1Propofol differentially induces unconsciousness and respiratory depression through distinct interactions between GABAa receptor and GABAergic neuron in corresponding nuclei显示文摘Propofol is the most commonly used intravenous anesthetic worldwide.It can induce loss of consciousness prior to the occurrence of severe respiratory suppression,which is also a pharmacodynamic feature of all general anesthetics.However,the neural mechanisms underlying this natural phenomenon are controversial and highly related to patient safety.In the present study,we demonstrated that the pharmacodynamic effects of propofol(50 and 100 μM)on suppression of consciousness-related excitatory postsynaptic currents in the medial prefrontal cortex(mPFC)and centromedian nucleus of the thalamus(CMT)were lower than those in the kernel respiratory rhythmogenesis nucleus pre-Botzinger complex(PrBo).Furthermore,we unexpectedly found that the GABAa receptor β3 subunit is the key target for propofol's action and that it is mutually and exclusively expressed in GABAergic neurons.It is also more abundant in the mPFC and CMT,but mainly co-localized with GABAergic neurons in the PrBo.As a result,the differentiated expression pattern should mediate more neuron suppression through the activation of GABAergic neurons in the mPFC and CMT at low doses of propofol(50 μM).However,PrBo GABAergic neurons were only activated by propofol at a high dose(100 μM).These results highlight the detailed pharmacodynamic effects of propofol on consciousness-related and respiration-related nuclei and provide the distinct interaction mechanism between the β3 subunit and GABAergic neurons in mediating the suppression of consciousness compared to the inhibition of respiration.Junli Jiang Yingfu Jiao Po Gao Wen Yin Wei Zhou Yunchun Zhang Yanjun Liu Daxiang Wen Yuan Wang Liang Zhou Tian Yu Weifeng Yu 2021Acta Biochimica et Biophysica Sinica2021,53,8:11

3High-performance quasi-2D perovskite light-emitting diodes: from materials to devices显示文摘Quasi-two-dimensional(quasi-2D)perovskites have attracted extraordinary attention due to their superior semiconducting properties and have emerged as one of the most promising materials for next-generation lightemitting diodes(LEDs).The outstanding optical properties originate from their structural characteristics.In particular,the inherent quantum-well structure endows them with a large exciton binding energy due to the strong dielectricand quantum-confinement effects;the corresponding energy transfer among different n-value species thus results in high photoluminescence quantum yields(PLQYs),particularly at low excitation intensities.The review herein presents an overview of the inherent properties of quasi-2D perovskite materials,the corresponding energy transfer and spectral tunability methodologies for thin films,as well as their application in high-performance LEDs.We then summarize the challenges and potential research directions towards developing high-performance and stable quasi-2D PeLEDs.The review thus provides a systematic and timely summary for the community to deepen the understanding of quasi-2D perovskite materials and resulting LED devices.Li Zhang Changjiu Sun Tingwei He Yuanzhi Jiang Junli Wei Yanmin Huang Mingjian Yuan 2021Light(Science & Applications)2021,10,4:5
4Activation of anterior thalamic reticular nucleus GABAergic neurons promotes arousal from propofol anesthesia in mice显示文摘Propofol is widely used for the induction and maintenance of anesthesia,which causes a rapid loss of consciousness.However,the mechanisms underlying the hypnosis effect of propofol are still not fully understood.The thalamic reticular nucleus(TRN)is crucial for regulating wakefulness,sleep rhythm generation,and sleep stability,while the role of TRN in the process of propofol-induced anesthesia is still unknown.Here,we investigated the function of the anterior TRN in propofol general anesthesia.Our results demonstrated that the neural activity of anterior TRN is suppressed during propofol anesthesia,whereas it is robustly activated from anesthesia by recording the calcium signals using fiber photometry technology.The results showed that the activation of anterior TRN neurons by chemogenetic and optogenetic methods shortens the emergency time without changing the induction time.Conversely,chemogenetic or optogenetic inhibition of the TRN neurons leads to a delay in the recovery time.Our study showed that anterior TRN is crucial for behavioral arousal without affecting the induction time of propofol anesthesia.Yanjun Liu Bing Chen Yirong Cai Yuan Han Ying Xia Nanqi Li Bingqian Fan Tianjie Yuan Junli Jiang Po Gao Weifeng Yu Yingfu Jiao Wenxian Li 2021Acta Biochimica et Biophysica Sinica2021,53,7:3
