1Comparative Immunization in BALB/c Mice with Recombinant Replication-Defective Adenovirus Vector and DNA Plasmid Expressing a SARS-CoV Nucleocapsid Protein Gene显示文摘In order to investigate immunogenicity in the induction of humoral and cellular immune responses, severe acute respiratory syndrome associated coronavirus (SARS-CoV)-N gene recombinant replication-defective adenoviral vector, rAd-N, was generated and immunized BALB/c mice in a pcDNA3.1-N prime-rAd-N boost regimen. After humoral and cellular immune response detection, different levels of SARS-CoV N protein specific antibodies and interferon-γ (IFN-γ) secretion are shown compared to controls. The humoral immune response was induced more effectively by the DNA priming and recombinant adenovirus boosting regimen. There is a significant difference between heterogeneous and homologous vaccinations. The heterogeneous combinations were all higher than those of the homologous combinations in the induction of anti-N antibody response. Among the three heterogeneous combinations, pcDNA3.1-N/pcDNA3.1-N/pcDNA3.1-N/rAd-N induced the strongest antibody response. In the induction of IFN-γ production, the homologous combination of rAd-N/rAd-N/rAd-N/rAd-N was significantly stronger than that of pcDNA3.1-N/pcDNA3.1-N/pcDNA3.1-N/pcDNA3.1-N, but was relatively weaker than the heterogeneous combination of pcDAN3.1-N/pcDAN3.1-N/pcDAN3.1-N/rAd-N. This combination was a most efficient immunization regimen in induction of SARS-CoV-N-specific (IFN-γ) secretion just as the antibody response. 2Autotuning algorithm of particle swarm PID parameter based on D-Tent chaotic model显示文摘An improved particle swarm algorithm based on the D-Tent chaotic model is put forward aiming at the standard particle swarm algorithm. The convergence rate of the late of proposed algorithm is improved by revising the inertia weight of global optimal particles and the introduction of D-Tent chaotic sequence. Through the test of typical function and the autotuning test of proportionalintegral-derivative(PID) parameter, finally a simulation is made to the servo control system of a permanent magnet synchronous motor(PMSM) under double-loop control of rotating speed and current by utilizing the chaotic particle swarm algorithm. 3Theoretical Calculations and Experimental Verification for the Pumping Effect Caused by the Dynamic Micro-tapered Angle显示文摘The atomizer with micro cone apertures has advantages of ultra-fine atomized droplets, low power consumption and low temperature rise. The current research of this kind of atomizer mainly focuses on the performance and its application while there is less research of the principle of the atomization. Under the analysis of the dispenser and its micro-tapered aperture's deformation, the volume changes during the deformation and vibration of the micro-tapered aperture on the dispenser are calculated by coordinate transformation. Based on the characters of the flow resistance in a cone aperture, it is found that the dynamic cone angle results from periodical changes of the volume of the micro-tapered aperture of the atomizer and this change drives one-way flows. Besides, an experimental atomization platform is established to measure the atomization rates with different resonance frequencies of the cone aperture atomizer. The atomization performances of cone aperture and straight aperture atomizers are also measured. The experimental results show the existence of the pumping effect of the dynamic tapered angle. This effect is usually observed in industries that require low dispersion and micro- and nanoscale grain sizes, such as during production of high-pressure nozzles and inhalation therapy. Strategies to minimize the pumping effect of the dynamic cone angle or improve future designs are important concerns. 5Integrating a model with remote sensing observations by a data assimilation approach to improve the model simulation accuracy of carbon flux and evapotranspiration at two flux sites显示文摘Model simulation and in situ observations are often used to research water and carbon cycles in terrestrial ecosystems, but each of these methods has its own advantages and limitations. Combining these two methods could improve the accuracy of quantifying the dynamics of the water and carbon fluxes of an ecosystem. Data assimilation is an effective means of integrating modeling with in situ observation. In this study, the ensemble Kalman filter(En KF) and the unscented Kalman filter(UKF) algorithms were used