2Development of a Label-Free Colorimetric Aptasensor with Rationally Utilized Aptamer for Rapid Detection of Okadaic Acid显示文摘Okadaic acid(OA)is a typical marine toxin with strong toxicity causing diarrheic shellfish poisoning(DSP).Aptamers show great advantages in toxin detection and attract increasing attentions in the field of food analysis.In this study,a label-free col-orimetric aptasensor was constructed for visual and rapid detection of OA in shellfish.To exploit the binding capability of the anti-OA aptamer,the inherent molecular recognition mechanism of aptamer and OA was studied,based on molecular docking,fluorescent assay,and biolayer interferometry.Consistent results showed that the stem-loop near the 3’terminal of the aptamer exhibit dominate binding capacity.Based on the revealed recognition information,the aptamer was thus rationally utilized and combined with AuNPs and cationic polymer polydiallyl dimethyl ammonium chloride(PDDA)for the development of the label-free colorimetric aptasensor,in which the 3’terminal was thoroughly exposed to OA.The aptasensor provided robust performance with a linear detection range of 100-1200 nmol L-1,a limit of detection of 41.30 nmol L-1,recovery rates of 91.6%-106.2%,as well as a high selectivity towards OA in shellfish samples.The whole detection process can be completed within 1 h.To our best knowledge,this is the first time that the anti-OA aptamer was thoroughly studied,and a label-free colorimetric aptasensor was rationally designed in this way.This study not only provides a rapid detection method for highly sensitive and specific detection of OA,but also serves as a reference for the design of efficient aptasensors in the future.YAN Xiaochen QI Xiaoyan ZHAO Yinglin LI Ling MA Rui WANG Lele WANG Sai MAO Xiangzhao 2022Journal of Ocean University of China2022,21,2:2

6Spontaneous immunomodulation and regulation of angiogenesis and osteogenesis by Sr/Cu-borosilicate glass (BSG) bone cement to repair critical bone defects显示文摘Injectable bone biomaterials like bone cement should be designed and fabricated with certain biological criteria,which include:1)recruitment and polarization of the macrophages from M1(pro-inflammatory)to M2(anti-inflammatory)phenotype,2)enhance vascularization,and 3)activate osteogenic differentiation of bone marrow-derived stem cells to promote bone healing.So far,no injectable biomaterials could spontaneously regulate the entire bone healing process that involves inflammation,angiogenesis,and osteogenesis.Therefore,in this study,we designed bone cement comprised of strontium and copper-incorporated borosilicate glass(Sr/Cu-BSG)in the liquid phase of chitosan to modulate bone healing.In vitro studies showed that the controlled release of Sr and Cu ions up-regulated anti-inflammatory genes(IL-1Ra and TGF-β1)while down-regulating pro-inflammatory genes(IL-1βand IL-6)in macrophages at 3 days.Sr and Cu ions also increased the expressions of angiogenic genes(VEGF and bFGF)in HUVECs at 5 days and osteogenic genes(Runx-2,OCN,and OPN)in hBMSCs at 7,14,and 21 days.5Sr3Cu-BSG bone cement exhibited the best anti-inflammatory,angiogenic,and osteogenic properties among the bone cement groups with different Sr and Cu ratios.Short-term and long-term implantation of Sr/Cu-BSGs in femoral condylar bone defects of rats and rabbits confirmed the in vitro results,where the degradation rate of Sr/Cu-BSG matched the bone healing rate.Similar to in vitro,the 5Sr3Cu-BSG group also showed the highest bone formation in vivo.Excellent physical and chemical properties,along with its bone repairing ability,make the Sr/Cu-BSG bone cement a good candidate biomaterial for treating bone defects.Shuaijie Li Liyan Zhang Chunyu Liu Jua Kim Kun Su Tingli Chen Limin Zhao Xiaomei Lu Hao Zhang Yinglin Cui Xu Cui Feng Yuan Haobo Pan 2023Bioactive Materials2023,,5:1
