomyelitis group and an olfactory ensheathing cell transplantation group,36 rats in each group.Physiological saline(2μL) or an olfactory ensheathing cell suspension(2μL) was separately injected along lateral cerebral ventricle at day 7 post-model induction. MAIN OUTCOME MEASURES:The migration and distribution of olfactory ensheathing cells were observed under fluorescence microscopy;myelin repair was detected using hematoxylin-eosin staining and luxol fast blue staining;synaptophysin expression was measured using immunohistochemical staining;motor function was evaluated using a motor function scale. RESULTS:Olfactory ensheathing cells could survive in vivo and migrate to the distal end of the transplant focus and spinal cord,and survived 21 days.Hematoxylin-eosin staining and luxol fast blue staining indicated that myelin in the transplantation group was intact,and the inflammatory focus gradually disappeared.Transplantation increased synaptophysin expression(P<0.05 versus control) and motor function(P<0.05). Basing on the compositional inhomogeneity inside the nanospheres of 3-aminophenol/formaldehyde (3-AF) polymerization, the internal 3-AF oligomer with low-molecular-weight is dissolved by acetone to form cavity. Then the dispersive 3-AF oligomer reassemble with silica oligomer as pore-forming agent and cetyltrimethyl-ammonium bromide as surfactant to form a mesoporous outer shell. In addition, the amount of acetone has a great influence on the morphology and structure of H(Micro/Meso)CS. | 11 | Expression and biological activity of double replica retrovirus carrier-mediated neurotrophin-3 in olfactory ensheathing cells显示文摘BACKGROUND:Previous studies have demonstrated that the combination of olfactory ensheathing cells(OECs) and neurotrophic factor-3(NT-3) in the rat lateral ventricle can promote nerve axonal regeneration and myelin sheath repair.However,this effect remains very short-lived, OBJECTIVE:To transfect NT-3 into OECs and to observe the biological activity of OEC-expressing NT-3. DESIGN,TIME AND SETTING:This genetic engineering,in vitro experiment was performed in the Provincial Hospital Affiliated to Shandong University between January 2007 and October 2008. MATERIALS:Trizol Reagent kit was purchased from Gibco,USA;reverse transcription kit,NT-3 Emax^(?)ImmunoAssay System reagent was purchased from Promega,USA. METHODS:Neonatal Wistar rat OECs were established as primary cultures and were transfected with pN2A-NT-3 viral vector.The OECs with the highest virus titer and stable cellular growth served as the transfection group;OECs transfected with NT-3-free retrovirus carrier pN2A served as the empty vector group;un-transfected OECs served as the control group.After adherence,the logarithmically cultured PC12-TrkC cells were plated in OECs supernatant from the transfection and empty vector groups,as well as 20μL PBS,and cultured for 4 days. MAIN OUTCOME MEASURES:NT-3 mRNA expression in OECs,fluorescence of NT-3-positive cells in the transfection group and control group;influence of OECs secreting NT-3 on the differentiation ratio of PC12-TrkC cells. RESULTS:NT-3 mRNA expression was observed 24 hours after transfection and lasted for 28 days, which was greater than the control and empty vector groups(P<0.01).A large number of NT-3-positive cells were observed in the transfection group,and immunofluorescence was greater than the control and empty vector groups.PC12-TrkC cells co-cultured with OECs from the transfection group exhibited a thick and long cell process,increased cell density,and the differentiation ratio was increased(P<0.01). The pSSCMV/human thioredoxin-PR39 vector was successfully constructed in this study and used to infect ECV304 cells. Transfected ECV304 cells were incubated at 1%, 5% hypoxic, and normal oxygen conditions. | 13 | Multiple factor analysis of symptomatic intracranial arterial stenosis显示文摘Objective: To discuss the correlation among intracranial arterial stenosis and its risk factors. Methods: A total of 486 patients with transient ischemic attack(TIA) or ischemic cerebral infarction were examined using color doppler flow imaging(CDFI) and transcranial doppler ultrosonography(TCD). According to the degrees of extracranial arterial stenosis, patients with mild-to-moderate extracranial stenosis were classified into group A(435 cases) while those with constant severe stenosis or occlusion were classified into group B(51 cases). The differences between the two groups of risk factors were compared, and the multi-factor logistic regression analysis of risk factors associated with moderately severe intracranial arterial stenosis was performed. Results:(1) The risk factors that were significantly associated with intracranial arterial stenosis included age(P = 0.034) and gender(P = 0.044).(2) Intracranial artery stenosis was observed in both anterior and posterior cerebral arteries in patients with hypertension, diabetes, and coronary heart disease respectively(P < 0.05).(3) Compared with group A, patients in group B were older(P = 0.000), with a higher proportion of men(P = 0.037), and the intracranial arterial stenosis degrees were significantly higher(P = 0.013).