| 1 | Tobacco curly shoot virus isolated in Yunnan is a distinct species of Begomovirus显示文摘Virus isolate Y1 was obtained from tobacco showing curly shoot symptoms in Baoshan, Yunnan Province. Whitefly transmission test and virion morphology observa-tion showed that it is a begomovirus. In reactions with 14 monoclonal antibodies raised against begomoviruses, Y1 was readily differentiated from begomoviruses reported in China, Pakistan and India. The complete nucleotide sequence of DNA-A was determined, it contains 2746 nucleotides, with two ORFs in virion-sense DNA and four ORFs in comple-mentary-sense DNA. Comparisons with total DNA-A, inter-genie region and deduced amino acid sequences of individual ORFs showed that Y1 is a distinct Begomovirus species, for which the name Tobacco curly shoot virus (TCSV) is pro-posed. The total DNA-A of TCSV is most closely related to that of Tomato leaf curl virus from India (85% sequence identity). | 6 | Cas9-NG Greatly Expands the Targeting Scope of the Genome-Editing Toolkit by Recognizing NG and Other Atypical PAMs in Rice显示文摘CRISPR technologies enabling precise genome manipulation are valuable for gene function studies and molecular crop breeding. However, the requirement of a protospacer adjacent motif (PAM)y such as NGG and TTN, for Cas protein recognition restricts the selection of targetable genomic loci in practical applications of CRISPR technologies. Recently Cas9-NG, which recognizes a minimal NG PAM, was reported to expand the targeting space of genome editing in human cells, but it remains unclear whether this Cas9 variant can be used in plants. In this study, we evaluated the nuclease activity of Cas9-NG toward various NGN PAMs by targeting endogenous genes in transgenic rice. We found that Cas9-NG edits all NGG, NGA, NGT, and NGC sites with impaired activity, while the gene-edited plants were dominated by monoallelic mutations. Cas9-NG-engineered base editors were then developed and used to generate O s B Z R I gainof- function plants that can not be created by other available Cas9-engineered base editors. Moreover, we showed that a Cas9-NG-based transcriptional activator efficiently upregulated the expression of endogenous target genes in rice. In addition, we discovered that Cas9-NG recognizes NAC, NTG, NTT, and NCG apart from NG PAM. | 9 | Molecular characterization of Tomato yellow leaf curl China virus and its satellite DNA isolated from tobacco显示文摘Virus isolates, Y8, Y36 and Y38, were obtained from tobacco plants showing leaf curl symptoms in Honghe, Yunnan Province. In reactions with 14 monoclonal antibod-ies raised against Begomovirus particles, Y8, Y36 and Y38had similar antigenic reaction in TAS-ELISA as Tomato yel-low leaf curl China virus (TYLCCNV). The complete DNA-A nucleotide sequences of Y8, Y36 and Y38 were determinedand they contain 2727, 2730 and 2730 nucleotides, respec-tively. Each of the DNA-A sequences has a typical Bego-movirus genome organization encoding 6 ORFs with 2 ORFs [AV1(CP) and AV2] in virion-sense DNA and 4 ORFs (AC1to AC4) in complementary-sense DNA. Comparisons withtotal DNA-A, intergenic region and deduced amino acid se-quences of individual ORFs show that Y8, Y36 and Y38 are isolates of TYLCCNV. | 14 | Microstructure and strengthening mechanism of hot-extruded ultralight Mg-Li-Al-Sn alloys with high strength显示文摘Duplex-structured Mg-7Li-2Al-1.5Sn alloys with high strength were fabricated and their strengthening mechanism was investigated.The Mg-7Li-2Al-1.5Sn alloys were prepared by casting and extruded at the temperature of 533 K with an extrusion ratio of 25:1.The microstructure and mechanical properties of Mg-7Li-2Al-1.5Sn alloys were systematically investigated by OM,XRD,SEM,TEM,and tensile tests.The results show that Mg-7Li-2Al-1.5Sn alloys are mainly composed ofα-Mg,β-Li,LiMgAl_(2),Mg_(2)Sn and Li_(2)MgSn phases.The yield strength(YS),ultimate tensile strength(UTS)and elongation(EL)of the extruded alloy at room temperature reach 250 MPa,324 MPa and 11.9%,respectively.A lot of Sn-rich precipitates(Mg_(2)Sn