1miR-4458 inhibits the epithelial-mesenchymal transition of hepatocellular carcinoma cells by suppressing the TGF-β signaling pathway via targeting TGFBR1显示文摘Hepatocellular carcinoma(HCC)is one of the most lethal cancers in the world.MicroRNAs play a pivotal role in the progression of various cancers.To date,very lttle attention has beerl paid to miR-4458.Therefore,the aim of our study was to explore the function and underlying molecular mech-anism of miR-4458 in HCC.We found that the expression of miR-4458 was reduced in HCC tissues and cell lines.Forced overexpression of miR-4458 inhibited the migration,invasion,and epithelial-mesenchymal transition(EMT)of HCC cells,while downregulation of miR-4458 promoted the aggressive phenotype.Furthermore,transforming growth factor beta receptor 1(TGFBR1),the modulator of the TGF-βsignaling pathway,was verified to be a novel target gene of miR-4458 by dual-luciferase reporter gene assay.Upregulated miR-4458 dramatically abolished TGFBR1 and p-Smad2/3 expression,thus blocking the TGF-βsig naling pathway.Moreover,restoration of TGFBR1 partially rescued the miR-4458-mediated suppressive effect on the migration,invasion,and EMT and reactivated the TGFβsignaling pathway in HCC cells.In summary,our findings first demonstrated a mechanism of miR 4458 in HCC cell migration,invasion,and EMT by regulating the TGFβsignaling pathway via directly targeting TGFBR1.Yuke Zhang Kun Shi Hang Liu Wei Chen Yunhai Luo Xufu Wei Zhongjun Wu 2020Acta Biochimica et Biophysica Sinica2020,52,5:4
2Nucleic acid nanoassembly-enhanced RNA therapeutics and diagnosis显示文摘RNAs are involved in the crucial processes of disease progression and have emerged as powerful therapeutic targets and diagnostic biomarkers.However,efficient delivery of therapeutic RNA to the targeted location and precise detection of RNA markers remains challenging.Recently,more and more attention has been paid to applying nucleic acid nanoassemblies in diagnosing and treating.Due to the flexibility and deformability of nucleic acids,the nanoassemblies could be fabricated with different shapes and structures.With hybridization,nucleic acid nanoassemblies,including DNA and RNA nanostructures,can be applied to enhance RNA therapeutics and diagnosis.This review briefly introduces the construction and properties of different nucleic acid nanoassemblies and their applications for RNA therapy and diagnosis and makes further prospects for their development.Mengnan Zhao Rujing Wang Kunmeng Yang Yuhong Jiang Yachen Peng Yuke Li Zhen Zhang Jianxun Ding Sanjun Shi 2023Acta Pharmaceutica Sinica B2023,13,3:2
3Seryl tRNA synthetase cooperates with POT1 to regulate telomere length and cellular senescence显示文摘Deregulated telomere length is a causative factor in many physiological and pathological processes,including aging and cancer.Many studies focusing on telomeres have revealed important roles for cooperation between the Shelterin protein complex and telomerase in maintaining telomere length.However,it remains largely unknown whether and how aging-related stresses,such as deregulated protein homeostasis,impact telomere length.Here,we explored the possible roles of aminoacyl tRNA synthetases(AARSs),key enzymes catalyzing the first reactions in protein synthesis,in regulating telomere length and aging.We selected seryl tRNA synthetase(SerRS)since our previous studies discovered expanded functions of SerRS in the nucleus in addition to its canonical cytoplasmic role in protein synthesis.In this study,we revealed that overexpression of SerRS promoted cellular senescence and inhibited the growth of cervical tumor xenografts in mice by triggering the senescence of tumor cells.In the nucleus,SerRS directly bound to telomeric DNA repeats and tethered more POT1 proteins to telomeres through a direct interaction between the UNE-S domain of SerRS and the OB1 domain of POT1.We further demonstrated that SerRS-induced enrichment of POT1 prevented the recruitment of telomerase to telomeres,resulting in progressive telomere shortening.Our data suggested a possible molecular link between protein synthesis and telomere length control,the deregulation of which may be associated with aging and cancer.Yingxi Li Xiyang Li Mei Cao Yuke Jiang Jie Yan Ze Liu Rongcun Yang Xu Chen Peiqing Sun Rong Xiang Longlong Wang Yi Shi 2019Signal Transduction and Targeted Therapy2019,4,1:1
4Behavior and functional modeling methods of doubly salient electromagnetic generators for aircraft electrical power system applications显示文摘The Doubly Salient Electromagnetic Generator(DSEG) is a promising candidate in aircraft generator application due to the simplicity, robustness and reliability. However, the field windings and the armature windings are strongly coupled, which makes the inductance characteristics non-linear and too complex to model. The complex model with low precision also leads to difficulties in modeling and analysis of the entire aircraft Electrical Power System(EPS). A behavior level modeling method based on modified inductance Support Vector Machine(SVM) is proposed. The Finite Element Analysis(FEA) inductance data are modified based on the experiment results to improve the precision. A functional level modeling method based on input–output characteristics SVM is also proposed. The two modeling methods are applied to a 9 kW DSEG prototype. The steady state and transient process precision of the proposed methods are proved by comparing with the experiment results. Meanwhile, the modeling time consumption, the application time consumption and the calculation resource demand are compared. 