1High nitrogen application rate and planting density reduce wheat grain yield by reducing filling rate of inferior grain in middle spikelets显示文摘Excessive use of nitrogen fertilizer and high planting density reduce grain weight in wheat.However,the effects of high nitrogen and planting density on the filling of grain located in different positions of the wheat spikelet are unknown.A two-year field experiment was conducted to investigate this question and the underlying mechanisms with respect to hormone and carbohydrate activity.Both high nitrogen application and planting density significantly increased spike density,while reducing kernel number per spike and 1000-kernel weight.However,the effects of high nitrogen and high plant density on kernel number per spike and 1000-kernel weight were different.The inhibitory effect of high nitrogen application and high planting density on kernel number per spike was achieved mainly by a reduction in kernel number per spikelet in the top and bottom spikelets.However,the decrease in 1000-kernel weight was contributed mainly by the reduced weight of grain in the middle spikelets.The grain-filling rate of inferior grain in the middle spikelets was significantly decreased under high nitrogen input and high planting density conditions,particularly during the early and middle grain-filling periods,leading to the suppression of grain filling and consequent decrease in grain weight.This effect resulted mainly from inhibited sucrose transport to and starch accumulation in inferior grain in the middle spikelets via reduction of the abscisic acid/ethylene ratio.This mechanism may explain how high nitrogen application and high planting density inhibit the grain filling of inferior wheat grain.Yang Liu Yuncheng Liao Wenzhao Liu 2021The Crop Journal2021,9,2:9
2Population Genomic Analysis and De Novo Assembly Reveal the Origin of Weedy Rice as an Evolutionary Game显示文摘Crop weediness,especially that of weedy rice(Oryza sativa f.spontanea),remains mysterious.Weedy rice possesses robust ecological adaptability;however,how this strain originated and gradually formed proprietary genetic features remains unclear?Here,we demonstrate that weedy rice at Asian high latitudes(WRAH)is phylogenetically well defined and possesses unselected genomic characteristics in many divergence regions between weedy and cultivated rice.We also identified novel quantitative trait loci underlying weedy-specific traits,and revealed that a genome block on the end of chromosome 1 is associated with rice weediness.To identify the genomic modifications underlying weedy rice evolution,we generated the first de novo assembly of a high-quality weedy rice genome(WR04-6),and conducted a comparative genomics study between WR04-6 with other rice reference genomes.Multiple lines of evidence,including the results of demographic scenario comparisons,suggest that differentiation between weedy rice and cultivated rice was initiated by genetic improvement of cultivated rice and that the essence of weediness arose through semi-domestication.A plant height model further implied that the origin of WRAH can be modeled as an evolutionary game and indicated that strategy-based selection driven by fitness shaped its genomic diversity.Jian Sun Dianrong Ma Liang Tang Minghui Zhao Guangchen Zhangh Wenjia Wang Jiayu Song Xiang Li Zimeng Liu Wenxing Zhang Quan Xu Yuncheng Zhou Jianzhong Wu Toshio Yamamoto Fei Dai Yan Lei Song Li Gang Zhou Hongkun Zheng Zhengjin Xu Wenfu Chen 2019Molecular Plant2019,12,5:9
3Inhibiting Hv1 channel in peripheral sensory neurons attenuates chronic inflammatory pain and opioid side effects显示文摘Both opioids and nonsteroidal anti-inflammatory drugs(NSAIDS)produce deleterious side effects and fail to provide sustained relief in patients with chronic inflammatory pain.Peripheral neuroinflammation(PN)is critical for initiation and development of inflammatory pain.A better understanding of molecular mechanisms underlying PN would facilitate the discovery of new analgesic targets and the development of new therapeutics.Emerging evidence suggests that peripheral sensory neurons are not only responders to painful stimuli,but are also actively engaged in inflammation