1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 2Growth, photosynthesis and podophyllotoxin accumulation of Dysosma versipellis in response to a light gradient and conservation implications显示文摘Dysosma versipellis (Hance) M. Cheng ex Ying (Berberidaceae) is a rare and vulnerable, perennial herb endemic to China with pharmaceutical significance. Increasing habitat loss and over-exploitation of the plant has severely affected the plant's in situ conservation, necessitating ex situ conservation and commercial cultivation. The light regime is a critical environmental factor contributing to successful ex situ conservation via efficient production of biomass and secondary metabolites. We investigated the responses of 2-year-old D. versipellis plants to a light gradient in terms of leaf morphology, growth, biomass production, photo- synthesis and podophyllotoxin (PTOX) accumulation. D. versipellis responded to the light gradient in terms of plant height, leaf size, leaf photosynthesis and PTOX accumulation. Plants demonstrated optimal vegetative growth, photosynthetic capability, and PTOX production, under a light intensity equal to 30% full sunlight. Half full sunlight led to the lowest production of PTOX despite having no obviously different effects on growth. Full sunlight was able to cause leaf death in mid-summer (July), although these plantlets did not significantly differ in growth or biomass production before July and new leaves emerged in August. D. versipellis is therefore proposed to be a shade-tolerant plant, well adapted to variations in irradiance. 3Compositions and sources of organic acids in fine particles(PM_(2.5)) over the Pearl River Delta region, south China显示文摘Organic acids as important constituents of organic aerosols not only influence the aerosols' hygroscopic property, but also enhance the formation of new particles and secondary organic aerosols. This study reported organic acids including C14–C32fatty acids, C4–C9dicarboxylic acids and aromatic acids in PM2.5collected during winter 2009 at six typical urban, suburban and rural sites in the Pearl River Delta region. Averaged concentrations of C14–C32fatty acids, aromatic acids and C4– C9 dicarboxylic acids were 157, 72.5 and 50.7 ng/m3, respectively. They totally accounted for 1.7% of measured organic carbon. C20–C32fatty acids mainly deriving from higher plant wax showed the highest concentration at the upwind rural site with more vegetation around, while C14–C18fatty acids were more abundant at urban and suburban sites, and dicarboxylic acids and aromatic acids except 1,4-phthalic acid peaked at the downwind rural site. Succinic and azelaic acid were the most abundant among C4–C9dicarboxylic acids, and 1,2-phthalic and 1,4-phthalic acid were dominant aromatic acids. Dicarboxylic acids and aromatic acids exhibited significant mutual correlations except for 1,4-phthalic acid, which was probably primarily emitted from combustion of solid wastes containing polyethylene terephthalate plastics. Spatial patterns and correlations with typical source tracers suggested that C14–C32fatty acids were mainly primary while dicarboxylic and aromatic acids were largely secondary. Principal component analysis resolved six sources including biomass burning, natural higher plant wax, two mixed anthropogenic and two secondary sources; further multiple linear regression revealed their contributions to individual organic acids. 4Template-free synthesis and characterization of K-phillipsite for use in potassium extraction from seawater显示文摘K-phillipsite was prepared using a hydrothermal method.Soluble glass and sodium aluminate were used as raw materials in the absence of an organic template,investigations regarding the K^+ ions were conducted at room temperature to determine the ion-exchange capacity in the seawater sample and the selectivity coefficient of the mixed K^+-Na^+solution.The sample was characterized by X-ray diffraction(XRD),scanning electron microscopy(SEM),and energy dispersion spectroscopy(EDS).The K^+ ionexchange capacity is 51 mg/g in seawater and the selectivity coefficient is 75.1 in the mixed K^+-Na^+solution.The sample has a selectivity preference for K^+,and therefore can be used to selectively extract potassium from seawater.The sample composed of Si,Al,K,Na,and O exhibits a cross-like shape and is a typical K-phillipsite structure.Jin Hou Junsheng Yuan Jia Xu Yunpeng Fu Chunxia Meng 2013Particuology2013,11,6:3