75-HMF attenuates inflammation and demyelination in experimental autoimmune encephalomyelitis mice by inhibiting the MIF-CD74 interaction显示文摘The neuroprotective role of 5-hydroxymethyl-2-furfural(5-HMF)has been demonstrated in a variety of neurological diseases.The aim of this study is to investigate the effect of 5-HMF on multiple sclerosis(MS).IFN-γ-stimulated murine microglia(BV2 cells)are considered a cell model of MS.With 5-HMF treatment,microglial M1/2 polarization and cytokine levels are detected.The interaction of 5-HMF with migration inhibitory factor(MIF)is predicted using online databases.The experimental autoimmune encephalomyelitis(EAE)mouse model is established,followed by a 5-HMF injection.The results show that 5-HMF facilitates IFN-γ-stimulated microglial M2 polarization and attenuates the inflammatory response.According to the network pharmacology and molecular docking results,5-HMF has a binding site for MIF.Further results show that blocking MIF activity or silencing CD74 enhances microglial M2 polarization,reduces inflammatory activity,and prevents ERK1/2 phosphorylation.5-HMF inhibits the MIF-CD74 interaction by binding to MIF,thereby inhibiting microglial M1 polarization and enhancing the anti-inflammatory response.5-HMF ameliorates EAE,inflammation,and demyelination in vivo.In conclusion,our research indicates that 5-HMF promotes microglial M2 polarization by inhibiting the MIF-CD74 interaction,thereby attenuating inflammation and demyelination in EAE mice.Dongsheng Guan Yingxia Li Yinglin Cui Huanghong Zhao Ning Dong Kun Wang Deqi Ren Tiantian Song Xiaojing Wang Shijie Jin Yinghe Gao Mengmeng Wang 2023Acta Biochimica et Biophysica Sinica2023,55,8:0
8Scientific Advances and Weather Services of the China Meteorological Administration’s National Forecasting Systems during the Beijing 2022 Winter Olympics显示文摘Since the Beijing 2022 Winter Olympics was the first Winter Olympics in history held in continental winter monsoon climate conditions across complex terrain areas,there is a deficiency of relevant research,operational techniques,and experience.This made providing meteorological services for this event particularly challenging.The China Meteorological Administration(CMA)Earth System Modeling and Prediction Centre,achieved breakthroughs in research on short-and medium-term deterministic and ensemble numerical predictions.Several key technologies crucial for precise winter weather services during the Winter Olympics were developed.A comprehensive framework,known as the Operational System for High-Precision Weather Forecasting for the Winter Olympics,was established.Some of these advancements represent the highest level of capabilities currently available in China.The meteorological service provided to the Beijing 2022 Games also exceeded previous Winter Olympic Games in both variety and quality.This included achievements such as the“100-meter level,minute level”downscaled spatiotemporal resolution and forecasts spanning 1 to 15 days.Around 30 new technologies and over 60 kinds of products that align with the requirements of the Winter Olympics Organizing Committee were developed,and many of these techniques have since been integrated into the CMA’s operational national forecasting systems.These accomplishments were facilitated by a dedicated weather forecasting and research initiative,in conjunction with the preexisting real-time operational forecasting systems of the CMA.This program represents one of the five subprograms of the WMO’s high-impact weather forecasting demonstration project(SMART2022),and continues to play an important role in their Regional Association(RA)II Research Development Project(Hangzhou RDP).Therefore,the research accomplishments and meteorological service experiences from this program will be carried forward into forthcoming highimpact weather forecasting activities.This article provides an overview and assessment of this program and the operational national forecasting systems.Guo DENG Xueshun SHEN Jun DU Jiandong GONG Hua TONG Liantang DENG Zhifang XU Jing CHEN Jian SUN Yong WANG Jiangkai HU Jianjie WANG Mingxuan CHEN Huiling YUAN Yutao ZHANG Hongqi LI Yuanzhe WANG Li GAO Li SHENG Da LI Li LI Hao WANG Ying ZHAO Yinglin LI Zhili LIU Wenhua GUO 2024Advances in Atmospheric Sciences2024,41,5:0
