| 2 | Very-High Color Rendering Index Hybrid White Organic Light-Emitting Diodes with Double Emitting Nanolayers显示文摘A very-high color rendering index white organic light-emitting diode(WOLED) based on a simple structure was successfully fabricated. The optimized device exhibits a maximum total efficiency of 13.1 and 5.4 lm/W at 1,000 cd/m2. A peak color rendering index of 90 and a relatively stable color during a wide range of luminance were obtained. | 3 | Expression, characterization, and antimicrobial ability of T4 lysozyme from methylotrophic yeast Hansenula polymorpha A16显示文摘Lysozyme is an enzyme that is essential for protection against bacterial infections.In this study,a T4 lysozyme gene was cloned into the yeast expression vector pPIC9K under the control of the Pichia pastoris glyceraldehyde-3-phosphate dehydrogenase promoter (pGAP).A Hansenula polymorpha-derived ribosomal DNA (rDNA)-targeting element was inserted into the expression vector and was critical for stable DNA integration into the H.polymorpha chromosome.Recombinant T4 lysozyme was successfully expressed in the yeast H.polymorpha A16;0.49 g L-1 secreted recombinant T4 lysozyme was obtained 72 h after incubation in culture broth that had an initial pH of 6.0.Recombinant T4 lysozyme showed lytic activity against the cell walls of the gram positive bacteria,Micrococcus lysodeikticus,and the gram negative bacteria Xanthomonas campestris pv.malvacearum and Xanthomonas oryzae pv.oryzae.The zone of inhibition assay was used to evaluate antimicrobial activity.Mass spectrometry showed the N-terminal sequence of recombinant T4 lysozyme was identical to that of the native enzyme.SDS-PAGE indicated that the molecular mass of recombinant T4 lysozyme was 18.7 kD which corresponds to a monomer of the native enzyme.SDS-PAGE without 0.2 mol L-1 dithiothreitol treatment detected two bands (15 and 31 kD) suggesting that some recombinant T4 lysozyme formed interand intra-molecular disulfide bonds which resulted in loss of enzyme activity. | WANG Nan WANG YueJu LI GangQiang SUN Ning LIU DeHu | 2011 | Science China(Life Sciences)2011,54,6: | 3 |
| 4 | Rapid and convenient transformation of cotton (Gossypium hirsutum L.) using in planta shoot apex via glyphosate selection显示文摘Cotton plants are recalcitrant with regards to transformation and induced regeneration.In the present study,5-enolpyruvylshikimate-3-phosphate(EPSPS),a glyphosate resistant gene from the bacterium Agrobacterium sp.strain CP4,was introduced into an elite Bt transgenic cotton cultivar with a modified technique involving in planta Agrobacteriummediated transformation of shoot apex.Primary transformants were initially screened using a 0.26%glyphosate spray and subsequently by PCR analysis.Five out of 4 000 transformants from T_1 seeds were obtained resulting in an in planta transformation rate of 0.125%.Four homozygous lines were produced by continuous self-fertilization and both PCR-based selection and glyphosate resistance.Transgene insertion was analyzed by Southern blot analysis.Gene transcription and protein expression levels in the transgenic cotton lines were further investigated by RT-PCR,Western blot,and ELISA methods.Transgenic T_3 plants were resistant to as much as 0.4% of glyphosate treatments in field trials.Our results indicate that the cotton shoot apex transformation technique which is both tissue-culture and genotype-independent would enable the exploitation of transgene technology in different cotton cultivars.Since this method does not require sterile conditions,the use of specialized growth media or the application of plant hormones,it can be conducted under the greenhouse condition. | GUO Wen-fang Kevin Yueju Wang WANG Nan LI Jun LI Gang-qiang LIU De-hu | 2018 | Journal of Integrative Agriculture2018,17,10: | 2 |
| 5 | A first-principles study of helium diffusion in quartz and coesite under high pressure up to 12GPa显示文摘Helium diffusion in mantle minerals is crucial for understanding mantle structure and the dynamic processes of Earth's degassing.In this paper,we report helium incorporation and the mechanism of its diffusion in perfect crystals of quartz and coesite.The diffusion pathways,activation energies(Ea),and frequency factors of helium under ambient and high pressure conditions were calculated using Density Functional Theory(DFT)and the climbing image nudged elastic band(CI-NEB)method.The calculated diffusive coefficients of He in the quartz in different orientations are:D[100]=1.24×10^(−6)exp.(−26.83 kJ/mol/RT)m^(2)/s D[010]=1.11×10^(−6)exp.(−31.60 kJ/mol/RT)m^(2)/s.and in the coesite:D[100]=3.00×10^(−7)exp.(−33.79 kJ/mol/RT)m^(2)/s D[001]=2.21×10^(−6)exp.(−18.33 kJ/mol/RT)m^(2)/s.The calculated results indicate that diffusivity of helium is anisotropic in both quartz and coesite and that the degree of anisotropy is much more pronounced in coesite.Helium diffusion behavior in coesite under high pressures was investigated.The activation energies increased with pressure:Ea[100]increased from 33.79 kJ/mol to 58.36 kJ/mol,and Ea[001]increased from 18.33 kJ/mol to 48.87 kJ/mol as pressure increased from0 GPa to 12 GPa.Our calculations showed that helium is not be quantitatively retained in silica at typical surface temperatures on Earth,which is consistent with the findings from previous studies.These results have implications for discussion of the Earth's mantle evolution and for recognition thermal histories of ultra-high pressure(UHP)metamorphic terranes. | Hong Liu Lingling Wang Shuchen Li Yaochun Yang Hua Tian Fengxia Sun Chaowen Xu Lei Liu Yi Li Yueju Cui Ying Li Jijun Zhao | 2021 | Geoscience Frontiers2021,12,2: | 1 |
| 6 | Expression, Characterization and Antimicrobial Ability of a Variant T4 Lysozyme in Pichia pastoris显示文摘T4 lysozyme was engineered with disulfide bonds and expressed in Pichia pastoris. The secreted proteins were purified and made into powder by lyophilization. Recombinant protein purity was more than 70% measured by HPLC. The lytic activity of variant T4-lysozyme was measured by the lysis of the cell wall of Xanthomonas oryzae pv. oryzae, X. oryzae pv. oryzicola, Ralstonia solanacearum comb.nov, Clavibacter michiganensis subsp. michiganensis, X. campestris pv. malvacearum, Fusarium oxysporium sp. vasinfectum, Verticillium dahliae kleb. Inhibition zone assay showed that variant T4 lysozyme significantly inhibited X. o. oryzicola and X. c. malvacearum.
