1Efficacy and safety of low-dose interleukin-2 in combination with methotrexate in patients with active rheumatoid arthritis:a randomized,double-blind,placebo-controlled phase 2 trial显示文摘Rheumatoid arthritis(RA)is an aggressive autoimmune arthritis,and current therapies remain unsatisfactory due to low remission rate and substantially adverse effects.Low-dose interleukin-2(Ld-IL2)is potentially a therapeutic approach to further improve the disease.This randomized,double-blind,placebo-controlled trial was undertaken to evaluate the efficacy and safety of Ld-IL2 in patients with active RA.Patients were randomly assigned(1:1)to receive Ld-IL2,defined as a dose of 1 million IU,or placebo in a 12-week trial with a 12-week follow-up.Three cycles of Ld-IL2 or placebo were administered subcutaneously every other day for 2 weeks(a total of 7 doses),followed by a 2-week break.All patients received a stable dose of methotrexate(MTX).The primary outcomes were the proportion of patients achieving the ACR20,DAS28-ESR<2.6,and the change from baseline in CDAI or SDAI at week 24.Secondary endpoints included other clinical responses and safety.The primary outcomes were achieved in the perprotocol population.The improvements from baseline in CDAI and SDAI were significantly greater across time points for the LdIL2+MTX group(n=17)than for the placebo+MTX group(n=23)(P=0.018 and P=0.015,respectively).More patients achieved ACR20 response in the Ld-IL2+MTX group than those in the placebo+MTX group at week 12(70.6%vs 43.5%)and at week 24(76.5%vs 56.5%)(P=0.014).In addition,low Treg and high IL-21 were associated with good responses to Ld-IL2.Ld-IL-2 treatment was well-tolerated in this study.These results suggested that Ld-IL2 was effective and safe in RA.ClinicalTrials.gov number:NCT 02467504.Xiaoying Zhang Miao Miao Ruijun Zhang Xu Liu Xiaozhen Zhao Miao Shao Tian Liu Yuebo Jin Jiali Chen Huixin Liu Xia Zhang Yun Li Yunshan Zhou Yue Yang Ru Li Haihong Yao Yanying Liu Chun Li Yuhui Li Limin Ren Yin Su Xiaolin Sun Jing He Zhanguo Li 2022Signal Transduction and Targeted Therapy2022,7,4:7
2Analysis and Prediction of Influence Imposed on Jiaozhou Bay Tidal Currents and Tidal Energy of M_2 Tidal System by Jiaozhou Bay Reclamation显示文摘The 3-D ECOMSED ocean model was applied to establish a time-dependent boundary model for Jiaozhou Bay (JZB), in which the operator-splitting technique was used and the ‘dry and wet’ method was introduced. The influence caused by JZB reclamation on the surface level, residual currents, tidal system and tidal energy of M2 tidal system were predicted and analyzed. The results show that JZB reclamation has slight impact on the M2 tidal system, in which the variation of amplitude and phase is less than 1%.The changes of the currents and residual currents in Qian Bay and near the reclamation areas are greater, but in other areas the changes are smaller, in which the currents have a change of around 1%, while the residual currents change ranges from 1.82%–9.61%. 3Genome-wide identification of RNA editing in seven porcine tissues by matched DNA and RNA high-throughput sequencing显示文摘Background: RNA editing is a co/posttranscriptional modification mechanism that increases the diversity of transcripts, with potential functional consequences. The advent of next-generation sequencing technologies has enabled the identification of RNA edits at unprecedented throughput and resolution. However, our knowledge of RNA editing in swine is still limited.Results: Here, we utilized RES-Scanner to identify RNA editing sites in the brain, subcutaneous fat, heart, liver,muscle, lung and ovary in three 180-day-old Large White gilts based on matched strand-specific RNA sequencing and whole-genome resequencing datasets. In total, we identified 74863 editing sites, and 92.1% of these sites caused adenosine-to-guanosine(A-to-G) conversion. Most A-to-G sites were located in noncoding regions and generally had low editing levels. In total, 151 A-to-G sites were detected in coding regions(CDS), including 94 sites that could lead to nonsynonymous amino acid changes. We provide further evidence supporting a previous observation that pig transcriptomes are highly editable at PRE-1 elements. The number of A-to-G editing sites ranged from 4155(muscle) to 25001(brain) across the seven tissues. The expression levels of the ADAR enzymes could explain some but not all of this variation across tissues. The functional analysis of the genes with tissuespecific editing sites in each tissue revealed that RNA editing might play important roles in tissue function.Specifically, more pathways showed significant enrichment in the fat and liver than in other tissues, while no pathway was enriched in the muscle.Conclusions: This study identified a total of 74863 nonredundant RNA editing sites in seven tissues and revealed the potential importance of RNA editing in tissue function. 