| 1 | Characteristics of the microbial communities in the integrated vertical-flow constructed wetlands显示文摘Microorganisms play an important role in removing pollutants from constructed wetlands.We investigated the microbial characteristics in a novel integrated vertical-flow constructed wetland(IVCW) ,which has been in operation in Wuhan,China since 1998.We used phospholipid fatty acid(PLFA) and amoA gene to analyze the structure and diversity of the microbial community within the IVCW.PLFA results suggested that the amount of bacterial PLFA was significantly higher than that of fungal PLFA,but the total microbial biomass represented by PLFA index was low in the system.Microbial spatial distribution showed significantly higher bacterial(both G+and G-) and fungal biomass in the surface than in the subsurface layers.The ratios of monounsaturated to branched PLFA demonstrated that an anaerobic layer sandwiched by two aerobic layers existed in the IVCW,consistent with the redox potential results.Analysis of the amoA revealed the presence of Nitrosomonas-like sequences in the surface substrate of the downflow chamber and apparent diversities of ammonia-oxidizing bacteria in the system.These results suggest that microorganisms,despite their relatively low biomass,have inhabited the IVCW,and the results will offer some valuable information on microbe to system designers and managers. | ZHOU Qiaohong,HE Feng,Zhang Liping,WANG Yanfen,WU Zhenbin State Key Laboratory of Freshwater Ecology and Biotechnology,Institute of Hydrobiology,Chinese Academy of Sciences,Wuhan 430072,China Department of Biology and Chemistry,City University of Hong Kong | 2009 | Journal of Environmental Sciences2009,21,9: | 14 |
| 2 | Comparative study of microbial community structure in different filter media of constructed wetland显示文摘Comparisons of microbial community structure, in eight filter media of zeolites, anthracite, shale, vermiculite, ceramic filter media, gravel, steel slag and bio-ceramic, were undertaken by analyzing the phospholipid fatty acid (PLFA) composition. A total of 20 fatty acids in the range of C11 to C20 were determined but only 13 PLFAs were detected in steel slag. They consist of saturated fatty acids, branched fatty acids, monounsaturated fatty acids, and polyunsaturated fatty acids. The variation of fatty acids was revealed in the relative proportions of these fatty acids in different media. The aerobic prokaryotes were the predominant group in all media. The PLFA composition showed significant differences among the eight different media by Tukey's honestly test. It was found that steel slag was significantly different in the microbial community as compared to other filter media, probably due to its alkaline efluent. Steel slag alone is probably not a good choice of substratum in constructed wetlands. | 4 | Detection and fine-mapping of SC7 resistance genes via linkage and association analysis in soybean显示文摘Soybean mosaic virus(SMV) disease is one of the most serious and broadly distributed soybean(Glycine max(L.) Merr.) diseases. Here, we combine the advantages of association and linkage analysis to identify and fine-map the soybean genes associated with resistance to SMV strain SC7.A set of 191 soybean accessions from different geographic origins and 184 recombinant inbred lines(RILs) derived from Kefeng No.1(resistant) Nannong 1138-2(susceptible) were used in this study. The SC7 resistance genes were previously mapped to a 2.65 Mb region on chromosome 2 and a 380 kb region on chromosome 13. Among 19 single nucleotide polymorphisms(SNPs) detected via association analysis in the study, the SNP BARC-021625-04157 was located in the2.65 Mb region, and the SNP BARC-041671-08065 was located near the 380 kb region; three genes harboring the SNPs were probably related to SC7 resistance. The resistance gene associated with BARC-021625-04157 was then finemapped to a region of approximately 158 kb on chromosome2 using 184 RILs. Among the 15 genes within this region, one NBS-LRR type gene, one HSP40 gene and one serine carboxypeptidase-type gene might be candidate SC7 resistance genes. | 5 | Expanded endoscopic therapy criteria should be cautiously used in intramucosal gastric cancer显示文摘Objective: Accurate estimation of lymph node metastasis(LNM) in intramucosal gastric cancer is essential to select less invasive treatment options and even avoid surgery. The aim of this study was to evaluate combined clinicopathological features to predict the presence of LNM.Methods: A retrospective review of data from 386 intramucosal gastric cancer patients who underwent gastrectomy with extended lymphadenectomy from 2003 to 2010 was conducted. The mutual relation between