| 1 | Gambogic acid induces mitochondria-dependent apoptosis by modulation of Bcl-2 and Bax in mantle cell lymphoma JeKo-1 cells显示文摘Aim: To study the mechanisms in gambogic acid (GA) -induced JeKo-1 human Mantle Cell Lymphoma cell apoptosis in vitro. Methods: The proliferation of GA-treated JeKo-1 cells was measured by CCK-8 assay and Ki-67 immunocytochemical detection. Apoptosis, cell cycle and mitochondrial membrane potential were measured by flow cytometric analysis. Caspase-3,-8 and -9 were detected by colorimetric assay. Bcl-2 and Bax were analyzed by Western blotting. Results: GA inhibited cell growth in a time-and dose-dependent manner. GA induces apoptosis in JeKo-1 cells but not in normal bone marrow cells, which was involved in reducing the membrane potential of mitochondria, activating caspases-3, -8 and -9 and decreasing the ratio of Bcl-2 and Bax without cell cycle arresting. | 2 | Relationship between the number of tapping-induced secondary laticifer lines and rubber yield among Hevea germplasm显示文摘The lack of suitable early selection parameters means that traditional rubber breeding for yield is time consuming and inefficient. Laticifer is a tissue specific fo natural rubber biosynthesis and storage in rubber tree. The number of the secondary laticifers in the trunk bark tissues is positively correlated with rubber yield in the matured rubber trees that are regularly tapped. In the present study the rubber yield from 280 of 4–5 year-old virgin trees from7 cross combinations was compared with the number o newly differentiated secondary laticifers caused by tapping. | 4 | Complementary transmissive ultra-thin meta-deflectors for broadband polarization-independent refractions in the microwave region显示文摘Polarization manipulation is a significant issue for artificial modulation of the electromagnetic(EM) wave, but general mechanisms all suffer the restriction of inherent symmetric properties between opposite handedness.Herein, a strategy to independently and arbitrarily manipulate the EM wave with orthogonal circular polarizations based on a metasurface is proposed, which effectually breaks through traditional symmetrical characteristics between orthogonal handedness. By synthesizing the propagation phase and geometric phase, the appropriate Jones matrix is calculated to obtain independent wavefront manipulation of EM waves with opposite circular polarizations. Two transmissive ultra-thin meta-deflectors are proposed to demonstrate the asymmetrical refraction of transmitted circularly polarized waves in the microwave region. Simulated transmitted phase front and measured far-field intensity distributions are in excellent agreement, indicating that the transmitted wave with different polarizations can be refracted into arbitrary and independent directions within a wide frequency band(relative bandwidth of 25%). | 5 | Transforming growth factor-β1-induced N-cadherin drives cell–cell communication through connexin43 in osteoblast lineage显示文摘Gap junction(GJ)has been indicated to have an intimate correlation with adhesion junction.However,the direct interaction between them partially remains elusive.In the current study,we aimed to elucidate the role of N-cadherin,one of the core components in adhesion junction,in mediating connexin 43,one of the functional constituents in gap junction,via transforming growth factor-β1(TGF-β1)induction in osteoblasts.We first elucidated the expressions of N-cadherin induced by TGF-β1 and also confirmed the upregulation of Cx43,and the enhancement of functional gap junctional intercellular communication(GJIC)triggered by TGF-β1 in both primary osteoblasts and MC3T3 cell line.Colocalization analysis and Co-IP experimentation showed that N-cadherin interacts with Cx43 at the site of cell–cell contact.Knockdown of N-cadherin by siRNA interference decreased the Cx43 expression and abolished the promoting effect of TGF-β1 on Cx43.Functional GJICs in living primary osteoblasts and MC3T3 cell line were also reduced.TGF-β1-induced increase in N-cadherin and Cx43 was via Smad3 activation,whereas knockdown of Smad3 signaling by using siRNA decreased the expressions of both N-cadherin and Cx43.Overall,these data indicate the direct interactions between N-cadherin and Cx43,and reveal the intervention of adhesion junction in functional gap junction in living osteoblasts. | Yueyi Yang Wenjing Liu JieYa Wei Yujia Cui Demao Zhang Jing Xie | 2021 | International Journal of Oral Science2021,13,2: | 4 |
| 6 | Runxl protects against the pathological progression of osteoarthritis显示文摘Runt-related transcription factor-1(Runxl)is required for chondrocyte-to-osteoblast lineage commitment by enhancing both chondrogenesis and osteogenesis during vertebrate development.However,the potential role of Runxl in joint diseases is not well known.In the current study,we aimed to explore the role of Runxl in osteoarthritis induced by anterior cruciate ligament transaction(ACLT)surgery.We showed that chondrocyte-specific Runxl knockout(Runx1f/fCol2a1-Cre)aggravated cartilage destruction by accelerating the loss of proteoglycan and collagen II in early osteoarthritis.Moreover,we observed thinning and ossification of the growth plate,a decrease in chondrocyte proliferative capacity and the loss of bone matrix around the growth plate in late osteoarthritis.We overexpressed Runxl by adeno-associated virus(AAV)in articular cartilage and identified its protective effect by slowing the destruction of osteoarthritis in cartilage in early osteoarthritis and alleviating the pathological progression of growth plate cartilage in late osteoarthritis.ChIP-seq analysis identified new targets that interacted with Runxl in cartilage pathology,and we confirmed the direct interactions of these factors with Runxl by ChIP-qPCR.This study helps us to understand the function of Runxl in osteoarthritis and provides new clues for targeted osteoarthritis therapy. | Chenchen Zhou Yujia Cui Yueyi Yang Daimo Guo Demao Zhang Yi Fan Xiaobing Li Jing Zou Jing Xie | 2021 | Bone Research2021,9,4: | 4 |
| 7 | Lipidomics reveals association of circulating lipids with body mass index and outcomes in IgA nephropathy patients显示文摘IgA nephropathy(IgAN)is a leading cause of chronic kidney disease(CKD),which are commonly accompanied by dyslipidemia.Obesity is also associated with dyslipidemia and risk of CKD,but the relation of the dyslipidemia patterns with obesity and disease progression in IgAN patients remains unknown.Traditional Chinese medicine(TCM)and the combined treatment with corticosteroids and TCM have been shown to be of benefit for IgAN patients,but predictive markers for guiding these treatments are lacking.Here,we quantified 545 lipid species in the plasma from 196 participants,including 140 IgAN patients and 56 healthy volunteers,and revealed an altered plasma lipidome in IgAN patients as compared to healthy participants.Association analysis showed that a subgroup of glycerides,particularly triacylglycerols(TGs)containing docosahexaenoic acid,were positively associated with high body mass index(BMI)in under-or normal-weight IgAN patients,while several free fatty acids and sphingomyelins were positively associated with high BMI in overweight or obese IgAN patients.Further,our study suggested that elevated levels of eight lipids,mainly TG species containing linolenic acid,were independent risk factors for IgAN progression and also reported the prospective association of circulating lipids with treatment outcomes in IgAN.Taken together,our findings may not only help to achieve precision medicine but also provide a knowledge base for dietary intervention in the treatment of IgAN. | Yueyi Deng Qingqing Wu Wanjia Chen Li Zhu Wangyi Liu Fangying Xia Liang Sun Xu Lin Rong Zeng | 2021 | Journal of Molecular Cell Biology2021,13,8: | 2 |
