| 4 | A 2-kb Mycovirus Converts a Pathogenic Fungus into a Beneficial Endophyte for Brassica Protection and Yield Enhancement显示文摘Mycoviruses are viruses that infect fungi,and hypovirulence-associated mycoviruses have the potential to control fungal diseases.However,it is unclear how mycovirus-mediated hypovirulent strains live and survive in the field,and no mycovirus has been applied for field crop protection.In this study,we found that a previously identified small DNA mycovirus(SsHADV-1)can convert its host,Sclerotinia sclerotiorum,from a typical necrotrophic pathogen to a beneficial endophytic fungus.SsHADV-1 downregulates the expression of key pathogenicity factor genes in S.sclerotiorum during infection.When growing in rapeseed,the SsHADV-1-infected strain DT-8 significantly regulates the expression of rapeseed genes involved in defense,hormone signaling,and circadian rhythm pathways.As a result,plant growth is promoted and disease resistance is enhanced.Field experiments showed that spraying DT-8 at the early flowering stage can reduce the disease severity of rapeseed stem rot by 67.6%and improve yield by 14.9%.Moreover,we discovered that SsHADV-1 could also infect other S.sclerotiorum strains on DT-8-inoculated plants and that DT-8 could be recovered from dead plants.These findings suggest that the mycoviruses may have the ability to shape the origin of endophytism.Our discoveries suggest that mycoviruses may influence the origin of endophytism and may also offer a novel strategy for disease control in which mycovirus-infected strains are used to improve crop health and release mycoviruses into the field. | Hongxiang Zhang Jiatao Xie Yanping Fu Jiasen Cheng Zheng Qu Zhenzhen Zhao Shufen Cheng Tao Chen Bo Li Qianqian Wang Xinqiang Liu Binnian Tian David B.Collinge Daohong Jiang | 2020 | Molecular Plant2020,13,10: | 9 |
| 5 | Separation and Precise Measurement of Lithium Isotopes in Three Reference Materials Using Multi Collector-Inductively Coupled Plasma Mass Spectrometry显示文摘Lithium separation technique for three reference materials has been established together with precise determination of lithium isotope using a Neptune multi collector-inductively coupled plasma mass spectrometry (MC-ICP-MS). The solutions of lithium element standard reference materials, potassium, calcium, sodium, magnesium and iron single element, were used to evaluate analytical methods applied. Three separate stages of ion-exchange chromatography were carried out using organic cation-exchange resin (AG 50W-X8). Lithium was enriched for the three stages using different eluants, which are 2.8 M HCl, 0.15 M HCl and 0.5 M HCl in 30% ethanol, respectively. The columns for the first and second stages are made of polypropylene, and those for the third stage are made of quartz. Total reagent volume for the entire chemical process was 35 mL for three reference materials. The recovery yielded for the three stages is 98.9-101.2% with an average of 100.0%, 97.6- 101.9% with an average of 99.9%, and 99.8-103.3% with an average of 100.6%, respectively. The precision of this technique is conservatively estimated to be ±0.72-1.04‰ (2σ population), which is similar to the precision obtained by different authors in different laboratories with MC-ICP-MS. The δ 7 Li values ( 7 Li/ 6 Li relative to the IRMM-016 standard) determined for andesite (AGV-2) and basalt (BHVO-2) are 5.68‰ (n=18), 4.33‰ (n=18), respectively. The δ 7 Li value ( 7 Li/ 6 Li relative to the L-SVEC standard) determined for IRMM-016 is -0.01‰ (n=15). All these analytical results are in good agreement with those previously reported. In addition, the results for the same kinds of samples analyzed at the MLR Key Laboratory of Metallogeny and Mineral Assessment, Institute of Mineral Resources, Chinese Academy of Geological Sciences, are consistent with those obtained at the Plasma Laboratory, University of Maryland, within analytical uncertainty. | 6 | Personalized Recommendation Algorithm Based on Preference Features显示文摘A hybrid collaborative filtering algorithm based on the user preferences and item features is proposed. A thorough investigation of Collaborative Filtering(CF) techniques preceded the development of this algorithm. The proposed algorithm improved the user-item similarity approach by extracting the item feature and applying various item features' weight to the item to confirm different item features. User preferences for different item features were obtained by employing user evaluations of the items. It is expected that providing better recommendations according to preferences and features would improve the accuracy and efficiency of recommendations and also make it easier to deal with the data sparsity. In addition, it is expected that the potential semantics of the user evaluation model would be revealed. This would explain the recommendation results and increase accuracy. A portion of the MovieLens database was used to conduct a comparative experiment among the proposed algorithms,i.e., the collaborative filtering algorithm based on the