5Effect of cytotoxic T-lymphocyte antigen-4,TNF-alpha polymorphisms on osteosarcoma: evidences from a meta-analysis显示文摘Objective:Previous studies have investigated the role of cytotoxic T-lymphocyte antigen-4(CTLA-4)and tumor necrosis factor-alpha(TNF-α)in carcinogenesis of osteosarcoma,but their results were inconsistent.We aimed to clarify the associations between CTLA-4,TNF-αpolymorphism and osteosarcoma risk by using meta-analysis.Methods:We searched relevant studies without language restriction in PubMed,EMbase,Cochrane Library,Google Scholar databases,Chinese National Knowledge Infrastructure(CNKI)and conference literature in humans published prior to March 2013.The strengths of the associations between genetic variants and osteosarcoma risk were estimated by odds ratio(OR)with 95%confidence interval(95%CI).Results:A total of seven studies with 1,198 osteosarcoma patients and 1,493 controls were selected.Four studies were eligible for CTLA-4(1,003 osteosarcoma and 1,162 controls),and three studies for TNF-α(195 osteosarcoma and 331 controls).Pooled results showed that rs231775 polymorphism of CTLA-4 was associated with osteosarcoma risk(GG vs.AA:OR=1.63,95%CI=1.24-2.13;GG+GA vs.AA:OR=1.56,95%CI=1.21-2.01;AA+GA vs.GG:OR=0.83,95%CI=0.71-0.97;G vs.A:OR=1.21,95%CI=1.08-1.36).No significant heterogeneity was observed across the studies.No significant associations were found between rs5742909 polymorphism of CTLA-4 or rs1800629 polymorphism of TNF-αand osteosarcoma risk.Conclusions:These results suggest that the rs231775 polymorphism of CTLA-4 may play an important role in carcinogenesis of osteosarcoma.Jianwei Liu Junli Wang Weiping Jiang Yujin Tang 2013Chinese Journal of Cancer Research2013,25,6:3

8Correlation between radiologic features on contrastenhanced CT and pathological tumor grades in pancreatic neuroendocrine neoplasms显示文摘Contrast-enhanced computed tomography(CT)contributes to the increasing detection of pancreatic neuroendocrine neoplasms(PNENs).Nevertheless,its value for differentiating pathological tumor grades is not well recognized.In this report,we have conducted a retrospective study on the relationship between the 2017 World Health Organization(WHO)classification and CT imaging features in 94 patients.Most of the investigated features eventually provided statistically significant indicators for discerning PNENs G3 from PNENs G1/G2,including tumor size,shape,margin,heterogeneity,intratumoral blood vessels,vascular invasion,enhancement pattern in both contrast phases,enhancement degree in both phases,tumor-to-pancreas contrast ratio in both phases,common bile duct dilatation,lymph node metastases,and liver metastases.Ill-defined tumor margin was an independent predictor for PNENs G3 with the highest area under the curve(AUC)of 0.906 in the multivariable logistic regression and receiver operating characteristic curve analysis.The portal enhancement ratio(PER)was shown the highest AUC of 0.855 in terms of quantitative features.Our data suggest that the traditional contrastenhanced CT still plays a vital role in differentiation of tumor grades and heterogeneity analysis prior to treatment.Wenbin Xu Han Yan Lulu Xu Mingna Li Wentao Gao Kuirong Jiang Junli Wu Yi Miao 2021The Journal of Biomedical Research2021,35,3:2
9Dexmedetomidine maintains blood-brain barrier integrity by inhibiting Drp1-related endothelial mitochondrial dysfunction in ischemic stroke显示文摘Stroke is the second leading cause of death and long-term disability worldwide,which lacks effective treatment.Perioperative stroke is associated with much higher rates of mortality and disability.The neuroprotective role of dexmedetomidine(Dex),a highly selective agonist of alpha2-adrenergic receptor,has been reported in a stroke rat model,and it was found that pretreatment of Dex before stroke could alleviate blood–brain barrier(BBB)breakdown.However,the underlying mechanisms are still unknown.As the brain endothelial cells are the main constituents of BBB and in high demand of energy,mitochondrial function of endothelial cells plays an important role in the maintenance of BBB.Given that dynamin-related protein 1(Drp1)is a protein mediating mitochondrial fission,with mitochondrial fusion that balances mitochondrial morphology and ensures mitochondria function,the present study was designed