RNA-binding proteins in neurological diseases显示文摘Emerging studies support that RNA-binding proteins(RBPs)play critical roles in human biology and pathogenesis.RBPs are essential players in RNA processing and metabolism, including pre-mRNA splicing, polyadenylation, transport, surveillance,mRNA localization,mRNA stability control, translational control and editing of various types of RNAs.Aberrant expression of and mutations in RBP genes affect various steps of RNA processing, altering target gene function.RBPs have been associated with various diseases, including neurological diseases. Here,we mainly focus on selected RNA-binding proteins including Nova-1/Nova-2,HuR/HuB/HuC/HuD,TDP-43,Fus,Rbfox1/Rbfox2,QKI and FMRP,discussing their function and roles in human diseases. | ZHOU HuaLin MANGELSDORF Marie LIU JiangHong ZHU Li WU Jane Y | 2014 | Science China(Life Sciences)2014,57,4: | 9
The nucleoprotein of severe fever with thrombocytopenia syndrome virus processes a stable hexameric ring to facilitate RNA encapsidation显示文摘Severe fever with thrombocytopenia syndrome virus(SFTSV),a member of the Phlebovirus genus from the Bunyaviridae family endemic to China,is the causative agent of life-threatening severe fever with thrombocyto-penia syndrome(SFTS),which features high fever and hemorrhage.Similar to other negative-sense RNA viruses,SFTSV encodes a nucleocapsid protein(NP)that is essen-tial for viral replication.NP facilitates viral RNA encapsida-tion and is responsible for the formation of ribonucleopro-tein complex. However, recent studies have indicated that NP from Phlebovirus members behaves in inhomogene-ous oligomerization states. In the present study,we report the crystal structure of SFTSV NP at 2.8Åresolution and demonstrate the mechanism by which it processes a ring-shaped hexameric form to accomplish RNA encapsida-tion.Key residues essential for oligomerization are identi-fi ed through mutational analysis and identifi ed to have a signifi cant impact on RNA binding, which suggests that correct formation of highly ordered oligomers is a criti-cal step in RNA encapsidation. The fi ndings of this work provide new insights into the discovery of new antiviral reagents for Phlebovirus infection. | Honggang Zhou Yuna Sun Ying Wang Min Liu Chao Liu Wenming Wang Xiang Liu Le Li Fei Deng Hualin Wang Yu Guo Zhiyong Lou | 2013 | Protein & Cell2013,4,6: | 4
A Comparative Study of the Seismic Performances and Failure Mechanisms of Slopes Using Dynamic Centrifuge Modeling显示文摘In this study, dynamic centrifuge model tests were performed for sand slopes under different earthquake ground motions and slope angle to characterize the seismic performance of slopes. Four groups of tests under varying seismic input amplitude were conducted. Under the action of increasing earthquake intensity, the rigidity of the soil decreases and the damping ratio increases, both of the dynamic response and the predominant period of slopes are increased. Three types of seismic waves with the same seismic intensity were applied in the model tests. It shows that the variability in the ground motion leads to the acceleration response spectra of the slopes being completely different and the Northridge seismic wave with low-frequency component is closest to the predominant period of the slope model. In addition, the effect of slope angle on the seismic performance of slopes were also clarified. The results reveal how the slope angle affects the acceleration recorded on the ground surface of the slope, both in terms of the peak ground-motion acceleration(PGA) amplification factor and the predominant period. Finally, the permanent displacement of the model slopes under different earthquake intensities were further analyzed. Based on the nonlinear growth of the permanent displacement of the slope, the test results demonstrated the failure process of the slope, which can further provide a basis for theperformance-based seismic design of slopes. | Hualin Cheng Jiamin Zhou Zhiyi Chen Yu Huang | 2021 | Journal of Earth Science2021,32,5: | 3
Effects of the chemical structure of surfactants on the stability of naphthenic oil-based metalworking fluids显示文摘In this study, various nonionic surfactants(NS) with different ethylene oxide(EO) numbers and tail lengths and its binary blends with anionic surfactants(AS) were used as emulsifiers for naphthenic oil to form the microemulsion metalworking fluids(MWFs),and the effects of them on the stability of the emulsion system were investigated by formulation triangle method. The results indicated that binary complex surfactants of NS and AS as emulsifiers exhibited better emulsifying effect than that of single NS.NS with different EO numbers and tail lengths presented various emulsifying effects.NS(EO=10)exhibited the greatest number of stable formulations, especially the TX-10,but no linear relationship existed between the number of stable formulations and the tail length of NS.In addition, aromatic primary alcohol ethoxy late(APAE) series surfactants containing benzene groups similar to the cycloalkanes in the naphthenic oil so that presented the best emulsifying affect and the greatest number of stable formulations. The co-surfactant of sodium dodecyl benzene sulfonate(SDBS) binary blends with NS exerted the best synergistic effect, and the stable formulations numbers were ranged from 5 to 7,next sodium stearate(SS) comes last followed by sodium dodecyl sulfate(SDS-1) and sodium dodecyl sulfonate(SDS-2). | Yuan Xue Jinbao Liu Maiying Xie Fengfei Chen Mingan Zhou Hualin Lin Fan Zhang Shizhou Liu Zhiqiao Wu Sheng Han | 2020 | Chinese Chemical Letters2020,31,2: | 2

