1Endothelium-specific SIRT1 overexpression inhibits hyperglycemia-induced upregulation of vascular cell senescence显示文摘The rapidly increasing prevalence of diabetes mellitus worldwide is one of the most serious and challenging health problems in the 21st century.Mammalian sirtuin 1(SIRT1) has been shown to decrease high-glucose-induced endothelial cell senescence in vitro and prevent hyperglycemia-induced vascular dysfunction.However,a role for SIRT1 in prevention of hyperglycemia-induced vascular cell senescence in vivo remains unclear.We used endothelium-specific SIRT1 transgenic(SIRT1-Tg) mice and wild-type(WT) mice to construct a 40-week streptozotocin(STZ)-induced diabetic mouse model.In this mode,42.9% of wild-type(WT) mice and 38.5% of SIRT1-Tg mice were successfully established as diabetic.Forty weeks of hyperglycemia induced significant vascular cell senescence in aortas of mice,as indicated by upregulation of expression of senescence-associated markers including p53,p21 and plasminogen activator inhibitor-1(PAI-1).However,SIRT1-Tg diabetic mice displayed dramatically decreased expression of p53,p21 and PAI-1 compared with diabetic WT mice.Moreover,manganese superoxide dismutase expression(MnSOD) was significantly downregulated in the aortas of diabetic WT mice,but was preserved in diabetic SIRT1-Tg mice.Furthermore,expression of the oxidative stress adaptor p66Shc was significantly decreased in aortas of SIRT1-Tg diabetic mice compared with WT diabetic mice.Overall,these findings suggest that SIRT1-mediated inhibition of hyperglycemia-induced vascular cell senescence is mediated at least partly through the reduction of oxidative stress.CHEN HouZao WAN YanZhen ZHOU Shuang LU YunBiao ZHANG ZhuQin ZHANG Ran CHEN Feng HAO DeLong ZHAO Xiang GUO ZhiChen LIU DePei LIANG ChihChuan 2012Science China(Life Sciences)2012,55,6:14

5Possible mechanisms underlying treatment of Alzheimer’s disease with Traditional Chinese Medicine: active components, potential targets and synthetic pathways of Bulao Elixir显示文摘OBJECTIVE: To elucidate the mechanisms underlying the treatment of Alzheimer’s disease(AD) with Traditional Chinese Medicine(TCM), by examining the active components, potential targets and synthetic pathways of Bulao Elixir(BLE).METHODS: The Absorption, Distribution, Metabolism, Excretion(ADME)/Toxicology(T) calculation was used to screen the active components of Bulao Elixir. Based on the TCM Systems Pharmacology Analysis Platform(TCMSP database) and a text mining tool(GoPubMed database), we predicted and screened the active components of Bulao Elixir and its therapeutic targets for AD. Using the Database for Annotation, Visualization and Integrated Discovery(DAVID), we obtained the targets for AD. Cytoscape software was used to establish a network map of the active component-target and target-pathway of Bulao Elixir. Gene function, related biological processes and signaling pathways were analyzed using the DAVID database.RESULTS: Twelve active components were selected from 196 components of Bulao Elixir. Among 2209 targets, 102 effective targets were selected, and 30 important targets were identified via matching with the disease targets. After further analysis, 14 core targets were identified. Enrichment analysis revealed that most of these important targets were involved in multiple biological processes, including apoptosis, inflammatory reactions, and cell regulation cycles. 