5Enriched endoplasmic reticulum-mitochondria interactions result in mitochondrial dysfunction and apoptosis in oocytes from obese mice显示文摘Background: Maternal obesity alters oocytes and subsequent fetal metabolism. An increasing number of studies have shown that the endoplasmic reticulums(ER) or mitochondria have important effects on oocyte quality, but there has been no study of the effect of mitochondria-associated ER membranes(MAMs) on oocyte quality. The present study was designed to assess whether the level of MAM and MAM-related proteins were different in oocytes from obese and control mice.Results: First, oocytes from mice with high-fat-diet(HFD)-induced obesity had higher levels(either greater numbers or a higher proportion for the same numbers) of MAM than oocytes from control mice. The abundance of MAM-related proteins in oocytes from obese mice was significantly greater at both the messenger RNA and protein levels, including inositol 1,4,5-trisphosphate receptor, type 1(IP3R1), inositol 1,4,5-trisphosphate receptor,type 2(IP3 R2) and phosphofurin acidic cluster sorting protein 2(PACS-2). Further, there was an increase in mitochondrial Ca^(2+)([Ca^(2+)]_m) which was associated with increased apoptosis and compromised cytoplasmic maturation in oocytes from obese mice. Down-regulation of MAM-related protein IP3R1 in oocytes from obese mice decreased [Ca^(2+)]_mand apoptosis and improved cytoplasmic maturation but did not reduce the overal MAM level. However, down-regulating MAM-related protein PACS-2 in oocytes from obese mice did reduce the level of MAM and [Ca^(2+)]_m, which decreased the rate of apoptosis and improved cytoplasmic maturation of oocytes from obese mice.Conclusions: It is possible that enriched MAM could increase [Ca^(2+)]_m, and this increase has been found to be associated with increased apoptosis and compromised cytoplasmic maturation in oocytes from obese mice. 6The preoperative neutrophil-to-lymphocyte ratio predicts the outcomes of patients with hepatocellular carcinoma and cirrhosis after hepatectomy显示文摘Objective The aim of the study was to investigate the prognostic value of the preoperative peripheral neutrophil-to-lymphocyte ratio(NLR) in patients with hepatocellular cancer(HCC) and cirrhosis after hepatectomy. Methods This retrospective study included 321 patients with HCC who underwent resection. The NLR was calculated using the neutrophil and lymphocyte counts in routine preoperative blood tests. Receiver operating characteristic curve analysis was performed to select the most appropriate NLR cutoff value. The preoperative NLR, patient demographics, and clinical and pathological data, including disease-free survival(DFS) and overall survival(OS), were analyzed. Results The NLR was correlated with alpha-fetoprotein levels(χ2 = 5.876, P = 0.015), tumor size(χ2 = 32.046, P < 0.001), portal vein tumor thrombus(PVTT; χ2 = 4.930, P = 0.026), tumor encapsulation(χ2 = 7.243, P = 0.007), and recurrence(χ2 = 7.717, P = 0.005). Multivariate analyses illustrated that the number of tumors, PVTT, tumor size, and the NLR were independent factors for predicting DFS and OS. In patients with HCC and cirrhosis, but not among those without cirrhosis, a larger NLR predicted poorer postoperative DFS and OS(both P < 0.001). 7Antioxidant procyanidin B2 protects oocytes against cryoinjuries via mitochondria regulated cortical tension显示文摘Background:Irreversible cryodamage caused by oocyte vitrification limited its wild application in female fertility preservation.Antioxidants were always used to antagonist the oxidative stress caused by vitrification.However,the comprehensive mechanism underlying the protective role of antioxidants has not been studied.Procyanidin B2(PCB2)is a potent natural antioxidant and its functions in response to vitrification are still unknown.In this study,the effects of PCB2 on vitrified-thawed oocytes and subsequent embryo development were explored,and the mechanisms underlying the protective role of PCB2 were systematically elucidated.Results:Vitrification induced a marked decline in oocyte quality,while PCB2 could improve oocyte viability and further development after parthenogenetic activation.A subsequent study indicated that PCB2 effectively attenuated vitrification-induced oxidative stress,rescued mitochondrial dysfunction,and improved cell viability.Moreover,PCB2 also acts as a cortical tension regulator apart from strong antioxidant properties.Increased cortical tension caused by PCB2 would maintain normal spindle morphology and promote migration,ensure correct meiosis progression and finally reduce the aneuploidy rate in vitrified oocytes.Further study reveals that ATP biosynthesis