1A novel colloidal gold immunochromatography assay strip for the diagnosis of schistosomiasis japonica in domestic animals显示文摘Background:Schistosomiasis remains a major public health concern in China and an epidemiological survey has revealed that schistosome-infected bovines and goats are the main transmission sources for the disease.Therefore,development of a sensitive technique for the diagnosis of schistosomiasis in domestic animals is necessary.Method:A novel colloidal gold immunochromatography assay(GICA)strip was developed for detecting Schistosoma japonicum in domestic animals.The colloidal gold was conjugated with recombinant streptococcal protein G(rSPG).As the test and control lines,the schistosome soluble egg antigen and rSPG,respectively,were blotted on nitrocellulose membrane.Results:The lowest detectable serum dilution was 1∶640 for schistosome-infected buffaloes.The cross-reaction rate of GICA was 14.29%with Paramphistomum sp.in buffaloes,16.67%with Haemonchus sp.in goats,and 33.33%with Orientobilharzia sp.in goats.These results were slightly lower and similar to those obtained through ELISA.Moreover,the strips for detecting S.japonicum in mice,rabbits,buffaloes,and goats showed high sensitivity(100.00%,100.00%,100.00%,and 100.00%,respectively)and specificity(100.00%,100.00%,94.23%,and 88.64%,respectively).And the sensitivity or specificity of the GICA strips did not present any significant differences after storage for 12 months at room temperature.When compared with ELISA,the GICA strips exhibited similar sensitivity and specificity in the diagnosis of schistosomiasis in mice,rabbits,buffaloes,and goats.Besides,only 5μl of serum are required for the test and the detection can be completed within 5 min.Conclusion:This study is the first to develop a GICA strip using gold-rSPG conjugate for the diagnosing of schistosomiasis in domestic animals,and preliminary results showed that the developed strip may be suitable for large-scale screening of schistosomiasis in endemic areas.Rui Xu Jintao Feng Yang Hong Chao Lv Dengyun Zhao Jiaojiao Lin Ke Lu Hao Li Jinming Liu Xiaodan Cao Tao Wang Jinli Zai Zhaozhe Wang Bingguang Jia Qian Han Chuangang Zhu 2017Infectious Diseases of Poverty2017,6,1:12
2Tissue-specific transcription reprogramming promotes liver metastasis of colorectal cancer显示文摘Metastasis,the development of secondary malignant growths at a distance from a primary tumor,is the cause of death for 90%of cancer patients,but little is known about how metastatic cancer cells adapt to and colonize new tissue environments.Here,using clinical samples,patient-derived xenograft(PDX)samples,PDX cells,and primary/metastatic cell lines,we discovered that liver metastatic colorectal cancer(CRC)cells lose their colon-specific gene transcription program yet gain a liver-specific gene transcription program.We showed that this transcription reprogramming is driven by a reshaped epigenetic landscape of both typical enhancers and super-enhancers.Further,we identified that the liver-specific transcription factors FOXA2 and HNF1A can bind to the gained enhancers and activate the liver-specific gene transcription,thereby driving CRC liver metastasis.Importantly,similar transcription reprogramming can be observed in multiple cancer types.Our data suggest that reprogrammed tissue-specific transcription promotes metastasis and should be targeted therapeutically.Shuaishuai Teng Yang Eric Li Ming Yang Rui Qi Yiming Huang Qianyu Wang Yanmei Zhang Shanwen Chen Shasha Li Kequan Lin Yang Cao Qunsheng Ji Qingyang Gu Yujing Cheng Zai Chang Wei Guo Pengyuan Wang Ivan Garcia-Bassets Zhi John Lu Dong Wang 2020Cell Research2020,30,1:11

4Immunity-and-matrix-regulatory cells derived from human embryonic stem cells safely and effectively treat mouse lung injury and fibrosis显示文摘Lung injury and fibrosis represent the most significant outcomes of severe and acute lung disorders,including COVID-19.However,there are still no effective drugs to treat lung injury and fibrosis.In this study,we report the generation of clinical-grade human embryonic stem cells(hESCs)-derived immunity-and matrix-regulatory cells(IMRCs)produced under good manufacturing practice requirements,that can treat lung injury and fibrosis in vivo.We generate IMRCs by sequentially differentiating hESCs with serum-free reagents.IMRCs possess a unique gene expression profile distinct from that of umbilical cord mesenchymal stem cells(UCMSCs),such as higher expression levels of proliferative,immunomodulatory and anti-fibrotic genes.Moreover,intravenous delivery of IMRCs inhibits both pulmonary inflammation and fibrosis in mouse models of lung injury,and significantly improves the survival rate of the recipient mice in a dose-dependent manner,likely through paracrine regulatory mechanisms.IMRCs are superior to both primary UCMSCs and the FDA-approved drug pirfenidone,with an excellent efficacy and safety profile in mice and monkeys.In light of public health crises involving pneumonia,acute lung injury and acute respiratory distress syndrome,our findings suggest that IMRCs are ready for clinical trials on lung disorders.Jun Wu Dingyun Song Zhongwen Li Baojie Guo Yani Xiao Wenjing Liu Lingmin Liang Chunjing Feng Tingting Gao Yanxia Chen Ying Li Zai Wang Jianyan Wen Shengnan Yang Peipei Liu Lei Wang Yukai Wang Liang Peng Glyn Nigel Stacey Zheng Hu Guihai Feng Wei Li Yan Huo Ronghua Jin Ng Shyh-Chang Qi Zhou Liu Wang Baoyang Hu Huaping Dai Jie Hao 2020Cell Research2020,30,9:7
