1Key technologies for the construction of the Xiluodu high arch dam on the Jinsha River in the development of hydropower in western China显示文摘Hydropower development in China is concentrated in the country's western regions.Among all the rivers in China,the lower course of the Jinsha River contains the richest hydro-energy resource,and therefore,4 mammoth hydropower plants are under construction on this particular section of the river at Wudongde,Baihetan,Xiluodu,and Xiangjiaba.The water-blocking structures of the hydropower facilities at Wudongde,Baihetan and Xiluodu are all arch dams of around 300 m high.In view of changes in the geological conditions at the foundation of the Xiluodu dam on the riverbed after excavation started,the designs of expanding foundation surface excavation and dovetailing the dam body and foundation rock on both upstream and downstream sides were introduced,allowing the arch dam and foundation to fit each other and improving the stress conditions of the dam body and foundation.By dividing the dam body into various concrete sections,the dynamic properties of concrete were adequately adjusted to the distribution of stress in the dam body.In addition,the use of the most optimal concrete material and mixture ratio allowed thermodynamics of concrete to satisfy the requirements of the strength,durability,temperature control and crack prevention of the concrete.Moreover,rigorous temperature control measures were introduced to prevent harmful cracking,thus enhancing the integrity of the arch dam.Furthermore,sophisticated construction machinery,scientific testing methods,and sound construction techniques were employed to ensure the uniformity and reliability of concrete placement.The 'Digital Dam' for the Xiluodu project,which is based on the theory of total life cycle,has supplied strong support for construction process control and decision-making.Lu Youmei Fan Qixiang Zhou Shaowu Li Bingfeng Li Wenwei 2012Engineering Sciences2012,10,2:3

6Design of a novel chimeric peptide via dual blockade of CD47/SIRPαand PD-1/PD-L1 for cancer immunotherapy显示文摘Although immune checkpoint inhibition has been shown to effectively activate antitumor immunity in various tumor types,only a small subset of patients can benefit from PD-1/PD-L1 blockade.CD47 expressed on tumor cells protects them from phagocytosis through interaction with SIRPαon macrophages,while PD-L1 dampens T cell-mediated tumor killing.Therefore,dual targeting PD-L1 and CD47 may improve the efficacy of cancer immunotherapy.A chimeric peptide Pal-DMPOP was designed by conjugating the double mutation of CD47/SIRPαblocking peptide(DMP)with the truncation of PD-1/PD-L1 blocking peptide OPBP-1(8-12)and was modified by a palmitic acid tail.Pal-DMPOP can significantly enhance macrophage-mediated phagocytosis of tumor cells and activate primary T cells to secret IFN-γin vitro.Due to its superior hydrolysis-resistant activity as well as tumor tissue and lymph node targeting properties,Pal-DMPOP elicited stronger anti-tumor potency than Pal-DMP or OPBP-1(8-12)in immune-competent MC38 tumor-bearing mice.The in vivo anti-tumor activity was further validated in the colorectal CT26 tumor model.Furthermore,Pal-DMPOP mobilized macrophage and T-cell anti-tumor responses with minimal toxicity.Overall,the first bispecific CD47/SIRPαand PD-1/PD-L1 dual-blockade chimeric peptide was designed and exhibited synergistic anti-tumor efficacy via CD8^(+)T cell activation and macrophage-mediated immune response.The strategy could pave the way for designing effective therapeutic agents for cancer immunotherapy.Zheng Hu Wanqiong Li Shaomeng Chen Danhong Chen Ran Xu Danlu Zheng Xin Yang Shuzhen Li Xiuman Zhou Xiaoshuang Niu Youmei Xiao Zhuoying He Huihao Li Juan Liu Xinghua Sui Yanfeng Gao 2023Science China(Life Sciences)2023,66,10:1