to assimilate remotely sensed leaf area index(LAI) data with the Biome-BGC model to simulate water and carbon fluxes at the Harvard Forest Environmental Monitoring Site(EMS) and the Dinghushan site. After MODIS LAI data from 2000–2004 were assimilated into the improved Biome-BGC model using the En KF algorithm at the Harvard Forest site, the R2 between the simulated and observed results for NEE and evapotranspiration increased by 7.8% and 4.7%, respectively. In addition, the sum of the absolute error(SAE) and the root mean square error(RMSE) of NEE decreased by an average of 21.9% and 26.3%, and the SAE and RMSE of evapotranspiration decreased by 24.5% and 25.5%, respectively. MODIS LAI data of 2003 were assimilated into the Biome-BGC model for the Dinghushan site, and the R2 values between the simulated and observed results for NEE and evapotranspiration were increased by 6.7% and 17.3%, respectively. In addition, the SAE values of NEE and ET were decreased by 11.3% and 30.7%, respectively, and the RMSE values of NEE and ET decreased by 10.1% and 30.9%, respectively. 6Comparative transcriptome analysis of different heat stress responses between self-root grafting line and heterogeneous grafting line in rose显示文摘Rose is sensitive to high temperature which will make the rose go into a semi-dormancy state.Grafting is an excellent way to enhance rose heat tolerance.Here,heat-tolerant Rosa multiflora‘Huanong Wuci 1'(W)and heat-sensitive Rosa chinensis‘Old Blush’(X)were selected as experimental materials.The RNA-seq technique was used to investigate the transcriptomes of self-root grafting line(XX0),heterogeneous grafting line(XW0),self-root grafting line under 6 h heat stress(XX6),and heterogeneous grafting line under 6 h heat stress(XW6).Under high temperature stress,multiple signaling pathways were activated,moreover,a large number of transcription factors and functional genes were induced,especially the HSFs and HSPs with remarkably upregulated expression levels.The GO analysis showed that the differences in the expressions of the genes related to fatty acids and carbohydrates were observed between self-root grafting line and heterogeneous grafting line.In addition,14 P450s were differentially expressed,and one lectin gene was up-regulated in XW0 vs XW6,but down-regulated in XX0 vs XX6.Considering physiological and biochemical traits such as relative electrolyte leakage,SOD activity,proline,and total soluble sugars,DEGs involved in these processes may be key factors to resist high temperature.The present study provides an insight into the complex mechanism underlying grafting in response to heat stress.Our results indicate that grafting is an effective way to improve rose heat resistance.Wenquan Qi Chunling Zhang Wenjing Wang Zhe Cao Song Li Hang Li Wan Zhu Yongqi Huang Manzhu Bao Yanhong He Riru Zheng 2021Horticultural Plant Journal2021,7,3:3
7On the boundedness of running-in attractors based on recurrence plot and recurrence qualification analysis显示文摘A feature parameter was proposed to quantitatively explore the boundedness of running-in attractors;its variation throughout the friction process was also investigated. The enclosing radius R was built with recurrence plots(RPs) and recurrence qualification analysis(RQA) by using the time delay embedding and phase space reconstruction. Additionally, the typology of RPs and the recurrence rate(RR) were investigated to verify the applicability of R in characterizing the friction process. Results showed that R is larger at the beginning, but exhibits a downward trend in the running-in friction process;R becomes smooth and trends to small steady values during the steady-state friction period, and finally shows an upward trend until failure occurs. The evolution of R, which corresponded with the typology of RPs and RR during friction process, can be used to quantitatively analyze the variation of the running-in attractors and friction state identifacation. 