| 3 | SARS-CoV-2 triggers inflammatory responses and cell death through caspase-8 activation显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection can lead to respiratory illness and multi-organ failure in critically ill patients.Although the virus-induced lung damage and inflammatory cytokine storm are believed to be directly associated with coronavirus disease 2019(COVID-19)clinical manifestations,the underlying mechanisms of virus-triggered inflammatory responses are currently unknown.Here we report that SARS-CoV-2 infection activates caspase-8 to trigger cell apoptosis and inflammatory cytokine processing in the lung epithelial cells.The processed inflammatory cytokines are released through the virus-induced necroptosis pathway.Virus-induced apoptosis,necroptosis,and inflammation activation were also observed in the lung sections of SARS-CoV-2-infected HFH4-hACE2 transgenic mouse model,a valid model for studying SARS-CoV-2 pathogenesis.Furthermore,analysis of the postmortem lung sections of fatal COVID-19 patients revealed not only apoptosis and necroptosis but also massive inflammatory cell infiltration,necrotic cell debris,and pulmonary interstitial fibrosis,typical of immune pathogenesis in the lung.The SARS-CoV-2 infection triggered a dual mode of cell death pathways and caspase-8-dependent inflammatory responses may lead to the lung damage in the COVID-19 patients.These discoveries might assist the development of therapeutic strategies to treat COVID-19. | Shufen Li Yulan Zhang Zhenqiong Guan Huiling Li Meidi Ye Xi Chen Jun Shen Yiwu Zhou Zheng-Li Shi Peng Zhou Ke Peng | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 3 |
| 4 | The staphylococcal nuclease prevents biofilm formation in Staphylococcus aureus and other biofilm-forming bacteria显示文摘The staphylococcal nuclease, encoded by the nuc1 gene, is an important virulence factor of Staphylococcus aureus. However, the physiological role of the nuclease has not been fully characterized. The current study observed that biofilm development could be prevented in staphylococcal nuclease-producing strains of S. aureus; however, when the nuc1 gene was knocked out, the ability to form a biofilm significantly increased. Scanning electron and confocal scanning laser microscopy were used to evaluate the role of the nuc1 gene in biofilm formation. Moreover, the nuc1 gene product, staphylococcal nuclease, and recombinant NUC1 protein were found to have a visible effect on other biofilm-forming bacteria, such as Pseudomonas aeruginosa, Actinobacillus pleuropneumoniae, and Haemophilus parasuis. The current study showed a direct relationship between staphylococcal nuclease production and the prevention of biofilm development. The findings from this study underscore the important role of staphylococcal nuclease activity to prevent biofilm formation in S. aureus. | 6 | Intrusions of Kuroshio and Shelf Waters on Northern Slope of South China Sea in Summer 2015显示文摘The northern slope region of the South China Sea(SCS) is a biological hot spot characterized by high primary productivity and biomasses transported by cross-shelf currents, which support the spawning and growth of commercially and ecologically important fish species. To understand the physical and biogeochemical processes that promote the high primary production of this region, we conducted a cruise from June 10 and July 2, 2015. In this study, we used fuzzy cluster analysis and optimum multiparameter analysis methods to analyze the hydrographic data collected during the cruise to determine the compositions of the upper 55-m water masses on the SCS northern slope and thereby elucidate the cross-slope transport of shelf water(SHW) and the intrusions of Kuroshio water(KW). We also analyzed the geostrophic currents derived from acoustic Doppler current profiler measurements and satellite data. The results reveal the surface waters on the northern slope of the SCS to be primarily composed of waters originating from South China Sea water(SCSW), KW, and SHW. The SCSW dominated a majority of the study region at percentages ranging between 60% and 100%. | 11 | The Upper 1000-m Slope Currents North of the South Shetland Islands and Elephant Island Based on Ship Cruise Observations显示文摘While the Antarctic Slope Current(ASC) has been intensively studied for the East Antarctica slope area and the Weddell Sea, its fate in the western Antarctic Peninsula(WAP) region remains much less known. Data from two cruises conducted near the South Shetland Islands(SSIs) and the Elephant Island(EI), one in austral summer of 2004 and one in austral winter of 2006, were analyzed to provide a broad picture of the circulation pattern over the continental slope of