and immunity,whereas the intrinsic regulatory mechanism is poorly understood.Here we report the expression of proton-selective ion channel Hv1 in peripheral sensory neurons in rodents and humans,which was previously shown as selectively expressed in microglia in mammalian central nervous system.Neuronal Hv1 was up-regulated by PN or depolarizing stimulation,which in turn aggravates inflammation and nociception.Inhibiting neuronal Hv1 genetically or by a newly discovered selective inhibitor YHV98-4 reduced intracellular alkalization and ROS production in inflammatory pain,mitigated the imbalance in downstream SHP-1-pAKT signaling,and also diminished pro-inflammatory chemokine release to alleviate nociception and morphine-induced hyperalgesia and tolerance.Thus,our data reveal neuronal Hv1 as a novel target in analgesia strategy and managing opioids-related side effects.Qiansen Zhang Yimin Ren Yiqing Mo Peipei Guo Ping Liao Yuncheng Luo Jie Mu Zhuo Chen Yang Zhang Ya Li Linghui Yang Daqing Liao Jie Fu Juwen Shen Wei Huang Xuewen Xu Yanyan Guo Lianghe Mei Yunxia Zuo Jin Liu Huaiyu Yang Ruotian Jiang 2022Cell Research2022,32,5:3
4Transcriptional control by HNF-1:Emerging evidence showing its role in lipid metabolism and lipid metabolism disorders显示文摘The present review focuses on the roles and underlying mechanisms of action of hepatic nuclear factor-1(HNF-1)in lipid metabolism and the development of lipid metabolism disorders.HNF-1 is a transcriptional regulator that can form homodimers,and the HNF-1αand HNF-1βisomers can form heterodimers.Both homo-and heterodimers recognize and bind to specific cis-acting elements in gene promoters to transactivate transcription and to coordinate the expression of target lipid-related genes,thereby influencing the homeostasis of lipid metabolism.HNF-1 was shown to restrain lipid anabolism,including synthesis,absorption,and storage,by inhibiting the expression of lipogenesis-related genes,such as peroxisome proliferator-activated receptorγ(PPARγ)and sterol regulatory element-binding protein-1/2(SREBP-1/2).Moreover,HNF-1 enhances the expression of various genes,such as proprotein convertase subtilisin/kexin type 9(PCSK9),glutathione peroxidase 1(GPx1),and suppressor of cytokine signaling-3(SOCS-3)and negatively regulates signal transducer and activator of transcription(STAT)to facilitate lipid catabolism in hepatocytes.HNF-1 reduces hepatocellular lipid decomposition,which alleviates the progression of nonalcoholic fatty liver disease(NAFLD).HNF-1 impairs preadipocyte differentiation to reduce the number of adipocytes,stunting the development of obesity.Furthermore,HNF-1 reduces free cholesterol levels in the plasma to inhibit aortic lipid deposition and lipid plaque formation,relieving dyslipidemia and preventing the development of atherosclerotic cardiovascular disease(ASCVD).In summary,HNF-1 transcriptionally regulates lipid-related genes to manipulate intracorporeal balance of lipid metabolism and to suppress the development of lipid metabolism disorders.Fang Liu Xiao Zhu Xiaping Jiang Shan Li Yuncheng Lv 2022Genes & Diseases2022,9,5:3

7Temporal-spatial distributions of road silt loadings and fugitive road dust emissions in Beijing from 2019 to 2020显示文摘Road silt loading(sL)is an important parameter in the fugitive road dust(FRD)emissions.In this study,the improved Testing Re-entrained Aerosol Kinetic Emissions from Roads(TRAKER)combined with the AP-42 method was firstly developed to quickly measure and estimate the sLs of paved roads in Beijing,China.The annual average sLs in Beijing was 0.59±0.31 g/m^(2)in 2020,and decreased by 22.4%compared with that in 2019.The seasonal variations of sLs followed the order of spring>winter>summer>autumn in the two years.The seasonal mean road sLs on the same type road in the four seasons presented a decline trend from^(2)019 to 2020,especially on the Expressway,decreasing 47.4%-72.7%.The road sLs on the different type roads in the same season followed the order of Major arterial∼Minor arterial∼Branch road>Express road,and Township road∼Country highway>Provincial highway∼National highway.The emission intensities of PM10 and PM_(2.5)from FRD in Beijing in 2020 were lower than those in 2019.The PM10 and PM_(2.5)emission intensities at the four planning areas in the two years all presented the order of the capital functional core area>the urban functional expansion area>the urban development new area>the ecological conservation and development area.The annual emissions of PM10 and PM_(2.5)from FRD in Beijing in 2020 were 74,886 ton and 18,118 ton,respectively,decreasing by∼33.3%compared with those in 2019.Haibin Wang Lihui Han Tingting Li Song Qu Yuncheng Zhao Shoubin Fan Tong Chen Haoran Cui Junfang Liu 2023Journal of Environmental Sciences2023,,10:1

