SARS-CoV-2 spike protein interacts with and activates TLR41显示文摘Dear Editor,Accumulating clinical data suggest the main causes of death by COVID-19 include respiratory failure and the onset of sepsis.1 Importantly, sepsis has been observed in nearly all deceased patients.2,3,4,5 It remains elusive how SARS-CoV-2 infection results in viral sepsis in humans.Toll-like receptor 4(TLR4)mediates anti-gram-negative bacterial immune responses by recognizing lipopolysaccharide(LPS)from bacteria.6 We recently found that SARS-CoV-2 infection provoked an anti-bacterial like response at the very early stage of infection via TLR4.However,the identity of the original trigger initiating these abnormal immune responses during SARS-CoV-2 infection is unknown. | Yingchi Zhao Ming Kuang Junhong Li Ling Zhu Zijing Jia Xuefei Guo Yaling Hu Jun Kong Hang Yin Xiangxi Wang Fuping You | 2021 | Cell Research2021,31,7: | 3
Molecular imaging of advanced atherosclerotic plaques with folate receptor-targeted 2D nanoprobes显示文摘Vulnerable atherosclerotic plaques are responsible for most cardiovascular diseases(CVDs).Folate receptor(FR)positive activated macrophages were thought to be a prominent component in the development of vulnerable plaque. The objective of this study is to develop folate conjugated two-dimensional(2D)Pd@Au nanomaterials(Pd@Au-PEG-FA)for targeted multimodal imaging of the FRs in advanced atherosclerotic plaques.Pharmacokinetic and imaging studies(single photon emission computed tomography(SPECT),computed tomography(CT)and photoacoustic(PA)imaging)were performed to confirm the prolonged blood half-life and enrichment of radioactivity in atherosclerotic plaques.Strong signals were detected in vivo with SPECT,CT and PA imaging in heavy atherosclerotic plaques, which were significantly higher than those of the normal aortas after injection of Pd@Au-PEG-FA.Blocking studies with preinjection of excess FA could effectively reduce the targeting ability of Pd@Au-PEG-FA in atherosclerotic plaques, further demonstrating the specific binding of Pd@Au-PEG-FA for plaque lesions.Histopathological characterization revealed that the signal of probe was in accordance with the high-risk plaques. In summary, the Pd@Au-PEG-FA has favorable pharmacokinetic properties and provides a valuable approach for detecting high-risk plaques in the presence of FRs in atherosclerotic plaques. | Zhide Guo Liu Yang Mei Chen Xuejun Wen Huanhuan Liu Jingchao Li Duo Xu Yuanyuan An Changrong Shi Jindian Li Xinhui Su Zijing Li Ting Liu Rongqiang Zhuang Nanfeng Zheng Haibo Zhu Xianzhong Zhang | 2020 | Nano Research2020,13,1: | 2

Gravitational wave astronomy: the current status显示文摘In the centenary year of Einstein's General Theory of Relativity, this paper reviews the current status of gravitational wave astronomy across a spectrum which stretches from attohertz to kilohertz frequencies. Sect. 1 of this paper reviews the historical development of gravitational wave astronomy from Einstein's first prediction to our current understanding the spectrum. It is shown that detection of signals in the audio frequency spectrum can be expected very soon, and that a north-south pair of next generation detectors would provide large scientific benefits. Sect. 2 reviews the theory of gravitational waves and the principles of detection using laser interferometry. The state of the art Advanced LIGO detectors are then described. These detectors have a high chance of detecting the first events in the near future. Sect. 3 reviews the KAGRA detector currently under development in Japan, which will be the first laser interferometer detector to use cryogenic test masses. Sect. 4 of this paper reviews gravitational wave detection in the nanohertz frequency band using the technique of pulsar timing. Sect. 5 reviews the status of gravitational wave detection in the attohertz frequency band, detectable in the polarisation of the cosmic microwave background, and discusses the prospects for detection of primordial waves from the big bang. The techniques described in sects. 1–5 have already placed significant limits on the strength of gravitational wave sources. Sects. 