| 2 | Panax notoginseng saponins influence on transplantation of neural stem cell-derived dopaminergic neurons in a rat model of Parkinson’s disease显示文摘BACKGROUND: Dopaminergic neurons differentiated from neural stem cells have been successfully used in the treatment of rat models of Parkinson’s disease; however, the survival rate of transplanted cells has been low. Most cells die by apoptosis as a result of overloaded intracellular calcium and the formation of oxygen free radicals. OBJECTIVE: To observe whether survival of transplanted cells, transplantation efficacy, and dopaminergic differentiation from neural stem cells is altered by Panax notoginseng saponins (PNS) in a rat model of Parkinson’s disease. DESIGN, TIME AND SETTING: Cellular and molecular biology experiments with randomized group design. The experiment was performed at the Animal Experimental Center, First Hospital of Sun Yat-sen Uni- versity from April to October 2007. MATERIALS: Thirty-two adult, healthy, male Sprague Dawley rats, and four healthy Sprague Dawley rat embryos at gestational days 14-15 were selected. The right ventral mesencephalon was injected with 6-hydroxydopamine to establish a model of Parkinson’s disease. 6-hydroxydopamine and apomorphine were purchased from Sigma, USA. METHODS: Neural stem cells derived from the mesencephalon of embryonic rats were cultivated and passaged in serum-free culture medium. Lesioned animals were randomly divided into four groups (n = 8): dopaminergic neuron, dopaminergic neuron + PNS, PNS, and control. The dopaminergic neuron group was injected with 3 μL cell suspension containing dopaminergic neurons differentiated from neural stem cells. The dopaminergic neurons + PNS group received 3 μL dopaminergic cell suspension combined with PNS (250 mg/L). The PNS group received 3 μL PNS (250 mg/L), and the control group received 3 μL DMEM/F12 culture medium. MAIN OUTCOME MEASURES: The rats were transcardially perfused with 4% paraformaldehyde at 60 days post-grafting for immunohistochemistry. The rats were intraperitoneally injected with apomorphine (0.5 mg/kg) to induce rotational behavior. RESULTS: Cell counts of tyrosine hydroxylase-positive neurons in the dopaminergic neuron + PNS group were (732±82.6) cells/400-fold field. This was significantly greater than the dopaminergic neuron group [(326 ± 34.8) cells/400-fold field, P < 0.01]. Compared to the control group, the rotational asymmetry of rats that received dopaminergic neuron transplants was significantly decreased, beginning at 20 days after opera- tion (P < 0.01). Rotational asymmetry was further reduced between 10-60 days post-surgery in the dopaminergic neuron + PNS group, compared to the dopaminergic neuron group (P < 0.01). | 5 | The telomere-to-telomere gap-free reference genome of wild blueberry (Vaccinium duclouxii) provides its high soluble sugar and anthocyanin accumulation显示文摘Vaccinium duclouxii,endemic to southwestern China,is a berry-producing shrub or small tree belonging to the Ericaceae family,with high nutritive,medicinal,and ornamental value,abundant germplasm resources,and good edible properties.In addition,V.duclouxii exhibits strong tolerance to adverse environmental conditions,making it a promising candidate for research and offering wide-ranging possibilities for utilization.However,the lack of V.duclouxii genome sequence has hampered its development and utilization.Here,a high-quality telomere-to-telomere genome sequence of V.duclouxii was de novo assembled and annotated.All of 12 chromosomes were assembled into gap-free single contigs,providing the highest integrity and quality assembly reported so far for blueberry.The V.duclouxii genome is 573.67 Mb,which encodes 41953 protein-coding genes.Combining transcriptomics and metabolomics analyses,we have uncovered the molecular mechanisms involved in sugar and acid accumulation and anthocyanin biosynthesis in V.duclouxii.This provides essentialmolecular information for further research on the quality of V.duclouxii.Moreover,the high-quality telomere-to-telomere assembly of the V.duclouxii genome will provide insights into the genomic evolution of Vaccinium and support advancements in blueberry genetics and molecular breeding. | Tuo Zeng Zhijiao He Jiefang He Wei Lv Shixiang Huang Jiawen Li Liyong Zhu Shuang Wan Wanfei Zhou Zhengsong Yang Yatao Zhang Chong Luo Jiawei He Caiyun Wang Liangsheng Wang | 2023 | Horticulture Research2023,10,11: | 0 |