11Early Diagnosis of Bipolar Disorder Coming Soon:Application of an Oxidative Stress Injury Biomarker(BIOS)Model显示文摘Early distinction of bipolar disorder(BD)from major depressive disorder(MDD)is difficult since no tools are available to estimate the risk of BD.In this study,we aimed to develop and validate a model of oxidative stress injury for predicting BD.Data were collected from 1252 BD and 1359 MDD patients,including 64 MDD patients identified as converting to BD from 2009 through 2018.30 variables from a randomly-selected subsample of 1827(70%)patients were used to develop the model,including age,sex,oxidative stress markers(uric acid,bilirubin,albumin,and prealbumin),sex hormones,cytokines,thyroid and liver function,and glycolipid metabolism.Univariate analyses and the Least Absolute Shrinkage and Selection Operator were applied for data dimension reduction and variable selection.Multivariable logistic regression was used to construct a model for predicting bipolar disorder by oxidative stress biomarkers(BIOS)on a nomogram.Internal validation was assessed in the remaining 784 patients(30%),and independent external validation was done with data from 3797 matched patients from five other hospitals in China.10 predictors,mainly oxidative stress markers,were shown on the nomogram.The BIOS model showed good discrimination in the training sample,with an AUC of 75.1%(95%CI:72.9%–77.3%),sensitivity of 0.66,and specificity of 0.73.The discrimination was good both in internal validation(AUC 72.1%,68.6%–75.6%)and external validation(AUC 65.7%,63.9%–67.5%).In this study,we developed a nomogram centered on oxidative stress injury,which could help in the individualized prediction of BD.For better real-world practice,a set of measurements,especially on oxidative stress markers,should be emphasized using big data in psychiatry.Zhiang Niu Xiaohui Wu Yuncheng Zhu Lu Yang Yifan Shi Yun Wang Hong Qiu Wenjie Gu Yina Wu Xiangyun Long Zheng Lu Shaohua Hu Zhijian Yao Haichen Yang Tiebang Liu Yong Xia Zhiyu Chen Jun Chen Yiru Fang 2022Neuroscience Bulletin2022,38,9:1

15Removal of superior wheat kernels promotes filling of inferior kernels by changing carbohydrate metabolism and sink strength显示文摘Poor filling and low weight of inferior kernels limit the further improvement of wheat yield. Two cultivars, Shuangda 1 and Xinong 538, with different grain weights, were selected to investigate the physiological changes of inferior kernels by removal of superior kernels(RS) at the flowering stage. i TRAQ combined with physiological indexes was used to identify factors limiting the filling of inferior kernels.Removal of superior kernels significantly increased the mean filling rate of inferior kernels and thus increased their weight. A set of 6012 proteins in inferior wheat kernels were differentially expressed between the RS and control. These differentially expressed proteins were involved mainly in carbon metabolism and energy metabolism. The main reason for the promoting effect of RS on the filling of inferior kernels may be that RS downregulated proteins involved in glycolysis and pyruvate metabolism while upregulating proteins involved in carbon fixation and photosynthesis. Consequently, RS greatly increased the ATP content in inferior kernels, supplying energy for them to absorb photosynthetic assimilates. Removal of superior kernels increased the activities of sucrose synthase, soluble starch synthase,adenosine diphosphate glucose pyrophosphorylase, and starch branching enzyme in inferior kernels and promoted starch accumulation in them. Thus, RS promoted the filling of inferior kernels and increased their weight. 