| 13 | Application of StrucGP in medical immunology:site-specific N-glycoproteomic analysis of macrophages显示文摘The structure of N-glycans on specific proteins can regulate innate and adaptive immunity via sensing environmental signals.Meanwhile,the structural diversity of N-glycans poses analytical challenges that limit the exploration of specific glycosylation functions.In this work,we used THP-1-derived macrophages as examples to show the vast potential of a N-glycan structural interpretation tool StrucGP in N-glycoproteomic analysis.The intact glycopeptides of macrophages were enriched and analyzed using mass spectrometry(MS)-based glycoproteomic approaches,followed by the large-scale mapping of site-specific glycan structures via StrucGP.Results revealed that bisected GlcNAc,core fucosylated,and sialylated glycans(e.g.,HexNAc4Hex5Fuc1Neu5Ac1,N4H5F1S1)were increased in M1 and M2 macrophages,especially in the latter.The findings indicated that these structures may be closely related to macrophage polarization.In addition,a high level of glycosylated PD-L1 was observed in M1 macrophages,and the LacNAc moiety was detected at Asn-192 and Asn-200 of PD-L1,and Asn-200 contained Lewis epitopes.The precision structural interpretation of site-specific glycans and subsequent intervention of target glycoproteins and related glycosyltransferases are of great value for the development of new diagnostic and therapeutic approaches for different diseases. | Pengfei Li Zexuan Chen Shanshan You Yintai Xu Zhifang Hao Didi Liu Jiechen Shen Bojing Zhu Wei Dan Shisheng Sun | 2023 | Frontiers of Medicine2023,17,2: | 0 |
| 14 | Celastrol targeting Nedd4 reduces Nrf2-mediated oxidative stress in astrocytes after ischemic stroke显示文摘Stroke is the second leading cause of death worldwide,and oxidative stress plays a crucial role.Celastrol exhibits strong antioxidant properties in several diseases;however,whether it can affect oxidation in cerebral ischemic-reperfusion injury(CIRI)remains unclear.This study aimed to determine whether celastrol could reduce oxidative damage during CIRI and to elucidate the underlying mechanisms.Here,we found that celastrol attenuated oxidative injury in CIRI by upregulating nuclear factor E2-related factor 2(Nrf2).Using alkynyl-tagged celastrol and liquid chromatography-tandem mass spectrometry,we showed that celastrol directly bound to neuronally expressed developmentally downregulated 4(Nedd4)and then released Nrf2 from Nedd4 in astrocytes.Nedd4 promoted the degradation of Nrf2 through K48-linked ubiquitination and thus contributed to astrocytic reactive oxygen species production in CIRI,which was significantly blocked by celastrol.Furthermore,by inhibiting oxidative stress and astrocyte activation,celastrol effectively rescued neurons from axon damage and apoptosis.Our study uncovered Nedd4 as a direct target of celastrol,and that celastrol exerts an antioxidative effect on astrocytes by inhibiting the interaction between Nedd4 and Nrf2 and reducing Nrf2 degradation in CIRI. | Zexuan Hong Jun Cao Dandan Liu Maozhu Liu Mengyuan Chen Fanning Zeng Zaisheng Qin Jigang Wang Tao Tao | 2023 | Journal of Pharmaceutical Analysis2023,13,2: | 0 |
| 15 | Mechanical properties and strengthening mechanism of silty sands stabilized with steel slag-based geopolymer binder显示文摘Geopolymer binder has the advantages of early strength,fast solidification,high volume stability,and low permeability.It is beneficial to improve the mechanical performance of silty sands,saving cement consumption and being environmentally friendly.However,the strength improvement of silty sand stabilized with steel slag-based geopolymer was significantly controlled by their material composition and technical parameters.This study conducted a series of unconfined compression tests to investigate the material composition of steel slag-based geopolymer binders and their reasonable mixing ratio for silty sand stabilization.The optimum mixing ratio of precursor(steel slag)to alkaline activator(the combination of Na2SiO3 and CaO)and the optimum dosage of steel slag-based geopolymer for silty sand stabilization were explored.The strengthening mechanism of geopolymer-stabilized silty sands was discussed based on microstructural images and elemental concentrations of primary components observed by SEM and EDS.The results show that when the mass ratio of steel slag:Na2SiO3:CaO was 80:35:21,and the steel slag-based geopolymer material was 15%,the silty sand could achieve the best mechanical performance improvement.The microstructural characteristics of geopolymer-stabilized silty sands at different curing ages illustrated that the compactness and integrity of silty sand structures were enhanced over the curing age.The improving cementitious contact among particles and enlarging particle size was responsible for the strength improvement of silty sand.This research can provide a reference for applying steel slag-based geopolymer in silty sand stabilization in engineering practices. | Shengnian Wang Zewei Chen Zhijian Wu Leilei Gu Jun Su Zexuan Zhao | 2022 | Emergency Management Science and Technology2022,2,1: | 0 |
