1Fucoidan protects hepatocytes from apoptosis and inhibits invasion of hepatocellular carcinoma by up-regulating p42/44 MAPK-dependent NDRG-1/CAP43显示文摘Fucoidan is a traditional Chinese medicine suggested to possess anti-tumor effects.In this study the anti-metastatic effects of fucoidan were investigated in vitro in human hepatocellular carcinoma(HCC) cells(Huh-7 and SNU-761) under normoxic and hypoxic conditions and in vivo using a distant liver metastasis model involving injection of MH134 cells into spleen via the portal vein.Its ability to protect hepatocytes against bile acid(BA)-induced apoptosis was investigated in primary hepatocytes.Fucoidan was found to suppress the invasion of HCC cells through up-regulation of p42/44 MAPKdependent NDRG-1/CAP43 and partly,under normoxic conditions,through up-regulation of p42/44MAPK-dependent VMP-1 expression.It also significantly decreased liver metastasis in vivo.As regards its hepatoprotective effect,fucoidan decreased BA-induced hepatocyte apoptosis as shown by the attenuation of caspase-8,and-7 cleavages and suppression of the mobilization of caspase-8 and Fas associated death domain(FADD) into the death-inducing signaling complex.In summary,fucoidandisplays inhibitory effects on proliferation of HCC cells and protective effects on hepatocytes.The results suggest fucoidan is a potent suppressor of tumor invasion with hepatoprotective effects.Yuri Cho Jung-Hwan Yoon Jeong-ju Yoo Minjong Lee Dong Hyeon Lee Eun Ju Cho Jeong-Hoon Lee Su Jong Yu Yoon Jun Kim Chung Yong Kim 2015Acta Pharmaceutica Sinica B2015,5,6:3

5Reactive oxygen species-induced activation of Yes-associated protein-1 through the c-Myc pathway is a therapeutic target in hepatocellular carcinoma显示文摘BACKGROUND The Hippo signaling pathway regulates organ size by regulating cell proliferation and apoptosis with terminal effectors including Yes-associated protein-1(YAP-1).Dysregulation in Hippo pathway has been proposed as one of the therapeutic targets in hepatocarcinogenesis.The levels of reactive oxygen species(ROS)increase during the progression from early to advanced hepatocellular carcinoma(HCC).AIM To study the activation of YAP-1 by ROS-induced damage in HCC and the involved signaling pathway.METHODS The expression of YAP-1 in HCC cells(Huh-7,HepG2,and SNU-761)was quantified using real-time polymerase chain reaction and immunoblotting.Human HCC cells were treated with H2O2,which is a major component of ROS in living organisms,and with either YAP-1 small interfering RNA(siRNA)or control siRNA.To investigate the role of YAP-1 in HCC cells under oxidative stress,MTS assays were performed.Immunoblotting was performed to evaluate the signaling pathway responsible for the activation of YAP-1.Eighty-eight surgically resected frozen HCC tissue samples and 88 nontumor liver tissue samples were used for gene expression analyses.RESULTS H2O2 treatment increased the mRNA and protein expression of YAP-1 in HCC cells(Huh-7,HepG2,and SNU-761).Suppression of YAP-1 using siRNA transfection resulted in a significant decrease in tumor proliferation during H2O2 treatment both in vitro and in vivo(both P<0.05).The oncogenic action of YAP-1 occurred via the activation of the c-Myc pathway,leading to the upregulation of components of the unfolded protein response(UPR),including 78-kDa glucoseregulated protein and activating transcription factor-6(ATF-6).The YAP-1 mRNA levels in human HCC tissues were upregulated by 2.6-fold compared with those in nontumor tissues(P<0.05)and were positively correlated with the ATF-6 Levels(Pearson’s coefficient=0.299;P<0.05).CONCLUSION This study shows a novel connection between YAP-1 and the UPR through the c-Myc pathway during oxidative stress in HCC.The ROS-induced activation of YAP-1 via the c-Myc pathway,which leads to the activation of the UPR pathway,might be a therapeutic target in HCC.Yuri Cho Min Ji Park Koeun Kim Sun Woong Kim Wonjin Kim Sooyeon Oh Joo Ho Lee 2020World Journal of Gastroenterology2020,26,42:1

8cell2mol:encoding chemistry to interpret crystallographic data显示文摘The creation and maintenance of crystallographic data repositories is one of the greatest data-related achievements in chemistry.Platforms such as the Cambridge Structural Database host what is likely the most diverse collection of synthesizable molecules.If properly mined,they could be the basis for the large-scale exploration of new regions of the chemical space using quantum chemistry(QC).Yet,it is currently challenging to retrieve all the necessary information for QC codes based exclusively on the available structural data,especially for transition metal complexes.To overcome this limitation,we present cell2mol,a software that interprets crystallographic data and retrieves the connectivity and total charge of molecules,including the oxidation state(OS)of metal atoms.We demonstrate that cell2mol outperforms other popular methods at assigning the metal OS,while offering a comprehensive interpretation of the unit cell.The code is made available,as well as reliable QC-ready databases totaling 31k transition metal complexes and 13k ligands that contain incomparable chemical diversity.Sergi Vela Ruben Laplaza Yuri Cho Clémence Corminboeuf 2022npj Computational Materials2022,,1:0
