2The Characterization, Authentication, and Gene Expression Pattern of the MdCER Family in Malus domestica显示文摘Plant cuticular wax is an important determinant factor of fruit quality. Cuticular wax can protect the fruit from UV radiation, drought, and disease and increase the storage period of fruit. In many species, CER gene is associated with wax and can affect plant response to stress, but no characterization of CER gene and its family has been reported in apples. In this study, we identified 10 MdCER genes in Malus domestica based on the sequences of 10 CER genes in Arabidopsis thaliana. Three-dimensional structures were then defined, and root-mean-square deviation(RMSD)scoring matrixes were used to evaluate the matches. These 10 genes were divided into three classes using phylogenetic methods; namely,class I, II, and III. The predicted MdCER genes were distributed across 7 out of 17 chromosomes with different densities. Furthermore, the gene structures and motif compositions of the MdCER genes were analyzed. The quantitative real-time PCR results indicated that MdCER family genes were mainly expressed in the leaves and stems and rarely in the roots. 3MdDREB2A in apple is involved in the regulation of multiple abiotic stress responses显示文摘Abiotic stress has a serious effect on plant growth.The transcription factor DREB2A is a member of the AP2/ERF family,which is widely involved in abiotic stress response.However,the function of apple MdDREB2A has not been systematically investigated.In this study,MdDREB2A was isolated from the cultivar‘Royal Gala’.The open reading frame of MdDREB2A was 1197 bp in length and it encoded a protein of 398 amino acidswithmolecularweight of 43.8 kD.As a transcription factor,MdDREB2Awas located in the nucleus.qRT-PCR analysis showed that MdDREB2A was involved in responses to drought,salt,and ABA stresses.Under these stress treatments,the relative electrical conductivity,superoxide anion and malondialdehyde(MDA)in transgenic materials significantly decreased,and the content of proline increased in MdDREB2A transgenic plants,compared to the controls,indicating that MdDREB2A transgenic apple calli and transgenic Arabidopsis exhibited improved resistance to abiotic stress.This study introduces a candidate gene for the genetic improvement of crop resistance and reveals important function of MdDREB2A in the regulation of abiotic stress response.Xinyu Lian Xinyu Zhao Qiang Zhao Guiluan Wang Yuanyuan Li Yujin Hao 2021Horticultural Plant Journal2021,7,3:6
4Molecular Cloning and Expression Analysis of a Hexokinase Gene,MdHXK1 in Apple显示文摘A hexokinase gene named MdHXK1(MDP0000309677) was cloned from ‘Gala' apple(Malus × domestica Borkh.). Sequence analysis showed that the MdHXK1 gene was 1 497 bp long and encoded 499 amino acids. The predicted molecular mass of this protein was 54.05 kD, and the pI was 5.76. A phylogenetic tree indicated apple MdHXK1 exhibited the highest sequence similarity to Pyrus bretschneideri PbHXK1. Analysis of the functional domain showed that the MdHXK1 protein included two conserved kinase domains. The prediction of subcellular localization suggested that the MdHXK1 protein was mainly localized in the cytoplasm. There was an indication that MdHXK1 existed as one copy in the apple genome by Southern blotting. Silico analysis suggested that the promoter sequence contained several typical cis-acting elements, including defense, sugar signaling and phytohormone responsive elements. Quantitative real-time PCR analysis demonstrated that the MdHXK1 gene was mainly expressed in stem and flower tissues. During the development of apple fruits, the expression of the MdHXK1 gene initially increased and then decreased. The changes on Glc phosphorylation relative activity and glucose concentration showed the same trend. In addition, the expression of this gene was induced by salt stress, low temperature, and abscisic acid(ABA). Finally, we obtained and purified the fused MdHXK1 protein by recombinant prokaryotic expression. Studies have demonstrated that MdHXK1 may participate in sugar metabolism in apple fruits. Enzyme encoded by MdHXK1 is a key factor in the mediation of sugar accumulation. Recently,researchers on hexokinase at home and abroad mainly focused on model plants, such as Arabidopsis, tobacco and rice, but orchard fruit like apple were underresearched. 5Functional Characterization of the Apple RING E3 Ligase MdMIEL1 in Transgenic Arabidopsis显示文摘E3 ubiquitin ligases are involved in various physiological processes,and they play pivotal roles in growth and development.In this study,we identified a previously unknown gene in the apple fruit(Malus×domestica)and named it MdMIEL1.The MdMIEL1 gene encoded a protein that contained a zinc-finger domain at its N-terminus and a RING-finger motif at its C-terminus.To investigate MdMIEL1 functions,we generated transgenic Arabidopsis lines expressing the MdMIEL1 gene under the control of the Cauliflower mosaic virus 35S promoter.Interestingly,ectopic expression of MdMIEL1 in Arabidopsis produced multiple phenotypes,including early germination,early flowering and a lateral root number increase relative to wild-type plants.Further analysis indicated that MdMIEL1 regulated lateral root initiation by increasing auxin accumulation in the roots.In a word,these results suggest that,MdMIEL1 as a novel RING-finger ubiquitin ligase influences plant growth and development,and highlight that MdMIEL1 regulates lateral root growth.AN Jianping LIU Xin SONG Laiqing YOU Chunxiang WANG Xiaofei HAO Yujin 2017Horticultural Plant Journal2017,3,2:3
