| 1 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 2 | Cystathionine-γ-lyase ameliorates the histone demethylase JMJD3-mediated autoimmune response in rheumatoid arthritis显示文摘Cystathionine-γ-lyase(CSE),an enzyme associated with hydrogen sulfide(H2S)production,is an important endogenous regulator of inflammation.Jumonji domain-containing protein 3(JMJD3)is implicated in the immune response and inflammation.Here,we investigated the potential contribution of JMJD3 to endogenous CSE-mediated inflammation in rheumatoid arthritis(RA).Upregulated CSE and JMJD3 were identified in synovial fibroblasts(SFs)from RA patients as well as in the joints of arthritic mice.Knocking down CSE augmented inflammation in IL-1β-induced SFs by increasing JMJD3 expression.In addition,CSE−/−mice with collagen-induced arthritis(CIA)developed severe joint inflammation and bone erosion.Conversely,overexpressing CSE inhibited JMJD3 expression by the transcription factor Sp-1 and was accompanied by reduced inflammation in IL-1β-treated SFs.Furthermore,JMJD3 silencing or the administration of the JMJD3 inhibitor GSK-J4 significantly decreased the inflammatory response in IL-1β-treated SFs,mainly by controlling the methylation status of H3K27me3 at the promoter of its target genes.GSK-J4 markedly attenuated the severity of arthritis in CIA mice.In conclusion,suppressing JMJD3 expression by the transcription factor Sp-1 is likely responsible for the ability of CSE to negatively modulate the inflammatory response and reduce the progression of RA. | Weijun Wu Ming Qin Wanwan Jia Zheng Huang Zhongzheng Li Di Yang Mengwei Huang Chenxi Xiao Fen Long Jianchun Mao Philip K.Moore Xinhua Liu Yi Zhun Zhu | 2019 | Cellular & Molecular Immunology2019,16,8: | 9 |
| 3 | Personalized neoantigen pulsed dendritic cell vaccine for advanced lung cancer显示文摘Neoantigens are considered to be ultimate target of tumor immunotherapy due to their high tumor specificity and immunogenicity.Dendritic cell(DCs)vaccines based on neoantigens have exciting effects in treatment of some malignant tumors and are a promising therapeutic modality.Lung cancer is a lethal disease with the highest morbidity and mortality rate in the world.Despite the rapid development of targeted therapy and immune checkpoint inhibitors for lung cancer in recent years,their efficacy is still unsatisfactory overall.Therefore,there is an urgent unmet clinical need for lung cancer treatment.Here,we attempted to treat lung cancer using a personalized neoantigen peptide-pulsed autologous DC vaccine and conducted a single-arm,2 medical centers,pilot study initiated by the investigator(ChiCTR-ONC 16009100,NCT02956551).The patients enrolled were patients with heavily treated metastatic lung cancer.Candidate neoantigens were derived from whole-exome sequencing and RNA sequencing of fresh biopsy tissues as well as bioinformatics analysis.A total of 12 patients were enrolled in this study.A total of 85 vaccine treatments were administered with a median value of 5 doses/person(range:3-14 doses/person).In total,12-30 peptide-based neoantigens were selected for each patient.AIl treatment-related adverse events were grade 1-2 and there were no delays in dosing due to toxic effects.The objective effectiveness rate was 25%;the disease control rate was 75%;the median progression-free survival was 5.5 months and the median overall survival was 7.9 months.This study provides new evidence for neoantigen vaccine therapy and new therapeutic opportunities for lung cancer treatment. | Zhenyu Ding Qing Li Rui Zhang Li Xie Yang shu Song Gao Peipei Wang Xiaoqing Su Yun Qin Yuelan Wang Juemin Fang Zhongzheng Zhu Xuyang Xia Guochao Wei Hui Wang Hong Qian Xianling Guo Zhibo Gao Yu Wang Yuquan Wei Qing Xu Heng Xu Li Yang | 2021 | Signal Transduction and Targeted Therapy2021,6,2: | 3 |

| 9 | New Method of Image Retrieval Based on Interest Image Region Blocks and Comprehensive Features显示文摘We propose an image retrieval method based on interest image region by asymmetrical blocking. An image is segmented into the interest region and background region on a certain rule. For the interest image regions, the color histogram of the uneven blocks is extracted as the color characteristic. We also collect the mean and variance value of the Gabor filtering results of background blocks as texture features of the background image. Then, the images can be retrieved by synthesizing the image color and texture features. We test our approaches by analyzing the results of recall and precision indicators for the Corel image data-base. | 10 | Ring-Opening Polymerization of a Benzyl-Protected Cyclic Ester towards Functional Aliphatic Polyester显示文摘Monomer design strategy has become a powerful tool to access polymers with desired and diverse functionalities. Here, we designed a novel monomer 2-((benzyloxy)methyl)-1,4-oxathiepan-7-one (BTO) via installing a benzyl ether side chain to the structure of 1,4-oxathiepan-7-one (OTO). The ring-opening polymerization of BTO with Zn1 as the catalyst demonstrated the characteristics of living polymerization with turnover frequency (TOF) up to 2520 h^(−1). With a [BTO]_(0)/[Zn1]_(0)/[I]_(0) feed ratio of 2000/2/1, polymer with high number-average molecular weight (Mn = 536 kDa) and narrow dispersity (Ð = 1.06) was obtained. The produced polymer with a glass transition temperature of -17℃ behaved as an elastomer at room temperature. Consequently, the monomer BTO was copolymerized with _(L)-LA to modulate the mechanical properties of P(L-LA). When the BTO content is 10%, the copolymer exhibits excellent strength (24 MPa) and elongation at break (270%), affording a crystalline, hard, and tough plastic material that combines the high ductility of P(BTO) and the high modulus of P(_(L)-LA). In addition, the oxidation of P(BTO) to P(BTO)-SO_(2) led to an improvement of T_(g) from -17℃ to 38℃. Debenzylation of P(BTO)-SO_(2) afforded P(BTO)-SO_(2)-OH containing free hydroxyl groups. | 11 | Pd–Ni–P metallic glass nanoparticles for nonenzymatic glucose sensing显示文摘Metallic glass nanoparticles hold great promise as nonenzymatic glucose sensors due to their rich low-coordinated active sites and high biocompatibility.However,their non-periodic atomic structure and unclear structure-property relationship pose significant challenges for realizing and optimizing their sensing performance.In this work,Pd–Ni–P metallic glass nanoparticles with variable compositions were successfully prepared as nonenzymatic glucose sensors via a laser-evaporated inert-gas condensation method.The electrochemical tests show that the sensor based on Pd_(41·25)Ni_(41·25)P_(17.5)nanoparticles shows a wide linear detection range(0.003–1.31mM),high sensitivity(516μAmM^(−1)·cm^(−2)),and high stability(∼97.8%current retention after 1000 cycles).Local structural investigations using synchrotron pair distribution function and high-resolution microscopic techniques reveal a strong structural correlation within short-to medium-range orders in the Pd_(41·25)Ni_(41·25)P_(17.5)nanoparticles,which can be well retained after electrochemical cycling.These atomic-scale structural characteristics might be responsible for the high sensing performance.This study demonstrates the high applicability of Pd–Ni–P metallic glass nanoparticles as sensitive and stable non-enzymatic glucose sensors. | Yu Lou Zhongzheng Yao Shu Fu Sinan Liu Xindong Zhu Wen Huang a Min Dong Jianrong Zeng He Lin He Zhu Si Lan | 2023 | Progress in Natural Science:Materials International2023,33,2: | 0 |
