| 1 | Randomized Latent Factor Model for High-dimensional and Sparse Matrices from Industrial Applications显示文摘Latent factor(LF) models are highly effective in extracting useful knowledge from High-Dimensional and Sparse(HiDS) matrices which are commonly seen in various industrial applications. An LF model usually adopts iterative optimizers,which may consume many iterations to achieve a local optima,resulting in considerable time cost. Hence, determining how to accelerate the training process for LF models has become a significant issue. To address this, this work proposes a randomized latent factor(RLF) model. It incorporates the principle of randomized learning techniques from neural networks into the LF analysis of HiDS matrices, thereby greatly alleviating computational burden. It also extends a standard learning process for randomized neural networks in context of LF analysis to make the resulting model represent an HiDS matrix correctly.Experimental results on three HiDS matrices from industrial applications demonstrate that compared with state-of-the-art LF models, RLF is able to achieve significantly higher computational efficiency and comparable prediction accuracy for missing data.I provides an important alternative approach to LF analysis of HiDS matrices, which is especially desired for industrial applications demanding highly efficient models. | Mingsheng Shang Xin Luo Zhigang Liu Jia Chen Ye Yuan MengChu Zhou | 2019 | IEEE/CAA Journal of Automatica Sinica2019,6,1: | 11 |

| 4 | Generation and antitumor effects of an engineered and energized fusion protein VL-LDP-AE composed of single-domain antibody and lidamycin显示文摘Type IV collagenase plays a pivotal role in invasion, metastasis and angiogenesis of tumor. Single domain antibodies are attractive as tumor-targeting vehicle because of their much smaller size com-pared with antibody molecules produced by conventional methods. Lidamycin (LDM) is a potent enediyne-containing antitumor antibiotic. In this study an engineered and energized fusion protein VL-LDP-AE composed of lidamycin and VL domain of mAb 3G11 directed against type IV collagenase was prepared using a novel two-step method. First a VL-LDP fusion protein was constructed by DNA recombination. Secondly VL-LDP-AE was obtained by molecular reconstitution. In MTT assay, VL-LDP-AE showed potent cytotoxicity to HT-1080 cells and KB cells with IC50 values of 8.55×10-12 and 1.70×10-11 mol/L, respectively. VL-LDP-AE showed antiangiogenic activity in chick chrorioallantoic membrane (CAM) assay and tube formation assay. In in vivo experiments, VL-LDP-AE was proved to be more effective than free LDM against the growth of subcutaneously transplanted hepatoma 22 in mice. Drugs were given intravenously on day 3 and 10 after tumor transplantation. Compared in terms of maximal tolerated doses, VL-LDP-AE at 0.25 mg/kg suppressed the tumor growth by 89.5%, LDM at 0.05 mg/kg by 69.9%, and mitomycin at 1 mg/kg by 35%. Having a molecular weight of 25.2 kDa, VL-LDP-AE was much smaller than other reported antibody-based drugs. The results suggested that VL-LDP-AE would be a promising candidate for tumor targeting therapy. | 12 | Genomic footprints of sorghum domestication and breeding selection for multiple end uses显示文摘Domestication and diversification have had profound effects on crop genomes.Originating in Africa and subsequently spreading to different continents,sorghum(Sorghum bicolor)has experienced multiple onsets of domestication and intensive breeding selection for various end uses.However,how these processes have shaped sorghum genomes is not fully understood.In the present study,population genomics analyses were performed on a worldwide collection of 445 sorghum accessions,covering wild sorghum and four end-use subpopulations with diverse agronomic traits.Frequent genetic exchanges were found among various subpopulations,and strong selective sweeps affected 14.68%(∼107.5 Mb)of the sorghum genome,including 3649,4287,and 3888 genes during sorghum domestication,improvement of grain sorghum,and improvement of sweet sorghum,respectively.Eight different models of haplotype changes in domestication genes