Blast responses of shallow-buried prefabricated modular concrete tunnels reinforced by BFRP-steel bars显示文摘This paper reports the anti-blast performance of shallow-buried prefabricated modular tunnel reinforced by basalt fiber-reinforced polymer(BFRP)-steel bars.Three concrete arch members with steel bars and three concrete arch members with BFRP-steel bars were fabricated, with the other arch parameters kept constant. The three identical arches were assembled into an integral structure and then buried in soil for field anti-blast experiments.Through the experiment, the pressure on the vault, the displacement and acceleration of the vault, the strain in the reinforcement bars and the macroscopic damage of the arches under the blast load were determined. To evaluate the damage of the arch tunnel,a residual load-bearing capacity test was conducted on the arch members after the explosion experiment. The experimental results showed that the BFRP-steel bars reinforced concrete arch exhibited a higher load-bearing capacity and more safety redundancy than the steel bars reinforced concrete arch, and that the BFRP-steel bars could inhibit the occurrence of concrete cracks to a certain extent.A comparison between the arches assembled at different positions showed that the prefabricated modular tun-nel can be simplified directly as a two-dimensional arch structure under the blast load for analysis and calculation. | Chengjie Zhao Zexun Tang Peng Wang Jiang Feng Jiannan Zhou Xinli Kong Hansheng Geng Fengnian Jin Hualin Fan | 2022 | Underground Space2022,7,2: | 1

Spatiotemporal Dynamics of the Molecular Expression Pattern显示文摘Nerve regeneration in adult mammalian spinal cord is poor because of the lack of intrinsic regeneration of neurons and extrinsic factors–the glial scar is triggered by injury and inhibits or promotes regeneration.Recent technological advances in spatial transcriptomics(ST)provide a unique opportunity to decipher most genes systematically throughout scar formation, which remains poorly understood. Here,we frst constructed the tissue-wide gene expression patterns of mouse spinal cords over the course of scar formation using ST after spinal cord injury from 32 samples.Locally,we profled gene expression gradients from the leading edge to the core of the scar areas to further understand the scar microenvironment, such as neurotransmitter disorders, activation of the pro-infammatory response, neurotoxic saturated lipids, angiogenesis, obstructed axon extension, and extracellular structure re-organization.In addition,we described 21 cell transcriptional states during scar formation and delineated the origins, functional diversity, and possible trajectories of subpopulations of fbroblasts, glia, and immune cells.Specifcally,we found some regulators in special cell types, such as Thbs1 and Col1a2 in macrophages,CD36 and Postn in fbroblasts,Plxnb2 and Nxpe3 in microglia,Clu in astrocytes, and CD74 in oligodendrocytes.Furthermore, salvianolic acid B,a blood–brain barrier permeation and CD36 inhibitor, was administered after surgery and found to remedy fbrosis.Subsequently,we described the extent of the scar boundary and profled the bidirectional ligand-receptor interactions at the neighboring cluster boundary, contributing to maintain scar architecture during gliosis and fbrosis, and found that GPR37L1_PSAP,and GPR37_PSAP were the most signifcant gene-pairs among microglia, fbroblasts, and astrocytes.Last,we quantifed the fraction of scar-resident cells and proposed four possible phases of scar formation:macrophage infltration, proliferation and diferentiation of scar-resident cells, scar emergence, and scar stationary.Together, these profles delineated the spatial heterogeneity of the scar, confrmed the previous concepts about scar architecture, provided some new clues for scar formation, and served as a valuable resource for the treatment of central nervous system injury. | Leilei Gong Yun Gu Xiaoxiao Han Chengcheng Luan Chang Liu Xinghui Wang Yufeng Sun Mengru Zheng Mengya Fang Shuhai Yang Lai Xu Hualin Sun Bin Yu Xiaosong Gu Songlin Zhou | 2023 | Neuroscience Bulletin2023,39,2: | 0
Non-cell-autonomous manner of AAV administration to attenuate cardiomyocyte hypertrophy by targeting paracrine signaling on ECM to reduce viral dosage显示文摘Dear Editor,Adeno-associated virus(AAV)is one of the most powerful vectors for exogenous gene delivery.1 However, the risks associated with the high dosages of AAV administration that are required to achieve meaningful effects limit the applicability of this method.Generally,AAV only presents cell autonomous manner in infected cells.Besides, mosaic genetic editing or gene therapy would trigger global biological function restore by above 30–40%infective ratio in heart diseases. | Lei Liu Peng Yue Yue Zhang Yimin Hua Wenwei Bi Hualin Yan Hongyu Liao Jiawen Li Kaiyu Zhou Yifei Li | 2022 | Signal Transduction and Targeted Therapy2022,7,2: | 0
