1Pressure balance and imbalance in the optic nerve chamber: The Beijing Intracranial and Intraocular Pressure (iCOP) Study显示文摘To determine the interdependence of intracranial pressure(ICP) and intraocular pressure(IOP) and how it affects optic nerve pressures, eight normal dogs were examined using pressure-sensing probes implanted into the left ventricle, lumbar cistern, optic nerve subarachnoid space in the left eye, and anterior chamber in the left eye. This allowed ICP, lumbar cistern pressure(LCP), optic nerve subarachnoid space pressure(ONSP) and IOP to be simultaneously recorded. After establishing baseline pressure levels, pressure changes that resulted from lowering ICP(via shunting cerebrospinal fluid(CSF) from the ventricle) were recorded. At baseline, all examined pressures were different(ICP>LCP>ONSP), but correlated(P<0.001). As ICP was lowered during CSF shunting, IOP also dropped in a parallel time course so that the trans-lamina cribrosa gradient(TLPG) remained stable(ICP-IOP dependent zone). However, once ICP fell below a critical breakpoint, ICP and IOP became uncoupled and TLPG changed as ICP declined(ICP-IOP independent zone). The optic nerve pressure gradient(ONPG) and trans-optic nerve pressure gradient(TOPG) increased linearly as ICP decreased through both the ICP-IOP dependent and independent zones. We conclude that ICP and IOP are coupled in a specific pressure range, but when ICP drops below a critical point, IOP and ICP become uncoupled and TLPG increases. When ICP drops, a rise in the ONPG and TOPG creates more pressure and reduces CSF flow around the optic nerve. 2Reliable screening of pesticide residues in maternal and umbilical cord sera by gas chromatography-quadrupole time of flight mass spectrometry显示文摘The widespread use of pesticides induces heavy adverse effects on human health,especially for the pregnant women and the newborns.In this study,a screening method has been developed for the determination of multi-pesticides in maternal and umbilical cord sera.All pesticides in sera were collected using solid phase extraction(SPE),and analyzed by gas chromatography-quadrupole time of flight mass spectrometry(GC-QTOF MS).To set up the quality criteria,a database of 50 pesticides was created and the accurate masses of 3 up to 5 representative ions with their intensity ratios were included for each pesticide.In addition,a novel'identification points'(IPs)system relying on the accurate MS1 and MS2 spectra was used to interpret the data for each suspected pesticide.The methodology was then applied to a pair of maternal and umbilical cord sera.A total of six pesticide residues were screened out successfully.In conclusion,GC-QTOF MS combined with an accurate mass database seemed to be one of the most efficient tools for systematic pesticide analysis.FAN RuoJing ZHANG Fang WANG HaoYang ZHANG Li ZHANG Jing ZHANG Ying YU ChongTian GUO YinLong 2014Science China Chemistry2014,57,5:3
3Separation of injectable salidroside by column chromatography of macroporous resins for treating myocardial ischemia显示文摘The objective of the present study is to develop a method for large-scale separating and purifying salidroside from rhodiola kirilowii roots and for preparing injectable medicinal ingredient.Crude extract of salidroside was prepared by water-ethanol system,and purified by column chromatography of macroporous resins.Static adsorption and desorption studies were performed on six kinds of macroporous resins,and SP825 resin was chosen,followed by optimizing process parameters.The optimum sample volume,feed concentration,ratio of diameter to height,and feeding flow rate were 1.5 bed volumes(BV),15 mg/mL,1:10 and 1 BV/h,respectively.Dynamic desorption was performed consecutively with 8 BV of distilled water,3 BV of 5% ethanol and 8 BV of 10% ethanol at a flow rate of 2 BV/h.After three cycles in separating 3.5 tons of rhodiola kirilowii roots,salidroside purity was increased from 3.4% in the crude extract to 93.6% in purified salidroside product.This study provides a novel method to separate salidroside for injectable use.JU RuiJun HUANG RenJie ZHOU Jia LI RuoJing ZHOU Peng ZHANG ZaoHua XIANG FeiJun XU DongJin LIU WeiXiang MA XingTian ZHANG Qiang LU WanLiang 2012Science China Chemistry2012,55,7:2

8Modeling of micropipette aspiration and optical tweezers stretching of erythrocytes with or without Malaria parasite显示文摘The erythrocytes play an important role in the human body.The healthy erythrocytes can undergo extremely large deformation while passing through small capillaries.Their infection by Malaria Plasmodium falcipurum(P.f.) will lead to capillary blockage and blood flow obstruction.Many experimental and computational methods have been applied to study the increase in stickiness and decrease in deformability of the Malaria(P.f.) infected erythrocytes.The novelty of this paper lies in the establishment of an multi-component model for investigating mechanical properties of Malaria(P.f.) infected erythrocytes,especially of their enclosed parasites.Finite element method was applied to simulate the erythrocytes' deformation in micropipette aspiration and optical tweezers stretching using the computational software ABAQUS.The comparisons between simulations and experiments were able to quantitatively conclude the effects of stiffness and stickiness of the parasitophorous vacuole membrane on the cells' deformation,which could not be obtained from experiments directly.Guyue Jiao Ruojing Zhang 2013Theoretical & Applied Mechanics Letters2013,3,3:0