4Urban Plant Diversity in Relation to Land Use Types in Built-up Areas of Beijing显示文摘Urban plants provide various ecosystem services and biodiversity for human well-being. It is necessary to examine the plant species and functional traits composition and the influencing factors. In this study, a field survey was conducted using the tessellation-randomized plot method to assess the plant species and functional traits variability in greenspaces across eight land use types(LUTs) in the built-up areas of Beijing, China. Results showed that the woody plants in the built-up areas of Beijing comprised 85 non-native species(57%), 21 pollen-allergenic species(14%), and 99 resistant species(67%). Residential areas, community parks and institutional areas had higher woody plant species richness than other LUTs. Native and extralimital native species were more widespread than exotic species. Proportions of species with resistances were low except for cold-and drought-resistance; consequently, a high intensity of management and maintenance is essential for survival of plants in this urban area. Caution should be exerted in selecting plant species with resistance to harsh conditions in different LUTs. Housing prices, distances from the urban center, years since the establishment of LUTs and greening rate were strongly correlated with the plant functional traits and species diversity. 5Multi-target localization for bistatic MIMO radar in the presence of unknown mutual coupling显示文摘A decoupling-estimation signal parameters via rotarional invariance technique(ESPRIT) method is presented for multi-target localization with unknown mutual coupling in bistatic multiple-input multiple-output(MIMO) radar.Two steps are carried out in this method.The decoupling operation between angle and mutual coupling estimates is realized by choosing the auxiliary elements on both sides of the transmit and receive uniform linear arrays(ULAs).Then the ESPRIT method is resilient against the unknown mutual coupling matrix(MCM) and can be directly utilized to estimate the direction of departure(DOD) and the direction of arrival(DOA).Moreover,the mutual coupling coefficient is estimated by finding the solution of the linear constrained optimization problem.The proposed method allows an efficient DOD and DOA estimates with automatic pairing.Simulation results are presented to verify the effectiveness of the proposed method.Zhidong Zheng Jianyun Zhang Yuebo Wu 2012Journal of Systems Engineering and Electronics2012,23,5:3

8观察参芪扶正注射液在卵巢癌化疗中减轻毒副作用的效果(英文)显示文摘Objective: The aim of the study was to observe the clinical effects of Shenqi Fuzheng injection decreasing sideeffects of chemotherapy for patients with ovarian epithelial cancer. Methods: The 36 cases of ovarian epithelial cancer in The Third Affiliated Hospital of SUN Yat-sen University(Guangzhou, China) from June 2010 to June 2013, were randomly divided into the study group and the control group. The study group contained 18 cases using Shenqi Fuzheng injection combined with TP(Taxol + Carboplatin/cisplatin) chemotherapy, and the control group contained 18 cases only using TP chemotherapy without Shenqi Fuzheng injection. During and after chemotherapy, the side-effects and therapy effects were observed. Results: The grade II of nausea and vomit were less in the study group than that in the control group, which was significantly different(P < 0.05). But there were no significantly differences in grade I of nausea and vomit between the two groups(P > 0.05). There was less degree of decrease of lymphocyte in the study group than that in the control group, which was significantly different(P < 0.05). But between two groups, there were no significant differences in the drop of white blood cells and red blood cells, the incidence of allergic reaction, liver and renal functions, nerve lesion and phalacrosis(P > 0.05). There was no significant difference in chemotherapy effect between the two groups(P > 0.05). 