10Identification of natural allelic variation in TTL1 controlling thermotolerance and grain size by a rice super pan-genome∞显示文摘Continuously increasing global temperatures present great challenges to food security.Grain size,one of the critical components determining grain yield in rice(Oryza sativa L.),is a prime target for genetic breeding.Thus,there is an immediate need for genetic improvement in rice to maintain grain yield under heat stress.However,quantitative trait loci(QTLs)endowing heat stress tolerance and grain size in rice are extremely rare.Here,we identified a novel negative regulator with pleiotropic effects,Thermo‐Tolerance and grain Length 1(TTL1),from the super pan‐genomic and transcriptomic data.Loss‐of‐function mutations in TTL1 enhanced heat tolerance,and caused an increase in grain size by coordinating cell expansion and proliferation.TTL1 was shown to function as a transcriptional regulator and localized to the nucleus and cell membrane.Furthermore,haplotype analysis showed that hapL and hapS of TTL1 were obviously correlated with variations of thermotolerance and grain size in a core collection of cultivars.Genome evolution analysis of available rice germplasms suggested that TTL1 was selected during domestication of the indica and japonica rice subspecies,but still had much breeding potential for increasing grain length and thermotolerance.These findings provide insights into TTL1 as a novel potential target for the development of high‐yield and thermotolerant rice varieties.Yarong Lin Yiwang Zhu Yuchao Cui Hongge Qian Qiaoling Yuan Rui Chen Yan Lin Jianmin Chen Xishi Zhou Chuanlin Shi Huiying He Taijiao Hu Chenbo Gu Xiaoman Yu Xiying Zhu Yuexing Wang Qian Qian Cuijun Zhang Feng Wang Lianguang Shang 2023Journal of Integrative Plant Biology2023,65,12:0

12miR-139-5p mediates the palmitate-induced inhibition of insulin secretion by targeting neuronal pentraxin 1 in INS-1 cells显示文摘High fatty acid reduces insulin secretion in pancreatic β-cells and miR-139-5p is increased in diabetic pancreatic tissues and induces islet β-cell apoptosis.However,to date,there is no study exploring whether or not miR-139-5p is involved in high fatty acid-induced insulin secretion.In the present study,INS-1 cells were exposed to different concentrations(0.1,0.2,and 0.4 mM)of palmi-tate for different time periods(12,24,and 48 h).The expression levels of miR-139-5p and neuronal pentraxin 1(NPTX1)were evaluated by real-time PCR and western blot analysis.The regulation of NPTX1 by miR-139-5p was examined by luciferase assay.Cell transfection was conducted using Lipo8000 or Lipofectamine RNAiMAX.Potassium or glucose-stimulated insulin secretion levels were used to verify the function of miR-139-5p or NPTX1 in insulin secretion.Insulin secretion levels were detected by radioimmunoassay.We found that miR-139-5p was increased in INS-1 cells stimulated with palmitate.In addition,miR-139-5p was also elevated in islets of high-fat diet-fed mice and db/db mice compared to those in islets of normal diet-fed mice and wild-type mice.Knockdown of miR-139-5p could reverse high fatty acid-induced insulin secretion defects in INS-1 cells.Furthermore,we demonstrated that NPTX1 is a target of miR-139-5p.miR-139-5p mediated palmitate-induced insulin secretion defects by targeting NPTX1.Moreover,palmitate treatment declined the expression of NPTX1 and the NPTX1 expression was also decreased in islets of high-fat diet-fed mice and db/db mice.Impaired NPTX1 expression is involved in fatty acid-induced insulin secretion defects.Collectively,our results illustrate that the induction of β-cell insulin secretion defects by fatty acids is mediated,at least in part,by miR-139-5p via downregulation of NPTX1 expression.Kai Zhang Yijian Zhang Cheng Chen Yuexing Yuan Xiaotian Jiang Xiangjiang Yuan Yao Wang 2021Acta Biochimica et Biophysica Sinica2021,53,8:0
13Insulin-resistance and depression cohort data mining to identify nutraceutical related DNA methylation biomarker for type 2 diabetes显示文摘Insulin-resistance(IR)is one of the most important precursors of type 2 diabetes(T2D).Recent evidence suggests an association of depression with the onset of T2D.Accumu-lating evidence shows that depression and T2D share common biological origins,and DNA methylation examination might reveal the link between lifestyle,disease risk,and potential therapeutic targets for T2D.Here we hypothesize that integrative mining of IR and depression cohort data will facilitate predictive biomarkers identification for T2D.We utilized a newly proposed method to extract gene-level information from probe level data on genome-wide DNA methylation array.We identified a set of genes associated with IR and depression in clin-ical cohorts.By overlapping the IR-related nutraceutical-gene network with depression net-works,we identified a common subnetwork centered with Vitamin D Receptor(VDR)gene.Preliminary clinical validation of gene methylation set in a small cohort of T2D patients and controls was established using the Sequenome matrix-assisted laser desorption ionization-time flight mass spectrometry.A set of sites in the promoter regions of VDR showed a signifi-cant difference between T2D patients and controls.Using a logistic regression model,the optimal prediction performance of these sites was AUC Z 0.902,and an odds ratio Z 19.76.Thus,monitoring the methylation status of specific VDR promoter region might help stratify the high-risk individuals who could potentially benefit from vitamin D dietary sup-plementation.Our results highlight the link between IR and depression,and the DNA methyl-ation analysis might facilitate the search for their shared mechanisms in the etiology of T2D.Fengji Liang Yuan Quan Andong Wu Ying Chen Ruifeng Xu Yuexing Zhu Jianghui Xiong 2021Genes & Diseases2021,8,5:0
