1TRAP1 Shows Clinical Significance and Promotes Cellular Migration and Invasion through STAT3/MMP2 Pathway in Human Esophageal Squamous Cell Cancer显示文摘Tumor necrosis factor receptor-associated protein 1(TRAP1), an important member of mitochondrial heat shock protein 90 family, is involved in multiple biological processes in several types of tumors. However, its pathological role in esophageal squamous cell cancer(ESCC) remains unknown. Herein, we demonstrated the clinical value of TRAP1, and its role in apoptosis and motility in ESCC. The clinical potential of TRAP1 was investigated through immunohistochemical analysis in 328 ESCC samples, which revealed that strong TRAP1 expression was associated with increased risk of lymph node metastasis, while high TRAP1 expression correlated with poor prognosis. Expression of TRAP1 was found to be an independent prognostic factor for patients with ESCC. Additionally, the upregulation of TRAP1 antagonized cisplatin-induced apoptosis while its downregulation sensitized cells to cisplatin-induced apoptosis. As revealed by the transwell assay, TRAP1 overexpression promoted cellular migration and invasion as compared to the control groups. In contrast,silencing of endogenous TRAP1 expression attenuated the ability of migration and invasion. Finally, the molecular mechanism investigated in the present study demonstrated that TRAP1-mediated migration and invasion occurred through STAT3/MMP2 signaling pathway. 2Suppressor of RNA silencing encoded by Rice gall dwarf virus genome segment 11显示文摘Rice gall dwarf virus(RGDV)is an important rice pathogen in China and Southeast Asia.However,little is known about the molecular mechanisms of RGDV interactions with plant cells.Here,we have identi-fied an RGDV protein,Pns11,which acts as a suppressor of RNA silencing in coinfiltration assays with the reporter,green fluorescent protein(GFP)in transgenic Nicotiana benthamiana line 16c carrying GFP.Pns11 suppressed local and systemic silencing induced by sense RNA.The spread of mobile RNA si-lencing signals was blocked or inactivated by Pns11.Expression of Pns11 also enhanced Potato virus X pathogenicity in N.benthamiana.This suppressor could reduce,but not eliminate,siRNA in the local and systemic RNA silencing suppression assays,suggesting that Pns11 functions by interfering with initial stages of RNA silencing.LIU FuXiu ZHAO Qin RUAN XiaoLei HE YunWei LI HuaPing 2008Chinese Science Bulletin2008,53,3:8
3Migfilin promotes migration and invasion in glioma by driving EGFR and MMP-2 signalings:A positive feedback loop regulation显示文摘Glioma is the most common type of primary brain tumors in the central nervous system(CNS). Migfilin occurs in human glioma and enhances cellular motility via the epidermal growth factor receptor(EGFR)pathway. However, the underlying molecular mechanism is not fully understood. In this study, we found that Migfilin promoted matrix metalloproteinase-2(MMP-2) activity, and restrained the expression of tissue inhibitor of metalloproteinase 2(TIMP2), which is an MMP-2 inhibitor. Functional and structural studies showed that the LIM1 domain of Migfilin was required for Migfilin-mediated TIMP2 expression inhibition and MMP-2 activity, and was also necessary in promoting cell motility. Furthermore, Migfilininduced EGFR phosphorylation was greatly reduced by MMP-2 inhibitor(GM6001) or si RNA, while Migfilin-induced MMP-2 activation was also blocked by the EGFR inhibitor(AG1478) or si RNA. MMP-2 and EGFR inhibitors and their si RNAs can block Migfilin-induced migration and invasion, respectively.These results demonstrated that EGFR and MMP-2 signalings may form a positive feedback loop to enhance Migfilin-induced migration and invasion. Finally, we detected that the expression of Migfilin,EGFR phosphorylation(Tyr1173) and MMP-2 activity had a positive correlation in the clinical glioma sample. 6Strength-Toughening Model of Eutectic Ceramic Composite with Inherent Defects显示文摘Strengthening and toughening mechanisms in composite ceramics is complex. A change in a single parameter induces multiple property variations. The multiple changes in properties are often incompletely represented in theoretical models. This incompleteness in the parameter chosen fails to explain the mechanism of failure in composite ceramics. The exponential toughness function is used to represent the pull-out toughening mechanism, which dominates the crack growth resistance curve(R-curve). The strengthening-toughening model is established based on the Mori-Tanaka method(M-T method). The influence of inherent defects on toughness function and strength is analyzed by using this model. The theoretical result is compared with the experiment data. This model exactly reflects the change in strength. The theoretical result indicates that defects change the toughness function. Moreover, micro-cracks increase toughness size ac, and the strength of crack instable extensions acutely decreases as defect content increases. 