2PDGFRαreporter activity identifies periosteal progenitor cells critical for bone formation and fracture repair显示文摘The outer coverings of the skeleton,which is also known as the periosteum,are arranged in concentric layers and act as a reservoir for tissue-specific bone progenitors.The cellular heterogeneity within this tissue depot is being increasingly recognized.Here,inducible PDGFRαreporter animals were found to mark a population of cells within the periosteum that act as a stem cell reservoir for periosteal appositional bone formation and fracture repair.During these processes,PDGFRαreporter^(+)progenitors give rise to Nestin+periosteal cells before becoming osteoblasts and osteocytes.The diphtheria toxin-mediated ablation of PDGFRαreporter^(+)cells led to deficits in cortical bone formation during homeostasis and a diminutive hard callus during fracture repair.After ossicle transplantation,both mouse PDGFRαreporter^(+)periosteal cells and human Pdgfrα+periosteal progenitors expand,ossify,and recruit marrow to a greater extent than their counterpart periosteal cells,whereas PDGFRαreporter^(−)periosteal cells exhibit a predisposition to chondrogenesis in vitro.Total RNA sequencing identified enrichment of the secreted factors Fermt3 and Ptpn6 within PDGFRαreporter^(+)periosteal cells,which partly underlie the osteoblastogenic features of this cell population.Jiajia Xu Yiyun Wang Zhu Li Ye Tian Zhao Li Amy Lu Ching-Yun Hsu Stefano Negri Cammy Tang Robert J.Tower Carol Morris Aaron W.James 2022Bone Research2022,10,1:2

5The proline synthesis enzyme P5CS forms cytoophidia in Drosophila显示文摘Compartmentation of enzymes via filamentation has arisen as a mechanism for the regulation of metabolism.In 2010,three groups independently reported that CTP synthase(CTPS)can assemble into a filamentous structure termed the cytoophidium.In searching for CTPS-interacting proteins,here we perform a yeast two-hybrid screening of Drosophila proteins and identify a putative CTPS-interacting protein,△~1-pyrroline-5-carboxylate synthase(P5CS).Using the Drosophila follicle cell as the in vivo model,we confirm that P5CS forms cytoophidia,which are associated with CTPS cytoophidia.Overexpression of P5CS increases the length of CTPS cytoophidia.Conversely,filamentation of CTPS affects the morphology of P5CS cytoophid ia.Finally,in vitro analyses confirm the filament-fo rming property of P5CS.Our work links CTPS with P5CS,two enzymes involved in the rate-limiting steps in pyrimidine and proline biosynthesis,respectively.Bo Zhang Omür Y.Tastan Xian Zhou Chen-Jun Guo Xuyang Liu Aaron Thind Huan-Huan Hu Suwen Zhao Ji-Long Liu 2020Journal of Genetics and Genomics2020,47,3:1

10Neuron-to-vessel signaling is a required feature of aberrant stem cell commitment after soft tissue trauma显示文摘The functional interdependence of nerves and blood vessels is a well-established concept during tissue morphogenesis, yet the role of neurovascular coupling in proper and aberrant tissue repair is an emerging field of interest. Here, we sought to define the regulatory relationship of peripheral nerves on vasculature in a severe extremity trauma model in mice, which results in aberrant cell fate and heterotopic ossification(HO). First, a high spatial degree of neurovascular congruency was observed to exist within extremity injury associated heterotopic ossification. Vascular and perivascular cells demonstrate characteristic responses to injury,as assessed by single cell RNA sequencing. This vascular response to injury was blunted in neurectomized mice, including a decrease in endothelial proliferation and type H vessel formation, and a downregulation of key transcriptional networks associated with angiogenesis. Independent mechanisms to chemically or genetically inhibit axonal ingrowth led to similar deficits in HO site angiogenesis, a reduction in type H vessels, and heterotopic bone formation. Finally, a combination of single cell transcriptomic approaches within the dorsal root ganglia identified key neural-derived angiogenic paracrine factors that may mediate neuron-to-vascular signaling in HO. 12Tppp3~+synovial/tendon sheath progenitor cells contribute to heterotopic