6Genome-Wide Identification, Characterization, and Expression Analysis of Calmodulin-Like Proteins(CMLs) in Apple显示文摘Calcium(Ca^(2+)), a crucial second messenger in plants, is involved in diverse signaling pathways including biotic and abiotic stress responses. However, the functions of most calcium sensors including calcium-dependent protein kinases(CDPKs), calmodulins(Ca Ms), calmodulinlike proteins(CMLs), and calcineurin B-like proteins(CBLs) remain limited in plants, particularly in woody plants. Currently, a total of 83 CMLs and seven Ca Ms were discovered in the apple genome in this study. Functional domain analysis showed that these CMLs and Ca Ms contain a highly conserved EF-hand motif. q RT-PCR assays indicated that these CMLs were expressed ubiquitously in apple, including in the roots, stems,leaves, flowers, and fruits, and also possessed spatial specificity. Finally, most of these CMLs were induced by hormonal and abiotic stress, suggesting their potential roles in the regulation of growth, development, and the stress response in apple. 7The Growth-Promoting Mechanism of Brevibacillus laterosporus AMCC100017 on Apple Rootstock Malus robusta显示文摘Plant growth-promoting rhizobacteria(PGPR) located in rhizobacteria soil are beneficial to plant growth and development. A PGPR strain AMCC100017 of Brevibacillus laterosporus synthesizes the plant hormone IAA in a tryptophan-dependent manner. In this study, the AMCC100017 strain was used to treat Malus robusta, an excellent natural rootstock for apple production, and assess its ability to promote growth. The fresh weight, dry weight, plant height, and lateral root growth of M. robusta were significantly increased with treatment. The presence of the AMCC100017 strain increased IAA content in M. robusta and promoted root secretion of tryptophan. Colonization of the strain in the roots allowed continuous synthesis of IAA and promoted plant growth. In addition, the photosynthetic efficiency in leaves increased after microbial treatment, and the utilization of nitrogen, zinc, iron, copper and magnesium in leaves was increased, which was conducive to photosynthesis.Interestingly, the activities of CAT and SOD, as well as the contents of ROS in plants colonized by AMCC100017 were increased compared to control plants, but the activities of POD and MDA contents were decreased. AMCC100017 strain affected the antioxidant capacity and stress resistance of plants. AMCC100017 strain increased the content of soluble protein and soluble sugar in plants, improved plant metabolic activity and stress resistance. 8Expression Analysis of the MdCIbHLH1 Gene in Apple Flower Buds and Seeds in the Process of Dormancy显示文摘A bHLH transcription factor that is induced by low temperature was found in apple(Malus×domestica Borkh.). To understand the sequence characteristics of the gene, bioinformatics analysis was performed. Furthermore, gene expression patterns of the laminated apple seeds and lateral flower buds were analyzed during the period of dormancy release with semi-quantitative RT-PCR. Based on secondary structure predictions, the results showed that the MdCIbHLH1 protein structure mainly included α-helix and random coil, while β-sheet and extended strand content was less. Semi-quantitative RT-PCR analysis showed that the expression patterns of MdCIbHLH1 were similar in laminated apple seeds and lateral flower buds during the period of dormancy release. Before dormancy release, expression levels of MdCIbHLH1 were high and gradually decreased during the period of dormancy release. 9Genome-wide identification and functional characterization of the MdCLE peptide family in apple(Malus×domestica)显示文摘The CLAVATA3/EMBRYO SURROUNDING REGION-related(CLE)peptides are critical for stem cell homeostasis in plant shoot and root apical meristem.Although CLE genes have been reported in numerous plants,there is limited information for apple.Here,twenty-five MdCLE genes were identified fromapple genome(Apple Genome V1.0 predicted peptides).Analysis of chromosomal location showed that the 25 MdCLE genes were located on 12 of 17 apple chromosomes.Genetic structure analysis showed that 21 of 25 the MdCLE genes were intron-free.Expression patterns showed thatmost of the MdCLE genes showed higher expression levels in leaves and root compared with the other tissues.In addition,expression analysis demonstrated that MdCLE genes had different gene expression patterns for abiotic stress treatment,suggesting their potential roles in acclimation of apple to adverse environments.The 25 MdCLE genes encoded 19 different CLE peptides,and they were divided into two groups depending on their effects on the inhibition of Arabidopsis root growth.Our results suggest that MdCLE genesmay have an important but redundant function in regulating plant growth and development,and this work provides valuable references for further investigation of the biological functions of MdCLE genes.Tianen Zhang Xiuming Li Qiang Zhao Yan Shi Yujin Hao Chunxiang You 2022Horticultural Plant Journal2022,8,3:2
