2Cloning of a gene-edited macaque monkey by somatic cell nuclear transfer显示文摘Cloning of macaque monkeys by somatic cell nucleus transfer(SCNT) allows the generation of monkeys with uniform genetic backgrounds that are useful for the development of non-human primate models of human diseases. Here, we report the feasibility of this approach by SCNT of fibroblasts from a macaque monkey(Macaca fascicularis), in which a core circadian transcription factor BMAL1 was knocked out by clustered regularly interspaced short palindromic repeat/Cas9 gene editing(see accompanying paper).Out of 325 SCNT embryos transferred into 65 surrogate monkeys, we cloned five macaque monkeys with BMAL1 mutations in both alleles without mosaicism, with nuclear genes identical to that of the fibroblast donor monkey. Further peripheral blood m RNA analysis confirmed the complete absence of the wild-type BMAL1 transcript. 3Sandwiching Sulfur into the Dents Between N,O Co-Doped Graphene Layered Blocks with Strong Physicochemical Confinements for Stable and High-Rate Li-S Batteries显示文摘The development of lithium-sulfur batteries(LSBs)is restricted by their poor cycle stability and rate performance due to the low conductivity of sulfur and severe shuttle effect.Herein,an N,O co-doped graphene layered block(NOGB)with many dents on the graphene sheets is designed as effective sulfur host for high-performance LSB s.The sulfur platelets are physically confined into the dents and closely contacted with the graphene scaffold,ensuring structural stability and high conductivity.The highly doped N and O atoms can prevent the shuttle effect of sulfur species by strong chemical adsorption.Moreover,the micropores on the graphene sheets enable fast Li^+transport through the blocks.As a result,the obtained NOGB/S composite with 76 wt%sulfur content shows a high capacity of 1413 mAh g^-1 at 0.1 C,good rate performance of 433 mAh g^-1 at 10 C,and remarkable stability with 526 mAh g^-1 at after 1000 cycles at 1 C(average decay rate:0.038%per cycle).Our design provides a comprehensive route for simultaneously improving the conductivity,ion transport kinetics,and preventing the shuttle effect in LSBs.Mengjiao Shi Su Zhang Yuting Jiang Zimu Jiang Longhai Zhang Jin Chang Tong Wei Zhuangjun Fan 2020Nano-Micro Letters2020,12,11:3

5Quadrupole-Linear Ion Trap Tandem Mass Spectrometry System for Clinical Biomarker Analysis显示文摘The accurate and efficient measurement of small molecule disease markers for clinical diagnosis is of great importance.In this study,a quadrupole-linear ion trap(Q-LIT)tandem mass spectrometer was designed and built in our laboratory.Target precursor ions were first selected in the quadrupole,and then injected,trapped,and fragmented simultaneously in the linear ion trap(LIT)to reduce the space charge effect,enrich the target product ions,and promote sensitivity.The targeted analytes were measured with selected reaction monitoring using a positive scan mode with electrospray ionization(ESI).Ions with a mass-to-charge ratio(m/z)ranging from 195 to 2022 were demonstrated.When scanning at 1218amu.s^(-1),unit resolution and an accuracy of higher than m/z 0.28 was obtained for m/z up to 2000.The dimensionless Mathieu parameter(q)value used in this study was 0.40 for collision-induced dissociation(CID),which was activated by resonance excitation.And an overall CID efficiency of 64%was achieved(activation time,50 ms).Guanidinoacetic acid(GAA)and creatine(CRE)were used as model compounds for small molecule clinical biomarkers.The limits of quantification were 1.0 and 0.2 nmol.L^(-1)for GAA and CRE,respectively.A total of 77 actual samples were successfully analyzed by the home-built ESI-Q-LIT tandem mass spectrometry system.The developed method can reduce matrix interference,minimize space charge effects,and avoid the chromatographic separation of complex samples to simplify the pretreatment process.This novel Q-LIT system is expected to be a good candidate for the determination of biomarkers in clinical diagnosis and therapeutics.Xiang Fang Jie Xie Shiying Chu You Jiang Yuting An Chang Li Xiaoyun Gong Rui Zhai Zejian Huang Chunling Qiu Xinhua Dai 2022Engineering2022,,9:1