8Geographic Information and Node Selfish-Based Routing Algorithm for Delay Tolerant Networks显示文摘In Delay Tolerant Networks(DTNs), some routing algorithms ignore that most nodes are selfish, i.e.,nodes are willing to use their own resources to forward messages to nodes with whom they have a relationship. In view of this phenomenon, we propose a routing algorithm based on Geographic Information and Node Selfishness(GINS). To choose a forwarding node, GINS combines nodes' willingness to forward and their geographic information to maximize the possibility of contacting the destination. GINS formulates the message forwarding process as a 0-1 Knapsack Problem with Assignment Restrictions to satisfy node demands for selfishness.Extensive simulations were conducted, and results show that GINS can achieve a high delivery ratio and a lower hop count compared with GRONE and LPHU. 15Targeting cAMP in D1-MSNs in the nucleus accumbens, a new rapid antidepressant strategy显示文摘Studies have suggested that the nucleus accumbens(NAc)is implicated in the pathophysiology of major depression;however,the regulatory strategy that targets the NAc to achieve an exclusive and outstanding anti-depression benefit has not been elucidated.Here,we identified a specific reduction of cyclic adenosine monophosphate(cAMP)in the subset of dopamine D1 receptor medium spiny neurons(D1-MSNs)in the NAc that promoted stress susceptibility,while the stimulation of cAMP production in NAc D1-MSNs efficiently rescued depression-like behaviors.Ketamine treatment enhanced cAMP both in D1-MSNs and dopamine D2 receptor medium spiny neurons(D2-MSNs)of depressed mice,however,the rapid antidepressant effect of ketamine solely depended on elevating cAMP in NAc D1-MSNs.We discovered that a higher dose of crocin markedly increased cAMP in the NAc and consistently relieved depression 24 h after oral administration,but not a lower dose.The fast onset property of crocin was verified through multicenter studies.Moreover,crocin specifically targeted at D1-MSN cAMP signaling in the NAc to relieve depression and had no effect on D2-MSN.These findings characterize a new strategy to achieve an exclusive and outstanding anti-depression benefit by elevating cAMP in D1-MSNs in the NAc,and provide a potential rapid antidepressant drug candidate,crocin.Yue Zhang Jingwen Gao Na Li Peng Xu Shimeng Qu Jinqian Cheng Mingrui Wang Xueru Li Yaheng Song Fan Xiao Xinyu Yang Jihong Liu Hao Hong Ronghao Mu Xiaotian Li Youmei Wang Hui Xu Yuan Xie Tianming Gao Guangji Wang Jiye Aa 2024Acta Pharmaceutica Sinica B2024,14,2:0

19A Simple Clear Technique in Observing Vascular Development of Grape Ovary显示文摘The vascular system of the grapevine(Vitis vinifera L.)flower is a channel for transporting water and nutrients to the ovary.It plays an important role in the development of the ovary and fertilization through pollination.However,the vascular bundles in the flower are so tiny that they are difficult to sample and observe by traditional slicing techniques.In this study,‘Summer Black’grape flowers were selected as the test materials,and the tissue samples were treated by the optical clearing technique.After simple compaction,the structure and development of the vasculature were observed by common microscopy,fluorescence microscopy and laser confocal microscopy.The results showed that the transparency effects of 3%NaOH and a saturated trichloroacetaldehyde composite agreed well with the observations of the vascular structure and the developmental process of the flower in different periods.Moreover,the samples after optical clearing could be reconstructed in 3D,which helped us know more about its development and function.According to these observations,the vasculature of the‘Summer Black’flower can be divided into ovule vascular bundles,peripheral vascular bundles and central vascular bundles.The peripheral vascular bundles were composed of the first-order vascular bundles and the inferior vascular bundles which branched from the superior vascular bundles.These bundles branched in different directions with no discernible pattern.The two different branching methods were as follows.First,the inferior vascular bundle was directly connected to a superior vascular bundle.Secondly,some of the superior vascular bundles bent in different ways,forming the inferior vascular bundle connecting the superior vascular bundles by a metamorphosed vessel with a triangular shape.In a comparison of the developmental changes in various periods,the growth of vascular bundles at each period was directly proportional to the growth of the flower.Laser confocal scanning was used to explore the three-dimensional morphology of the peripheral vascular bundle and showed that the peripheral vascular bundle of grapes was not completely parallel to the flower’s epidermal cells.As a result,the optical clearing technique was convenient and authentic compared with the traditional slicing operation for tiny flower organs.With these advantages according to the observations,this study provides a feasible technique and useful information for the study of vascular bundle development in grape flower organs.Teng Fei Youmei Li Bo Li Zhaosen Xie 2023Phyton-International Journal of Experimental Botany2023,92,7:0