clinicopathological characteristics and LNM was analyzed.Results: LNM was detected in 40(10.4%) of the 386 patients. Histological type and vascular or lymphatic invasion presence showed a positive correlation with LNM occurrence by univariate analysis. Multivariate analysis revealed that histological type was the only factor associated with LNM. Combined clinicopathologic characteristics would be more predictable for LNM. We found no LNM when we used combined clinicopathological characteristics conforming to Japanese absolute indications for endoscopic therapy. The LNM rate was as high as 8.7% when Japanese expanded criteria were used. Univariate analysis in cancer conformity to expand endoscopic submucosal dissection(ESD) indication also revealed that the undifferential type was the only significant factor for LNM.Conclusions: It was possible to predict intramucosal gastric cancer cases without LNM using combined clinicopathological characteristic analysis. | 6 | Comparison of lymph node number and prognosis in gastric cancer patients with perigastric lymph nodes retrieved by surgeons and pathologists显示文摘Objective: To compare the numbers of positive and total lymph nodes and prognosis in gastric cancer patients whose perigastric lymph node retrieval was performed by surgeons and pathologists.Methods: We conducted a retrospective analysis of clinical and follow-up data from 1,056 patients who underwent gastric cancer D2 radical lymph node resection between January 2008 and December 2010 in the Gastrointestinal Surgery Department of Yantai Yuhuangding Hospital.The follow-up ended in December 2015.Patients were divided into two groups according to the specialty of physicians who performed the postoperative perigastric lymph node retrieval: the surgeon group(475 cases) and the pathologist group(581 cases).The numbers of positive and total perigastric lymph nodes and the 3- and 5-year survival were compared between gastric cancer patients in the two groups overall and stratified by TNM stage(AJCC 7th Edition).Results: Overall, the numbers of positive and total lymph nodes were significantly higher in the surgeon group than in the pathologist group(6.53±4.07 vs.4.09±3.70, P=0.021; 29.64±11.50 vs.20.71±8.56, P<0.001).Further analysis showed that the total number of lymph nodes in stage Ⅰ patients(19.40±9.62 vs.15.45±8.59, P=0.011) and the numbers of positive and total lymph nodes in stage Ⅱ(1.38±1.08 vs.0.87±1.55, P=0.031; 25.35±10.80 vs.16.75±8.56, P<0.001) and stage Ⅲ patients(8.11±6.91 vs.6.66±5.12, P=0.026; 32.34±12.55 vs.25.45±8.31, P<0.001)were significantly higher in the surgeon group than in the pathologist group.The survival analysis showed that the3- and 5-year survival of stage Ⅱ and Ⅲ patients was significantly higher in the surgeon group than in the pathologist group(82.0% vs.73.1%, 69.5% vs.61.2%, P=0.038; 49.2% vs.38.9%, 36.3% vs.28.0%; P=0.045).Conclusions: Compared with retrieval performed by pathologists, postoperative perigastric lymph node retrieval performed by surgeons was associated with significant increase in the total lymph node number of stage Ⅰ patients, the numbers of positive and total lymph nodes of stage Ⅱ and Ⅲ patients, and the survival of stage Ⅱ and stage Ⅲ gastric cancer patients. | Lixin Jiang Zengwu Yao Yifei Zhang Jinchen Hu Dawei Zhao Huiyuan Zhai Xixun Wang Zhenbin Zhang Dong Wang | 2016 | Chinese Journal of Cancer Research2016,28,5: | 5 |
| 7 | Functional analysis of a novel C-glycosyltransferase in the orchid Dendrobium catenatum显示文摘Flavonoids,which are a diverse class of phytonutrients,are used by organisms to respond to nearly all abiotic stresses and are beneficial for human health.Glycosyltransferase,used during the last step of flavonoid biosynthesis,is important in flavonoid enrichment.However,little is known about glycosyltransferase in the orchid Dendrobium catenatum(D.officinale).In this study,we isolated a novel C-glycosyltransferase(designated DcaCGT)from the orchid D.catenatum by identifying and analyzing 82 putative genes in the GT1 family.DcaCGT could specifically catalyze not only di-C-glycosylation but also O-glycosylation.Apart from the normal function of catalyzing 2-hydroxynaringenin and phloretin to the respective di-C-glycosides,DcaCGT also catalyzes apigenin to cosmosiin.Targeted metabolic profiling of the substrates(2-hydroxynaringenin,phloretin,and apigenin)and products(vitexin,isovitexin,vicenin-2,nothofagin,3’,5’-di-C-glucosylphloretin,and cosmosiin)in different tissues showed that vicenin-2 was the most abundant product of this novel enzyme.Cosmosiin was detected in flowers and flower buds.We also established that DcaCGT functions expanded throughout the evolution of D.catenatum.Residual OGT activity may help D.catenatum resist drought stress.Our study illustrates the function,origin,and differentiation of DcaCGT and provides insights into glycosylation and molecular propagation processes,which can be used to improve the production of flavonoids by the cultivated medicinal plant D.catenatum. | Zhiyao Ren Xiaoyu Ji Zhenbin Jiao Yingyi Luo Guo-Qiang Zhang Shengchang Tao Zhouxi Lei Jing Zhang Yuchen Wang Zhong-Jian Liu Gang Wei | 2020 | Horticulture Research2020,7,1: | 5 |
