1Adaptive and regulatory mechanisms in aged rats with postoperative cognitive dysfunction显示文摘Inflammation may play a role in postoperative cognitive dysfunction. 5′ Adenosine monophosphate-activated protein kinase, nuclear factor-kappa B, interleukin-1β, and tumor necrosis factor-α are involved in inflammation. Therefore, these inflammatory mediators may be involved in postoperative cognitive dysfunction. Western immunoblot analysis revealed 5′ adenosine monophosphate-activated protein kinase and nuclear factor-kappa B in the hippocampus of aged rats were increased 1–7 days after splenectomy. Moreover, interleukin-1β and tumor necrosis factor-α were upregulated and gradually decreased. 2Personalized neoantigen pulsed dendritic cell vaccine for advanced lung cancer显示文摘Neoantigens are considered to be ultimate target of tumor immunotherapy due to their high tumor specificity and immunogenicity.Dendritic cell(DCs)vaccines based on neoantigens have exciting effects in treatment of some malignant tumors and are a promising therapeutic modality.Lung cancer is a lethal disease with the highest morbidity and mortality rate in the world.Despite the rapid development of targeted therapy and immune checkpoint inhibitors for lung cancer in recent years,their efficacy is still unsatisfactory overall.Therefore,there is an urgent unmet clinical need for lung cancer treatment.Here,we attempted to treat lung cancer using a personalized neoantigen peptide-pulsed autologous DC vaccine and conducted a single-arm,2 medical centers,pilot study initiated by the investigator(ChiCTR-ONC 16009100,NCT02956551).The patients enrolled were patients with heavily treated metastatic lung cancer.Candidate neoantigens were derived from whole-exome sequencing and RNA sequencing of fresh biopsy tissues as well as bioinformatics analysis.A total of 12 patients were enrolled in this study.A total of 85 vaccine treatments were administered with a median value of 5 doses/person(range:3-14 doses/person).In total,12-30 peptide-based neoantigens were selected for each patient.AIl treatment-related adverse events were grade 1-2 and there were no delays in dosing due to toxic effects.The objective effectiveness rate was 25%;the disease control rate was 75%;the median progression-free survival was 5.5 months and the median overall survival was 7.9 months.This study provides new evidence for neoantigen vaccine therapy and new therapeutic opportunities for lung cancer treatment.Zhenyu Ding Qing Li Rui Zhang Li Xie Yang shu Song Gao Peipei Wang Xiaoqing Su Yun Qin Yuelan Wang Juemin Fang Zhongzheng Zhu Xuyang Xia Guochao Wei Hui Wang Hong Qian Xianling Guo Zhibo Gao Yu Wang Yuquan Wei Qing Xu Heng Xu Li Yang 2021Signal Transduction and Targeted Therapy2021,6,2:3
3Petrography and chronology of lunar meteorite Northwest Africa 6950显示文摘Northwest Africa(NWA)6950 is an olivine-gabbro lunar meteorite that has a distinctly different petrographic texture from other lunar basalts.In this contribution,we report the petrography,mineralogy,and U-Pb geochronology of baddeleyite and phosphate in this lunar meteorite and make a comparison with other NWA 773 paired meteorites.NWA 6950 consists mainly of coarse-grained olivine,pyroxene,and plagioclase,with minor ilmenite,chromite,troilite,baddeleyite,taenite,and apatite.The abundant rounded to euhedral olivine has a limited Fo ranging from 68 to 69.Pyroxenes include both augite and pigeonite,with a narrow range of Mg#=71–80,and composition,respectively,En46-52Fs12-16Wo32-41 and En58-68Fs19-28Wo7-20.Plagioclase is mainly anhedral,conforming to the crystal margins of olivine and pyroxene.The plagioclase grains are elongated and several hundreds of microns in length.Raman spectra shows that plagioclase has been transformed into maskelynite,which is very calcic(An87-95)with low sodium and very low potassium(Ab4-9Or0.6-3.7).The weighted mean 207Pb/206Pb ages of baddeleyite and phosphate measured using a sensitive high resolution ion micro probe(SHRIMP II)are 