1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 2The directional migration and differentiation of mesenchymal stem cells toward vascular endothelial cells stimulated by biphasic calcium phosphate ceramic显示文摘Osteoinductivity of porous calcium phosphate(CaP)ceramics has been widely investigated and confirmed,and it might be attributed to the rapid formation of the vascular networks after in vivo implantation of the ceramics.In this study,to explore the vascularization mechanism within the CaP ceramics,the migration and differentiation of bone marrow-derived mesenchymal stem cells(BMSCs)under the stimulation of porous biphasic calcium phosphate(BCP)ceramic with excellent osteoinductivity were systematically investigated.The results indicated that the directional migration of BMSCs toward BCP ceramic occurred when evaluated by using a transwell model,and the BMSCs migration was enhanced by the seeded macrophages on the ceramic in advance.Besides,by directly culturing BMSCs on BCP ceramic discs under both in vitro and in vivo physiological environment,it was found that the differentiation of BMSCs toward vascular endothelial cells(VECs)happened under the stimulation of BCP ceramic,as was confirmed by the up-regulated gene expressions and protein secretions of VECs-related characteristic factors,including kinase insert domain receptor,von willebrand factor,vascular cell adhesion molecule-1 and cadherin 5 in the BMSCs.This study offered a possibility for explaining the origin of VECs during the rapid vascularization process after in vivo implantation of porous CaP ceramics and could give some useful guidance to reveal the vascularization mechanism of the ceramics.Ying Chen Jing Wang Xiangdong Zhu Xuening Chen Xiao Yang Kai Zhang Yujiang Fan Xingdong Zhang 2018Regenerative Biomaterials2018,5,3:8

4The effect of slight to minor biodegradation on C_6 to C_7 light hydrocarbons in crude oils: a case study from Dawanqi Oilfield in the Tarim Basin,NW China显示文摘Light hydrocarbons(LHs) are one of the main petroleum fractions in crude oils, and carry much information regarding the genetic origin and alteration of crude oils. But secondary alterations—especially biodegradation—have a significant effect on the composition of LHs in crude oils. Because most of the LHs affected in oils underwent only slight biodegradation(rank 1 on the biodegradation scale), the variation of LHs can be used to describe more the refined features of biodegradation. Here,23 crude oils from the Dawanqi Oilfield in the Tarim Basin, NW China, eleven of which have been biodegraded to different extents, were analyzed in order to investigate the effect of slight to minor biodegradation on C_6–C_7 LHs.The study results showed that biodegradation resulted in the prior depletion of straight-chained alkanes, followed by branched alkanes. In slight and minor biodegraded oils,such biodegradation scale could not sufficiently affect C_6–C_7cycloalkanes. For branched C_6–C_7 alkanes, generally,monomethylalkanes are biodegraded earlier than dimethylalkanes and trimethylalkanes, which indicates that branched alkanes are more resistant to biodegradation, with the increase of substituted methyl groups on parent rings.The degree of alkylation is one of the primary controlling factors on the biodegradation of C_6–C_7 LHs. There is a particular case: although 2,2,3-trimethylbutane has a relative higher alkylation degree, 2,2-dimethylpentane is more resistant to biodegradation than 2,2,3-trimethylbutane. 2,2-Dimethylpentane is the most resistant to biodegradation in branched C_6–C_7 alkanes. Furthermore, the 2-methylpentane/3-methylpentane and 2-methylhexane/3-methylhexane ratios decreased steadily with increasing biodegradation,which implies that isomers of bilateral methyl groups are more prone to bacterial attack relative to mid-chain isomers. The position of the alkyls on the carbon skeleton is also one of the critical factors controlling the rate of biodegradation. With increasing biodegradation, Mango's LH parameters K1 values decrease and K2 values increase,the values of n-heptane and isoheptane decrease, and the indices of methylcyclohexane and cyclohexane increase.LH parameters should be applied cautiously for the biodegraded oils. Because biodegraded samples belong to slight or minor biodegraded oils, the values of n-heptane and isoheptane from Dawanqi Oilfield can better reflect and determine the ‘‘Biodegraded'' zone. 5TAA family contributes to auxin production during de novo regeneration of adventitious roots from Arabidopsis leaf explants显示文摘Many differentiated plant organs have the ability to regenerate into a new plant after detachment via de novo organogenesis. During de novo root organogenesis from Arabidopsis thaliana leaf explants, wounding first induces endogenous auxin production in mesophyll cells. Auxin is then polar transported to, and accumulates in, regenerationcompetent cells near the wound to trigger the cell-fate transition. The TRYPTOPHAN AMINOTRANSFERASE OF ARABIDOPSIS(TAA) family proteins and the YUCCA(YUC) family proteins catalyze two successive biochemical steps in auxin biogenesis, and YUCs have been shown to be involved in auxin production in mesophyll cells during de novo root organogenesis. In thisstudy, we show that the TAA family is also required for adventitious rooting. Inhibition of TAA blocked adventitious root formation from leaf explants. Intriguingly,whereas YUC1 and YUC4 have been shown to be highly induced by wounding, TAA genes retained consistent expression levels before and after leaf detachment.Therefore, we suggest that TAAs and YUCs are both required for auxin biogenesis in leaf explants, but they play different roles in regeneration. 6Effects of modified kaolin on the crystallization property of PP/Kaolin nanocomposites显示文摘Modified kaolin-filled polypropylene was prepared by blending different kinds of modified kaolin intercalated master batches with polypropylene.The microstructure of PP/Kaolin was studied with X-ray diffraction (XRD) patterns and transmission electron microscopy (TEM) photos.The effects of the organic kaolin master batches on the crystallization property of polypropylene were investigated by means of non-isothermal differential scanning calorimetry analysis (DSC) and polarized light microscopy (PLM).Non-isothermal crystallization kinetics of polypropylene was studied by the use of modified Avrami Equation improved by Jeziorny.The results indicate that some organic kaolin can be exfoliated by polypropylene,proved by an in-visible 001 peak on XRD pattern and individual layer on transmission electron microscopy (TEM) photos,and the heterogeneous nucleus increased with the addition of modified kaolin,proved by the PLM photos.The filled polypropylene crystallizes fastest at 395 K.The results also show that organic kaolin master batches not only can effectively accelerate the heterogeneous nucleation of polypropylene but also can increase the rate of crystallizing point and the temperature of crystallization.However,the rate constant of crystallization remains unchanged.LIU Xuening HU Nan ZHANG Hongtao HA Chengyong YANG Zhizhong 2005Science China Chemistry2005,48,4:3