(4) Multi-factor logistic regression analysis showed that diabetes is a risk factor for moderately severe intracranial arterial stenosis(P < 0.05), and hyperlipidemia is a protective factor(P = 0.012). Conclusions: Age, gender, hypertension, diabetes, coronary heart disease, and smoking are risk factors for the distribution of intracranial arterial stenosis. The degrees of intracranial arterial stenosis are related with extracranial arterial stenosis. | 14 | Generation of genetically modified olfactory ensheathing cells that secrete VEGF: Potential application in cell-based treatment of leukoaraiosis显示文摘Objective: We aimed to provide an alternative cell source for cell therapy in leukoaraiosis(LA). Methods: Olfactory ensheathing cells(OECs) from the olfactory bulb were isolated,cultured, and purified. Next, the lentivirus carrying human VEGF165 gene was constructed and transfected into OECs. Results: The proliferative capacity of primary OECs was strong. OECs were infected with different multiplicity of infection. The expression level of VEGF was confirmed by real-time PCR with specific primers for GAPDH and VEGF, indicating that the genetically engineered OECs-VEGF produced VEGF with functional activity. | 16 | Fine-Tuning Pyridinic Nitrogen in Nitrogen-Doped Porous Carbon Nanostructures for Boosted Peroxidase-Like Activity and Sensitive Biosensing显示文摘Carbon materials have been widely used as nanozymes in bioapplications,attributing to their intrinsic enzyme-like activities.Nitrogen(N)-doping has been explored as a promising way to improve the activity of carbon material-based nanozymes(CMNs).However,hindered by the intricate N dopants,the real active site of N-doped CMNs(N-CMNs)has been rarely investigated,which subsequently retards the further progress of high-performance N-CMNs.Here,a series of porous N-CMNs with well-controlled N dopants were synthesized,of which the intrinsic peroxidase(POD)like activity has a positive correlation with the pyridinic N content.Density functional theory calculations also reveal that pyridinic N boosts the intrinsic POD-like activity of N-CMNs.Pyridinic-N dopant can effectively promote the first H_(2)O desorption process in comparison with the graphitic and pyrrolic N,which is the key endothermic reaction during the catalytic process.Then,utilizing the optimized nanozymes with high pyridinic N content(NP-CMNs)and superior POD-like activity,a facile total antioxidant capacity(TAC)assay was developed,holding great promise in the quality assessment of medicine tablets and antioxidant food for healthcare and healthy diet. | Hongye Yan Linzhe Wang Yifeng Chen Lei Jiao Yu Wu Weiqing Xu Wenling Gu Weiyu Song Dan Du Chengzhou Zhu | 2020 | Research2020,,1: | 0 |
| 17 | S-scheme heterojunction/Schottky junction tandem synergistic effect promotes visible-light-driven catalytic activity显示文摘Designing photocatalysts with high light utilization and efficient photogenerated carrier separation for pollutant degradation is one of the important topics for sustainable development.In this study,hierarchical core–shell materialα-Fe_(2)O_(3)@ZnIn_(2)S_(4)with a step-scheme(S-scheme)heterojunction is synthesized by in situ growth technique,and MXene Ti_(3)C_(2)quantum dots(QDs)are introduced to construct a double-heterojunction tandem mechanism.The photodegradation efficiency ofα-Fe_(2)O_(3)@ZnIn_(2)S_(4)/Ti_(3)C_(2)QDs to bisphenol A is 96.1%and its reaction rate constant attained 0.02595 min^(−1),which is 12.3 times that of pureα-Fe_(2)O_(3).Meanwhile,a series of characterizations analyze the reasons for the enhanced photocatalytic activity,and the charge transport path of the S-scheme heterojunction/Schottky junction tandem is investigated.The construction of the S-scheme heterojunction enables the photo-generated electrons ofα-Fe_(2)O_(3)and the holes of ZnIn2S4 to transfer and combine under the action of the reverse built-in electric field.Due to the metallic conductivity of Ti_(3)C_(2)QDs,the photogenerated electrons of ZnIn_(2)S_(4)are further transferred to Ti_(3)C_(2)QDs to form a Schottky junction,which in turn forms a double-heterojunction tandem mechanism,showing a remarkable charge separation efficiency.This work provides a new opinion for the construction of tandem double heterojunctions to degrade harmful pollutants. | Shuai Wang Xin Du Changhao Yao Yifeng Cai Huiyuan Ma Baojiang Jiang Jun Ma | 2023 | Nano Research2023,16,2: | 0 |
| 18 | Ablation of Zfhx4 results in early postnatal lethality by disrupting the respiratory center in mice显示文摘Breathing is an integrated motor behavior that is driven and controlled by a network of brainstem neurons.Zfhx4 is a zinc finger transcription factor and our results showed that it was specifically expressed in several regions of the mouse brainstem.Mice lacking Zfhx4 died shortly after birth from an apparent inability to initiate respiration.We also found that the electrical rhythm of brainstem‒spinal cord preparations was significantly depressed in Zfhx4-null mice compared to wild-type mice.Immunofluorescence staining revealed that Zfhx4 was coexpressed with Phox2b and Math1 in the brainstem and that Zfhx4 ablation greatly decreased the expression of these proteins,especially in the retrotrapezoid nucleus.Combined ChIP‒seq and mRNA expression microarray analysis identified Phox2b as the direct downstream target gene of Zfhx4,and this finding was validated by ChIP‒qPCR.Previous studies have reported that both Phox2b and Math1 play key roles in the development of the respiratory center,and Phox2b and Math1 knockout mice are neonatal lethal due to severe central apnea.On top of this,our study revealed that Zfhx4 is a critical regulator of Phox2b expression and essential for perinatal breathing. | Meiqin Zhang Sichen Du Huayuan Ou Renjie Cui Nan Jiang Yifeng Lin Runsheng Ge Duan Ma Jin Zhang | 2021 | Journal of Molecular Cell Biology2021,13,3: | 0 |