17Sortilin-induced lipid accumulation and atherogenesis are suppressed by HNF1b SUMOylation promoted by flavone of Polygonatum odoratum显示文摘This study aims to investigate the impact of hepatocyte nuclear factor 1β(HNF1b)on macrophage sortilin-mediated lipid metabolism and aortic atherosclerosis and explore the role of the flavone of Polygonatum odoratum(PAOA-flavone)-promoted small ubiquitin-related modifier(SUMO)modification in the atheroprotective efficacy of HNF1b.HNF1b was predicted to be a transcriptional regulator of sortilin expression via bioinformatics,dual-luciferase reporter gene assay,and chromatin immunoprecipitation.HNF1b overexpression decreased sortilin expression and cellular lipid contents in THP-1 macrophages,leading to a depression in atherosclerotic plaque formation in low-density lipoprotein(LDL)receptor-deficient(LDLR−/−)mice.Multiple SUMO1-modified sites were identified on the HNF1b protein and co-immunoprecipitation confirmed its SUMO1 modification.The SUMOylation of HNF1b protein enhanced the HNF1b-inhibited effect on sortilin expression and reduced lipid contents in macrophages.PAOA-flavone treatment promoted SUMO-activating enzyme subunit 1(SAE1)expression and SAE1-catalyzed SUMOylation of the HNF1b protein,which prevented sortilin-mediated lipid accumulation in macrophages and the formation of atherosclerotic plaques in apolipoprotein E-deficient(ApoE−/−)mice.Interference with SAE1 abrogated the improvement in lipid metabolism in macrophage cells and atheroprotective efficacy in vivo upon PAOA-flavone administration.In summary,HNF1b transcriptionally suppressed sortilin expression and macrophage lipid accumulation to inhibit aortic lipid deposition and the development of atherosclerosis.This anti-atherosclerotic effect was enhanced by PAOA-flavone-facilitated,SAE1-catalyzed SUMOylation of the HNF1b protein.Fang LIU Shirui CHEN Xinyue MING Huijuan LI Zhaoming ZENG Yuncheng LV 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,11:0
18Acousto-optic scanning spatial-switching multiphoton lithography显示文摘Nano-3D printing has obtained widespread attention owing to its capacity to manufacture end-use components with nano-scale features in recent years.Multiphoton lithography(MPL)is one of the most promising 3D nanomanufacturing technologies,which has been widely used in manufacturing micro-optics,photonic crystals,microfluidics,meta-surface,and mechanical metamaterials.Despite of tremendous potential of MPL in laboratorial and industrial applications,simultaneous achievement of high throughput,high accuracy,high design freedom,and a broad range of material structuring capabilities remains a long-pending challenge.To address the issue,we propose an acousto-optic scanning with spatial-switching multispots(AOSS)method.Inertia-free acousto-optic scanning and nonlinear swept techniques have been developed for achieving ultrahigh-speed and aberration-free scanning.Moreover,a spatial optical switch concept has been implemented to significantly boost the lithography throughput while maintaining high resolution and high design freedom.An eight-foci AOSS system has demonstrated a record-high 3D printing rate of 7.6×10^(7)voxel s^(-1),which is nearly one order of magnitude higher than earlier scanning MPL,exhibiting its promise for future scalable 3D nanomanufacturing.Binzhang Jiao Fayu Chen Yuncheng Liu Xuhao Fan Shaoqun Zeng Qi Dong Leimin Deng Hui Gao Wei Xiong 2023International Journal of Extreme Manufacturing2023,5,3:0
19IMPULSIVE VACCINATION OF SIRS EPIDEMIC MODEL WITH STAGE-STRUCTURE显示文摘The period of infection is partitioned into the early and later stages according to the development process of infection. The infected individuals in the different stages have the different ability of transmitting disease. The constant recruitment rate and exponential natural death, as well as the disease-related death and pulse vaccination are incorporated into the model. Through the discrete dynamical system determined by the stroboscopic map, we obtain the exact infection-free periodic solution to the system. Further more, by the comparison theorem, we prove that the infection-free periodic solution is globally stable.
