1Essential roles of exosome and circRNA_101093 on ferroptosis desensitization in lung adenocarcinoma显示文摘Background:Resistance to ferroptosis,a regulated cell death caused by irondependent excessive accumulation of lipid peroxides,has recently been linked to lung adenocarcinoma(LUAD).Intracellular antioxidant systems are required for protection against ferroptosis.The purpose of the present studywas to investigate whether and how extracellular system desensitizes LUAD cells to ferroptosis.Methods:Established human lung fibroblasts MRC-5,WI38,and human LUAD H1650,PC9,H1975,H358,A549,and H1299 cell lines,tumor and matched normal adjacent tissues of LUAD,and plasma from healthy individuals and LUAD patients were used in this study.Immunohistochemistry and immunoblotting were used to analyze protein expression,and quantitative reverse transcription-PCR was used to analyze mRNA expression.Cell viability,cell death,and the lipid reactive oxygen species generationwere measured to evaluate the responses to ferroptosis.Exosomes were observed using transmission electron microscope.The localization of arachidonic acid(AA)was detected using click chemistry labeling followed by confocal microscopy.Interactions between RNAs and proteins were detected using RNA pull-down,RNA immunoprecipitation and photoactivatable ribonucleoside-enhanced crosslinking and immunoprecipitation methods.Proteomic analysis was used to investigate RNA-regulated proteins,and metabolomic analysis was performed to analyze metabolites.Cellderived xenograft,patient-derived xenograft,cell-implanted intrapulmonary LUAD mouse models and plasma/tissue specimens from LUAD patients were used to validate the molecular mechanism.Results:Plasma exosome from LUAD patients specifically reduced lipid peroxidation and desensitized LUAD cells to ferroptosis.A potential explanation is that exosomal circRNA_101093(cir93)maintained an elevation in intracellular cir93 in LUAD to modulate AA,a poly-unsaturated fatty acid critical for ferroptosisassociated increased peroxidation in the plasma membrane.Mechanistically,cir93 interacted with and increased fatty acid-binding protein 3(FABP3),which transported AA and facilitated its reaction with taurine.Thus,global AA was reduced,whereas N-arachidonoyl taurine(NAT,the product of AA and taurine)was induced.Notably,the role of NAT in suppressing AA incorporation into the plasma membrane was also revealed.In pre-clinical in vivo models,reducing exosome improved ferroptosis-based treatment.Conclusion:Exosome and cir93 are essential for desensitizing LUAD cells to ferroptosis,and blocking exosome may be helpful for future LUAD treatment.Xiao Zhang Yunhua Xu Lifang Ma Keke Yu Yongjie Niu Xin Xu Yi Shi Susu Guo Xiangfei Xue Yikun Wang Shiyu Qiu Jiangtao Cui Hong Wang Xiaoting Tian Yayou Miao Fanyu Meng Yongxia Qiao Yongchun Yu Jiayi Wang 2022Cancer Communications2022,42,4:10
2The generation and accumulation of natural gas from Yinan 2 gas pool in Kuqa Depression显示文摘By using the methods of hydrocarbon generation kinetics and carbon isotope kinetics, combined with geological background of natural gas pool formation, the generation and accumulation of natural gas from Yinan 2 gas pool was studied in Kuqa Depression of the Tarim Basin. Natural gas of Yinan 2 gas pool is mainly derived from Middle and Lower Jurassic coal-bearing source rocks, and generally belongs to long time-accumulated gas. It is suggested that Yinan 2 gas is chiefly accumulated in the last 5 Ma, its Ro ranges from 1.25% to 1.95%, and the loss rate of natural gas is about 25%-30%. 3Secondary aerosol formation and oxidation capacity in photooxidation in the presence of Al_2O_3 seed particles and SO_2显示文摘To investigate the sensitivity of secondary aerosol formation and oxidation capacity to NOx in homogeneous and heterogeneous reactions, a series of irradiated toluene/NOx/air and ?-pinene/NOx/air experiments were conducted in smog chambers in the absence or presence of Al2O3 seed particles. Various concentrations of NOx and volatile organic compounds(VOCs) were designed to simulate secondary aerosol formation under different scenarios for NOx. Under 'VOC-limited' conditions, the increasing NOx concentration suppressed secondary aerosol formation, while the increasing toluene concentration not only contributed to the increase in secondary aerosol formation, but also led to the elevated oxidation degree for the organic aerosol. Sulfate formation was suppressed with the increasing NOx due to a decreased oxidation capacity of the photooxidation system. Secondary organic aerosol(SOA) formation also decreased with the presence of high concentration of NOx, because organo-peroxy radicals(RO2) react with NOx instead of with peroxy radicals(RO2 or HO2), resulting in the formation of volatile organic products. The increasing concentration of NOx enhanced the formation of sulfate, nitrate and SOA under 'NOx-limited' conditions, in which the heterogeneous reactions played an important role. In the presence of Al2O3 seed particles, a synergetic promoting effect of mineral dust and NOx on secondary aerosol formation in heterogeneous reactions was observed in the photooxidation. This synergetic effect strengthened the positive relationship between NOx and secondary aerosol formation under 'NOx-limited' conditions but weakened or even overturned the negative relationship between NOx and secondary aerosol formation under 'VOC-limited' conditions. Sensitivity of secondary aerosol formation to NOx seemed different in homogeneous and heterogeneous reactions, and should be both taken into account in the sensitivity study. The sensitivity of secondary aerosol formation to NOx was further investigated under 'winter-like' and NH3-rich conditions. 