item and the collaborative filtering algorithm based on the item feature. The Mean Absolute Error(MAE) was utilized to conduct performance testing. | 7 | Biomechanical properties of acellular sciatic nerves treated with a modified chemical method显示文摘Nerve grafts are able to adapt to surrounding biomechanical environments if the nerve graft itself exhibits appropriate biomechanical properties (load, elastic modulus, etc.). The present study was designed to determine the differences in biomechanical properties between fresh and chemically acellularized sciatic nerve grafts. Two different chemical methods were used to establish acellular nerve grafts. The nerve was chemically extracted in the Sondell method with a combination of Triton X-100 (nonionic detergent) and sodium deoxycholate (anionic detergent), and in the modified method with a combination of Triton X-200 (anionic detergent), sulfobetaine-10 (SB-10, amphoteric detergents), and sulfobetaine-16 (SB-16, amphoteric detergents). Following acellularization, hematoxylin-eosin staining and scanning electron microscopy demonstrated that the effect of acellularization via the modified method was similar to the traditional Sondell method. However, effects of demyelination and nerve fiber tube integrity were superior to the traditional Sondell method. Biomechanical testing showed that peripheral nerve graft treated using the chemical method resulted in decreased biomechanical properties (ultimate load, ultimate stress, ultimate strain, and mechanical work to fracture) compared with fresh nerves, but the differences had no statistical significance (P > 0.05). These results demonstrated no significant effect on biomechanical properties of nerves treated using the chemical method. | 8 | The Arabidopsis B-box protein BZS1/BBX20 interacts with HY5 and mediates strigolactone regulation of photomorphogenesis显示文摘Plant growth is controlled by integration of hormonal and light-signaling pathways. BZS1 is a B-box zinc finger protein previously characterized as a negative regulator in the brassinosteroid(BR)-signaling pathway and a positive regulator in the light-signaling pathway. However, the mechanisms by which BZS1/BBX20 integrates light and hormonal pathways are not fully understood. Here, using a quantitative proteomic workflow, we identified several BZS1-associated proteins, including light-signaling components COP1 and HY5. Direct interactions of BZS1 with COP1 and HY5 were verified by yeast two-hybrid and co-immunoprecipitation assays. Overexpression of BZS1 causes a dwarf phenotype that is suppressed by the hy5 mutation, while overexpression of BZS1 fused with the SRDX transcription repressor domain(BZS1-SRDX) causes a long-hypocotyl phenotype similar to hy5, indicating that BZS1's function requires HY5. BZS1 positively regulates HY5 expression, whereas HY5 negatively regulates BZS1 protein level, forming a feedback loop that potentially contributes to signaling dynamics. In contrast to BR,strigolactone(SL) increases BZS1 level, whereas the SL responses of hypocotyl elongation, chlorophyll and HY5 accumulation are diminished in the BZS1-SRDX seedlings, indicating that BZS1 is involved in these SL responses. | 20 | Anti-hypertensive and endothelia protective effects of Fufang Qima capsule(复方芪麻胶囊) on primary hypertension via adiponectin/adenosine monophosphate activated protein kinase pathway显示文摘OBJECTIVE:To investigate the potential mechanism of the vascular remodeling effect and provide additional information about anti-hypertension activity of Fufang Qima capsule(复方芪麻胶囊,QM).METHODS:Spontaneous hypertensive rats(SHRs)were used to study the underlying mechanism of the anti-hypertension activity of QM.In this study,SHRs were randomly divided into 5 groups:model group,Telmisartan group(7.2 mg/kg,p.o.),and three QM groups(0.9298,1.8596,and 3.7192 g/kg,p.o.).Wistar Kyoto rats(WKY)were used as normal control group.Blood pressure(BP),aorta,perivascular adipose tissue(PVAT)histology were investigated to evaluate the effect of QM.Nitric oxide(NO)and endothelial nitric oxide synthase(eNOS)phosphorylation were measured.Adiponectin(APN)secretion,as well as APN signal pathway proteins including APN,adiponectin receptors(R1 and R2)and adenosine 5’-monophosphate-activated protein kinase(AMPK)were all analyzed.RESULTS:QM significantly reduced BP and ameliorated the vascular pathological change,i.e.intima media thicken and collagen fiber hyperplasia.Meanwhile,QM increased concentration of NO and the phosphorylation of eNOS in the aorta.The anti-hypertensive and endothelia-protective effect of QM could be attributed to activating APN/AMPK pathway by up-regulating the expression of APN in PVAT and APN Receptor 2,AMPKαand phosphorylated AMPKαin the aorta.CONCLUSION:The QM alleviation effect mechanism for primary hypertension was via modulating the APN/AMPK signal pathway. | ZHAO Zhiyue SHI Zhenyu ZHANG Zhenzhen LI Yinghong ZENG Xiaohui CHEN Yuxing YAO Nan ZHOU Min SU Hui WANG Qinghai JIN LiLi | 2021 | Journal of Traditional Chinese Medicine2021,41,6: | 2 |
