| 4 | Chemical compound cinobufotalin potently induces FOXO1-stimulated cisplatin sensitivity by antagonizing its binding partner MYH9显示文摘In this study,we present novel molecular mechanisms by which FOXO1 functions as a tumor suppressor to prevent the pathogenesis of nasopharyngeal carcinoma(NPC).First,we observed that FOXO1 not only controlled tumor stemness and metastasis,but also sensitized NPC cells to cisplatin(DDP)in vitro and in vivo.Mechanistic studies demonstrated that FOXO1-induced miR-200b expression through the GSK3β/β-catenin/TCF4 network-mediated stimulation of ZEB1,which reduced tumor stemness and the epithelial–mesenchymal transition(EMT)signal.Furthermore,we observed FOXO1 interaction with MYH9 and suppression of MYH9 expression by modulating the PI3K/AKT/c-Myc/P53/miR-133a-3p pathway.Decreased MYH9 expression not only reduced its interactions with GSK3β,but also attenuated TRAF6 expression,which then decreased the ubiquitin-mediated degradation of GSK3βprotein.Increased GSK3βexpression stimulated theβ-catenin/TCF4/ZEB1/miR-200b network,which increased the downstream tumor stemness and EMT signals.Subsequently,we observed that chemically synthesized cinobufotalin(CB)strongly increased FOXO1-induced DDP chemosensitivity by reducing MYH9 expression,and the reduction in MYH9 modulated GSK3β/β-catenin and its downstream tumor stemness and EMT signal in NPC.In clinical samples,the combination of low FOXO1 expression and high MYH9 expression indicated the worst overall survival rates.Our studies demonstrated that CB potently induced FOXO1-mediated DDP sensitivity by antagonizing its binding partner MYH9 to modulate tumor stemness in NPC. | YongHao Li Xiong Liu Xian Lin Menyang Zhao Yanyi Xiao Chen Liu Zixi Liang Zelong Lin Renhui Yi Zibo Tang Jiahao Liu Xin Li Qingping Jiang Libo Li Yinyin Xie Zhen Liu Weiyi Fang | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 6 |
| 5 | Immobilization of insulin-like growth factor-1 onto thermosensitive hydrogels to enhance cardiac progenitor cell survival and differentiation under ischemic conditions显示文摘Stem cell therapy is a promising approach to treat myocardial infarction. However, direct delivery of stem cells into hearts experiences poor cell engraftment and differentiation, due to ischemic conditions(low nutrient and oxygen) in the infarct hearts. Development of suitable cell carriers capable of supporting cell survival and differentiation under these harsh conditions is critical for improving the efficacy of current stem cell therapy. In this work, we created a family of novel cell carriers based on thermosensitive hydrogels and insulin-like growth factor 1(IGF-1), and investigated if these cell carriers can improve cell survival and differentiation under ischemic conditions. The thermosensitive hydrogels were synthesized from N-isopropylacryla- mide, acrylic acid, acrylic acid N-hydroxysuccinicimide ester, and 2-hydroxyethyl methacrylate-oligo(hydroxybutyrate). The hydrogel solutions can be readily injected through 26G needles, and can quickly solidify at 37 oC to form highly flexible hydrogels. IGF-1 was immobilized into the hydrogels in order to support long-term cell survival and differentiation. Different amount of IGF-1 was immobilized by using hydrogels with different content of N-hydroxysuccinicimide ester groups. Cardiosphere derived cells were encapsulated in the hydrogels and cultured under ischemic conditions. The results demonstrated that a significant improvement of cell survival and differentiation was achieved after IGF-1 immobilization. | 6 | Copy number alteration profiling facilitates differential diagnosis between ossifying fibroma and fibrous dysplasia of the jaws显示文摘Ossifying fibroma(OF)and fibrous dysplasia(FD)are two fibro-osseous lesions with overlapping clinicopathological features,making diagnosis challenging.In this study,we applied a whole-genome shallow sequencing approach to facilitate differential diagnosis via precise profiling of copy number alterations(CNAs)using minute amounts of DNA extracted from morphologically correlated microdissected tissue samples.Freshly frozen tissue specimens from OF(n=29)and FD(n=28)patients were obtained for analysis.Lesion fibrous tissues and surrounding normal tissues were obtained by laser capture microdissection(LCM),with~30–50 cells(5000–10000µm2)per sample.We found that the rate of recurrent CNAs in OF cases was much higher(44.8%,13 of 29)than that in FD cases(3.6%,1 of 28).Sixty-nine percent(9 of 13)of the CNA-containing OF cases involved segmental amplifications and deletions on Chrs 7 and 12.We also identified eight CNA-associated genes(HILPDA,CALD1,C1GALT1,MICALL2,PHF14,AIMP2,MDM2,and CDK4)with amplified expression,which was consistent with the copy number changes.We further confirmed a jaw lesion with a previous uncertain diagnosis due to its ambiguous morphological features and the absence of GNAS mutation as OF based on the typical Chr 12 amplification pattern in its CNA profile.Moreover,analysis of a set of longitudinal samples collected from an individual with a cellular lesion in suspicion of OF at the first surgery,recurrence and the latest malignant transformation revealed identical CNA patterns at the three time points,suggesting that copy number profiling can be used as an important tool to identify borderline lesions or lesions with malignant potential.Overall,CNA profiling of fibro-osseous lesions can greatly improve differential diagnosis between OF and FD and help predict disease progression. | Ming Ma Lu Liu Ruirui Shi Jianyun Zhang Xiaotian Li Xuefen Li Jiaying Bai Jianbin Wang Yanyi Huang Tiejun Li | 2021 | International Journal of Oral Science2021,13,2: | 3 |