8Effects of subsoiling on working quality and total power consumption of high stubble straw returning machine显示文摘Straw returning into field is a direct and effective measure to reduce the straw burning and improve the soil organic matter content.Straw returning directly to field needs higher performance machines,especially under the condition of large amount of straw in the field is more difficult.Therefore,the model of conservation tillage by combination of subsoiling and straw returning was studied.Experiments on combined tillage machine for effect of subsoiling on working quality and total power consumption for high stubble straw returning were carried out.The high stubble rape field was used as the test field;forward speed and PTO speed of tractor were taken as the test factors.Straw coverage rate and straw proportion of the lower half burying layer were taken as the test indexes of the working quality.Subsoiling and rotary burying(SRB)returning operation was used as experimental group and direct rotary burying(DRB)returning operation was the control group.The results showed that under different working conditions,the mean value of straw coverage rate of SRB was 93.0%,straw proportion of the lower half burying layer was 52.8%,these values were better than DRB.The straw proportion of the lower half burying layer of SRB compared with DRB increased by 10.5%.Two factors all had a significant effect on it under the SRB and DRB conditions.Subsoiling could significantly reduce the PTO torque.Under low speed,the total power consumption of SRB was slightly smaller,while under high speed,the total power consumption of DRB was slightly smaller.Under the SRB and DRB conditions,two factors both had a significant effect on total power consumption.The optimal working combination(working quality as the primary index)was 1.5 km/h of forward speed and 720 r/min of PTO speed.Under this condition,the straw coverage rate was 94.1%,the straw proportion of the lower half burying layer was 59.0%,and the total power consumption was 35.62 kW.The research confirmed that subsoiling is beneficial to the working quality and total power consumption of high stubble straw returning machine.It could meet the working requirements,and provide a reference for optimizing straw returning machine and improving working quality.Hua Zhou Jumin Zhang Junfang Xia H.M.Tahir Yinghao Zhu Chunling Zhang 2019International Journal of Agricultural and Biological Engineering2019,12,4:2

10An oral second-generation proteasome inhibitor oprozomib significantly inhibits lung cancer in a p53 independent manner in vitro显示文摘The destruction of proteins via the ubiquitin-proteasome system is amulti-step,complex process involving polyubiquitination of substrate proteins,follow ed by proteolytic degradation by the m acrom olecular 26S proteasome complex.Inhibitors of the proteasome prom ote the accum ulation of proteins that are deleterious to cell survival and are prom ising anticancer agents.Oprozomib(OPZ),an oral second-generation proteasome inhibitor,has been shown to inhibit the grow th of several cancers in preclinical and clinical trials,including m ultiple m yelom a and head and neck cancers,but its effects on lung cancer has not yet been determ ined.In this study,we evaluated the inhibitory effects of OPZ on lung cancer cell lines in vitro.The results showed that OPZ significantly suppressed cell proliferation and strongly induced apoptosis in both tested lung cancer cells independent of p53 expression.OPZ was able to cause obvious caspase 3 and PARP cleavages and stabilize p53 and its transcriptional targets p21,PUMA,and Noxa.Moreover,OPZ was capable of sensitizing lung cancer cells to the conventional chem otherapeutic drug cisplatin.Our study provides preclinical data and sheds light on the potential applications of proteasome inhibitor OPZ in lung cancer treatment.Hongge Zhu Tianhai Wang Zhou Xin Yiyi Zhan Guoming Gu Xiaoqin Li Xiuli Wang Shune Yang Chunling Liu 2019Acta Biochimica et Biophysica Sinica2019,51,10:2
11Qualitative analysis for state/event fault trees using formal model checking显示文摘A state/event fault tree(SEFT)is a modeling technique for describing the causal chains of events leading to failure in software-controlled complex systems.Such systems are ubiquitous in all areas of everyday life,and safety and reliability analyses are increasingly required for these systems.SEFTs combine elements from the traditional fault tree with elements from state-based techniques.In the context of the real-time safety-critical systems,SEFTs do not describe the time properties and important timedependent system behaviors that can lead to system failures.Further,SEFTs lack the precise semantics required for formally modeling time behaviors.In this paper,we present a qualitative analysis method for SEFTs based on transformation from SEFT to timed automata(TA),and use the model checker UPPAAL to verify system requirements’properties.The combination of SEFT and TA is an important step towards an integrated design and verification process for real-time safety-critical systems.Finally,we present a case study of a powerboat autopilot system to confirm our method is viable and valid after achieving the verification goal step by step.JIANG Quan ZHU Chunling WANG Siqi 2019Journal of Systems Engineering and Electronics2019,30,5:2