6Liver resection versus intensity-modulated radiation therapy for treatment of hepatocellular carcinoma with hepatic vein tumor thrombus:a propensity score matching analysis显示文摘Background:The presence of hepatic vein tumor thrombus(HVTT)is a major determinant of survival outcomes in hepatocellular carcinoma(HCC)patients.This study compared survival outcomes between liver resection(LR)and intensity-modulated radiation therapy(IMRT)in HCC patients with HVTT.Methods:Data from patients who underwent LR or IMRT for HCC with HVTT at the Eastern Hepatobiliary Surgery Hospital were retrospectively analyzed.Their survival outcomes were compared before and after propensity score matching(PSM).Results:Three hundred and seven HCC patients with HVTT who underwent either LR(n=140)or IMRT(n=167)were enrolled.PSM matched 82 pairs of patients.The overall survival(OS)and recurrence-free survival(RFS)rates were significantly higher for patients in the LR group than those in the IMRT group.On subgroup analysis,significantly better survival outcomes were obtained after LR than IMRT in patients with peripheral type of HVTT(pHVTT)and major type of HVTT(mHVTT).However,similar survival outcomes were obtained after LR and IMRT when the HVTT had developed into inferior vena cava tumor thrombus(IVCTT).Conclusions:LR resulted in significantly better survival outcomes in HCC patients with HVTT when compared to IMRT.Once the HVTT had developed IVCTT,LR and IMRT resulted in similarly bad survival outcomes.Zhen-Hua Chen Xiu-Ping Zhang Shuang Feng Jing-Kai Feng Zong-Tao Chai Wei-Xing Guo Jie Shi Wan Yee Lau Yan Meng Shu-Qun Cheng 2021Hepatobiliary Surgery and Nutrition2021,10,5:3
7Preliminary Study on Drug Susceptibility Profile and Resistance Mechanisms to Macrolides of Clinical Isolates of Non-tuberculous Mycobacteria from China显示文摘Objective Macrolide susceptibility and drug resistance mechanisms of clinical non-tuberculous mycobacteria(NTM) isolates were preliminarily investigated for more accurate diagnosis and treatment of the infection in China. Methods Four macrolides, including clarithromycin(CLAR), azithromycin(AZM), roxithromycin(ROX), and erythromycin(ERY), were used to test the drug susceptibility of 310 clinical NTM isolates from six provinces of China with the broth microdilution method. Two resistance mechanisms, 23 S r RNA and erm, were analyzed with nucleotide sequence analysis. Results Varied effectiveness of macrolides and species-specific resistance patterns were observed. Most Mycobacterium abscessus subsp. massiliense were susceptible and all M. fortuitum were highly resistant to macrolides. All the drugs, except for erythromycin, exhibited excellent activities against slow-growing mycobacteria, and drug resistance rates were below 22.2%. Only four highly resistant strains harbored 2,058/2,059 substitutions on rrl and none of other mutations were related to macrolide resistance. G2191 A and T2221 C on rrl were specific for the M. abscessus complex(MABC). Seven sites, G2140 A, G2210 C, C2217 G, T2238 C, T2322 C, T2404 C, and A2406 G, were specifically carried by M. avium and M. intracellulare. Three sites, A2192 G, T2358 G, and A2636 G, were observed only in M. fortuitum and one site G2152 A was specific for M. gordonae. The genes erm(39) and erm(41) were detected in M. fortuitum and M. abscessus and inducible resistance was observed in relevant sequevar. Conclusion The susceptibility profile of macrolides against NTM was demonstrated. 8Cramer-Rao bound of joint estimation of target location and velocity for coherent MIMO radar显示文摘The optimal estimation performance of target parameters is studied. First, the general form of Cramer-Rao bound(CRB)for joint estimation of target location and velocity is derived for coherent multiple input multiple output(MIMO) radars. To gain some insight into the behavior of the CRB, the CRB with a set of given orthogonal waveforms is studied as a specific case. Second, a maximum likelihood(ML) estimation algorithm is proposed. 