9Chemical Control of Ceratovacuna lanigera Zehntner with Multi-rotor Unmanned Aerial Vehicle显示文摘[Objective] The paper was to explore chemical control of Ceratovacuna lanigera Zehntner with multi-rotor unmanned aerial vehicle. [Method] According to the outbreak characteristics of C. lanigera,multi-rotor unmanned aerial vehicle was applied for flying control test. Referred to the spraying characteristics of multi-rotor unmanned aerial vehicle,two kinds of microcapsule pesticides,ALV-1501 and ALV-1502,and two kinds of spraying additives,SPA-01 and SPA-02,were designed to control C. lanigera. [Result] The control effect of ALV-1501 at the dose of 2. 25 L/hm^2 was 60. 02% at 1 d post administration and 54. 14%at 5 d post administration; the control effects of ALV-1502 at the dose of 2. 1 L/hm2 were 76. 35% and 81. 35% at 1 and 5 d post administration,respectively.Compared to individual pesticide,the control effects of ALV-1501 were improved 1. 42-1. 47 times and 1. 16-1. 14 times by adding 0. 6 L/hm^2 SPA-01 and SPA-02 in pesticide liquid,respectively. The control effects of ALV-1502 were improved 1. 23-1. 25 times and 1. 15-1. 16 times by adding 0. 6 L/hm2 SPA-01 and SPA-02,respectively. The control effects against C. lanigera at three flying speeds of 3,5 and 8 m/s were 99. 72%-99. 97%,81. 6%-99. 81% and63. 52%-68. 77%,respectively. 10A Label-Free Colorimetric Aptasensor Containing DNA Triplex Molecular Switch and AuNP Nanozyme for Highly Sensitive Detection of Saxitoxin显示文摘Saxitoxin(STX),one of the most toxic paralytic shellfish poisons discovered to date,is listed as a required item of aquatic product safety inspection worldwide.However,conventional detection methods for STX are limited by various issues,such as low sensitivity,complicated operations,and ethical considerations.In this study,an aptamer-triplex molecular switch(APT-TMS)and gold nanoparticle(AuNP)nanozyme were combined to develop a label-free colorimetric aptasensor for the rapid and highly sensitive de-tection of STX.An anti-STX aptamer designed with pyrimidine arms and a purine chain was fabricated to form an APT-TMS.Specific binding between the aptamer and STX triggered the opening of the switch,which causes the purine chains to adsorb onto the surface of the AuNPs and enhances the peroxidase-like activity of the AuNP nanozyme toward 3,3’,5,5’-tetramethylbenzidine.Under optimized conditions,the proposed aptasensor showed high sensitivity and selectivity for STX,with a limit of detection of 335.6 pmol L^(−1) and a linear range of 0.59-150 nmol L^(−1).Moreover,good recoveries of 82.70%-92.66%for shellfish and 88.97%-106.5%for seawater were obtained.The analysis could be completed within 1 h.The proposed design also offers a robust strategy to achieve detection of other marine toxin targets by altering the corresponding aptamers.QI Xiaoyan LI Ling YAN Xiaochen ZHAO Yinglin WANG Lele MA Rui WANG Sai MAO Xiangzhao 2022Journal of Ocean University of China2022,21,5:0

12Mass transport of a mesoscale eddy in the South China Sea identified by a simulated passive tracer显示文摘To quantitatively investigate the water mass transport of mesoscale eddies,the mass transport induced by a simulated anticyclonic eddy in the South China Sea was evaluated by using the Regional Ocean Modelling System(ROMS)and a built-in passive tracer module.The results indicate that the eddy can trap and transport 51%of the initial water in the eddy core to 689 km from its origin during its lifetime of 100 days,with a stable loss rate of 6‰per day.During propagation,there is drastic horizontal water exchange between the inside and outside of the eddy.Meanwhile,the vertical mass transport is signifi cant,and 65%of the water initially in the mixed layer of the eddy is eventually detrained into the subsurface.A tracer budget analysis of eddy shows that advection is the dominant dynamic process of transport,while the eff ect of mixing is weak,and horizontal process plays a controlling role.Horizontal and vertical advection exhibit opposite patterns and strongly off set each other.Particularly,a distinct dipole pattern is found in the local velocity fi eld of the eddy,with signifi cant convergence(downwelling)and divergence(upwelling)zones in the anterior and posterior of the eddy,respectively,which is likely related to the driving mechanism of the westward propagation of the eddy.The dipole further induces a vertical overturning cell,through which the surface water in the anterior of the eddy detrains into the subsurface by downwelling and resurface from the posterior of the eddy by upwelling and gradually spreads out of the eddy.The temporal variability in the tracer budget is signifi cant,in which horizontal advection is dominant.The propagation acceleration and temporal derivative of the deformation rate are highly correlated with tracer transport,suggesting the potential eff ect of the temporal instability of eddies on the eddy mass transport.Jun ZHAO Fan WANG Shan GAO Yinglin HOU Kai LIU 2022Journal of Oceanology and Limnology2022,40,2:0
