1Whole-genome sequencing of 508 patients identifies key molecular features associated with poor prognosis in esophageal squamous cell carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC)is a poor-prognosis cancer type with limited understanding of its molecular etiology.Using 508 ESCC genomes,we identified five novel significantly mutated genes and uncovered mutational signature clusters associated with metastasis and patients’outcomes.Several functional assays implicated that NFE2L2 may act as a tumor suppressor in ESCC and that mutations in NFE2L2 probably impaired its tumor-suppressive function,or even conferred oncogenic activities.Additionally,we found that the NFE2L2 mutations were significantly associated with worse prognosis of ESCC.We also identified potential noncoding driver mutations including hotspot mutations in the promoter region of SLC35E2 that were correlated with worse survival.Approximately 5.9%and 15.2%of patients had high tumor mutation burden or actionable mutations,respectively,and may benefit from immunotherapy or targeted therapies.We found clinically relevant coding and noncoding genomic alterations and revealed three major subtypes that robustly predicted patients’outcomes.Collectively,we report the largest dataset of genomic profiling of ESCC useful for developing ESCC-specific biomarkers for diagnosis and treatment.Yongping Cui Hongyan Chen Ruibin Xi Heyang Cui Yahui Zhao Enwei Xu Ting Yan Xiaomei Lu Furong Huang Pengzhou Kong Yang Li Xiaolin Zhu Jiawei Wang Wenjie Zhu Jie Wang Yanchun Ma Yong Zhou Shiping Guo Ling Zhang Yiqian Liu Bin Wang Yanfeng Xi Ruifang Sun Xiao Yu Yuanfang Zhai Fang Wang Jian Yang Bin Yang Caixia Cheng Jing Liu Bin Song Hongyi Li Yi Wang Yingchun Zhang Xiaolong Cheng Qimin Zhan Yanhong Li Zhihua Liu-Show 2020Cell Research2020,30,10:19

3BAALC-AS1/G3BP2/c-Myc feedback loop promotes cell proliferation in esophageal squamous cell carcinoma显示文摘Background:Long non-codingRNAs(lncRNAs)have been found to be involved in the development of many cancers.In this study,we aimed to identify the molecular mechanisms of lncRNA BAALC antisense RNA 1(BAALC-AS1)in regulating the malignancy of esophageal squamous cell carcinoma(ESCC).Methods:The expression of BAALC-AS1 in cancer patients was analyzed using a tissue microarray.The protein and RNA levels of BAALC-AS1 were determined by Western blotting analysis and quantitative reverse transcription-PCR(RT-qPCR),respectively.The cell proliferation was determined by cell viability assays,bromodeoxyuridine incorporation,and flow cytometry.The relationships among BAALC-AS1,RasGAPSH3 domain-binding protein 2(G3BP2),and c-Myc were determined using RNA immunoprecipitation,RNA pull-down assays,and luciferase assays.Results:The expression of BAALC-AS1 was highly up-regulated and associated with malignant phenotypes in ESCC tissues and cell lines.In vivo and in vitro assays showed that BAALC-AS1 promoted ESCC cell proliferation,migration,and invasion.BAALC-AS1 directly interacted with G3BP2,and thereby inhibited the degradation of c-Myc RNA 3’-UTR by G3BP2,thus leading to the accumulation of c-Myc expression.Additionally,c-Myc acted as a transcription factor that can induce the expression of BAALC-AS1 by directly binding to its promoter region.Conclusions:BAALC-AS1/G3BP2/c-Myc feedback loop plays a critical role in the development of ESCC,which might provide a novel therapeutic target and facilitate the development of new therapeutic strategies for the treatment of ESCC.Hongyue Zhang Yan Wang Weimin Zhang Qingnan Wu Jiawen Fan Qimin Zhan 2021Cancer Communications2021,41,3:7

5Dynamic analysis of DNA damage induced miRNAs in colon cancer cells显示文摘It is known that microRNAs(miRNAs)expression profile shows substantial changes in cells under DNA damage.Here,we did miRNA microarray and quantitative real-time PCR to comprehensively identify the differentially expressed miRNAs in colon cancer cell lines HCT116 p53/and HCT116 p53-/-.Cluster analysis revealed a panel of differentially expressed miRNAs which are regulated by p53and/or UV-C induced DNA damage.These altered miRNAs tend to be located in chromosomes 13,X and 17.Moreover,pathways enrichment analysis estimated that MAPK pathway,focal adheren pathway,p53 pathway and Wnt pathway were mediated by these miRNAs to exert their functions in DNA damage response.Additionally,we found that miR-320a,one of the UV-C induced miRNAs,play a role in protecting cells from DNA damage.Taken together,our results show that miRNAs are dynamic regulated in p53-dependent or-independent manners in different cell contexts and different situations following DNA damage.Weimin Zhang Fang Yang Qimin Zhan 2014Chinese Science Bulletin2014,59,19:4