from wild sorghum to landraces and improved sorghum were observed,and Sh1-and SbTB1-type genes were representative of two prominent models,one of soft selection or multiple origins and one of hard selection or an early single domestication event.We also demonstrated that the Dry gene,which regulates stem juiciness,was unconsciously selected during the improvement of grain sorghum.Taken together,these findings provide new genomic insights into sorghum domestication and breeding selection,and will facilitate further dissection of the domestication and molecular breeding of sorghum. | Xiaoyuan Wu Yuanming Liu Hong Luo Li Shang Chuanyuan Leng Zhiquan Liu Zhigang Li Xiaochun Lu Hongwei Cai Huaiqing Hao Hai-Chun Jing | 2022 | Molecular Plant2022,15,3: | 1 |

| 16 | Characterization of SOX3 Gene in an Ovoviviparous Teleost, Black Rockfish(Sebastes schlegeli)显示文摘Evolutionary evidence suggests that Sox3, a member of the high-mobility-group(HMG) family of transcription factors, is an ancestral precursor of Sry and is involved in sex determination similar to Sry. However, there is limited information regarding the SOX3 gene of the black rockfish(Sebastes schlegeli). In this study, we first isolated SOX3 gene from the gonads of S. schlegeli by homology cloning. The full-length of S. schlegeli SOX3(SsSOX3) c DNA was 1386 bp, comprising a 906-bp open reading frame, which encodes a peptide showing 93.6% and 93.9% homology with the Sox3 proteins of Epinephelus coioides and Oryzias latipe, respectively. Comparison of the cDNA sequence of the Ss SOX3 gene with the corresponding genomic DNA fragment revealed that the SsSOX3 gene consists of a single exon. Phylogenetic analysis demonstrated the evolutionary relationship of Ss SOX3 with other known SOXB1 genes in fish and tetrapods. The promoter region contains binding sites of several transcriptional factors that might participate in the regulation of Ss SOX3 expression. Quantitative real-time PCR analysis indicated that SsSOX3 was expressed in all the investigated larval developmental stages from 1 to 35 days after birth and the level of expression gradually decreased as the development proceeded. SsSOX3 exhibited sexually dimorphic expression in adult gonads, with high expression in the ovary but low expression in the testis. In situ hybridization revealed that SsSOX3 was strongly expressed in oocytes and follicular cells of ovaries but slightly expressed in germ cells of testicular tissues. | 20 | Evidence of Glacial Erratic Rollover Revealed by ^(10)Be and ^(26)Al Concentration Variations显示文摘Cosmogenic nuclide exposure dating is one of the most intensively applied dating methods with which to study glacial geomorphology.Glacial erratics have been the major dating objective in many studies.Some research has proposed that glacial erratics may undergo rollover and re-transportation during the late exposure stage,which can affect the dating results.However,there is no direct evidence to confirm this possibility.In this study,we collected seven samples from a vertical section inside a glacial erratic in the paleo-Daocheng ice cap in the southeastern Tibetan Plateau,measuring their contents of the cosmogenic nuclides ^(10)Be and ^(26)Al.The results show that from the top to the bottom,the concentrations of 10Be were(1.21±0.05)×10^(6),(1.00±0.02)×10^(6),(0.88±0.03)×10^(6),(0.77±0.02)×10^(6),(0.75±0.03)×10^(6),(0.95±0.03)×10^(6) and(1.46±0.04)×10^(6) atoms/g.The ^(10)Be concentrations decreased from(1.21±0.05)×10^(6) atoms/g to(0.75±0.03)×10^(6) atoms/g and then increased to(1.46±0.04)×10^(6) atoms/g,which is not consistent with the theoretical prediction of a gradual decrease.This phenomenon indicates that the glacial erratic may have rolled over at least once.The lower surface of the erratic could have been on top at some time in the past.Therefore,its exposure age was greater than the exposure age that was expected,based on its current orientation.This study provides numerical evidence for an erratic rollover event. | ZHANG Zhigang WANG Jian XU Xiaobin CHANG Zhiyang CUI Haitao LIANG Zhong CHEN Xia SHANG Zhiyuan | 2022 | Acta Geologica Sinica(English Edition)2022,96,1: | 0 |