bone after trauma显示文摘Heterotopic ossification(HO)is a pathological process resulting in aberrant bone formation and often involves synovial lined tissues.During this process,mesenchymal progenitor cells undergo endochondral ossification.Nonetheless,the specific cell phenotypes and mechanisms driving this process are not well understood,in part due to the high degree of heterogeneity of the progenitor cells involved.Here,using a combination of lineage tracing and single-cell RNA sequencing(sc RNA-seq),we investigated the extent to which synovial/tendon sheath progenitor cells contribute to heterotopic bone formation.For this purpose,Tppp3(tubulin polymerization-promoting protein family member 3)-inducible reporter mice were used in combination with either Scx(Scleraxis)or Pdgfra(platelet derived growth factor receptor alpha)reporter mice.Both tendon injury-and arthroplasty-induced mouse experimental HO models were utilized.Sc RNA-seq of tendon-associated traumatic HO suggested that Tppp3 is an early progenitor cell marker for either tendon or osteochondral cells.Upon HO induction,Tppp3 reporter^(+)cells expanded in number and partially contributed to cartilage and bone formation in either tendon-or joint-associated HO.In double reporter animals,both Pdgfra^(+)Tppp3^(+)and Pdgfra^(+)Tppp3^(-) progenitor cells gave rise to HO-associated cartilage.Finally,analysis of human samples showed a substantial population of TPPP3^(-) expressing cells overlapping with osteogenic markers in areas of heterotopic bone.Overall,these data demonstrate that synovial/tendon sheath progenitor cells undergo aberrant osteochondral differentiation and contribute to HO after trauma.Ji-Hye Yea Mario Gomez-Salazar Sharon Onggo Zhao Li Neelima Thottappillil Masnsen Cherief Stefano Negri Xin Xing Qizhi Qin Robert Joel Tower Chen-Ming Fan Benjamin Levi Aaron W.James 2023Bone Research2023,11,3:0

14Electrolyte/Structure‑Dependent Cocktail Mediation Enabling High‑Rate/Low‑Plateau Metal Sulfide Anodes for Sodium Storage显示文摘As promising anodes for sodium-ion batteries,metal sulfides ubiquitously suffer from low-rate and high-plateau issues,greatly hindering their application in full-cells.Herein,exemplifying carbon nanotubes(CNTs)-stringed metal sulfides superstructure(CSC)assembled by nano-dispersed SnS_(2) and CoS_(2) phases,cocktail mediation effect similar to that of high-entropy materials is initially studied in ether-based electrolyte to solve the challenges.The high nano-dispersity of metal sulfides in CSC anode underlies the cocktail-like mediation effect,enabling the circumvention of intrinsic drawbacks of different metal sulfides.By utilizing ether-based electrolyte,the reversibility of metal sulfides is greatly improved,sustaining a long-life effectivity of cocktail-like mediation.As such,CSC effectively overcomes low-rate flaw of SnS_(2) and highplateau demerit of CoS_(2),simultaneously realizes a high rate and a low plateau.In half-cells,CSC delivers an ultrahigh-rate capability of 327.6 mAh g^(−1) anode at 20 A g^(−1),far outperforming those of monometallic sulfides(SnS_(2),CoS_(2))and their mixtures.Compared with CoS_(2) phase and SnS_(2)/CoS_(2) mixture,CSC shows remarkably lowered average charge voltage up to ca.0.62 V.As-assembled CSC//Na1.5VPO4.8F0.7 full-cell shows a good rate capability(0.05~1.0 A g^(−1),120.3 mAh g^(−1) electrode at 0.05 A g^(−1))and a high average discharge voltage up to 2.57 V,comparable to full-cells with alloy-type anodes.Kinetics analysis verifies that the cocktail-like mediation effect largely boosts the charge transfer and ionic diffusion in CSC,compared with single phase and mixed phases.Further mechanism study reveals that alternative and complementary electrochemical processes between nano-dispersed SnS_(2) and CoS_(2) phases are responsible for the lowered charge voltage of CSC.This electrolyte/structure-dependent cocktail-like mediation effect effectively enhances the practicability of metal sulfide anodes,which will boost the development of high-rate/-voltage sodium-ion full batteries.Yongchao Tang Yue Wei Anthony F.Hollenkamp Mustafa Musameh Aaron Seeber Tao Jin Xin Pan Han Zhang Yanan Hou Zongbin Zhao Xiaojuan Hao Jieshan Qiu Chunyi Zhi 2021Nano-Micro Letters2021,13,11:0
