| 3 | Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. Further evaluation showed that p SM3-CAR effectively caused tumor necrosis, providing new options for improved CAR-T therapy in solid tumors. | Fengtao You Licui Jiang Bozhen Zhang Qiang Lu Qiao Zhou Xiaoyang Liao Hong Wu Kaiqi Du Youcai Zhu Huimin Meng Zhishu Gong Yunhui Zong Lei Huang Man Lu Jirong Tang Yafen Li Xiaochen Zhai Xiangling Wang Sisi Ye Dan Chen Lei Yuan Lin Qi Lin Yang | 2016 | Science China(Life Sciences)2016,59,4: | 16 |

| 5 | A novel pathway regulates social hierarchy via lncRNA AtLAS and postsynaptic synapsin Ⅱb显示文摘Dominance hierarchy is a fundamental phenomenon in grouped animals and human beings, however, the underlying regulatory mechanisms remain elusive. Here,we report that an antisense long non-coding RNA(lncRNA)of synapsinⅡ;named as AtLAS,plays a crucial role in the regulation of social hierarchy.AtLAS is decreased in the prefrontal cortical excitatory pyramidal neurons of dominant mice;consistently, silencing or overexpression of AtLAS increases or decreases the social rank, respectively.Mechanistically,we show that AtLAS regulates alternative polyadenylation of synapsinⅡgene and increases synapsin 2b(syn2b)expression.Syn2b reduces AMPA receptor(AMPAR)-mediated excitatory synaptic transmission through a direct binding with AMPAR at the postsynaptic site via its unique C-terminal sequence. Moreover,a peptide disrupting the binding of syn2b with AMPARs enhances the synaptic strength and social ranks. These findings reveal a novel role for lncRNA AtLAS and its target syn2b in the regulation of social behaviors by controlling postsynaptic AMPAR trafficking. | Mei Ma Wan Xiong Fan Hu Man-Fei Deng Xian Huang Jian-Guo Chen Heng-Ye Man Youming Lu Dan Liu Ling-Qiang Zhu | 2020 | Cell Research2020,30,2: | 3 |
| 6 | Scoring the collective effects of SNPs: association of minor alleles with complex traits in model organisms显示文摘It has long been assumed that most parts of a genome and most genetic variations or SNPs are non-functional with regard to reproductive fitness. However, the collective effects of SNPs have yet to be examined by experimental science. We here developed a novel approach to examine the relationship between traits and the total amount of SNPs in panels of genetic reference populations. We identified the minor alleles(MAs)in each panel and the MA content(MAC)that each inbred strain carried for a set of SNPs with genotypes determined in these panels.MAC was nearly linearly linked to quantitative variations in numerous traits in model organisms, including life span, tumor susceptibility, learning and memory, sensitivity to alcohol and anti-psychotic drugs, and two correlated traits poor reproductive fitness and strong immunity. These results suggest that the collective effects of SNPs are functional and do affect reproductive fitness. | YUAN DeJian ZHU ZuoBin TAN XiaoHua LIANG Jie ZENG Chen ZHANG JieGen CHEN Jun MA Long DOGAN Ayca BROCKMANN Gudrun GOLDMANN Oliver MEDINA Eva RICE Amanda D. | 9 | Stimulated Brillouin scattering induced all-optical modulation in graphene microfiber显示文摘Graphene microfibers are burgeoning modulators with great potential in all-optical communication. One of the critical issues that remains to be understood is the dynamic mechanism of light–graphene interaction. Here, we propose a power dependent modulation by using 980 nm pump light and 1064 nm signal light via graphene microfiber, and the results show a strong transmission reduction and frequency blue shift with the increase of pump power. The experimental observation is attributed to a stimulated Brillouin scattering process induced by the pump light. Power and frequency variations are a result of energy transition of the scattered phonon in the fiber. This work reveals the nonlinear effect process in the light–graphene interaction and provides a new method for power and frequency control with graphene all-optical modulation. | JIWEN ZHU XUEMEI CHENG YALI LIU RUIDUO WANG MAN JIANG DIAO LI BAOLE LU ZHAOYU REN | 2019 | Photonics Research2019,7,1: | 1 |