| 8 | Characteristic analysis of phospholipid fatty acids (PLFAs) in typical nutrient polluted lake sediment in Wuhan显示文摘The process of urbanization aggravates the endogenous pollution of urban lake sediment,and polluted sediment may seriously affect the quality of the water in lakes.At present,it is difficult to distinguish the difference between sediment that is heavily polluted by nitrogen(N)and phosphorus(P)when using an analysis based on a physicochemical index classification.The current study applied phospholipid fatty acid(PLFA)technology to further analyze the sediment characteristics from the perspective of microbiology.Surface sediment from five urban lakes that are heavily polluted with N and P in Wuhan were sampled.Statistical approaches were used to analyze the microbial community structure in the sampled sediment,and to determine the correlations between the microorganisms and physicochemical indices.The most severely polluted lake sediment had similar PLFA structures,the highest saturated fatty-acid content,and bacteria as the dominant microorganism.However,there were differences between the microbial biomass of the various sediment samples,which may have been related to the degree of N and P pollution.Analysis of the microbial diversity in the sediment samples indicated that the bacteria were experiencing starvation and nutrient pressure,which may have been due to the dissolved oxygen concentration of the heavily polluted lake sediment.A correlation analysis showed that the endogenous N and P had different effects on the microbes of the polluted sediment.A redundancy analysis(RDA)demonstrated that the N/P ratio had the greatest influence on the PLFA species,accounting for 83%of the cumulative interpretation.To effectively promote the role of sediment microorganisms on circulating elements,it is necessary to regulate the N/P ratio of the sediment to some extent.When the N/P ratio in sediment exceeds 6,N pollution should be prioritized. | Xia Zhang Qianru Chen Chuan Wang Hongpei Zhang Yuqing Zhao Liping Zhang Biyun Liu Zhenbin Wu Qiaohong Zhou | 2021 | International Journal of Sediment Research2021,36,2: | 5 |

| 11 | Structural and metabolic responses of microbial community to sewage-borne chlorpyrifos in constructed wetlands显示文摘Long-term use of chlorpyrifos poses a potential threat to the environment that cannot be ignored, yet little is known about the succession of substrate microbial communities in constructed wetlands(CWs) under chlorpyrifos stress. Six pilot-scale CW systems receiving artificial wastewater containing 1 mg/L chlorpyrifos were established to investigate the effects of chlorpyrifos and wetland vegetation on the microbial metabolism pattern of carbon sources and community structure, using BIOLOG and denaturing gradient gel electrophoresis(DGGE) approaches. Based on our samples, BIOLOG showed that Shannon diversity(HV) and richness(S) values distinctly increased after 30 days when chlorpyrifos was added. At the same time, differences between the vegetated and the non-vegetated systems disappeared. DGGE profiles indicated that H Vand S had no significant differences among four different treatments. The effect of chlorpyrifos on the microbial community was mainly reflected at the physiological level. Principal component analysis(PCA) of both BIOLOG and DGGE showed that added chlorpyrifos made a difference on test results.Meanwhile, there was no difference between the vegetation and no-vegetation treatments after addition of chlorpyrifos at the physiological level. Moreover, the vegetation had no significant effect on the microbial community at the genetic level. Comparisons were made between bacteria in this experiment and other known chlorpyrifos-degrading bacteria. For the difference of electro-migration, the atom of solute and solution will accumulate around the cluster, and then the smaller cluster may reorganize and grow up under the action of the coupled field. We also apply the electrostatic induction theory to analyze the bearing behavior of the half side of the cluster. The bigger the cluster is, the stronger the electrostatic force is, therefore, the bigger cluster's stability is weak apparently.