7Damage and fracture model for eutectic composite ceramics显示文摘Study of damage and fracture models to analyze the fracture mechanism of eutectic composite ceramics is of considerable importance because no accurate fracture models are available for these materials. Eutectic composite ceramics are composed of microcells with random direction. We present herein a model that predicts the damage and fracture of eutectic composite ceramics based on analysis of defect stability and the damage localization band. Firstly, given the microstructure of eutectic composite ceramics, a mesodomain and a microcell model are constructed. The local stress field in the mesodomain is then analyzed based on the interaction direct derivative estimate. Secondly, the stability of a defect around particles in a microcell is analyzed, and the stress intensity factor of an annular defect under the applied stress field and the residual stress field in the particle are calculated. The stress intensity factor of a defect is controlled by the residual stress when the defect extension is small. However, it is controlled by the applied stress when the defect extension is large. Finally, a model for the damage localization band at the crack tip is constructed based on the Dugdale-Barenblatt model. The residual intensity is the important factor affecting the length of the damage localization band. When the damage variables reach their largest value, the residual in ten sity and the length of the damage localization band attain their minimum value. 15Influence of two inlets of the Luzon overflow on the deep circulation in the northern South China Sea显示文摘An inverse reduced-gravity model is used to simulate the deep South China Sea(SCS)circulation.A set of experiments are conducted using this model to study the influence of the Luzon overflow through the two inlets on the deep circulation in the northern SCS.Model results suggest that the relative contribution of these inlets largely depends on the magnitude of the input transport of the overflow,but the northern inlet is more efficient than the southern inlet in driving the deep circulation in the northern SCS.When all of the Luzon overflow occurs through the northern inlet the deep circulation in the northern SCS is enhanced.Conversely,when all of the Luzon overflow occurs through the southern inlet the circulation in the northern SCS is weakened.A Lagrangian trajectory model is also developed and applied to these cases.The Lagrangian results indicate that the location of the Luzon overflow likely has impacts upon the sediment transport into the northern SCS.Muping Zhou Changlin Chen Yunwei Yan Wenhu Liu 2020Acta Oceanologica Sinica2020,39,11:1

18Hepatitis B virus X protein induces ALDH2 ubiquitin-dependent degradation to enhance alcoholic steatohepatitis显示文摘Background:Excessive alcohol intake with hepatitis B virus(HBV)infection accelerates chronic liver disease progression and patients with HBV infection are more susceptible to alcohol-induced liver disease.Hepatitis B virus X protein(HBx)plays a crucial role in disease pathogenesis,while its specific role in alcoholic liver disease(ALD)progression has not yet been elucidated.Here,we studied the role of HBx on the development of ALD.Methods:HBx-transgenic(HBx-Tg)mice and their wild-type littermates were exposed to chronic plus binge alcohol feeding.Primary hepatocytes,cell lines,and human samples were used to investigate the interaction between HBx and acetaldehyde dehydrogenase 2(ALDH2).Lipid profiles in mouse livers and cells were assessed by using liquid chromatography–mass spectrometry.Results:We identified that HBx significantly aggravated alcohol-induced steatohepatitis,oxidative stress,and lipid peroxidation in mice.In addition,HBx induced worse lipid profiles with high lysophospholipids generation in alcoholic steatohepatitis,as shown by using lipidomic analysis.Importantly,serum and liver acetaldehyde were markedly higher in alcoholfed HBx-Tg mice.Acetaldehyde induced lysophospholipids generation through oxidative stress in hepatocytes.Mechanistically,HBx directly bound to mitochondrial ALDH2 to induce its ubiquitin–proteasome degradation,resulting in acetaldehyde accumulation.More importantly,we also identified that patients with HBV infection reduced ALDH2 protein levels in the liver.Conclusions:Our study demonstrated that HBx-induced ubiquitin-dependent degradation of mitochondrial ALDH2 aggravates alcoholic steatohepatitis.Haoxiong Zhou Sizhe Wan Yujun Luo Huiling Liu Jie Jiang Yunwei Guo Jia Xiao Bin Wu 2023Gastroenterology Report2023,11,1:0

20Research Trends and Emerging Hotspots of Lung Cancer Surgery during 2012-2021:A 10-Year Bibliometric and Network Analysis显示文摘Background. Lung cancer remains the leading cause of death because of cancer globally in the past years. To inspire researcherswith new targets and path-breaking directions for lung cancer research, this study is aimed at exploring the research trends andemerging hotspots in the lung cancer surgery literature in the recent decade. Methods. This cross-sectional study combinedbibliometric and network analysis techniques to undertake a quantitative analysis of lung cancer surgery literature. Dimensionsdatabase was searched using keywords in a 10-year period (2012-2021). Publications were characterized by publication year,research countries, field citation ratio, cooperation status, research area, and emerging hotspots. Results. Overall, globalscholarly outputs of lung cancer surgery had almost doubled during the recent decade, with China, Japan, and the UnitedStates leading the way, while Denmark and Belgium predominated in terms of scientific influence. Network analysis showedthat international cooperation accounted for a relatively small portion in lung cancer surgery research, and the United States,China, and Europe were the prominent centers of international cooperation network. In the recent decade, research of lungcancer surgery majored in prevention, biomedical imaging, rehabilitation, and genetics, and the emerging research hotspotstransformed into immunotherapy. Research on immunotherapy showed a considerable increase in scientific influence in thelatest year. Conclusions.