6An updating of landslide susceptibility prediction from the perspective of space and time显示文摘Due to the similarity of conditioning factors,the aggregation feature of landslides and the multi-temporal landslide inventory,the spatial and temporal effects of landslides need to be considered in landslide susceptibility prediction(LSP).The ignorance of this issue will result in some biases and time-invariance in landslide susceptibility.Hence,a novel framework has been proposed to update landslide susceptibility by simultaneously considering the spatial and temporal effects of landslides at the regional scale.In this framework,the landslide inventory of Chongyi County has been divided into pre-and fresh-landslide inventories.According to the LSP results predicted by the support vector machine(SVM)model using the slope unit-based conditioning factors and pre-landslide inventory,a normalized spatial distance index(NSDI)is calculated to quantitatively represent the spatial correlation between landslides and surrounding slope units to develop the SVM-NSDI model.Furthermore,the SVM-Updating model,which incorporates the LSP results of the SVM-NSDI model and fresh-landslide inventory,could be developed to update the LSP results.Subsequently,the confusion matrix,the area under the receiver operating characteristic curve(AUC)and frequency ratio(FR)accuracy are used to evaluate the prediction performance of the above LSP models.The F1-score values of the SVM,SVM-NSDI and SVM-Updating models are 0.776,0.816 and 0.831,respectively.The AUC values are 0.869,0.903 and 0.914 and the FR accuracies are 0.795,0.853 and 0.873.It can be concluded that landslide susceptibility is a time-variant variable,which can be updated by considering the spatial correlation between landslides and surrounding slope units as well as the temporal effects of multi-temporal landslide inventory.This study provides a new framework to update landslide susceptibility over time and also provides more accurate LSP results for decisionmakers.Zhilu Chang Faming Huang Jinsong Huang Shui-Hua Jiang Yuting Liu Sansar Raj Meena Filippo Catani 2023Geoscience Frontiers2023,14,5:1

10Two pseudo-polymorphic porous POM-pillared MOFs for sulfide-sulfoxide transformation: Efficient synergistic effects of POM precursors, metal sites and microstructures显示文摘Developing sustainable and powerful heterogeneous catalytic systems to convert sulfides into high-value sulfoxide products has become a particularly appealing field and an arduous challenge.In this work,two porous polyoxometalate-pillared metal-organic frameworks,formulated as H_(3n)[Cu_(3)(pidc)_(2)(H_(2)O)_(2.5)]_(2)[PW_(12)O_(40)]_n·x H_(2)O (n=1.5,x=6 for 1,n=1,x=12 for 2;and H_(3)pidc=2-(3-pyridinyl)-1H-imidazole-4,5-dicarboxylic acid),were consciously manufacture and employed for heterogeneously catalyzed sulfide-sulfoxide transformation.Structural analysis shows that 1 and 2 exhibit similar porous frameworks with nearly identical two-dimensional metal-organic layers further pillared by tetradentate POM ligands with different coordination modes,which also result in the porosity of 1 being almost twice that of 2.In catalyzing the conversion of methyl phenyl sulfide (MPS) to methyl phenyl sulfoxide (MPSO),1 can convert nearly 100%of MPS into MPSO within 30 min,while 2 achieved the similar results requires 50 min.The higher activity of 1 may be attributed to its larger channel that can provide more active sites and more efficient mass transfer process.Systematic structure-activity analyses and mechanistic studies revealed dual-reaction pathways driven by POM sites and metal sites assisted by the structural microenvironment.Yanhong Chen Haiyan An Shenzhen Chang Yanqin Li Tieqi Xu Qingshan Zhu Huiyun Luo Yaohui Huang Yuting Wei 2023Chinese Chemical Letters2023,34,7:0
11Differential bone metabolism and protein expression in mice fed a high-fat diet versus Daurian ground squirrels following natural pre-hibernation fattening显示文摘This study compared the effects on bone metabolism and morphology of pathological obesity induced by excessive fat intake in a non-hibernator(mice)versus healthy obesity due to pre-hibernation fattening in a hibernator(ground squirrels).Kunming mice were fed a high-fat diet to provide a model of pathological obesity(OB group).Daurian ground squirrels fattened naturally in their pre-hibernation season(PRE group)were used as a healthy obesity model.Micro-computed tomography(micro-CT)and three-point bending tests were used to determine the microstructure and mechanical properties of bone.Western blots were used to analyze protein expression levels related to bone metabolism(Runt-related transcription factor 2(RunX2),osteocalcin(OCN),alkaline phosphatase(ALP),osteoprotegerin(OPG),receptor activator of nuclear factor-κB ligand(RANKL),cathepsin