10Functional identification of MdSMXL8.2,the homologous gene of strigolactones pathway repressor protein gene in Malus×domestica显示文摘A homologous gene of strigolactones repressor protein gene SMXL7/D53,MdSMXL8.2(GenBank accession No.:MD07G1222400),was cloned from‘Royal Gala’apple(Malus×domestica Borkh.)in this study.The sequence analysis revealed that the length of this gene was 3243 bp,which encoded 1080 amino acids,and had a protein molecular mass of∼110 kD.The phylogenetic tree analysis indicated that the MdSMXL8.2 exhibited the highest sequence similarity with Arabidopsis AtSMXL7.The protein conserved domain analysis revealed that the MdSMXL8.2 contained two ClpA domains.The prediction of the secondary and tertiary structures of the MdSMXL8.2 indicated that it contained 34.54%αhelix,3.43%β-sheet,and 11.76%extended chain.The in-silico analysis suggested that the promoter sequence of MdSMXL8.2 contained several typical cisacting elements,including abscisic acid(ABA),gibberellin(GA),ethylene,auxin,jasmonic acid(JA),salicylic acid(SA),drought,and heat stressresponsive elements.Quantitative real-time(qRT)-PCR analyses revealed that MdSMXL8.2 was expressed in different apple tissues,with the highest transcript level found in the stem.The expression of MdSMXL8.2 was significantly induced by exogenous ABA,PEG and mannitol,while exogenous NaCl significantly inhibited MdSMXL8.2 expression.The growing status of MdSMXL8.2-overexpressed Orin apple callus was worse than the wild type(WT)after NaCl treatment and had a higher malondialdehyde(MDA)content and relative conductance(REC).Additionally,MdSMXL8.2-overexpressed Arabidopsis exhibited shorter root length and a reduction in fresh weight under salt stress,indicating that MdSMXL8.2 negatively regulated salt tolerance in apples.Weijian Sun Xinglong Ji Laiqing Song Xiaofei Wang Chunxiang You Yujin Hao 2021Horticultural Plant Journal2021,7,4:1

12Genome-wide identification and expression analyses of homeodomain-leucine zipper family genes reveal their involvement in stress response in apple(Malus×domestica)显示文摘The homeodomain-leucine zipper(HD-Zip)family has been shown to perform amultitude of functions during plant development and stress responses;however,the familymembers and functions have not been identified in apple(Malus×domestica).In this study,83 HD-Zips(MdHDZs)were identified in the apple genome.They were assembled into four subgroups according to the classification in Arabidopsis,where MdHDZs in the same subgroup had similar gene structures and conserved protein motifs.Putative cis-element analysis of MdHDZs promoter regions uncovered numerous elements related to the response of stress and plant hormones.In addition,twelve transcripts of the MdHDZs showed different expression patterns under salt,drought,low temperature and ABA stresses by quantitative reverse transcription-PCR(qRT-PCR)assay.To further explore the function of MdHDZs in apple,MdHDZ3 was selected to verify its function under salt,low temperature and ABA stresses;and genetic transformation was used to obtain MdHDZ3 transgenic apple calli.The results demonstrated that MdHDZ3 increased sensitivity to salt,low temperature and abscisic acid in apple calli,suggesting that MdHDZ3 plays an important role in response to stresses.Subcellular localization and three-dimensional structural analysis revealed that MdHDZ3 was a nuclear-localized protein.Taken together,these findings provide potential information for further identification of HD-Zip proteins in apple.Quanyan Zhang Tao Chen Xun Wang Jiahui Wang Kaidi Gu Jianqiang Yu Dagang Hu Yujin Hao 2022Horticultural Plant Journal2022,8,3:1
13Person-independent expression recognition based on person-similarity weighted expression feature显示文摘A new method to extract person-independent expression feature based on higher-order singular value decomposition (HOSVD) is proposed for facial expression recognition. Based on the assumption that similar persons have similar facial expression appearance and shape, the person-similarity weighted expression feature is proposed to estimate the expression feature of test persons. As a result, the estimated expression feature can reduce the influence of individuals caused by insufficient training data, and hence become less person-dependent. The proposed method is tested on Cohn-Kanade facial expression database and Japanese female facial expression (JAFFE) database. 