| 1 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29 |
| 2 | A fundamental theorem for eco-environmental surface modelling and its applications显示文摘We propose a fundamental theorem for eco-environmental surface modelling(FTEEM) in order to apply it into the fields of ecology and environmental science more easily after the fundamental theorem for Earth’s surface system modeling(FTESM). The Beijing-Tianjin-Hebei(BTH) region is taken as a case area to conduct empirical studies of algorithms for spatial upscaling, spatial downscaling, spatial interpolation, data fusion and model-data assimilation, which are based on high accuracy surface modelling(HASM), corresponding with corollaries of FTEEM. The case studies demonstrate how eco-environmental surface modelling is substantially improved when both extrinsic and intrinsic information are used along with an appropriate method of HASM. Compared with classic algorithms, the HASM-based algorithm for spatial upscaling reduced the root-meansquare error of the BTH elevation surface by 9 m. The HASM-based algorithm for spatial downscaling reduced the relative error of future scenarios of annual mean temperature by 16%. The HASM-based algorithm for spatial interpolation reduced the relative error of change trend of annual mean precipitation by 0.2%. The HASM-based algorithm for data fusion reduced the relative error of change trend of annual mean temperature by 70%. The HASM-based algorithm for model-data assimilation reduced the relative error of carbon stocks by 40%. | 5 | Refined chronology of prehistoric cultures and its implication for re-evaluating human-environment relations in the Hexi Corridor,northwest China显示文摘The reconstruction of high-resolution chronologies for prehistoric cultures is a prerequisite for understanding the history of human evolution and its relationship with environmental change, and is valuable for exploring the trajectory of transcontinental cultural exchanges in prehistoric time. The Hexi Corridor of northwest China was one of the earliest centers for long-distance culture exchange in the prehistoric world. The timing and sequence of cultural changes in this area remains poorly understood resulting from the lack of radiometric age control. This paper presents a refined radiocarbon(14 C) chronology to resolve the timing of human occupation and cultural evolution in the Hexi Corridor. Radiocarbon dating of crop remains, which have an annual life cycle, has the advantage of eliminating problems such as the carbon stored in ‘old wood’. As a result,14C dates from crop remains are used to test the validity of the14C dates derived from charcoal, bone and plant remains. The resultant database of ages is modelled using the Bayesian approach. The updated chronology presented in this paper shows generally good agreement with the original dates used to constrain cultural evolution in the Hexi Corridor. However, the occupation of the corridor by the Shajing and Shanma Late Bronze Age-Early Iron Age cultures appears to have taken place ~300 years later than previously thought. | 6 | Probing primordial gravitational waves: Ali CMB Polarization Telescope显示文摘In this paper, we will give a general introduction to the Ali CMB Polarization Telescope(Ali CPT) project,which is a Sino–US joint project led by the Institute of High Energy Physics and involves many different institutes in China. It is the first ground-based Cosmic Microwave Background(CMB) polarization experiment in China and an integral part of China's Gravitational-wave Program. The main scientific goal of the Ali CPT project is to probe the primordial gravitational waves(PGWs) originating from the very early Universe. The AliCPT project includes two stages. The first stage, referred to as AliCPT-1, is to build a telescope in the Ali region of Tibet at an altitude of 5250 meters. Once completed, it will be the highest ground-based CMB observatory in the world and will open a new window for probing PGWs in the northern hemisphere. The AliCPT-1 telescope is designed to have about 7000 transition-edge sensor detectors at 95 GHz and 150 GHz. The second stage is to have a more sensitive telescope(Ali CPT-2) with more than 20 000 detectors. Our simulations show that AliCPT will improve the current constraint on the tensor-to-scalar ratio r by one order of magnitude with three years' observation. Besides the PGWs, Ali CPT will also enable a precise measurement of the CMB rotation angle and provide a precise test of the CPT symmetry. | 7 | Microvascular endothelial cells derived from spinal cord promote spinal cord injury repair显示文摘Neural regeneration after spinal cord injury (SCI) closely relates to the microvascular endothelial cell (MEC)- mediated neurovascular unit formation. However, the effects of central nerve system-derived MECs on neovascularization and neurogenesis, and potential signaling involved therein, are unclear. Here, we established a primary spinal cord-derived MECs (SCMECs) isolation with high cell yield and purity to describe the differences with brain-derived MECs (BMECs) and their therapeutic effects on SCI. Transcriptomics and proteomics revealed differentially expressed genes and proteins in SCMECs were involved in angiogenesis, immunity, metabolism, and cell adhesion molecular signaling was the only signaling pathway enriched of top 10 in differentially expressed genes and proteins KEGG analysis. SCMECs and BMECs could be induced angiogenesis by different stiffness stimulation of PEG hydrogels with elastic modulus 50-1650 Pa for SCMECs and 50-300 Pa for BMECs, respectively. Moreover, SCMECs and BMECs promoted spinal cord or brain-derived NSC (SNSC/BNSC) proliferation, migration, and differentiation at different levels. At certain dose, SCMECs in combination with the NeuroRegen scaffold, showed higher effectiveness in the promotion of vascular reconstruction. The potential underlying mechanism of this phenomenon may through VEGF/AKT/eNOS- signaling pathway, and consequently accelerated neuronal regeneration and functional recovery of SCI rats compared to BMECs. | 16 | The Integration of Farmers and Nomads: Archaeological Evidence for the Human Subsistence Strategy in Northwestern China during the Han Dynasty显示文摘The integration of farmers and nomads in northwestern China during the Han Dynasty(206 BCE ~ 220 CE) provides a crucial opportunity to reconstruct the material exchanges, formation and development of the Silk Road in antiquity. The subsistence strategy is arguably an effective proxy for the integration of various groups of people(e.g. farmers and nomads). In this paper, we have reported new stable isotope data from the Huangwan tombs dated to the Han dynasty in middle Gansu, which was the key juncture between the Han and Xiongnu empire, in order to fill the gap and further understand the substance strategies employed by the local people. According to the results of plant remains and stable isotopic data, millet farming, the typical agricultural activities for the Han Chinese in the Central Plains, was also the primary lifestyle for the Huangwan people in the mid Gansu. More importantly, this shows fundamentally remarkable difference from the agricultural practices in the Bronze Age Gansu Corridor, which were based on a variety of crops, including wheat, barley and millet.