9Effects of baria on propane oxidation activity of Pd/Al_2O_3 catalyst:Pd–BaO interaction and reaction routes显示文摘The BaO-modified Pd/Al2O3exhibits much better catalytic activity than Pd/Al2O3for C3H8 oxidation both before and after the hydrothermal aging treatment. Further studies ascribe its good activity to the influence of BaO species on the physicochemical characteristics of the catalyst and the reaction routes. Firstly, the BaO species inhibits the phase transformation of alumina, resulting in higher surface area of the catalysts and hereby a better dispersion of Pd. Secondly, the basic nature and electron-withdrawing effect of barium oxide maintain palladium at high oxidation state, which leads to a higher PdO content on surface of the BaO-modified catalyst. 11Bifurcations in penetrative Rayleigh-Be′nard convection in a cylindrical container显示文摘The bifurcations of penetrative Rayleigh-B′enard convection in cylindrical containers are studied by the linear stability analysis(LSA) combined with the direct numerical simulation(DNS) method. The working ?uid is cold water near 4?C, where the Prandtl number P r is 11.57, and the aspect ratio(radius/height) of the cylinder ranges from 0.66 to 2. It is found that the critical Rayleigh number increases with the increase in the density inversion parameter θ_m. The relationship between the normalized critical Rayleigh number(Rac(θ_m)/Rac(0)) and θ_m is formulated, which is in good agreement with the stability results within a large range of θ_m. The aspect ratio has a minor effect on Rac(θ_m)/Rac(0). The bifurcation processes based on the axisymmetric solutions are also investigated. 12Correlation between serum miR-154-5p and urinary albumin excretion rates in patients with type 2 diabetes mellitus:a cross-sectional cohort study显示文摘This study aimed to investigate the correlation between serum miR-154-5p and urinary albumin to creatinine ratio(UACR)in patients with type 2 diabetes mellitus(T2DM)and the association with biomarkers of inflammation and fibrosis in diabetic kidney disease(DKD).A total of 390 patients with T2DM were divided into three groups:normal albuminuria(UACR<30 mg/g,n=136,NA),microalbuminuria(UACR at 30-300 mg/g,n=132,MA),and clinical albuminuria(UACR>300 mg/g,n=122,CA).Circulating miR-154-5p,inflammatory(C-reactive protein(CRP);erythrocyte sedimentation rate(ESR);and tumor necrosis factor-a(TNF-α)and fibrotic markers(vascular endothelial growth factor(VEGF);transforming growth factor-β1(TGF-β1);and fibronectin(FN),and other biochemical indicators were assessed via real-time PCR,enzyme-linked immunosorbent assay,and chemiluminescence assay in patients with T2DM and 138 control subjects(NC).UACR,miR-154-5p,glycated hemoglobin(HbA1c),serum creatinine(sCr),blood urea nitrogen(BUN),ESR,CRP,VEGF,TNF-α,TGF-β1,and FN were significantly higher and the estimated glomerular filtration rate(eGFR)was significantly lower in NA,MA,and CA groups than in NC subjects(P<0.05).Elevated levels of UACR and miR-154-5p were directly correlated with HbA1c,sCr,BUN,ESR,CRP,VEGF,TNF-α,TGF-β1,and FN and negatively correlated with eGFR(P<0.05).miR-154-5p,HbA1c,sCr,BUN,eGFR,ESR,CRP,VEGF,TNF-α,TGF-β1,and FN were important factors affecting UACR.These findings indicated that elevated serum miR-154-5p is significantly correlated with high UACR in patients with T2DM and may offer a novel reference for the early diagnosis of DKD.Huiwen Ren Can Wu Ying Shao Shuang Liu Yang Zhou Qiuyue Wan 2020Frontiers of Medicine2020,14,5:2
13QNE1 is a key flowering regulator determining the length of the vegetative period in soybean cultivars显示文摘The soybean E1 gene is a major regulator that plays an important role in flowering time and maturity.However,it remains unclear how cultivars carrying the dominant E1 allele adapt to the higher latitudinal areas of northern China.We mapped the novel quantitative trait locus QNE1(QTL near E1) for flowering time to the region proximal to E1 on chromosome 6 in two mapping populations.Positional cloning revealed Glyma.06G204300,encoding a TCP-type transcription factor,as a strong candidate