the surveyed area, and an insight into the dynamical balance of the circulation. The results indicate that southwestward currents are present over the upper slope in the study area, indicating the ASC in the WAP region. Near the Shackleton Gap(SG) north of the EI, the southwestward slope currents near the shelf break are characterized by a water mass colder and fresher than the ambient water, which produces cross-slope density gradients and then vertical shear of the along-slope(or along-isobath) velocity. The vertical shear is associated with a reversal of the along-slope current from northeastward at surface to southwestward in deeper layers, or a depth-intensification of the southwestward slope currents. The water mass with temperature and salinity characteristics similar to the observed cold and fresh water is also revealed on the southern slope of the Scotia Sea, suggesting that this cold and fresh water is originated from the Scotia Sea slope and flows southwestward through the SG. Over the shelf north of the SSIs, the cold and fresh water mass is also observed and originates mainly from the Bransfield Strait. | 14 | Hybridoma-derived neutralizing monoclonal antibodies against Beta and Delta variants of SARS-CoV-2 in vivo显示文摘Neutralizing monoclonal antibodies(mAb)are a major therapeutic strategy for the treatment of severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)infection.The continuous emergence of new SARS-CoV-2 variants worldwide has increased the urgency for the development of new mAbs.In this study,we immunized mice with the receptor-binding domain(RBD)of the SARS-CoV-2 prototypic strain(WIV04)and screened 35 RBDspecific mAbs using hybridoma technology.Results of the plaque reduction neutralization test showed that 25 of the mAbs neutralized authentic WIV04 strain infection.The 25 mAbs were divided into three categories based on the competitive enzyme-linked immunosorbent assay results.A representative mAb was selected from each category(RD4,RD10,and RD14)to determine the binding kinetics and median inhibitory concentration(IC_(50))of WIV04 and two variants of concern(VOC):B.1.351(Beta)and B.1.617.2(Delta).RD4 neutralized the B.1.617.2 variant with an IC50 of 2.67 ng/mL;however,it completely lost neutralizing activity against the B.1.351 variant.RD10 neutralized both variants with an IC50 exceeding 100 ng/mL;whereas RD14 neutralized two variants with a higher IC50(>1 mg/mL).Animal experiments were performed to evaluate the protective effects of RD4 and RD10 against various VOC infections.RD4 could protect Adv-hACE2 transduced mice from B.1.617.2 infection at an antibody concentration of 25 mg/kg,while RD10 could protect mice from B.1.351 infection at an antibody concentration of 75 mg/kg.These results highlight the potential for future modifications of the mAbs for practical use. | Qianran Wang Lu Peng Yanqiu Nie Yanni Shu Huajun Zhang Zidan Song Yufeng Li Hengrui Hu Liushuai Li Xi Wang Jia Liu Jiang Li Zhengli Shi Fei Deng Yu Guo Yiwu Zhou Bing Yan Zhihong Hu Manli Wang | 2023 | Virologica Sinica2023,38,2: | 0 |

| 16 | Effects of glucocorticoid and glucocorticoid receptors on stress-induced neurogenesis suppression显示文摘Studies have shown that cerebral ischemia activates neurogenesis and that stress inhibits neurogenesis. However, the role of stress hormone levels on neurogenesis following cerebral ischemia remains poorly understood. The present study explored the possible regulatory mechanisms of adult neurogenesis under pathological conditions by examining changes and regulation of glucocorticoid receptors in adult rats subjected to transient unilateral middle cerebral artery suture occlusion. Corticosterone levels gradually increased following middle cerebral artery occlusion, and the number of glucocorticoid receptor-positive cells decreased. The number of 5-bromodeoxyuridine- and nestin-positive cells significantly increased at 1 and 2 weeks after ischemia. A large number of doublecortin-positive cells migrated from the hippocampus to the cortex. At 3 weeks post-surgery, the number of 5-bromodeoxyuridine- and nestin-positive cells significantly reduced in the subventricular zone. Increased corticosterone levels decreased vascular endothelial cell proliferation and neurogenesis, and the number of glucocorticoid receptor-positive cells decreased. In the sham surgery group, vascular endothelial cell proliferation related to post-ischemic cerebral rehabilitation was not detected. Corticosterone levels increased, but the number and distribution of glucocorticoid receptor-positive cells were not changed. However, normal neurogenesis and migration of neural stem cells existed in the adult rat brain in the sham surgery group.