6 and 7 review ambitious plans for future space based gravitational wave detectors in the millihertz frequency band. Sect. 6 presents a roadmap for development of space based gravitational wave detectors by China while sect. 7 discusses a key enabling technology for space interferometry known as time delay interferometry. | BLAIR David JU Li ZHAO ChunNong WEN LinQing CHU Qi FANG Qi CAI RongGen GAO JiangRui LIN XueChun LIU Dong WU Ling-An ZHU ZongHong REITZE David H. | 5 | SnO-SnO_(2) modified two-dimensional MXene Ti3C2Tx for acetone gas sensor working at room temperature显示文摘Acetone,as widely used reagents in industry and laboratories, are extremely harmful to the human. So the detection of acetone gas concentrations and leaks in special environments at room temperature is essential.Herein, the nanocomposite combining SnO-SnO_(2)(p-n junction)and Ti_(3)C_(2)T_(x) MXene was successfully synthesized by a one-step hydrothermal method.Because of the existence of a small amount of oxygen during the hydrothermal conditions, part of the p-type SnO was oxidized to n-type SnO_(2),forming in-situ p-n junctions on the surface of Sn O.The hamburger-like SnO-SnO_(2)/Ti_(3)C_(2)T_(x) sensor exhibited improved acetone gas sensing response of 12.1(R_(g)/R_(a))at room temperature, which were nearly 11 and 4 times higher than those of pristine Ti_(3)C_(2)T_(x) and pristine SnO-SnO_(2),respectively.Moreover,it expressed a short recovery time(9 s)and outstanding reproducibility.Because of the different work functions, the Schottky barrier was formed between the SnO and the Ti_(3)C_(2)T_(x) nanosheets, acting as a hole accumulation layer(HALs)between Ti_(3)C_(2)T_(x) and tin oxides.Herein, the sensing mechanism based on the formation of hetero-junctions and high conductivity of the metallic phase of Ti_(3)C_(2)T_(x) MXene in SnO-SnO_(2)/Ti_(3)C_(2)T_(x) sensors was discussed in detail. | Zijing Wang Fen Wang Angga Hermawan Yusuke Asakura Takuya Hasegawa Hiromu Kumagai Hideki Kato Masato Kakihana Jianfeng Zhu Shu Yin | 2021 | Journal of Materials Science & Technology2021,,14: | 1
The repression of oncoprotein SET by the tumor suppressor p53 reveals a p53-SET-PP2A feedback loop for cancer therapy显示文摘The oncoprotein SET is frequently overexpressed in many types of tumors and contributes to malignant initiation and progression through multiple mechanisms, including the hijacking of the tumor suppressors p53 and PP2A.Targeting aberrant SET represents a promising strategy for cancer intervention. However, the mechanism by which endogenous SET is regulated in cancer cells remains largely unknown. Here,we identified the tumor suppressor p53 as a key regulator that transcriptionally repressed the expression of SET in both normal and cancer cells. In addition,p53 stimulated PP2A phosphatase activity via p53-mediated transcriptional repression of SET,whereby SET-mediated inhibition of PP2A was alleviated. Moreover, targeting the interaction between SET and PP2A catalytic subunit(PP2Ac)with FTY720 enhanced stress-induced p53 activation via PP2A-mediated dephosphorylation of p53 on threonine 55(Thr55).Therefore,our findings uncovered a previously unknown p53-SETPP2A regulatory feedback loop.To functionally potentiate this feedback loop,we designed a combined therapeutic strategy by simultaneously administrating a p53 activator and SET antagonist in cancer cells and observed a dramatic synergistic effect on tumor suppression. Our study reveals mechanistic insight into the regulation of the oncoprotein SET and raises a potential strategy for cancer therapy by stimulating the p53-SET-PP2A feedback loop. | Han Yao Wenbin Xu Yajing Liu Zhijie Cao Jia Wen Mi Zhang Zhen Wu Xiaojun Yan Zishan Jiao Zijing Zhang Jianyuan Chen Meng Zhang Wei-Guo Zhu Donglai Wang | 2023 | Science China(Life Sciences)2023,66,1: | 0
Multimedia distribution and health risk assessment of typical organic pollutants in a retired industrial park显示文摘The overall cross-media risk evaluation of organic pollutants in retired industrial parks is insufficiently recognized. In this study,11 semi-volatile organic compounds(SVOCs)and 27 volatile organic compounds(VOCs)were measured in 531 soil and groundwater samples taken from a retired industrial park by coast in Zhejiang Province,China.Total petroleum hydrocarbons(TPHs),Di(2-ethylhexyl)phthalate(DEHP),benzene,and ethylbenzene were identified as the critical pollutants in the soil, while TPHs,1,2-dichloropropane(1,2-DCP),toluene,benzo[a]anthracene(BaA),and benzo[b]fluoranthene(BbF)were identified as critical pollutants in the groundwater for exceeding China national standards. The spatial correlation between the concentrations of organic pollutants in soil and groundwater was explored by employing the Geodetector model.Based on the results of spatial interpolation,high-risk hotspots regarding soil and groundwater pollution were identified. Moreover, the possible harm to human health of the critical pollutants were also under evaluation.Among various critical pollutants, benzene, ethylbenzene, and DEHP in soil, and 1,2-DCP in groundwater, were the main contributors to the overall health risk of multimedia pollution. This study developed a comprehensive approach to assess the risks posed by specific organic toxicants in various environmental media. The findings of this work can serve as a valuable reference for future management strategies in retired industrial parks. | Shijin Wu Zijing Xiang Daohui Lin Lizhong Zhu | 2023 | Frontiers of Environmental Science & Engineering2023,17,11: | 0
