| 1 | Regional and Global Land Data Assimilation Systems: Innovations,Challenges, and Prospects显示文摘Since the North American and Global Land Data Assimilation Systems(NLDAS and GLDAS) were established in2004, significant progress has been made in development of regional and global LDASs. National, regional, projectbased, and global LDASs are widely developed across the world. This paper summarizes and overviews the development, current status, applications, challenges, and future prospects of these LDASs. We first introduce various regional and global LDASs including their development history and innovations, and then discuss the evaluation, validation, and applications(from numerical model prediction to water resources management) of these LDASs. More importantly, we document in detail some specific challenges that the LDASs are facing: quality of the in-situ observations, satellite retrievals, reanalysis data, surface meteorological forcing data, and soil and vegetation databases; land surface model physical process treatment and parameter calibration; land data assimilation difficulties; and spatial scale incompatibility problems. Finally, some prospects such as the use of land information system software, the unified global LDAS system with nesting concept and hyper-resolution, and uncertainty estimates for model structure, parameters, and forcing are discussed. | Youlong XIA Zengchao HAO Chunxiang SHI Yaohui LI Jesse MENG Tongren XU Xinying WU Baoqing ZHANG | 2019 | Journal of Meteorological Research2019,33,2: | 7 |

| 7 | Research progresses and prospects of multi-sphere compound extremes from the Earth System perspective显示文摘Compound extremes, whose socioeconomic and ecological impacts are severer than that caused by each event occurring in isolation, have evolved into a hot topic in Earth Science in the past decade. In the context of climate change, many compound extremes have exhibited increasing frequency and intensity, and shown novel fashions of combinations, posing more pressing demands and tougher challenges to scientific research and disaster prevention and response. This article, via a perspective of multi-sphere interactions within the Earth System, systematically reviews the status quo,new scientific understanding, and deficiencies regarding the definition, mechanism, change, attribution, and projection of compound extremes. This study also sorts out existing challenges and outlines a potential roadmap in advancing the study on compound extremes with respect to data requirement, mechanistic diagnosis, numerical modeling, attribution and projection, risk assessment, and adaptive response.Further directions of compound extremes studies and key research topics that warrant multi-disciplinary and multisectoral coordinated efforts are also proposed.Given that climate change has reshaped the type of extremes,a transformation from the traditional single-event perspective to a compound-event perspective is needed for scientific research, disaster prevention and mitigation, and climate change adaptation, calling for bottom-up innovation in research objects, ideas, and methods. This article will add value to promoting the research on compound extremes and interdisciplinary cooperations. | Zengchao HAO Yang CHEN | 2024 | Science China Earth Sciences2024,67,2: | 0 |
| 8 | Bio-assembled smart nanocapsules for targeted delivery of KRAS shRNA and cancer cell bioimage显示文摘The five-year survival rate for pancreatic cancer is less than 5%. However, the current clinical multimodal therapy combined with first-line chemotherapy drugs only increases the patient’s median survival from 5.0 months to 7.2 months. Consequently, a new strategy of cancer treatments is urgently needed to overcome this high-fatality disease. Through a series of biometric analyses, we found that KRAS is highly expressed in the tumor of pancreatic cancer patients, and this high expression is closely related to the poor prognosis of patients. It shows that inhibiting the expression of KRAS has great potential in gene therapy for pancreatic cancer. Given those above, we have exploited the possibility of targeted delivery of KRAS shRNA with the intelligent and bio-responsive nanomedicine to detect the special oxidative stress microenvironment of cancer cells and realize efficient cancer theranostics. Our observations demonstrate that by designing the smart self-assembled nanocapsules of melanin with fluorescent nanoclusters we can readily achieve the bio-recognition and bioimaging of cancer cells in biological solution or serum. The self-assembled nanocapsules can make a significant bio-response to the oxidative stress microenvironment of cancer cells and generate fluorescent zinc oxide Nanoclusters in situ for targeted cell bioimaging. Moreover, it can also readily facilitate cancer cell suppression through the targeted delivery of KRAS shRNA and low-temperature hyperthermia. This raises the possibility to provide a promising theranostics platform and self-assembled nanomedicine for targeted cancer diagnostics and treatments through special oxidative stress-responsive effects of cancer cells. | Maonan Wang Zengchao Guo Jiayu Zeng Liu Liu Yihan Wang Jinpeng Wang Hongbing Lu Haijun Zhang Hui Jiang Xuemei Wang | 2023 | Chinese Chemical Letters2023,34,1: | 0 |
| 9 | Intelligent bio-assembly imaging-guided platform for real-time bacteria sterilizing and infectious therapy显示文摘Bacterial infection is rising as a threatening health issue.Because of the present delay in early diagnosis of bacterial diseases as well as the abuse of antibiotics,it has become a vital issue in the development of in-time detection and therapy of bacterial infections.Herein,we designed a multifunctional nanotheranostics platform based on the unique micro-environment of bacterial infections to achieve specific bioimaging and simultaneous inactivation of the target bacteria. We showed that in bacterial infections, the metal precursors(i.e.,HAuCl_(4),FeCl_(2),and herring sperm DNA)could be readily bio-self-assembled to multifunctional nanoclusters(NCs)that exhibit luminescence,in which AuCl_(4)-was biosynthesized via reductive biomolecules such as NADPH to the fluorescent AuNCs.The DNA may assist as an encapsulation and delivery vector, and Fe^(2+)served as a fluorescence intensifier and reduced reactive oxygen species(ROS)to produce the iron oxides. While the bacteria were being visualized, the microenvironment-responsive NCs were enabled to sterilize bacteria efficiently due to electrostatic effect, cell membrane destruction, inhibition of biofilm formation, and ROS accumulation.Besides, the bio-responsive self-assembled NCs complexes contributed to accelerating bacteria-infected wound healing and showed negligible side effects in long-term toxicity tests in vivo. Also, intracellular molecules involved in microenvironmental response were investigated. The work may become an effective strategy for the detection and real-time sterilization of intractable bacterial infections. | Jiayu Zeng Zengchao Guo Yihan Wang Zhaojian Qin Yi Ma Hui Jiang Yossi Weizmann Xuemei Wang | 2022 | Nano Research2022,15,5: | 0 |