and Li_(2)MgSn)are precipitated during extrusion with an average size of∼14 nm,which is beneficial to the grain refinement.Dynamic recrystallization occurs during hot deformation and the nanoprecipitates effectively refine the dynamic recrystallized(DRXed)grains.Besides,the residual dislocations existed in DRXed and un-DRXed grains result in the dislocation strengthening in the extruded alloy.Mg7Li-2Al-1.5Sn alloys possess excellent high-temperature mechanical properties with the YS,UTS and EL of 200 MPa,237 MPa and 26.7%at 423 K,respectively.Sn-rich precipitates with good thermal stability can effectively prevent grain growth,which is good for the improvement of the high-temperature performance of Mg-Li-Al-Sn alloy. | Gang Zhou Yan Yang Hanzhu Zhang Faping Hu Xueping Zhang Chen Wen Weidong Xie Bin Jiang Xiaodong Peng Fusheng Pan | 2022 | Journal of Materials Science & Technology2022,,8: | 7 |
| 15 | Identification of a novel DNA molecule associated with Tobacco leaf curl virus显示文摘A circular DNA molecule, designated as DNAp, was identified in tobacco plants infected with Tobacco leaf curl virus (TLCV) isolates Y5 and Y8 by PCR using primers based on the conserved region of the two reported DNAp sequences of whitefly-transmitted geminiviruses (WTGs). The complete nucleotide sequences of DNAβ of Y5 and Y8 (TLCV DNAβ) were determined. Y5 DNAβ comprises 1333 nucleotides encoding 8 predicted ORFs with 4 ORFs in virion-sense DNA and 4 ORFs in complementary-sense DNA; Y8 DNAP consists of 1338 nucleotides encoding 7 predicted ORFs with 4 ORFs in virion-sense DNA and 3 ORFs in complementary-sense DNA. | 17 | The pathogenicity on legumes of Cucumber mosaic virus was determined by 243 nucleotides on 2a polymerase gene of viral RNA2显示文摘We had isolated and identified two Cucumber mosaic virus (CMV) isolates, the CMV red bean (CMV-RB) isolate and the CMV pea (CMV-P1) isolate. CMV-RB induces necrotic local lesions on inoculated leaves of broad bean, pea, cowpea and bean, and could not infect these hosts systemically. However, CMV-P1 was able to infect these legumes systemically. To study the difference of pathogenic-ity on the legumes induced by these two CMV isolates, the full-length infectious cDNA clones of CMV-Fny, which induced similar symptoms as CMV-RB in the four legumes, were used. The 243 nucleotides fragment, which encodes highly conserved GDD amino acid motif on 2a replicase gene of CMV-Fny RNA2, was replaced with that of CMV-P1. The constructed chimeric virus FP could infect these legumes systemically. | 20 | Monoclonal Antibody-Based Serological Detection of Rice Stripe Mosaic Virus Infection in Rice Plants or Leafhoppers显示文摘Rice stripe mosaic virus(RSMV) is a rhabdovirus recently found in southern part of China and can cause severe reduction in rice production. To establish serological methods for RSMV epidemiological studies and to establish a control strategy for this virus, we first purified RSMV virions from infected rice plants and then used them as an immunogen to produce four RSMV-specific monoclonal antibodies(MAbs)(i.e.,1D4, 4A8, 8E4 and 11F11). With these MAbs, we have developed a highly specific and sensitive antigen-coated plate enzyme-linked immunosorbent assay(ACP-ELISA), a Dot-ELISA and a Tissue print-ELISA for rapid detections of RSMV infection in rice plants or in leafhoppers. Our results showed that RSMV can be readily detected in RSMV-infected rice plant tissue crude extracts diluted at 1:20,971,520(w/v, g/m L)through ACP-ELISA or diluted at 1:327,680(w/v, g/m L) through Dot-ELISA. Both ACP-ELISA and Dot-ELISA can also be used to detect RSMV infection in individual RSMV viruliferous leafhopper(Recilia dorsalis) homogenate diluted at 1:307,200 and 1:163,840(individual leafhopper/l L), respectively. Detection of RSMV infection in field-collected rice samples or in RSMV viruliferous leafhoppers indicated that the three serological methods can produce same results with that produced by RT-PCR(19 of the 33 rice samples and 5 of the 16 leafhoppers were RSMV-positive).
