1Comparison of concentration methods for detection of hepatitis A virus in water samples显示文摘Hepatitis A virus is a pathogen associated with water pollution.Contaminated drinking water can cause hepatitis A outbreaks,lead to economic losses,and even threaten human lives.It is difficult to detect low levels of hepatitis A virus in water,so the virus must be concentrated in order to quantify it accurately.Here,we present a simple,rapid,efficient technique for the concentration and detection of hepatitis A virus in water.Our data showed that adding phosphate-buffered saline to the water,pre-filtering the water,and adding Trizol reagent directly to the filtration membrane can significantly improve concentration efficiency.Of three types of filtration membranes studied(mixed cellulose ester membrane,polyvinylidene fluoride membrane,and nylon membrane),the concentration efficiency using mixed cellulose ester membrane with a 0.1-μm pore size was the highest,reaching 92.62 ± 5.17%.This method was used to concentrate hepatitis A virus in water samples from Donghu Lake.Using SYBR Green real-time reverse transcription polymerase chain reaction analysis,the detection sensitivity of this method reached 10~1 copies/μL and its concentration efficiency reached 79.45 ± 9.88%.Yuting Qiao Zhiwei Sui Guoliang Hu Huabin Cao Guoxiang Yang Yong Li Yongsong Lei Lihua Zhao Quanjiao Chen 2016Virologica Sinica2016,31,4:1
2FOXG1的表达与人类神经胶质瘤的恶性肿瘤有关(英文)显示文摘Objective: Recent evidence indicates that the increased expression of FOXG1 is associated with tumor genesis. This study was designed to explore the expression and role which FOXG1 plays in human glioma. Methods: We detected the expression of FOXG1 by immunohistochemistry in glioma tissue samples. Following the down-regulation of FOXG1 in glioma cell lines by a specific short hairpin RNA, the function of FOXG1 in proliferation and apoptosis was assessed. Results: Glioma tissues exhibited notably higher expression of FOXG1 compared with control brain tissues and was positively correlated with histological malignancy. The down-regulation of FOXG1 in glioma cells led to a cell apoptosis in vitro. 4Hyperparameter Optimization for Capsule Network Based Modified Hybrid Rice Optimization Algorithm显示文摘Hyperparameters play a vital impact in the performance of most machine learning algorithms.It is a challenge for traditional methods to con-figure hyperparameters of the capsule network to obtain high-performance manually.Some swarm intelligence or evolutionary computation algorithms have been effectively employed to seek optimal hyperparameters as a com-binatorial optimization problem.However,these algorithms are prone to get trapped in the local optimal solution as random search strategies are adopted.The inspiration for the hybrid rice optimization(HRO)algorithm is from the breeding technology of three-line hybrid rice in China,which has the advantages of easy implementation,less parameters and fast convergence.In the paper,genetic search is combined with the hybrid rice optimization algorithm(GHRO)and employed to obtain the optimal hyperparameter of the capsule network automatically,that is,a probability search technique and a hybridization strategy belong with the primary HRO.Thirteen benchmark functions are used to evaluate the performance of GHRO.Furthermore,the MNIST,Chest X-Ray(pneumonia),and Chest X-Ray(COVID-19&pneumonia)datasets are also utilized to evaluate the capsule network learnt by GHRO.The experimental results show that GHRO is an effective method for optimizing the hyperparameters of the capsule network,which is able to boost the performance of the capsule network on image classification.Zhiwei Ye Ziqian Fang Zhina Song Haigang Sui Chunyan Yan Wen Zhou Mingwei Wang 2023Intelligent Automation & Soft Computing2023,37,8:0

7ARTICLE OPEN Viral dynamics and antibody responses in people with asymptomatic SARS-CoV-2 infection显示文摘Over 40% of the coronavirus disease 2019(COVID-19)COVID-19 patients were asymptomatically infected with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and the immune responses of these asymptomatic individuals is a critical factor for developing the strategy to contain the COVID-19 pandemic.Here,we determined the viral dynamics and antibody responses among 143 asymptomatic individuals identified in a massive screening of more than 5 million people in eight districts of Wuhan in May 2020.Asymptomatic individuals were admitted to the government-designated centralized sites in accordance with policy.The incidence rate of asymptomatic infection is〜2.92/100,000.These individuals had low viral copy numbers(peaked at 315 copies/mL)and short-lived antibody responses with the estimated diminish time of 69 days.The antibody responses in individuals with persistent SARS-CoV-2 infection is much longer with the estimated diminish time of 257 days.These results imply that the immune responses in the asymptomatic individuals are not potent enough for preventing SARS-CoV-2 re-infection,which has recently been reported in recovered COVID-19 patients.This casts doubt on the efficacy of forming'herd-immunity'through natural SARS-CoV-2 infection and urges for the development of safe and effective vaccines.Zhiwei Sui Xinhua Dai Qingbin Lu Yulan Zhang Min Huang Shufen Li Tao Peng Jie Xie Yongzhuo Zhang Chunchen Wu Jianbo Xia Lianhua Dong Jiayi Yang Wenfeng Huang Siyuan Liu Ziquan Wang Ke Li Qingfang Yang Xi Zhou Ying Wu Wei Liu Xiang Fang Ke Peng 2021Signal Transduction and Targeted Therapy2021,6,6:0