3119±16 Ma(n=9,MSWD=2.5)(mean squared weighted deviates,MSWD)and 3119±18 Ma(n=15,MSWD=2.1),respectively,giving the crystallization age of the meteorite.NWA 6950 has similar mineral compositions,geochemical characteristics,and an almost identical age of crystallization to the magnesian gabbro of the NWA 773 clan.We conclude that NWA 6950 and NWA 773 are paired meteorites,which crystallize at ca.3119 Ma,and represent magmatic activities in the KREEP(potassium(K),rare earth elements(REE)and phosphorus(P)rich lunar material)region of the Moon.Zemin BAO Yuruo SHI JLawford ANDERSON Allen KENNEDY Zuokai KE Xiangping GU Peizhi WANG Xiaochao CHE Yuelan KANG Huiyi SUN Chen WANG 2020Science China(Information Sciences)2020,63,4:2

5Effect of TGO on the tensile failure behavior of thermal barrier coatings显示文摘Thermally grown oxide(TGO)may be generated in thermal barrier coatings(TBCs)after high-temperature oxidation.TGO increases the internal stress of the coatings,leading to the spalling of the coatings.Scanning electron microscopy and energy-dispersive spectroscopy were used to investigate the growth characteristics,microstructure,and composition of TGO after high-temperature oxidation for 0,10,30,and 50 h,and the results were systematically compared.Acoustic emission(AE)signals and the strain on the coating surface under static load were measured with AE technology and digital image correlation.Results showed that TGO gradually grew and thickened with the increase in oxidation time.The thickened TGO had preferential multi-cracks at the interface of TGO and the bond layer and delayed the strain on the surface of the coating under tensile load.TGO growth resulted in the generation of pores at the interface between the TGO and bond layer.The pores produced by TGO under tensile load delayed the generation of surface cracks and thus prolonged the failure time of TBCs.Le WANG Yuelan DI Ying LIU Haidou WANG Haoxing YOU Tao LIU 2019Frontiers of Mechanical Engineering2019,14,4:1

7Effects of Ethyl Pyruvate on High Mobility Group Box1 gene expression in septic lung of rat显示文摘Objective: Ethyl Pyruvate (EP) has been shown to be an effective anti-inflammatory agent in a variety of model systems. The aim of this study was to investigate the effects of EP on High Mobility Group Box1(HMGB1) genes expression and the possible mechanisms of EP protecting against acute lung injury induced by sepsis. Methods: Forty Wistar rats were randomly divided into normal controls,sham operation,acute lung injury,and EP treatment (40 mg/kg intra-peritoneally every 6 hrs) groups. At the time points of 24 hours the animals in each group were sacrificed, and the lungs were harvested. Wet/dry lung weight ratio, the protein in the bronchoalveolar lavage fluid(BALF),and pulmonary permeability index(PPI) were determined. The histological morphology of lung was observed under microscope. The expression of HMGB1 mRNA was measured using semi-quantitative RT-PCR. Results: EP treatment decreased wet/dry lung weight ratio, the protein in the BALF,and PPI (P<0.01). The histological morphology of lung injury was ameliorated. EP significantly inhibited the HMGB1 mRNA expression(P<0.01). HMGB1 mRNA expression in lungs positively correlation with wet/dry lung weight ratio, the protein in the BALF,and PPI. 8The Effects of Electroacupuncture at the Heart Meridian on Myocardial Contractile Function in Rabbits with Myocardial Ischemia显示文摘 Acute myocardial ischemia was induced by intravenous injection of pituitrin, and electroacupuncture (EA) was applied at the Heart and Lung Meridians (HM and LM), 3 points on each meridian. The changes in the left intraventricular pressure (LVP), the maximum rise rate of intraventricular pressure (LVP dp/dtmax), the area of cardiac force loop (ACFL), and the maximum shortening velocity of myocardial contractile element (Vmax) were observed. As a result, there were significant differences in the improvement of LVP, LVP dp/dtmax, ACFL and Vmax between EA at HM and LM.