9Differential risk of 23 site-specific incident cancers and cancer-related mortality among patients withmetabolic dysfunction-associated fatty liver disease:a population-based cohort study with 9.7 million Korean subjects显示文摘Introduction:Although an association between metabolic dysfunctionassociated fatty liver disease(MAFLD)and cardiovascular disease or overall mortality has been reported,it is unclearwhether there is an association between MAFLD and cancer incidence or mortality.We aimed to investigate the differential risk of all-and site-specific cancer incidence and mortality according to MAFLD subgroups categorized by additional etiologies of liver disease.Methods:Using the Korean National Health Insurance Service database,we stratified the participants into three groups:(1)single-etiology MAFLD(SMAFLD)or MAFLD of pure metabolic origin;(2)mixed-etiology MAFLD(M-MAFLD)or MAFLD with additional etiological factor(s)(i.e.,concomitant liver diseases and/or heavy alcohol consumption);and(3)non-MAFLD.Hepatic steatosis and fibrosiswere defined using the fatty liver index and the BARDscore,respectively.Cox proportional hazards regression was performed to estimate the risk of cancer events.Results:Among the 9,718,182 participants,the prevalence of S-MAFLD and M-MAFLD was 29.2%and 6.7%,respectively.During the median 8.3 years of follow-up,510,330(5.3%)individuals were newly diagnosed with cancer,and 122,774(1.3%)cancer-related deaths occurred among the entire cohort.Compared with the non-MAFLD group,the risk of all-cancer incidence and mortality was slightly higher among patients in the S-MAFLD group(incidence,adjusted hazard ratio[aHR]=1.03;95%confidence interval[CI]:1.02−1.04;mortality,aHR=1.06;95%CI:1.04−1.08)and highest among patients with M-MAFLD group(incidence,aHR=1.31;95%CI:1.29−1.32;mortality,aHR=1.45;95%CI:1.42−1.48,respectively).The M-MAFLD with fibrosis group(BARD score≥2)showed the highest relative risk of all-cancer incidence(aHR=1.38,95%CI=1.36–1.39),followed by the M-MAFLD without fibrosis group(aHR=1.09,95%CI=1.06–1.11).Similar trends were observed for cancer-related mortality.Conclusions:MAFLD classification,by applying additional etiologies other than pure metabolic origin,can be used to identify a subgroup of patients with poor cancer-related outcomes.Goh Eun Chung Su Jong Yu Jeong-Ju Yoo Yuri Cho Kyu-na Lee Dong Wook Shin Yoon Jun Kim Jung-Hwan Yoon Kyungdo Han Eun Ju Cho 2023Cancer Communications2023,43,8:0
10Submergence promotes auxin-induced callus formation through ethylene-mediated post-transcriptional control of auxin receptors显示文摘Plant cells in damaged tissue can be reprogrammed to acquire pluripotency and induce callus formation.However,in the aboveground organs of many species,somatic cells that are distal to the wound site become less sensitive to auxin-induced callus formation,suggesting the existence of repressive regulatory mechanisms that are largely unknown.Here we reveal that submergence-induced ethylene signals promote callus formation by releasing post-transcriptional silencing of auxin receptor transcripts in non-wounded regions.We determined that short-term submergence of intact seedlings induces auxin-mediated cell dedifferentiation across the entirety of Arabidopsis thaliana explants.The constitutive triple response 1-1(ctr1-1)mutation induced callus formation in explants without submergence,suggesting that ethylene facilitates cell dedifferentiation.We show that ETHYLENE-INSENSITIVE 2(EIN2)post-transcriptionally regulates the abundance of transcripts for auxin receptor genes by facilitating microRNA393 degradation.Submergence-induced calli in non-wounded regions were suitable for shoot regeneration,similar to those near the wound site.We also observed submergence-promoted callus formation in Chinese cabbage(Brassica rapa),indicating that this may be a conserved mechanism in other species.Our study identifies previously unknown regulatory mechanisms by which ethylene promotes cell dedifferentiation and provides a new approach for boosting callus induction efficiency in shoot explants.Seung Yong Shin Yuri Choi Sang-Gyu Kim Su-Jin Park Ji-Sun Park Ki-Beom Moon Hyun-Soon Kim Jae Heung Jeon Hye Sun Cho Hyo-Jun Lee 2022Molecular Plant2022,15,12:0