to investigate the possible role of Drp1 in endothelial cells involved in the neuroprotective effects of Dex in ischemic stroke.Our results showed that preconditioning with Dex reduced infarction volume,alleviated brain water content and BBB damage,and improved neurological scores in middle cerebral artery occlusion rats.Meanwhile,Dex enhanced cell activity and decreased cell apoptosis in oxygen-glucose deprivation human brain microvascular endothelial cells in vitro.These protective effects of Dex were correlated with the mitochondrial morphology integrality of endothelial cells,mediated by increased phosphorylation of serine 637 in Drp1,and could be reversed byα2-adrenergic receptor antagonist Yohimbine and AMP-activated protein kinase inhibitor Compound C.These findings suggest new molecular pathways involved in the neuroprotective effects of Dex in ischemic stroke.As Dex is routinely used as a sedative drug clinically,our findings provide molecular evidence that it has perioperative neuroprotection from ischemic stroke.Wei Zhou Yunchun Zhang Yingfu Jiao Wen Yin Haiping Dong Saihong Xu Dan Tang Junli Jiang Jianlin Shao Zhenhong Wang Weifeng Yu 2021Acta Biochimica et Biophysica Sinica2021,53,9:2

18Transcriptome and co-expression network analyses provide insights into fruit shading that enhances carotenoid accumulation in pomelo(Citrus grandis)显示文摘Carotenoids are indispensable for human health,and citrus fruit are a crucial source of dietary carotenoids.Bagging,an important orchard practice to enhance fruit economic value,is widely used in many horticultural crops,including citrus fruit.The bagged‘Majiayou’pomelo(Citrus grandis)produces vivid deeper red pulp,a fantastic agronomic trait,but the underlying molecular regulatory mechanism remains largely unexplored.Here,the enhancement of carotenoids,especially lycopene,was confirmed by HPLC analysis of carotenoids in the pulp of bagged fruit and controls.qRT-PCR analysis of the 14 carotenoid pathway genes further revealed that upregulated PSY and downregulated CCD1 expression in bagged fruit could directly enhance the accumulation of carotenoids.In addition,RNA-seq analysis identified 311 differentially expressed genes(DEGs)in the bagged fruit and controls in five developmental stages.Weighted gene co-expression network analysis(WGCNA)identified 13 critical candidate genes among the DEGs,which are closely associated with lycopene accumulation.The underlying regulatory mechanism of these candidate genes on the transcription of carotenoid pathway genes in the bagged fruit was discussed.Considering that the candidate genes were involved in the corresponding metabolic pathways,the increase in sucrose content and decrease in ABA in bagged fruit were also identified,implying that these candidate genes may be indirectly related to carotenoid enhancement in pulp by regulating phytohormones,primary metabolism,and stress responses.The results provide new insights into the potential regulatory mechanism of lycopene enhancement in the pulp of bagged‘Majiayou’pomelo,facilitating breeding and orchard management efforts to improve the nutritional quality and esthetic value of citrus,and perhaps other fruit crops.Qihang Jiang Junli Ye Kaijie Zhu Fangfang Wu Lijun Chai Qiang Xu Xiuxin Deng 2022Horticultural Plant Journal2022,8,4:1
19Genetic Architecture and Functional Implications of the CSF‑Contacting Nucleus显示文摘We previously identified a unique nucleus,the cerebrospinal fluid(CSF)-contacting nucleus.This study aims to understand its gene architecture and preliminarily suggest its functions.The results showed that there were about 19,666 genes in this nucleus,of which 913 were distinct from the dorsal raphe nucleus(non-CSF contacting).The top 40 highly-expressed genes are mainly related to energy metabolism,protein synthesis,transport,secretion,and hydrolysis.The main neurotransmitter is 5-HT.The receptors of 5-HT and GABA are abundant.The channels for Cl–,Na+,K+,and Ca2+are routinely expressed.The signaling molecules associated with the CaMK,JAK,and MAPK pathways were identified accurately.In particular,the channels of transient receptor potential associated with nociceptors and the solute carrier superfamily members associated with cell membrane transport were significantly expressed.The relationship between the main genes of the nucleus and life activities is preliminarily verified.Siyuan Song Yumin Yuan Lingling Xu Jun Jiang Ying Li Yao Yan Qing Li Fang Zhou Junli Cao Licai Zhang 2023Neuroscience Bulletin2023,39,11:1