8Wireless/wired integrated transmission for industrial cyber-physical systems:risk-sensitive co-design of 5G and TSN protocols显示文摘The growing popularity of intelligent manufacturing is driven by deterministic transmission demands of applications in industrial cyber-physical systems(ICPS).However,the ossified shortages of industrial wireless communication such as diverse quality of service(Qo S)and complex signaling processes incur a severe long tail of transmission delay distribution.As a solution,the 5th generation(5 G)wireless communication technology provides ultra-reliable and low-latency communication(URLLC)for industry scenarios.Moreover,the newly proposed time-sensitive networking(TSN)standards guarantee the transmission determinacy by gate mechanism.In this paper,we propose a heterogeneous time-sensitive network(HTSN)co-designed by 5 G and TSN.We first develop a predictive multi-priority wireless scheduling mechanism based on semi-persistent scheduling(SPS)to reduce signaling delay by reserving resources in advance.Then we propose an adaptive data injection mechanism for TSN based on per-stream filtering and policing(PSFP),which dynamically adjusts the priority of data for queue injection in TSN.To further reduce the long tail of delay,we employ a risk-sensitive learning method to improve the worst-case delay.Simulations on a hot rolling production scenario demonstrate that the proposed mechanisms under HTSN achieve great performance in terms of integrated delay and resource utilization.Yajing ZHANG Qimin XU Xinping GUAN Cailian CHEN Mingyan LI 2022Science China(Information Sciences)2022,65,1:1

11CDK7-YAP-LDHD axis promotes D-lactate elimination and ferroptosis defense to support cancer stem cell-like properties显示文摘Reprogrammed cellular metabolism is essential for maintaining cancer stem cells(CSCs)state.Here,we report that mitochondrial D-lactate catabolism is a necessary initiating oncogenic event during tumorigenesis of esophageal squamous cell carcinoma(ESCC).We discover that cyclin-dependent kinase 7(CDK7)phosphorylates nuclear Yes-associated protein 1(YAP)at S127 and S397 sites and enhances its transcription function,which promotes D-lactate dehydrogenase(LDHD)protein expression.Moreover,LDHD is enriched significantly in ESCC-CSCs rather than differentiated tumor cells and high LDHD status is connected with poor prognosis in ESCC patients.Mechanistically,the CDK7-YAP-LDHD axis helps ESCC-CSCs escape from ferroptosis induced by D-lactate and generates pyruvate to satisfy energetic demands for their elevated self-renewal potential.Hence,we conclude that esophageal CSCs adopt a D-lactate elimination and pyruvate accumulation mode dependent on CDK7-YAP-LDHD axis,which drives stemness-associated hallmarks of ESCC-CSCs.Reasonably,targeting metabolic checkpoints may serve as an effective strategy for ESCC therapy.Mengzhu Lv Ying Gong Xuesong Liu Yan Wang Qingnan Wu Jie Chen Qingjie Min Dongyu Zhao Xianfeng Li Dongshao Chen Di Yang Danna Yeerken Rui Liu Jinting Li Weimin Zhang Qimin Zhan 2023Signal Transduction and Targeted Therapy2023,8,9:1

16Defect of SLC38A3 promotes epithelial-mesenchymal transition and predicts poor prognosis in esophageal squamous cell carcinoma显示文摘Objective: Solute carrier family 38(SLC38 s) transporters play important roles in amino acid transportation and signaling transduction. However, their genetic alterations and biological roles in tumors are still largely unclear.This study aimed to elucidate the genetic signatures of SLC38 s transporters and their implications in esophageal squamous cell carcinoma(ESCC).Methods: Analyses on somatic mutation and copy number alterations(CNAs) of SLC38 A3 were performed as described. Immunohistochemistry(IHC) assay and Western blot assay were used to detect the protein expression level. MTS assay, colony formation assay, transwell assay and wound healing assay were used to explore the malignant phenotypes of ESCC cells. Immunofluorescence assay was used to verify the colocalization of two indicated proteins and immunopreciptation assay was performed to confirm the interaction of proteins.Results: Our findings revealed that SLC38 s family was significantly disrupted in ESCC, with high frequent CNAs and few somatic mutations. SLC38 A3 was the most frequent loss gene among them and was linked to poor survival and lymph node metastasis. The expression of SLC38 A3 was lower in tumor tissues compared to that in normal tissues, which was also significantly associated with worse clinical outcome. Further experiments revealed that depletion of SLC38 A3 could promote EMT in ESCC cell lines, and the interaction of SLC38 A3 and SETDB1 might lead to the reduced transcription of Snail.