| 7 | Improvement in the risk assessment of oral leukoplakia through morphology-related copy number analysis显示文摘Oral leukoplakia is the most common type of oral potentially malignant disorders and considered a precursor lesion to oral squamous cell carcinoma.However,a predictor of oral leukoplakia prognosis has not yet been identified.We investigated whether copy number alteration patterns may effectively predict the prognostic outcomes of oral leukoplakia using routinely processed paraffin sections.Comparison of copy number alteration patterns between oral leukoplakia with hyperplasia(HOL,n=22)and dysplasia(DOL,n=21)showed that oral leukoplakia with dysplasia had a higher copy number alteration rate(86%)than oral leukoplakia with hyperplasia(46%).Oral leukoplakia with dysplasia exhibited a wider range of genomic variations across all chromosomes compared with oral leukoplakia with hyperplasia.We also examined a retrospective cohort of 477 patients with oral leukoplakia with hyperplasia with detailed follow-up information.The malignant transformation(MT,n=19)and leukoplakia recurrence(LR,n=253)groups had higher frequencies of aneuploidy events and copy number loss rate than the free of disease(FD,n=205)group.Together,our results revealed the association between the degree of copy number alterations and the histological grade of oral leukoplakia and demonstrated that copy number alteration may be effective for prognosis prediction in oral leukoplakia patients with hyperplasia. | Xiaotian Li Lu Liu Jianyun Zhang Ming Ma Lisha Sun Xuefen Li Heyu Zhang Jianbin Wang Yanyi Huang Tiejun Li | 2021 | Science China(Life Sciences)2021,64,9: | 2 |
| 8 | Experimental Study on Hydrogen Leakage and Emission of Fuel Cell Vehicles in Confined Spaces显示文摘Hydrogen safety is one of the most important safety indicators in fuel cell vehicles(FCVs)(unlike in other types of alternative energy vehicles).This indicator in FCVs is directly related to the user’s personal safety in daily vehicle usage.This paper analyzes the safety standards of FCVs in confined spaces.A sealed test chamber and an appropriate test method are devel-oped to evaluate vehicle safety based on specific test requirements.Two FCVs are subjected to static hydrogen leakage and hydrogen emission testing performed in a confined space.The results reveal that the hydrogen concentration in the vicinity of the vehicles approximates 0.0004%which is much lower than 1%while parked for 8 h during the hydrogen leakage test.In the hydrogen emission test under operating conditions,the concentration of the hydrogen gas emitted from the vehicles exceeds 2300 ppm in the vicinity,which requires careful consideration.Based on experiment and analysis,recommendations for the hydrogen safety standards of FCVs in confined spaces are proposed. | Dong Hao Xiaobing Wang Yanyi Zhang Renguang Wang Guang Chen Jun Li | 2020 | Automotive Innovation2020,3,2: | 2 |

| 12 | Optical frequency synthesizer referenced to an ytterbium optical clock显示文摘Optical clocks with an unprecedented accuracy of 10-18 promise innovations in precision spectroscopy and measurement. To harness the full power of optical clocks, we need optical frequency synthesizers(OFSs) to accurately convert the stabilities and accuracies of optical clocks to other desired frequencies. This work demonstrates such an OFS referenced to an ytterbium optical clock. The OFS is based on an optical frequency comb phase-locked to a commercial rubidium microwave clock;in this way most combs can operate robustly. Despite comb frequency instability at 10^(-11), the synthesis noise and uncertainty reach 6 × 10^(-18)(1 s) and 5 × 10^(-21), respectively, facilitating frequency synthesis of the best optical clocks. The analysis of nucleic acids and nucleic acids-based analysis have been widely applied in biological studies, clinical diagnosis, environmental analysis, food safety and forensic analysis.During the past decades, the field of nucleic acids analysis has been rapidly advancing with many technological breakthroughs.In this review, we focus on the methods developed for analyzing nucleic acids, nucleic acids-based analysis, device for nucleic acids analysis, and applications of nucleic acids analysis. | 20 | A virtual sequencer reveals the dephasing patterns in error-correction code DNA sequencing显示文摘An error-correction code(ECC)sequencing approach has recently been reported to effectively reduce sequencing errors by interrogating a DNA fragment with three orthogonal degenerate sequencing-by-synthesis(SBS)reactions.However,similar to other non-single-molecule SBS methods,the reaction will gradually lose its synchronization within a molecular colony in ECC sequencing.This phenomenon,called dephasing,causes sequencing error,and in ECC sequencing,induces distinctive dephasing patterns.To understand the characteristic dephasing patterns of the dual-base flowgram in ECC sequencing and to generate a correction algorithm,we built a virtual sequencer in silico.Starting from first principles and based on sequencing chemical reactions,we simulated ECC sequencing results,identified the key factors of dephasing in ECC sequencing chemistry and designed an effective dephasing algorithm.The results show that our dephasing algorithm is applicable to sequencing signals with at least 500 cycles,or1000-bp average read length,with acceptably low error rate for further parity checks and ECC deduction.Our virtual sequencer with our dephasing algorithm can further be extended to a dichromatic form of ECC sequencing,allowing for a potentially much more accurate sequencing approach. | Wenxiong Zhou Li Kang Haifeng Duan Shuo Qiao Louis Tao Zitian Chen Yanyi Huang | 2021 | National Science Review2021,8,5: | 0 |