plays a crucial role in cortical tension regulation,and PCB2 effectively increased the cortical tension through the electron transfer chain pathway.Additionally,PCB2 would elevate the cortical tension in embryo cells at morula and blastocyst stages and further improve blastocyst quality.What's more,targeted metabolomics shows that PCB2 has a beneficial effect on blastocyst formation by mediating saccharides and amino acids metabolism.Conclusions:Antioxidant PCB2 exhibits multi-protective roles in response to vitrification stimuli through mitochondria-mediated cortical tension regulation.Qingrui Zhuan Jun Li Xingzhu Du Luyao Zhang Lin Meng Yuwen Luo Dan Zhou Hongyu Liu Pengcheng Wan Yunpeng Hou Xiangwei Fu 2023Journal of Animal Science and Biotechnology2023,14,1:0
8An efficient method for the sanitary vitrification of bovine oocytes in straws显示文摘Background:At present,vitrification has been widely applied to humans,mice and farm animals.To improve the efficiency of vitrification in straw,bovine oocytes were used to test a new two-step vitrification method in this study.Results:When in vitro matured oocytes were exposed to 20%ethylene glycol(EG20) for 5 min and 40%ethylene glycol(EG40) for 30 s,followed by treatment with 30%glycerol(Gly30),Gly40 or Gly50,a volume expansion was observed in Gly30 and Gly40 but not Gly50.This indicates that the intracellular osmotic pressure after a 30 s differs between EG40 and ranged between Gly40(approximately 5.6 mol/L) and Gly50(approximately 7.0 mol/L).Since oocytes are in EG40 just for only a short period of time(30 s) and at a lower temperature(4℃),we hypothesize that the main function of this step in to induce dehydration.Based on these results,we omitted the EG40 step,before oocytes were pretreated in EG20 for 5 min,exposed to pre-cooled(4℃) Gly50,for 30 s,and then dipped into liquid nitrogen.After warming,81.1%of the oocytes survived,and the surviving oocytes developed into cleavage stage embryos(63.5%) or blastocysts(20.0%) after parthenogenetic activation.Conclusions:These results demonstrate that in a two-step vitrification procedure,the permeability effect in the second step is not necessary.It is possible that the second step is only required to provide adequate osmotic pressure to condense the intracellular concentration of CPAs to a level required for successful vitrification.Yanhua Zhou Xiangwei Fu Guangbin Zhou Baoyu Jia Yi Fang Yunpeng Hou Shien Zhu 2014Journal of Animal Science and Biotechnology2014,5,4:0
9Cryopreservation of farm animal gametes and embryos:recent updates and progress显示文摘Cryopreservation has undergone tremendous advances and is widely used in animal production based on decades of study of cellular permeability, freezability and empirical generalization. Several improvement are particularly important: the cryopreservation protocol has been continuously re?ned over the years to achieve greater reproductive performance; cryoprotective agents are more effective and less toxic than previously; there has been signi?cant innovation in advanced cryopreservation systems and carriers. 10Core-Sheath CeO_(2)/SiO_(2) Nanofibers as Nanoreactors for Stabilizing Sinter‑Resistant Pt,Enhanced Catalytic Oxidation and Water Remediation显示文摘One-dimensional(1D)oxide nanofibers have attracted much attention in recent years but are still hampered by the difficulty in the expansion to 2D or 3D dimensions.Herein,ultrathin CeO_(2)/SiO_(2)nanofibers with intriguing core-sheath structures were simply fabricated by a facile single-spinneret electrospinning method and were subsequently integrated as 2D nanofi-brous mats and 3D sponges.Introducing secondary oxide(i.e.,SiO_(2))could induce a unique fine structure and further inhibit the sintering of CeO_(2)nanocrystals,endowing the resultant dual-oxide nanofibers with high porosity,good flexibility,and enriched oxygen defects.Benefiting from the core-sheath structure and dual-oxide component,the CeO_(2)/SiO_(2)nanofibers could stabilize 2.59 nm-Pt clusters against sintering at 600℃.Once assembled into a 2D mat,the nanofibers could efficiently decrease the soot oxidation temperature by 63℃.Moreover,the core-sheath CeO_(2)/SiO_(2)nanofibers can be readily integrated with graphene nanosheets into a 3D sponge via a gas foaming protocol,showing 218.5 mg/g of adsorption capacity toward Rhodamine B molecules.This work shed lights on the versatile applications of oxide nanofibers toward clean energy ultili-zation and low-carbon development.Yunpeng Wang Zhihui Li Wanlin Fu Yueming Sun Yunqian Dai 2022Advanced Fiber Materials2022,4,5:0