4Vesicle-shaped ZIF-8 shell shielded in 3D carbon cloth for uniform nucleation and growth towards long-life lithium metal anode显示文摘Lithium metal has aroused extensive research interests as the anode for next-generation rechargeable batteries.However,the well-known dendritic Li growth and consequent safety issues still impair the long-term cycling performance.Herein,a hybrid structure composed of 3 D carbon cloth and vesicleshaped hollow ZIF-8 modification shell(HZS@CC)was prepared as a smart host for guiding uniform Li deposition.The long-range interconnected 3 D carbon fiber network enables the reduced local current density with homogeneous electrons distribution.Synergistically,abundant surface polar groups and the ultrastructure on ZIF-8 particles effectively guide a well-distributed Li-ions flow to promote the uniform Li nucleation and growth.As a result,stable Li plating/stripping for 2000 h with a low overpotential(≈15 mV)at 1 mA cm^(-2) were achieved in symmetric cells.Coupling with LiFePO_(4) cathode,the full cell delivered long life over 1200 cycles at 6 C.This research demonstrated that a homogenization guiding of Li-ions is of great importance to better make use of the structural advantage of 3 D hosts and achieve improved electrochemical performance.Yuting Fang Wenlong Cai Shanshan Zhu Kangli Xu Maogen Zhu Guannan Xiao Yongchun Zhu 2021Journal of Energy Chemistry2021,30,3:2

6Tunable optical second-order sideband effects in a parity-time symmetric optomechanical system显示文摘We theoretically investigate the optical second-order sideband generation(OSSG)in an optical parity-time(PT)symmetric system,which consists of a passive cavity trapping the atomic ensemble and an active cavity.Compared with the double-passive system,it is found that near the exceptional point(EP),the efficiency of the OSSG increases sharply not only for the blue probepump detuning resonant case but also for the red one.Using experimentally achievable parameters,we study the effect of the atomic ensemble on the efficiency of the OSSG in the PT-symmetric system.The numerical results show that the efficiency of the OSSG is 30%higher than that of the first-order sideband,which is realized easily by simultaneously modulating the atom-cavity coupling strength and detuning.Moreover,the efficiency of the OSSG can also be tuned effectively by the pump power,and the efficiency is robust when the pump power is strong enough.This study may have some guidance for modulating the nonlinear optical properties and controlling light propagation,which may stimulate further applications in optical communications.Xing Xiao QingHong Liao NanRun Zhou WenJie Nie YongChun Liu 2020Science China(Physics,Mechanics & Astronomy)2020,63,11:1

10Rapid and sensitive detection of cucumber mosaic virus by reverse transcription loop-mediated isothermal amplification显示文摘Cucumber mosaic virus(CMV),as a member of Cucumovirus genus,is one of the most common lily viruses and has been reported in many countries [1,2].The CMV-infected plants may show developmental abnormalities of leaves,flowers,and bulbs [1].The symptoms will be extremely severe in plants infected by lily symptomless virus(LSV)[1].So far,CMV has posed a threat to cultivars of lily production.Consequently,it is important to develop an efficient approach to achieve rapid and sensitive detection of CMV.Many visual and serological methods have been reported to detect CMV,such as double antibody sandwich enzyme-linked immunosorbent assay [1],immunosorbent electron microscopy [1],enzyme-linked immunosorbent assay(ELISA)and reverse transcriptase-polymerase chain reaction(RT-PCR)[2].However,these methods are time-consuming and require skilled operator and specialized equipment.Loop-mediated isothermal amplification(LAMP)method provides a possible solution for these problems [3].LAMP was developed by Notomi et al.[4] with high specificity and sensitivity because of Bst DNA polymerase.LAMP method not only is suitable for field and large-scale detection of animal virus with more cost-effective but also could be applied to detect foodborne bacterial pathogens and other viruses with high performances [5].For a naked-eye inspection,SYBR Green I was added to the LAMP reaction products to visualize the results.In this study,we developed a rapid and sensitive LAMP method to detect CMV in lily plants and a simple reverse transcription loop-mediated isothermal amplification(RT-LAMP)to detect field CMV samples in lily bulbs.Xiaorui Fan Yanan Du Youming Cai Yongchun Zhang Xiao Zhao Jieling Liang Dan Yang Qi Zhang Xiaoxia Zhang Wanjing Zhang Yan Xu Kai Zhao 2019Acta Biochimica et Biophysica Sinica2019,51,2:0
11The generation kinetics of natural gases in the Kela-2 gas field from the Kuqa Depression, Tarim Basin, northwestern China显示文摘The Kela-2 gas field, found in the Kuqa Depression of the Tarim Basin, northwestern China, is a large-sized dry gas field (C1 /C1-5 =0.992 0.999) and characterized by ultra-high pressure (pressure factor up to 2.0 2.2). The pyrolysis experiment was carried out under isothermal gold-tube closed system, with samples collected from the Jurassic coal, Jurassic mudstone and Triassic mudstone in the Kuqa Depression. The result of gas yield showed that the Middle and Lower Jurassic source rocks have higher gas generation potential than the Triassic source rocks. The kinetic modeling of gas generation and methane carbon isotope fractionation suggested that the Kela-2 gases belong to the products of high-over mature stages and were mainly derived from the Middle and Lower Jurassic coal-bearing strata. The Triassic source rocks made a minor contribution to the Kela-2 gases.