14Clinicopathological characteristics of 8697 patients with COVID-19 in China:a meta-analysis显示文摘Objective Our study aims to present a summary of the clinicopathological characteristics of patients affected by the coronavirus disease 2019(COVID-19)that can be used as a reference for further research and clinical decisions.Design Studies were included in the meta-analysis if they had cohort,case-control or case series designs and provided sufficient details on clinical symptoms,laboratory outcomes and asymptomatic patients.setting PubMed,Embase,Chinese Biomedical Literature Database,Wanfang,China Science and Technology Journal Database and China National Knowledge Infrastructure databases were electronically searched to identify related studies published between 1 January 2020 and 16 March 2020.Three reviewers independently examined the literature,extracted relevant data and assessed the risk of publication bias before including the studies in the meta-analysis.Participants The confirmed cases of COVID-19.results A total of 55 unique retrospective studies involving 8697 patients with COVID-19 were identified.Meta-analysis showed that a higher proportion of infected patients were male(53.3%),and the two major symptoms observed were fever(78.4%)and cough(58.3%).Other common symptoms included fatigue(34%),myalgia(21.9%),expectoration(23.7%),anorexia(22.9%),chest tightness(22.9%)and dyspnoea(20.6%).Minor symptoms included nausea and vomiting(6.6%),diarrhoea(8.2%),headache(11.3%),pharyngalgia(11.6%),shivering(15.2%)and rhinorrhea(7.3%).About 5.4%of the patients were asymptomatic.Most patients showed normal leucocyte counts(64.7%)and elevated C reactive protein levels(65.9%).Lymphopaenia was observed in about 47.6%of the infected patients,along with abnormal levels of myocardial enzymes(49.4%)and liver function(26.4%).Other findings included leucopenia(23.5%),elevated D-dimer(20.4%),elevated erythrocyte sedimentation rate(20.4%),leucocytosis(9.9%),elevated procalcitonin(16.7%)and abnormal renal function(10.9%).Conclusions The most commonly experienced symptoms of patients with COVID-19 were fever and cough.Myalgia,anorexia,chest tightness and dyspnoea were found in some patients.A relatively small percentage of patients were asymptomatic and could act as carriers of the disease.Most patients showed normal leucocyte counts,elevated levels of C reactive protein and lymphopaenia,confirming the viral origin of the disease.Jieyun Zhu Zhimei Zhong Pan Ji Hongyuan Li Bocheng Li Jielong Pang Jianfeng Zhang Chunling Zhao 2020Family Medicine and Community Health2020,8,2:1

19Understanding the macroscopical flexibility/fragility of nanoporous Ag:Depending on network connectivity and micro-defects显示文摘The effective engineering applications of nanoporous metals(NPMs)in flexible energy storage and wearable healthcare biosensor monitoring require its uniform ligaments network,specifically crack-free and flexible monolithic bodies.However,the macroscopic fragility of NPMs restricts their applications in wearable electronics fields.Here we focus on the synthetization of highly flexible NPMs.The effects of structural factors,e.g.ligament-network connectivity and micro-cracks on the mechanical properties of nanoporous Ag(np Ag)are investigated.The well-interconnected np Ag metal exhibits higher tensile strength,nanohardness and Vickers hardness than those for the ill-interconnected np Ag metal.The quality of the network connectivity dominates the strength and hardness of the np Ag.The flexibility/fragility is determined by the micro-crack in np Ag.The crack-free np Ag exhibits good flexible behavior.When micro-cracks are introduced,the np Ag becomes fragile.The control of soft volume shrinkage rate(Vsr)and slow surface diffusivity(Ds)effectively suppresses the crack initiation and propagation of as-formed np Ag.These results provide useful insights to synthesize more flexible and crack-free NPM materials for effective use in public wearable electronics and diverse flexible engineering applications in the future.Chaoyang Wang Shengli Zhu Yanqin Liang Zhenduo Cui Shuilin Wu Chunling Qin Shuiyuan Luo Akihisa Inoue 2020Journal of Materials Science & Technology2020,52,18:1