gene for QNE1.Association analysis further confirmed that functional single nucleotide polymorphisms(SNPs) at nucleotides 686 and 1,063 in the coding region of Glyma.06G204300 were significantly associated with flowering time.The protein encoded by the candidate gene is localized primarily to the nucleus.Furthermore,soybean and Brassica napus plants overexpressing Glyma.06G204300 exhibited early flowering.We conclude that despite their similar effects on flowering time,QNE1 and E4 may control flowering time through different regulatory mechanisms,based on expression studies and weighted gene co-expression network analysis of flowering time-related genes.Deciphering the molecular basis of QNE1 control of flowering time enriches our knowledge of flowering gene networks in soybean and will facilitate breeding soybean cultivars with broader latitudinal adaptation.Zhengjun Xia Hong Zhai Yanfeng Zhang Yaying Wang Lu Wang Kun Xu Hongyan Wu Jinglong Zhu Shuang Jiao Zhao Wan Xiaobin Zhu Yi Gao Yingxiang Liu Rong Fan Shihao Wu Xin Chen Jinyu Liu Jiayin Yang Qijian Song Zhixi Tian 2022Science China(Life Sciences)2022,65,12:1

16Low Li ion diffusion barrier on low-crystalline FeOOH nanosheets and high performance of energy storage显示文摘To obtain symmetric supercapacitors(SCs)with high energy density,it is critical to fabricate an electrode with wide potential window and excellent capacitive performance.Herein,by using the strong double hydrolysis reaction between anions and cations,the FeOOH nanosheets on the surface of activated carbon cloth(FeOOH@AC)are prepared through a simple hydrothermal process.The FeOOH@AC electrode exhibits maximum capacitance of 4,090 mF·cm^−2 at wider potential window-1–0 V and 3,250 mF·cm^−2 at 0–1 V versus SCE in 2 M LiNO3 electrolyte.With two pieces of FeOOH@AC electrodes the obtained symmetric SC can operate at the voltage window of 2 V.This FeOOH symmetric SC shows high energy density of 13.261 mWh·cm^−3 at a power density of 14.824 mW·cm^−3 and maintains 4.175 mWh·cm^−3 at a maximum power density of 118.564 mW·cm^−3,as well as excellent charge storage capacity and cyclic stability.Li ion adsorption and diffusion mechanism on the(200)facets of FeOOH are explained by the density functional theory(DFT)calculations.The simple synthesis process and excellent capacitance performance of the FeOOH@AC composite make it a very promising candidate for high performance symmetric SC electrodes.Jien Li Shuang Luo Congcong Wang Qian Tang Yanwei Wang Xiangyu Han Hao Ran Jing Wan Xiao Gu Xue Wang Chenguo Hu 2020Nano Research2020,13,3:1
17Transcriptome analysis of salt-stress response in three seedling tissues of common wheat显示文摘Xiaoyan 60(XY60) is a new wheat variety bred in the laboratory of Zhensheng Li.After salt treatment, seedlings of XY60 maintain green leaves and produce longer roots than the high yielding cultivar Zhongmai 175(ZM175).To explain these different phenotypes we carried out an RNA-Seq analysis using 12 samples from three tissues of both varieties subjected to salt and control treatments.By comparing data from the salt treated plants with the control, 703, 979, and 1197 differentially expressed genes(DEGs) were detected in new leaves, old leaves, and roots of XY60, respectively.The corresponding numbers for ZM175 were 613, 1401, and 1301.The most significantly enriched Gene Ontology(GO) terms and KEGG pathways were associated with polyunsaturated fatty acid(PUFA) metabolism in both new and old leaves from XY60.They were associated with photosynthesis and energy metabolism in ZM175.The most significantly enriched KEGG pathway in roots of both varieties was “glucosinolate biosynthesis”.In addition, jasmonic acid(JA) concentration in XY60 was higher than in ZM175, although it increased significantly in both varieties following salt treatment.Trends in relative expression levels of AOS, MYC2, and JAZ revealed by qRT-PCR were concordant with those from RNA-Seq.Our results suggest that PUFAs may contribute to salt tolerance in common wheat by enhancing the photosynthetic system and JA-related pathways.Qiaoling Luo Wan Teng Shuang Fang Hongwei Li Bin Li Jinfang Chu Zhensheng Li Qi Zheng 2019The Crop Journal2019,7,3:1