10Micropascal-sensitivity ultrasound sensors based on optical microcavities显示文摘Whispering gallery mode(WGM)microcavities have been widely used for high-sensitivity ultrasound detection,owing to their optical and mechanical dual-resonance enhanced sensitivity.The ultrasound sensitivity of the cavity optomechanical system is fundamentally limited by thermal noise.In this work,we theoretically and experimentally investigate the thermal-noise-limited sensitivity of a WGM microdisk ultrasound sensor and optimize the sensitivity by varying the radius and a thickness of the microdisk,as well as using a trench structure around the disk.Utilizing a microdisk with a radius of 300μm and thickness of 2μm,we achieve a peak sensitivity of 1.18μPa Hz^(-1/2)at 82.6 k Hz.To the best of our knowledge,this represents the record sensitivity among cavity optomechanical ultrasound sensors.Such high sensitivity has the potential to improve the detection range of air-coupled ultrasound sensing technology.HAO YANG XUENING CAO ZHI-GANG HU YIMENG GAO YUECHEN LEI MIN WANG ZHANCHUN ZUO XIULAI XU BEI-BEI LI 2023Photonics Research2023,11,7:0

12SARS-CoV-2-specific antibody response characteristics in COVID-19 patients of different ages显示文摘Age has been found to be one of the main risk factors for the severity and outcome of COVID-19.However,differences in SARS-CoV-2 specific antibody responses among COVID-19 patients of different age groups remain largely unknown.In this study,we analyzed the IgG/IgM responses to 21 SARS-CoV-2 proteins and 197 peptides that fully cover the spike protein against 731 sera collected from 731 COVID-19 patients aged from 1 to 92 years.We show that there is no overall difference in SARS-CoV-2 antibody responses in COVID-19 patients in the 4 age groups.By antibody response landscape maps,we find that the IgG response profiles of SARS-CoV-2 proteins are positively correlated with age.The S protein linear epitope map shows that the immunogenicity of the S-protein peptides is related to peptide sequence,disease severity and age of the COVID-19 patients.Furthermore,the enrichment analysis indicates that low S1 IgG responses are enriched in patients aged<50 and high S1 IgG responses are enriched in mild COVID-19 patients aged>60.In addition,high responses of non-structural/accessory proteins are enriched in severe COVID-19 patients aged>70.These results suggest the distinct immune response of IgG/IgM to each SARS-CoV-2 protein in patients of different age,which may facilitate a deeper understanding of the immune responses in COVID-19 patients.Linfang Lu Siqi Yu Min Liu Yang Li Qing Lei Mingxi Lin Danyun Lai Shujuan Guo Hewei Jiang Hongyan Hou Yunxiao Zheng Xuening Wang Mingliang Ma Bo Zhang Hong Chen Junbiao Xue Hainan Zhang Huan Qi Ziyong Sun Feng Wang Xionglin Fan Zhaowei Xu 2022Acta Biochimica et Biophysica Sinica2022,54,4:0
13Machine learning on properties of multiscale multisource hydroxyapatite nanoparticles datasets with different morphologies and sizes显示文摘Machine learning models for exploring structure-property relation for hydroxyapatite nanoparticles(HANPs)are still lacking.A multiscale multisource dataset is presented,including both experimental data(TEM/SEM,XRD/crystallinity,ROS,anti-tumor effects,and zeta potential)and computation results(containing 41,976 data samples with up to 9768 atoms)of nanoparticles with different sizes and morphologies at density functional theory(DFT),semi-empirical DFTB,and force field,respectively.Three geometric descriptors are set for the explainable machine learning methods to predict surface energies and surface stress of HANPs with satisfactory performance.To avoid the pre-determination of features,we also developed a predictive deep learning model within the framework of graph convolution neural network with good generalizability.Energies with DFT accuracy are achievable for largesized nanoparticles from the learned correlations and scale functions for mapping different theoretical levels and particle sizes.The simulated XRD spectra and crystallinity values are in good agreement with experiments.Ziteng Liu Yinghuan Shi Hongwei Chen Tiexin Qin Xuejie Zhou Jun Huo Hao Dong Xiao Yang Xiangdong Zhu Xuening Chen Li Zhang Mingli Yang Yang Gao Jing Ma 2021npj Computational Materials2021,,1:0