K,matrix metallopeptidase 9(MMP9),patched protein homolog 1(Ptch1),phosphorylatedβ-catenin(P-β-catenin),and glycogen synthase kinase-3β(GSK-3β)).Compared with controls,there was no obvious bone loss in the OB mice,and the stiffness of the femur was increased significantly.Compared with summer active squirrels,bone formation was enhanced but the mechanical properties did not change in the PRE group squirrels.In OB mice,western blots showed significantly increased expression levels of all proteins except RunX2,OPG,and Ptch1.PRE ground squirrels showed significantly increased expression of most proteins except OCN and Ptch1,which decreased significantly,and P-β-catenin and OPG,which did not change.In conclusion,for non-hibernating mice,moderate obesity had a certain protective effect on bones,demonstrating two-way regulation,increasing both bone loss and bone formation.For pre-hibernating ground squirrels,the healthy obesity acquired before hibernation had a positive effect on the microstructure of bones,and also enhanced the expression levels of proteins related to bone formation,bone resorption,and Wnt signaling.Xuli GAO Shenyang SHEN Qiaohua NIU Weilan MIAO Yuting HAN Ziwei HAO Ning AN Yingyu YANG Yu ZHANG Han ZHANG Kenneth BSTOREY Hui CHANG 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,12:0
12Therapeutic application of chick early amniotic fluid: effective rescue of acute myocardial ischemic injury by intravenous administration显示文摘Myocardial regeneration has been considered a promising option for the treatment of adult myocardial injuries.Previously,a chick early amniotic fluid(ceAF)preparation was shown to contain growth-related factors that pro-moted embryonic growth and cellular proliferation,though the nature of the components within ceAF were not fully defined.Here we tested whether this ceAF preparation is similarly effective in the promotion of myocardial regen-eration,which could provide an alternative therapeutic for intervening myocardial injury.In this study,a myocardial ischemic injury model was established in adult mice and pigs by multiple research entities,and we were able to show that ceAF can efficiently rescue damaged cardiac tissues and markedly improve cardiac function in both experimental models through intravenous administration.ceAF administration increased cell proliferation and improved angio-genesis,likely via down-regulation of Hippo-YAP signaling.Our data suggest that ceAF administration can effectively rescue ischemic heart injury,providing the key functional information for the further development of ceAF for use in attenuating myocardial injury.Baiping Cui Yufan Zheng Xiang Gao Lihong Zhang Borui Li Jia Chen Xinyan Zhou Mengyuan Cai Wenrui Sun Yuting Zhang Keejong Chang Jiayi Xu Fuyin Zhu Yan Luo Tao Sun Jin Qian Ning Sun 2022Cell Regeneration2022,11,1:0

14Polarizable Additive with Intermediate Chelation Strength for Stable Aqueous Zinc‑Ion Batteries显示文摘Aqueous zinc-ion batteries are promising due to inherent safety,low cost,low toxicity,and high volumetric capacity.However,issues of dendrites and side reactions between zinc metal anode and the electrolyte need to be solved for extended storage and cycle life.Here,we proposed that an electrolyte additive with an intermediate chelation strength of zinc ion—strong enough to exclude water molecules from the zinc metal-electrolyte interface and not too strong to cause a significant energy barrier for zinc ion dissociation—can benefit the electrochemical stability by suppressing hydrogen evolution reaction,overpotential growth,and den-drite formation.Penta-sodium diethylene-triaminepentaacetic acid salt was selected for such a purpose.It has a suitable chelating ability in aqueous solutions to adjust solvation sheath and can be readily polarized under electrical loading conditions to further improve the passivation.Zn||Zn symmetric cells can be stably operated over 3500 h at 1 mA cm^(-2).Zn||NH4V4O10 full cells with the additive show great cycling stability with 84.6%capacity retention after 500 cycles at 1 A g^(-1).Since the additive not only reduces H2 evolution and corrosion but also modifies Zn2+diffusion and deposition,highlyreversible Zn electrodes can be achieved as verified by the experimental results.Our work offers a practical approach to the logical design of reliable electrolytes for high-performance aqueous batteries.Yuting Xia Rongao Tong Jingxi Zhang Mingjie Xu Gang Shao Hailong Wang Yanhao Dong Chang‑An Wang 2024Nano-Micro Letters2024,16,5:0