5Multifractal characteristics of shale and tight sandstone pore structures with nitrogen adsorption and nuclear magnetic resonance显示文摘Based on the experiments of nitrogen gas adsorption(N_2 GA) and nuclear magnetic resonance(NMR),the multifractal characteristics of pore structures in shale and tight s andstone from the Chang 7 member of Trias sic Yanchang Formation in Ordos Basin,NW China,are investigated.The multifractal spectra obtained from N2 GA and NMR are analyzed with pore throat structure parameters.The results show that the pore size distributions obtained from N2 GA and NMR are different,and the obtained multifractal characteristics vary from each other.The specific surface and total pore volume obtained by N2 GA experiment have correlations with multifractal characteristics.For the core samples with the similar specific surface,the value of the deviation of multifractal spectra Rd increases with the increase in the proportion of large pores.When the proportion of macropores is small,the Rd value will increase with the increase in specific surface.The multifractal characteristics of pore structures are influenced by specific surface area,average pore size and adsorption volume measured from N2 GA experiment.The multifractal characteristic parameters of tight sandstone measured from NMR spectra are larger than those of shale,which may be caused by the differences in pore size distribution and porosity of shale and tight sandstone.Fu-Yong Wang Kun Yang Yun Zai 2020Petroleum Science2020,17,5:4
6The LRXs-RALFs-FER module controls plant growth and salt stress responses by modulating multiple plant hormones显示文摘Salt stress is a major environmental factor limiting plant growth and productivity.We recently discovered an important new salt tolerance pathway,where the cell wall leucine-rich repeat extensins LRX3/4/5,the RAPID ALKALINIZATION FACTOR(RALF)peptides RALF22/23 and receptor-like kinase FERONIA(FER)function as a module to simultaneously regulate plant growth and salt stress tolerance.However,the intracellular signaling pathways that are regulated by the extracellular LRX3/4/5-RALF22/23-FER module to coordinate growth,cell wall integrity and salt stress responses are still unknown.Here,we report that the LRX3/4/5-RALF22/23-FER module negatively regulates the levels of jasmonic acid(JA),salicylic acid(SA)and abscisic acid(ABA).Blocking JA pathway rescues the dwarf phenotype of the lrx345 and fer-4mutants,while disruption of ABA biosynthesis suppresses the salt-hypersensitivity of these mutants.Many salt stress-responsive genes display abnormal expression patterns in the lrx345 and fer-4 mutants,as well as in the wild type plants treated with epigallocatechin gallate(EGCG),an inhibitor of pectin methylesterases,suggesting cell wall integrity as a critical factor that determines the expression pattern of stress-responsive genes.Production of reactive oxygen species(ROS)is constitutively increased in the lrx345 and fer-4mutants,and inhibition of ROS accumulation suppresses the salt-hypersensitivity of these mutants.Together,our work provides strong evidence that the LRX3/4/5-RALF22/23-FER module controls plant growth and salt stress responses by regulating hormonal homeostasis and ROS accumulation.Chunzhao Zhao Wei Jiang Omar Zayed Xin Liu Kai Tang Wenfeng Nie Yali Li Shaojun Xie Yuan Li Tiandan Long Linlin Liu Yingfang Zhu Yang Zhao Jian-Kang Zhu 2021National Science Review2021,8,1:3

19Antioxidative Effect of Schiff Bases with o-Hydroxybenzylidene Groupon Free Radical Induced Hemolysis of Human Red Blood Cell显示文摘The hemolysis of human red blood cells was initiated by a water soluble free radical initiator, 2,2′ azobis (2 amidinopropane hydrochloride)(AAPH), at 37 ℃ in phosphate buffered saline(pH=7.4). The respective addition of 1 [ N (o hydroxybenzylidene)amino]tetradecane(TDCA), 1,2 di[ N (o hydroxybenzylidene)amino]cyclohexane(DACH), 4 [ N (o hydroxybenzylidene)amino]benzoic acid(PABA), 4 nitro[ N (o hydroxybenzylidene)]aniline(APNA) or N (o hydroxybenzylidene)aniline(APA) can all prolong the inhibition period of hemolysis, indicating that the above Schiff bases play an antioxidative role in free radical induced hemolysis. It can be concluded that Schiff base with an alkyl group or a conjugated system in the molecule protect red blood cells against free radical induced hemolysis efficiently.