5Transcriptome Analysis and Genes Function Verification of Root Development of Paeonia suffruticosa under Sandy Loam Cultivation显示文摘Relatively poor in vitro rooting has limited the large-scale commercial production of tree peony.In this study,on the basis of transcriptome sequencing,differentially expressed genes and the associated metabolic pathways were identified in tree peony roots at different stages of root formation under sandy loam cultivation.A total of 31.63 Gb raw data were generated and 120,188 unigenes(mean length of 911.98 bp)were annotated according to six databases(NR,NT,GO,KEGG,COG,and Swiss-Prot).Analyses of the ungerminated root primordium period,induced root primordium period,and root formation period detected 8,232,6,907,and 10,687 differentially expressed genes related to 133,132,and 133 metabolic pathways,respectively.Two significantly differentially expressed genes(Unigene13430_All and CL10096.Contig1_All)were associated with the auxin pathway.The full-length Unigene13430_All coding sequence(843 bp)encoded 280 amino acids,whereas the full-length CL10096.Contig1_All coding sequence(1,470 bp)encoded 489 amino acids.Unigene13430_All and CL10096.Contig1_All were identified as IAA gene family members and were respectively named PsIAA27 and PsARF19.The qRT-PCR analysis and functional verification indicated that the expressions of PsARF19 and PsIAA27 in tree peony seedlings,cuttings and grafted seedlings were significant different.PsARF19 promoted root development,it might be a regulatory gene related to the formation of tree peony roots,while PsIAA27 inhibited lateral root development,and it might be involved in controlling auxin sensitivity during root formation.The results of this study may form the basis of future investigations on the mechanism mediating peony root formation.The transcriptome data will be an excellent resource for researchers interested in characterizing the rooting-related tree peony genes.Yinglong Song Wenqian Shang Zheng Wang Songlin He Xinya Meng Liyun Shi Yuxiao Shen Dan He Xueyuan Lou Yuke Sun 2022Phyton-International Journal of Experimental Botany2022,91,12:0
6Liver kinase B1 attenuates liver ischemia/reperfusion injury via inhibiting the NLRP3 inflammasome显示文摘Liver ischemia/reperfusion injury(IRI),a serious inflammatory response driven by innate immunity,occurs in liver surgeries such as liver resection and liver transplantation,leading to liver dysfunction,liver failure,and even rejection after transplantation.Liver kinase B1(LKB1)plays a pivotal anti-inflammatory role in IRI.One of the most important factors involved in liver IRI is the aberrant activation of the nucleotide binding oligomerization domain like receptor(NLR)family,pyrin domain-containing 3(NLRP3)inflammasome in Kupffer cells.However,the mechanisms underlying the effect of LKB1 on the NLRP3 inflammasome in liver IRI remain elusive.In this study,we found that the expression of LKB1 was decreased in liver IRI,while the NLRP3 inflammasome level was increased as shown,as revealed by RT-qPCR and western blot analysis.Furthermore,upregulation of LKB1 abrogated the expression of the NLRP3 inflammasome,which improved liver function and liver pathology in the liver IRI model in vivo.In vitro,overexpression of LKB1 inhibited the activation of NLRP3 inflammasome and nuclear factor-κB,while the inhibitory effect was reversed by silencing the expression of the forkhead box protein O1 in the RAW264.7 macrophage hypoxia/reoxygenation model.In conclusion,our results suggest that LKB1 exerts a protective effect against liver IRI by downregulating the NLRP3 inflammasome.Jiangwen Dai Qingsong Chen Weifeng Huang Kun Shi Yuke Zhang Tingting Li Tong Mou Zuotian Huang Zhongjun Wu 2021Acta Biochimica et Biophysica Sinica2021,53,5:0
7Controllable construction of ionic frameworks for multi-site synergetic enhancement of CO_(2) capture显示文摘CO_(2) capture is one of the key technologies for dealing with the global warming and implementing lowcarbon development strategy.The emergence of ionic metal-organic frameworks(I-MOFs)has diversified the field of porous materials,which have been extensively applied for gas adsorption and separation.In this work,amino-functionalized imidazolium ionic liquid as organic monodentate ligand was used for one step synthesis microporous Cu based I-MOFs.Precise tuning of the adsorption properties was obtained by incorporating aromatic anions,such as phenoxy,benzene carboxyl,and benzene sulfonic acid group into the I-MOFs via a facile ion exchange method.The new I-MOFs showed high thermal stability and high capacity of 5.4 mmol·g^(-1) under atmospheric conditions for selective adsorption of CO_(2).The active sites of microporous Cu-MOF are the ion basic center and unsaturated metal,and electrostatic attraction and hydroxyl bonding between CO_(2) and modified functional sulfonic groups are responsible for the adsorption.This work provides a feasible strategy for the design of I-MOF for functional gas capture.Yuke Zhang Hongxue Xu Haonan Wu Lijuan Shi Jiancheng Wang Qun Yi 2024Frontiers of Chemical Science and Engineering2024,18,1:0
8Distance-Based Regression Analysis for Measuring Associations显示文摘Distance-based regression model,as a nonparametric multivariate method,has been widely used to detect the association between variations in a distance or dissimilarity matrix for outcomes and predictor variables of interest in genetic association studies,genomic analyses,and many other research areas.Based on it,a pseudo-F statistic which partitions the variation in distance matrices is often constructed to achieve the aim.To the best of our knowledge,the statistical properties of the pseudo-F statistic has not yet been well established in the literature.To fill this gap,the authors study the asymptotic null distribution of the pseudo-F statistic and show that it is asymptotically equivalent to a mixture of chi-squared random variables.Given that the pseudo-F test statistic has unsatisfactory power when the correlations of the response variables are large,the authors propose a square-root F-type test statistic which replaces the similarity matrix with its square root.The asymptotic null distribution of the new test statistic and power of both tests are also investigated.Simulation studies are conducted to validate the asymptotic distributions of the tests and demonstrate that the proposed test has more robust power than the pseudo-F test.Both test statistics are exemplified with a gene expression dataset for a prostate cancer pathway.SHI Yuke ZHANG Wei LIU Aiyi LI Qizhai 2023Journal of Systems Science & Complexity2023,36,1:0