12The COVID‐19 vaccine: Attitudes and vaccination in patients with autoimmune inflammatory rheumatic diseases显示文摘Background:We examined attitudes toward the COVID‐19 vaccine,potential factors underlying these attitudes,and ways to increase vaccination willingness in autoimmune inflammatory rheumatic diseases(AIIRD)patients.Methods:A multicenter,web‐based,observational survey using an online questionnaire was conducted among AIIRD patients aged≥18 years from May 24,2021,to June 3,2021.Participants were 3104 AIIRD patients(2921 unvaccinated and 183 vaccinated).Results:Of the unvaccinated patients,32.9%were willing to receive the COVID‐19 vaccine,45.0%were uncertain,and 14.8%were unwilling.When vaccination was recommended by physicians,patients'willingness increased to 93.8%.Participants'main concerns were that the vaccine may aggravate AIIRD disease(63.0%)and may cause vaccine‐related adverse events(19.9%).Female patients were less likely to be vaccinated.However,patients who had children aged≤18 years were more willing to be vaccinated.In addition,vaccination willingness was higher in patients with trust in the safety and efficacy of the COVID‐19 vaccine.Notably,183(5.9%)patients were vaccinated.The major vaccination side effects were injection reaction,myalgia,and fatigue.At a median follow‐up of 88(38,131)days,patients'disease activities were stable.Conclusions:The findings show that AIIRD patients were unwilling to receive the COVID‐19 vaccine because of fears of potential disease exacerbation and additional adverse events.Sociodemographic characteristics and concerns about COVID‐19 disease and vaccines had a significant effect on vaccination willingness.Jiali Chen Wenxin Cai Tian Liu Yunshan Zhou Yuebo Jin Yue Yang Shi Chen Kun Tang Chun Li 2022Rheumatology & Autoimmunity2022,2,2:1

15Porcine methionine sulfoxide reductase B3:molecular cloning,tissue-specific expression profiles,and polymorphisms associated with ear size in Sus scrofa显示文摘Background: In Sus scrofa, methionine sulfoxide reductase B3(MSRB3) is a crucial candidate gene for ear size, and an important conformational trait of pig breeds. However, challenges in MSRB3 c DNA amplification have prevented further identification of MSRB3 allelic variants influencing pig ear size.Results: We cloned a full-length c DNA sequence of porcine MSRB3 by rapid-amplification of c DNA ends. The3,765-bp gene contained a 5'-untranslated region(UTR)(190 bp), a coding region(552 bp), and a 3'-UTR(3,016 bp) and shared 84 %, 84 %, 87 %, 86 %, and 70 % sequence identities with human, orangutan, mouse, chicken, and zebrafish,respectively. The gene encoded a 183-amino acid protein, which shared 88 %, 91 %, 89 %, 86 %, and 67 % identities with human, orangutan, mouse, chicken, and zebrafish, respectively. Tissue expression analysis using q RT-PCR revealed that MSRB3 was expressed in the heart, liver, lung, kidney, spleen, ear, muscle, fat, lymph, skeletal, and hypothalamic tissues. Three single nucleotide polymorphisms(SNPs) were identified in MSRB3: c.-735 C > T in the 5' flanking region,c.2571 T > C in the 3'-UTR, and a synonymous mutation of c.484 T > C in the coding region. The SNPs c.-735 C > T and c.2571 T > C were significantly associated with ear size in a Large White × Minzhu F2 population other than in Beijing Black pigs. Subsequently, at SNP c.-735 C > T, the m RNA of MSRB3 was significantly higher expressed in ears of individuals with the TT genotype(Minzhu) than those with CC(Large White).Conclusions: The porcine MSRB3 owned a 3,765-bp full-length c DNA sequence and was detected to express in ear tissue. Two SNPs of this gene were shown to be significantly associated with ear size in a Large White × Minzhu intercross population instead of Beijing Black pig population. What's more, the individuals with higher m RNA expression of MSRB3 have larger ear sizes. 20The gap in research on polyploidization between plants and vertebrates:model systems and strategic challenges显示文摘Polyploidization via whole-genome duplications(WGD) is a common phenomenon in organisms.However, investigations into this phenomenon differ greatly between plants and animals. Recent research on polyploid plants illustrates the immediate changes that follow WGDs and the mechanisms behind in both genetic and epigenetic consequences. Unfortunately, equivalent questions remain to be explored in animals. Enlightened by botanical research, the study of polyploidization in vertebrates involves the identification of model animals and the establishment of strategies. Here we review and comparethe research on plants and vertebrates while considering intrageneric or intraspecific variation in genome size.Suitable research methods on recently established polyploidy systems could provide important clues for understanding what happens after WGDs in vertebrates. The approach yields insights into survival and the rarity of polyploidization in vertebrates.