17Changing profile of lung cancer clinical characteristics in China:Over 8-year population-based study显示文摘Background:Although examinations and therapies for bronchial lung cancer,also called lung cancer(LC),have become more effective and precise,the morbidity and mortality of LC remain high worldwide.Describing the changing profile of LC characteristics over time is indispensable.This study aimed to understand the changes in real-world settings of LC and its characteristics in China.Methods:In this study,119,785 patients were enrolled from 2012 to 2020 in the Shanghai Pulmonary Hospital.The patients’medical records were extracted from the hospital’s database.Demographic characteristics,general clinicopathological information,and blood coagulation indices at the initial diagnoses were analyzed using the Kruskal-Wallis,Nemenyi,chi-squared,and Bonferroni tests.Changes in demographic characteristics during the 8-year study period,namely dynamic changes among different stages and different pathological types,were evaluated.Results:The percentages of female(from 38.50%[323/839]in 2012 to 48.29%[5112/10,585]in 2020)and non-smoking LC(from 69.34%[475/685]to 80.48%[8055/10,009])patients increased significantly during the study period,with a trend toward a younger age at diagnosis(from 3.58%[30/839]to 8.99%[952/10,585]).Over the study period,the proportion and absolute number of lung adenocarcinoma cases increased(from 67.97%[433/637]to 76.31%[6606/8657])while the proportion of lung squamous cell carcinoma decreased(from 21.19%[135/637]to 12.08%[1046/8657]).Comprehensive driver gene mutation examination became more common,and epidermal growth factor receptor(EGFR)mutation occurred more frequently in female vs.male(62.03%[12793/20625]vs.29.90%[8207/27,447])and non-smoking vs.smoking(53.54%[17,203/32,134]vs.23.73%[3322/13,997])patients(both P<0.001).The distribution of the common driver genes differed among different stages of LC.EGFR mutation was detected most frequently at each stage,and other driver gene alterations were more common in advanced stages(P<0.001).The combination of chemotherapy,targeted ther-apy,and immunotherapy,as a comprehensive management regimen,gradually became predominant over the study period(P<0.001).A hypercoagulable state was shown in advanced-stage LC patients and patients with the anaplastic lymphoma kinase fusion,indicated by significantly elevated levels of d-dimer,fibrinogen,and fibrinogen degradation products.Conclusions:This study comprehensively depicted the changing characteristics of Chinese LC patients over an 8-year period to provide preliminary insights into LC treatment.Trial registration:ClinicalTrials.gov,NCT05423236.Kandi Xu Hao Wang Simin Li Lishu Zhao Xinyue Liu Yujin Liu Li Ye Xiaogang Liu Linfeng Li Yayi He 2023Chinese Medical Journal Pulmonary and Critical Care Medicine2023,1,3:0
18Unfolding the structure-property relationships of Li_(2)S anchoring on two-dimensional materials with high-throughput calculations and machine learning显示文摘Lithium-sulfur(Li-S)batteries are notable for their high theoretical energy density,but the‘shuttle effect’and the limited conversion kinetics of Li-S species can downgrade their actual performance.An essential strategy is to design anchoring materials(AMs)to appropriately adsorb Li-S species.Herein,we propose a new three-procedure protocol,named InfoAd(Informative Adsorption)to evaluate the anchoring of Li_(2)S on two-dimensional(2D)materials and disclose the underlying importance of material features by combining high-throughput calculation workflow and machine learning(ML).In this paradigm,we calculate the anchoring of Li_(2)S on 12552D A_(x)B_(y)(B in the VIA/VIIA group)materials and pick out 44(un)reported nontoxic 2D binary A_(x)B_(y)AMs,in which the importance of the geometric features on the anchoring effect is revealed by ML for the first time.We develop a new Infograph model for crystals to accurately predict whether a material has a moderate binding with Li_(2)S and extend it to all 2D materials.Our InfoAd protocol elucidates the underlying structure-property relationship of Li_(2)S adsorption on 2D materials and provides a general research framework of adsorption-related materials for catalysis and energy/substance storage.Lujie Jin Hongshuai Wang Hao Zhao Yujin Ji Youyong Li 2023Journal of Energy Chemistry2023,,7:0

20Preparation and Properties of Silver Colloids Produced by Laser-Induced Reduction of Silver Nitrate显示文摘Silver colloids have been prapared by laser-induced reduction of silver nitrate.The.properties of the colloids were studied with transmission electron microscopy,visible-ultraviolet absorption and Raman spectroscopy.We Rnd that the growth of the silver grains is dependent on the laser illumination time.Two characteristic absorption bands of the colloids are located near 390 and 560nm,respectively.Not only are the silver colloids much cleaner than those prepared by chemical reduction methods,but also surface enhanced Raman scattering active.The method may be of interests for ultrafine particle studies as well.LIANG Erjun WANG Yuntao WANG Yujin HAO Qingxiu ZHANG Pengxiang 1992Chinese Physics Letters1992,9,2:0