18Solution processable transition metal dichalcogenides-based hybrids for photodetection显示文摘Transition metal dichalcogenides(TMDs), as one of the most promising two-dimensional(2D) materials, have attracted considerable attention for use in photodetection applications over the past few years due to their distinct properties, such as atomic-scale thickness, tunable direct bandgaps, and decent carrier mobilities at room temperature. Compared with pure 2D TMDs, the construction of hybrids consisting of TMDs and other low-dimensional materials can further improve the performance of photodetectors including their spectral range, responsivity and detectivity, which significantly boosts interest in the development of TMDs-based photodetectors. On the other hand, solution-phase synthesis methods provide a facile strategy for the scalable production of TMD hybrids, opening an exciting avenue to develop low-cost devices. 19Analysis on Change Characteristics and Forecast Factors of the Fog in Beibei District of Chongqing显示文摘[Objective] The research aimed to analyze change characteristics and forecast factors of the fog in Beibei District of Chongqing from 1953 to 2010. [Method] By observation data of the fog in Beibei District from 1953 to 2010, interdecadal, interannual, seasonal and monthly variation characteristics of the fog days and formation-dispersion time of the fog were conducted statistical analysis. Meteorological conditions and forecast factors of the fog were also analyzed. [Result] Distribution of the fog days in Beibei District had obvious interdecadal characteristics. Fog days was at its maximum in the 1980s while minimum in the 1960s. Fog duration presented slow increase trend. Interannual characteristic of the fog days overall presented increase trend, and it had 9-year periodic oscillation characteristic. Fog mainly concentrated in autumn and winter. Fog was mainly formed at night (20:00-08:00) and dispersed in the daytime (08:00-13:00). Meteorological conditions which affected heavy fog in Beibei District were water vapor and stratification, wind field, temperature, relative humidity and so on. 20A facile strategy of in-situ anchoring of Co_(3)O_(4) on N doped carbon cloth for an ultrahigh electrochemical performance显示文摘Enhancement of supercapacitors(SCs)with high-energy density and high-power density is still a great challenge.In this paper,a facile strategy for in situ anchoring of Co_(3)O_(4) particles on N doped carbon cloth(pCoNCC)is reported.Due to the interaction of the doped N and Co_(3)O_(4),the electrochemical performance improves significantly,reaching 1,940.13 mF·cm^(−2) at 1 mA·cm^(−2) and energy density of 172.46μWh·cm^(−2) at the power density of 400μW·cm^(−2),much larger than that without N doping electrode of 28.5 mF·cm^(−2).An aqueous symmetric supercapacitor(ASSC)assembled by two pCoNCC electrodes achieves a maximum energy density of 447.42μWh·cm^(−2) and a highest power density of 8,000μW·cm^(−2).Utilizing such a high-energy storage ASSC,a digital watch and a temperature-humidity detector are powered for nearly 1 and 2 h,respectively.Moreover,the ASSC displays a superb electrochemical stability of 87.7%retention after 10,000 cycles at 40 mA·cm^(−2).This work would provide a new sight to enhance active materials performance and be beneficial for the future energy storage and supply systems.Junlin Lu Jien Li Jing Wan Xiangyu Han Peiyuan Ji Shuang Luo Mingxin Gu Dapeng Wei Chenguo Hu 2021Nano Research2021,14,7:0