| 6 | Characterization and Expression Profiling of Genes Encoding the Gibberellin 3-oxidase in Dasypyrum villosum Dwarf Mutant显示文摘Gibberellin 3-oxidase catalyzes the conversion of inactive gibberellin(GA) species into GAs with biological activity and it is subjected to strict developmental controls in the life cycle of a plant. In this study, 33 gene sequences, encoding the gibberellin 3-oxidase(GA3ox) from Dasypyrum villosum and its dwarf mutant, were obtained. Each contained a 1 107 bp coding sequence(CDS) that encoded a putative protein containing 369 amino acids. The GA3ox protein showed 77% to 97% homology and shared the major conserved structural domains of GA3ox proteins with rice, sorghum bicolor, oat, barley, and wheat. Sequence alignment showed that there were 20 single nucleotide polymorphisms(SNPs) and 22 Insertion/deletions(In Dels) among these sequences, which could be divided into 2 haplotypes, haplotypes Ⅰ and Ⅱ. Haplotype Ⅰ was found in the wild type and was1 495 bp in length, and haplotype Ⅱ was found in the dwarf mutant and was 1 485 bp in length. The Q-PCR results showed that GA3ox was expressed in the leaves, roots, internodes, and stem nodes, and that there was a significant difference in the transcript level of the GA3ox between the wild type and dwarf mutant. The transcript levels of GA3ox in the leaves at the seedling stage, stem elongation stage and the heading stage, in the root and stem nodes at the stem elongation stage and in the internodes at the heading stage of the wild type, were significantly higher than those in the dwarf mutant. However, GA3ox expression in the rest of the wild type tissues at the 3 stages was slightly higher than or not different from the dwarf mutant.The results suggested that the wild type and mutant allele sequences of GA3ox in D. villosum showed 2 amino acid changes in exons and variations in the lengths of introns or the SNPs in introns, which most probably impaired the function of the enzyme,affected the GA3ox expression level, and eventually gave rise to dwarfing. | Peng CAI Zaijun YANG Zhengsong PENG Maoqun YU | 2017 | Agricultural Science & Technology2017,18,9: | 0 |
| 7 | Coupled intramolecular/heterointerfacial electron transfer in polyelectrolyte-shielded Iso-type black phosphorus hetero-structure boosts oxygen reduction kinetics显示文摘Searching new structured black phosphorus(BP)and exploring intriguing functions and applications have become a hot topic so far.Here,we introduce a novel Iso-type black phosphorus heterostructure guided by first principle calculation,which features unique heterointerface and electronic coupling interaction via stacking assembly of exfoliated black phosphorus(EBP)and amine-functionalized EBP(N-EBP).Inspired by the theoretical results,we constructed the Iso-type heterostructure comprising of ultrathin exfoliated few-layered EBP and N-EBP,both of which were derived from identical bulk BP.The purposive amine-functionalization not only creates positively-charged P atoms on N-EBP as effective active sites via N-induced intramolecular electron transfer(IET)but also endows N-EBP with lower work function relative to EBP,while the unique EBP/N-EBP Iso-type heterostructure engenders directional heterointerfacial electron transfer(HET).The coupled IET/HET effects optimize the charge redistribution to afford favorable O_(2)adsorption.In this case,our unique strategy for the first time exploits the inherent catalytic capability of BP toward the oxygen reduction reaction(ORR)and enables the first use of BP as metal-free ORR catalysts for Zn-air cells.The newly-designed heterostructure facilitates a 4-e^(-)transfer ORR relative to inactive EBP or N-EBP.Importantly,the polymer-shielded heterostructure acts as efficient air electrodes to endow a primary Zn-air cell with high stability,large capacity and high energy density—superior to the commercial Pt/C-enabled cell.This study as the first report on metal-free BP-based ORR catalysts and air electrodes not only extends BP's application scopes but also renders new insight toward design of electronically-coupled superstructures for energy-related applications. | Zhongke Yuan Jing Li Zhengsong Fang Meijia Yang Kancheng Mai Dingshan Yu | 2021 | Journal of Energy Chemistry2021,30,12: | 0 |
