2Taurolidine improved protection against highly pathogenetic avian influenza H5N1 virus lethal-infection in mouse model by regulating the NF-κB signaling pathway显示文摘Taurolidine(TRD),a derivative of taurine,has anti-bacterial and anti-tumor effects by chemically reacting with cell-walls,endotoxins and exotoxins to inhibit the adhesion of microorganisms.However,its application in antiviral therapy is seldom reported.Here,we reported that TRD significantly inhibited the replication of influenza virus H5N1 in MDCK cells with the half-maximal inhibitory concentration(EC_(50))of 34.45μg/mL.Furthermore,the drug inhibited the amplification of the cytokine storm effect and improved the survival rate of mice lethal challenged with H5N1(protection rate was 86%).Moreover,TRD attenuated virus-induced lung damage and reduced virus titers in mice lungs.Administration of TRD reduced the number of neutrophils and increased the number of lymphocytes in the blood of H5N1 virus-infected mice.Importantly,the drug regulated the NF-κB signaling pathway by inhibiting the separation of NF-κB and IκBa,thereby reducing the expression of inflammatory factors.In conclusion,our findings suggested that TRD could act as a potential anti-influenza drug candidate in further clinical studies.Chaoxiang Lv Yuanguo Li Tiecheng Wang Qiqi Zhang Jing Qi Mingwei Sima Entao Li Tian Qin Zhuangzhuang Shi Fangxu Li Xuefeng Wang Weiyang Sun Na Feng Songtao Yang Xianzhu Xia Ningyi Jin Yifa Zhou Yuwei Gao 2023Virologica Sinica2023,38,1:0
3Histones of Neutrophil Extracellular Traps Induce CD11b Expression in Brain Pericytes Via Dectin-1 after Traumatic Brain Injury显示文摘The brain pericyte is a unique and indispensable part of the blood-brain barrier(BBB),and contributes to several pathological processes in traumatic brain injury(TBI).However,the cellular and molecular mechanisms by which pericytes are regulated in the damaged brain are largely unknown.Here,we show that the formation of neutrophil extracellular traps(NETs)induces the appearance of CD11b^(+)pericytes after TBI.These CD11b^(+)pericyte subsets are characterized by increased permeability and pro-inflammatory profiles compared to CD11b-pericytes.Moreover,histones from NETs by Dectin-1 facilitate CD11b induction in brain pericytes in PKC-c-Jun dependent manner,resulting in neuroinflammation and BBB dysfunction after TBI.These data indicate that neutrophil-NET-pericyte and histone-Dectin-1-CD11b are possible mechanisms for the activation and dysfunction of pericytes.Targeting NETs formation and Dectin-1 are promising means of treating TBI.Yang-Wuyue Liu Jingyu Zhang Wanda Bi Mi Zhou Jiabo Li Tiantian Xiong Nan Yang Li Zhao Xing Chen Yuanguo Zhou Wenhui He Teng Yang Hao Wang Lunshan Xu Shuang-Shuang Dai 2022Neuroscience Bulletin2022,38,10:0
4Domain decomposition based on the spectral element method for frequency-domain computational elastodynamics显示文摘We propose a domain decomposition method based on the spectral element method(DDM-SEM)for elastic wave computation in frequency domain.It combines the high accuracy of the spectral element method and the high degree of parallelism of a domain decomposition technique,which makes this method suitable for accurate and efficient simulations of large scale problems in elastodynamics.In the DDM-SEM,the original large-scale problem is divided into a number of well designed subdomains.We use the spectral element method independently for each subdomain,and the neighboring subdomains are connected by a frequency-domain version of Riemann transmission condition(RTC)for elastic waves.For the proposed method,we can employ the non-conforming meshes and different interpolation orders in different subdomains to maximize the efficiency.By separating the internal and boundary unknowns of each subdomain,an efficient and naturally parallelizable block LDU direct solver is developed to solve the final system matrix.Numerical experiments verify its accuracy and efficiency,and show that the proposed DDM-SEM can be a promising numerical tool for accurately and effectively solving large and multi-scale problems of elastic waves.It is potentially valuable for the frequency domain seismic inversion where multiple source illuminations are required.Linlin SHI Mingwei ZHUANG Yuanguo ZHOU Na LIU Qinghuo LIU 2021Science China Earth Sciences2021,64,3:0
5Construction of Hsp90β gene specific silencing plasmid and its transfection efficiency显示文摘The purpose of this work was to construct the plasmid that could direct the synthesis of siRNA-like transcripts and thus mediate strong and specific repression of human heat shock protein 90β(Hsp90β)gene expression and to compare the transfection efficiency of the plasmids in varying conditions of transfection.Three 64 nt oligos corresponding to different regions of the target gene were chemically synthesized and annealed and were then ligated with pSUPER EGFP1 plasmid and double-digested with HindIII and BglII.Recombinant plasmids were transformed into Escherichia coli,DH5a,and the colonies were picked and grown in the Amp-agarose.The presence of positive clones was checked by the means of endodigestion and sequencing.Three cell strains,HepG2,Human umbilicus vein endothelium cells(HUVEC)and HeK293,were cultured.Then the plasmids were transfected into the cells at different ratios of plasmid to Lipofectamine.The transfection efficiency was measured by detection of enhanced green fluorescence protein(EGFP).The presence of positive recombinant clones were verified by double-digestion and sequencing.The bases inserted into the plasmids were correct and the positive colonies were named pSuper-Hsp90β1,pSuper-Hsp90β2 and pSuper-Hsp90β3.After optimizing the ratio of plasmid to Lipofectamine,we achieved high transfection efficiency in HeK293 cells.Transfection efficiency was still low in the HepG2 cells.In conclusion,the si-RNA-synthesizing plasmids targeting Hsp90βwere constructed and transfected into cells with different transfection efficiency.JI Yewei NIE Bin LI Ping XU Xiaoyu ZHOU Yuanguo 2007Frontiers of Medicine2007,1,3:0