15Modeling Common-Mode Current Due to Asymmetric Aging of Machine Winding Insulation显示文摘Machine stator winding insulation degradation is one of the main results of machine aging.It is non-negligible once this degradation process becomes asymmetric between phases.The traditional way to determine the insulation state of health is a partial discharge test.However,this method requires the system offline,which causes production loss and extra administrative burden.This paper presents an idea for better characterizing the insulation machine’s state of health using common-mode(CM)behavior in the machine-drive system.With the help of circuit decomposition methods and modeling tools,the CM quantities due to asymmetric aging show a unique characteristic that distinguishes itself from other differential-mode(DM)quantities in the equivalent circuit.It is shown effective to represent the asymmetric aging effect from the detection of system leakage current.This paper provides an analytical method to quantify this characteristic from mathematical approaches,and a proper approximation has been made on the CM equivalent model(CEM)such that the CM behavior is accurately characterized.The proposed method will serve the purpose of predicting machine abnormal behavior using the simple RLC circuit.Researchers can adapt this method to quantify and characterize the machine insulation state of health(SOH).Jin Zhao Aaron D.Brovont 2023Complex System Modeling and Simulation2023,3,2:0
16Frequency dispersion effect and parameters extraction method for novel HfO_2 as gate dielectric显示文摘The electric characteristic of MOS capacitor with HfO_2/SiO_2/p-Si grown by ALCVD (atom layer chemical vapor deposition) is investigated. The C-V curves show that the accumulation capacitances take on the frequency dispersion at high frequency. For MOS capacitor with ultra thin HfO_2/SiO_2 gate stack, different fabrication processes and measurement equipment will cause parasitic effect. Here an equivalent circuit model that can eliminate the frequency dispersion effect is proposed. The C-V characteristics curve at high frequency shows some distortion because of the bulk defects and the interface states. This paper discusses the distortion of the high frequency MOS C-V characteristic curve. A data processing method is advanced and interface trap density distribution in the band gap is presented. 17Fibromodulin reduces scar formation in adult cutaneouswounds by eliciting a fetal-like phenotype显示文摘Blocking transforming growth factor(TGF)β1 signal transduction has been a central strategy for scar reduction;however,this approach appears to be minimally effective.Here,we show that fibromodulin(FMOD),a 59-kD small leucine-rich proteoglycan critical for normal collagen fibrillogenesis,significantly reduces scar formation while simultaneously increasing scar strength in both adult rodent models and porcine wounds,which simulate human cutaneous scar repair.Mechanistically,FMOD uncouples promigration/contraction cellular signals from pro-fibrotic signaling by selectively enhancing SMAD3-mediated signal transduction,while reducing AP-1-mediated TGFβ1 auto-induction and fibrotic extracellular matrix accumulation.Consequently,FMOD accelerates TGFβ1-responsive adult fibroblast migration,myofibroblast conversion,and function.Furthermore,our findings strongly indicate that,by delicately orchestrating TGFβ1 activities rather than indiscriminately blocking TGFβ1,FMOD elicits fetal-like cellular and molecular phenotypes in adult dermal fibroblasts in vitro and adult cutaneous wounds in vivo,which is a unique response of living system undescribed previously.Taken together,this study illuminates the signal modulating activities of FMOD beyond its structural support functions,and highlights the potential for FMOD-based therapies to be used in cutaneous wound repair.Zhong Zheng Aaron W James Chenshuang Li Wenlu Jiang Joyce Z Wang Grace X Chang Kevin S Lee Feng Chen Emily A Berthiaume Yao Chen Hsin Chuan Pan Eric C Chen Weiming Li Zhihe Zhao Xinli Zhang Kang Ting Chia Soo 2017Signal Transduction and Targeted Therapy2017,2,1:0
