| 1 | Complement C3 activation regulates the production of tRNA-derived fragments Gly-tRFs and promotes alcohol-induced liver injury and steatosis显示文摘Complement is known to play a role in alcoholic fatty liver disease (AFLD), but the underlying mechanisms are poorly understood, thereby constraining the development of a rational approach for therapeutic intervention in the complement system. C3 deficiency has been shown to impart protective effects against ethanol-induced hepatic steatosis and inflammation. Here we demonstrate a protection effect in wild-type mice by treatment with CR2-Crry, a specific inhibitor of C3 activation. The expression of glycine transfer (t) RNA-derived fragments (Gly-tRFs) is upregulated in ethanol-fed mice and inhibition of Gly-tRFs in vivo decreases chronic ethanol feeding-induced hepatosteatosis without affecting inflammation. The expression of Gly-tRF was downregulated in C3-deficient or CR2-Crry-treated mice, but not in C5-deficient mice;Gly-tRF expression was restored by the C3 activation products C3a or Asp (C3a-des-Arg) via the regulation of CYP2E1. Transcriptome profiling of hepatic tissues showed that Gly-tRF inhibitors upregulate the expression of sirtuin1 (Sirt1) and subsequently affect downstream lipogenesis and β-oxidation pathways. Mechanistically, Gly-tRF interacts with AGO3 to downregulate Sirt1 expression via sequence complementarity in the 3′ UTR. Notably, the expression levels of C3d, CYP2E1 and Gly-tRF are upregulated, whereas Sirt1 is decreased in AFLD patients compared to healthy controls. Collectively, our findings suggest that C3 activation products contribute to hepatosteatosis by regulating the expression of Gly-tRF. | 9 | Experiments on the influences of well pattern on water flooding characteristics of dissolution vug-cave reservoir显示文摘Based on the similarity criterion, volcanic rock samples were taken from outcrops to make experimental models. Water flooding experiments of five-spot well pattern, nine-spot well pattern, five-spot to nine-spot well pattern, the relationship between relative well and fracture positions, and injection rate in dissolution vug-cave reservoirs with/without fractures were carried out to analyze variation regularities of development indexes, find out development characteristics of water flooding with different well patterns and sort out the optimal water flooding development mode. For dissolution vug-cave reservoirs without fractures, five-spot well pattern waterflooding has very small sweeping area, serious water channeling and low oil recovery. When the well pattern was adjusted from five-spot to nine-spot well pattern, oil recovery could be largely improved, but the corner well far from the injector is little affected. In dissolution vug-cave reservoirs with fractures, when the injector and producer are not connected by fractures, the fractures could effectively connect the poorly linked vugs to improve the development effect of water flooding. Whether there are fractures or not in dissolution vug-cave reservoirs, the development effect of nine-spot well-pattern is much better than that of five-spot well pattern and five-spot to nine-spot well pattern, this is more evident when there are fractures, and the edge well has better development indexes than corner well. At the high-water cut stage of water flooding with nine-spot well pattern, the oil recovery can be further improved with staggered line-drive pattern by converting the corner well into injection well. | 10 | Controlling factors of remaining oil distribution after water flooding and enhanced oil recovery methods for fracturecavity carbonate reservoirs in Tahe Oilfield显示文摘Based on comprehensive analysis of core, well logging, seismic and production data, the multi-scale reservoir space, reservoir types, spatial shape and distribution of fractures and caves, and the configuration relationship with production wells in fracture-cavity carbonate reservoirs were studied systematically, the influence of them on the distribution of residual oil was analyzed, and the main controlling factors mode of residual oil distribution after water flooding was established. Enhanced oil recovery methods were studied considering the development practice of Tahe oilfield. Research shows that the main controlling factors of residual oil distribution after water flooding in fracture-cavity carbonate reservoirs can be classified into four categories: local high point, insufficient well control, flow channel shielding and weak hydrodynamic. It is a systematic project to improve oil recovery in fracture-cavity carbonate reservoirs. In the stage of natural depletion, production should be well regulated to prevent bottom water channeling. In the early stage of waterflooding, injection-production relationship should be constructed according to reservoir type, connectivity and spatial location to enhance control and producing degree of waterflooding and minimize remaining oil. In the middle and late stage, according to the main controlling factors and distribution characteristics of remaining oil after water flooding, remaining oil should be tapped precisely by making use of gravity differentiation and capillary force imbibition, enhancing well control, disturbing the flow field and so on. | 11 | Theories and practices of carbonate reservoirs development in China显示文摘Carbonate reservoirs in China have the characteristics of diversified accumulation pattern, complex structure and varying reservoir conditions. Concerning these characteristics, this article tracks the technical breakthroughs and related practices since the 1950 s, summarizes the developed theory and technologies of carbonate reservoir development, analyzes their adaptability and problems, and proposes their development trend. The following theory and technologies have come into being:(1) carbonate reservoir formation mechanisms and compound flow mechanisms in complex medium;(2) reservoir identification and description technologies based on geophysics and discrete fracture-vuggy modeling method;(3) well testing analysis technology and numerical simulation method of coupling free flow and porous media flow;(4) enhanced oil recovery techniques for nitrogen single well huff and puff, and water flooding development techniques with well pattern design in spatial structure, changed intensity water injection, water plugging and channel blocking as the core;(5) drilling and completion techniques, acid fracturing techniques and its production increasing techniques.