Weakened humoral immune responses of inactivated SARS-CoV-2 vaccines in patients with solid tumors显示文摘Dear Editor Coronavirus disease 19(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),remains a pandemic.Cancer patients have a higher risk of poor outcomes for SARS-CoV-2 infection than the general population[1].Vaccines were shown to effectively prevent SARS-CoV-2 infection, severe disease progression, and mortality.Inactivated SARS-CoV-2 vaccines(BBIBPCorV and CoronaVac)have been approved and widely used in China, with the former shown to be more effective than the latter[2].Hence,there is an urgent need to investigate the safety and humoral immune responses of inactivated vaccines in cancer patients. | Zhiwei Chen Peng Zhu Zuojin Liu Bin Zhu Guobing Yin Jia Ming Rui Song Qingbo Pan Tong Li Xingwei Jiang Bing Wang Shuaibin Liu Hongxing Cai Jingjie Wang Yuling Han Zijing Lin Yang Hong Min Chen Mingli Peng Lina Hu Dachuan Cai Hong Ren | 2023 | Cancer Communications2023,43,2: | 0
Highly pathogenic coronavirus N protein aggravates inflammation by MASP-2-mediated lectin complement pathway overactivation显示文摘Excessive inflammatory responses contribute to the pathogenesis and lethality of highly pathogenic human coronaviruses, but the underlying mechanism remains unclear. In this study, the N proteins of highly pathogenic human coronaviruses, including severe acute respiratory syndrome coronavirus(SARS-CoV),middle east respiratory syndrome coronavirus(MERS-CoV)and severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),were found to bind MASP-2,a key serine protease in the lectin pathway of complement activation, resulting in excessive complement activation by potentiating MBL-dependent MASP-2 activation, and the deposition of MASP-2,C4b,activated C3 and C5b-9.Aggravated inflammatory lung injury was observed in mice infected with adenovirus expressing the N protein.Complement hyperactivation was also observed in SARS-CoV-2-infected patients.Either blocking the N protein:MASP-2 interaction,MASP-2 depletion or suppressing complement activation can significantly alleviate N protein-induced complement hyperactivation and lung injury in vitro and in vivo.Altogether, these data suggested that complement suppression may represent a novel therapeutic approach for pneumonia induced by these highly pathogenic coronaviruses. | Ting Gao Lin Zhu Hainan Liu Xiaopeng Zhang Tingting Wang Yangbo Fu Hongzhen Li Qincai Dong Yong Hu Zhang Zhang Jing Jin Zijing Liu Weihong Yang Yaoning Liu Yanwen Jin Kaitong Li Yongjiu Xiao Junli Liu Huailong Zhao Yue Liu Ping Li Jibo Song Lu Zhang Yuwei Gao Sisi Kang Shoudeng Chen Qingjun Ma Xiuwu Bian Wei Chen Xuan Liu Qing Mao Cheng Cao | 2022 | Signal Transduction and Targeted Therapy2022,7,10: | 0
Vitamin D receptor (VDR) mediates the quiescence of activated hepatic stellate cells (aHSCs) by regulating M2 macrophage exosomal smooth muscle cell-associated protein 5 (SMAP-5)显示文摘An effective therapeutic regimen for hepatic fibrosis requires a deep understanding of the pathogenesis mechanism.Hepatic fibrosis is characterized by activated hepatic stellate cells(aHSCs)with an excessive production of extracellular matrix.Although promoted activation of HSCs by M2 macrophages has been demonstrated, the molecular mechanism involved remains ambiguous.Herein,we propose that the vitamin D receptor(VDR)involved in macrophage polarization may regulate the communication between macrophages and HSCs by changing the functions of exosomes. We confirm that activating the VDR can inhibit the effect of M2 macrophages on HSC activation. The exosomes derived from M2 macrophages can promote HSC activation, while stimulating VDR alters the protein profiles and reverses their roles in M2 macrophage exosomes.Smooth muscle cell-associated protein 5(SMAP-5)was found to be the key effector protein in promoting HSC activation by regulating autophagy flux.Building on these results,we show that a combined treatment of a VDR agonist and a macrophage-targeted exosomal secretion inhibitor achieves an excellent anti-hepatic fibrosis effect. In this study,we aim to elucidate the association between VDR and macrophages in HSC activation. The results contribute to our understanding of the pathogenesis mechanism of hepatic fibrosis, and provide potential therapeutic targets for its treatment. | Xuwentai LIU Yue WU Yanyi LI Kaiming LI Siyuan HOU Ming DING Jingmin TAN Zijing ZHU Yingqi TANG Yuming LIU Qianhui SUN Cong WANG Can ZHANG | 2023 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,3: | 0