Regulation of hypoxic stress and oxidative stress in bone grafting: Current trends and future perspectives显示文摘Tissue engineering aims to offer large-scale replacement of damaged organs using implants with the com-bination of cells, growth factors and scaffolds. However, the intra/peri-implant region is exposed to se-vere hypoxic stress and oxidative stress during the early stage of implantation with bone graft materials, which endangers the survival, proliferation and differentiation of seed cells within the implants as well as the host cells surrounding the implants. If the bone graft material could spontaneously and intelligently regulate the hypoxic stress and oxidative stress to a moderate level,it will facilitate the vascularization of the implants and the rapid regeneration of the bone tissue. In this review,we will first introduce the signaling pathways of cellular response under hypoxic stress and oxidative stress, then present the clas-sical material designs and examples in response to hypoxic stress and oxidative stress. And finally,we will address the important role of epigenetic mechanisms in the regulation of hypoxic stress and oxida-tive stress and describe the potential applications and prospective smart bone graft materials based on novel epigenetic factors against hypoxic stress and oxidative stress in bone repair. The main content of this review is summarized in the following graphical abstract. | Hao Hu Xiao Liu Jun Chen Shangbin Cui Hualin Yi Gang Wang Renxian Wang Tiansheng Zheng Ben Wan Zhiyu Zhou Yong Wan Manman Gao Dafu Chen Xuenong Zou | 2023 | Journal of Materials Science & Technology2023,,26: | 0

The Method for Inferring a Buried Fault from Resistivity Tomograms and Its Typical Electrical Features显示文摘Electrical resistivity tomography (ERT) has been used to experimentally detect shallow buried faults in urban areas in the past a few years, with some progress and experience obtained. According to the results from Olympic Park, Beijing, Shandong Province, Gansu Province and Shanxi Province, we have generalized the method and procedure for inferring the discontinuity of electrical structures (DES) indicating a buried fault in urban areas from resistivity tomograms and its typical electrical features. In general, the layered feature of the electrical structure is first analyzed to preliminarily define whether or not a DES exists in the target area. Resistivity contours in resistivity tomograms are then analyzed from the deep to the shallow. If they extend upward from the deep to the shallow and shape into an integral dislocation, sharp flexure (convergence) or gradient zone, it is inferred that the DES exists, indicating a buried fault. Finally, horizontal tracing is be carried out to define the trend of the DES. The DES can be divided into three types-type AB, ABA and AC. In the present paper, the Zhangdian-Renhe fault system in Zibo city is used as an example to illustrate how to use the method to infer the location and spatial extension of a target fault. Geologic drilling holes are placed based on our research results, and the drilling logs testify that our results are correct. However, the method of this paper is not exclusive and inflexible. It is expected to provide reference and assistance for inferring the shallow buried faults in urban areas from resistivity tomograms in the future. | Zhu Tao Feng Rui Zhou Jianguo Hao Jinqi Wang Hualin Wang Shuoqing | 2009 | Earthquake Research in China2009,23,4: | 0

Thermodynamics and kinetics insights into naphthalene hydrogenation over a Ni-Mo catalyst显示文摘Hydrocracking represents an important process in modern petroleum refining industry, whose performance mainly relies on the identity of catalyst. In this work,we perform a combined thermodynamics and kinetics study on the hydrogenation of naphthalene over a commercialized NiMo/HY catalyst. The reaction network is constructed for the respective production of decalin and methylindane via the intermediate product of tetralin, which could further undergo hydrogenation to butylbenzene, ethylbenzene, xylene, toluene, benzene, methylcyclohexane and cyclohexane. The thermodynamics analysis suggests the optimum operating conditions for the production of monoaromatics are 400℃,8.0 MPa,and 4.0 hydrogen/naphthalene ratio.Based on these, the influences of reaction temperature,pressure,hydrogen/-naphthalene ratio, and liquid hourly space velocity(LHSV)are investigated to fit the Langmuir-Hinshelwood model. It is found that the higher temperature and pressure while lower LHSV favors monoaromatics production, which is insensitive to the hydrogen/naphthalene ratio.Furthermore, the high consistence between the experimental and simulated data further validates the as-obtained kinetics model on the prediction of catalytic performance over this kind of catalyst. | Chong Peng Zhiming Zhou Xiangchen Fang Hualin Wang | 2021 | Chinese Journal of Chemical Engineering2021,34,11: | 0
