| 1 | First identification of kdr allele F1534S in VGSC gene and its association with resistance to pyrethroid insecticides in Aedes albopictus populations from Haikou City,Hainan Island,China显示文摘Background:Aedes albopictus is distributed widely in China,as a primary vector of Dengue fever and Chikungunya fever in south of China.Chemical insecticide control is one of the integrated programmes to prevent mosquito-borne diseases.Long-term applications of pyrethroids have resulted in the development of resistance in Ae.albopictus populations in China.However,the susceptibility of Ae.albopictus to pyrethroids in Hainan Island was unclear.Knockdown resistance(kdr),caused by point mutations in the VGSC gene,is one of the mechanisms that confer resistance to DDT and pyrethroids.This study was to investigate the resistance level of Ae.albopictus populations in Haikou City to three pyrethroid insecticides,and elucidate the relationship between the resistant phenotype and kdr mutations.Methods:The Aedes albopictus samples were collected in Xinbu Island(XI),Longtang Town(LT),Shishan Town(ST),Baishamen Park(BP),and Flower Market(FM)from Haikou City,Hainan Island,China.The larval susceptibility to deltamethrin,permethrin and beta-cypermethrin was tested by larval bioassays,and adult susceptibility to deltamethrin and DDT was determined by adult bioassays.The degree of resistance was determined by resistance ratio value(RR50>3)for larvae and by mortality for adult.The kdr alleles at codon 1534 of the VGSC gene were genotyped.The relationship between kdr genotypes and resistant phenotypes was analyzed by Chi-square test.Results:Out of five populations,assessed by larval bioassays,XI was susceptible to deltamethrin and permethrin;LT was susceptible to permethrin and beta-cypermethrin;and ST was susceptible to permithrin.FM and BP both were resistant to all of the three pyrethroids,and FM showed the highest degree of resistance,with RR50 values from 65.17 to 436.36.A total of 493 individuals from the larval bioassays were genotyped for kdr alleles.Five alleles were detected,including two wildtype alleles,TTC(F)(67.04%)and TTT(F)(0.41%),and three mutant alleles,TGC(C)(0.30%),TCC(S)(31.54%)and TTG(L)(0.71%).There was a clear correlation between mutant alleles(or F1534S)and resistant phenotypes(P<0.01).Conclusion:Two novel kdr mutant alleles F1534S and F1534L were detected in the pyrethroid resistant populations of Ae.albopictus in Haikou Hainan,China.For the first time,the mutant F1534S was associated with pyrethroid resistance in Ae.albopictus. | Huiying Chen Kaili Li Xiaohua Wang Xinyan Yang Yi Lin Fang Cai Wenbin Zhong Chunyan Lin Zhongling Lin Yajun Ma | 2016 | Infectious Diseases of Poverty2016,5,1: | 19 |

| 3 | Evolution of Thermoplastic Shear Localization and Related Microstructures in Al/SiC_p Composites Under Dynamic Compression显示文摘The localized shear deformation in the 2024 and 2124 Al matrix composites reinforced with SiC particles was investigated with a split Hopkinson pressure bar (SHPB) at a strain rate of about 2.0×103 s-1. The results showed that the occurrence of localized shear deformation is sensitive to the size of SiC particles. It was found that the critical strain, at which the shear localization occurs, strongly depends on the size and volume fraction of SiC particles. The smaller the particle size, the lower the critical strain required for the shear localization. TEM examinations revealed that Al/SiCp interfaces are the main sources of dislocations. The dislocation density near the interface was found to be high and it decreases with the distance from the particles. The Al matrix in shear bands was highly deformed and severely elongated at iow angle boundaries. The Al/SiCp interfaces, particularly the sharp corners of SiC particles,provide the sites for microcrack initiation. Eventual fracture is caused by the growth and coalescence of microcracks along the shear bands. | 6 | Nerve growth factor alleviates cerebral infarction and neurologic deficits by regulating VEGF,SDF-1 and S100A12 expression through PI3K pathway显示文摘Stroke remains the leading cause of death and disability worldwide,which destroys the quality of patients’lives and thus is becoming a heavy burden to the society.However,the current therapeutic approaches are far from satisfaction.The objective of this study is to elucidate the impact of nerve growth factor(NGF)on the brain damage induced by cerebral ischemia and its potential molecular mechanism.Middle cerebral artery occlusion(MCAO)rats were used as animal models and neurological functions were evaluated by modified Neurological Severity Score(NSS).Brain cell apoptosis was analyzed by TUNEL-positive staining while brain infarct size was determined according to 2% 2,3,5-triphenyltetrazolium chloride(TCC)staining volume.Rats receiving NGF demonstrated significantly alleviated brain damage,reflected by a substantial improvement in the neurobehavioral outcome,a decrease in brain cell apoptosis and shrinkage of brain infarct volume.Further analysis revealed a markedly elevated circulating vascular endothelial growth factor(VEGF)and stromal cell-derived factor 1(SDF-1)levels as well as a significant downregulation of SA10012 expression in NGF treated group compared with the untreated group.Strikingly,the protective effect of NGF on cerebral ischemic injury was abolished in rats treated with both NGF and PI3K inhibitors,indicating that phosphoinositide-3-kinase(PI3K)signaling is essential for NGF function.In conclusion,NGF treatment might be a potential therapeutic approach against cerebral infarction by downregulating SA10012 expression and upregulating VEGF,SDF-1 in a PI3K signaling dependent manner. | ZHENG LI ZHONGLE ZHAO PENG ZHANG | 2019 | BIOCELL2019,43,3: | 3 |
| 7 | Expression of hypoxia inducible factor-1 alpha and ischemic erythropoietin tolerance in the brain of cerebral ischemic tolerance model rats显示文摘BACKGROUND: Hypoxia inducible factor-1 alpha (HIF-1 α) and erythropoietin(EPO), possessing neuroprotective effect in the cerebral ischemia, might play an important role in the formation of cerebral ischemic tolerance (IT). OBJECTIVE:To observe the neuroprotective effect of cerebral ischemic preconditioning(IPC) of rats, and the expression and mechanism of HIF-1α and target gene erythropoietin in the brain tissue following the formation of cerebral IT. DESIGN:A randomized and controlled observation. SETTING: Department of Neurology, the Affiliated Hospital of Medical College, Qingdao University. MATERIALS: Totally 84 enrolled adult healthy male Wistar rats of clean grade, weighing 250 to 300 g, were provided by the Animal Experimental Department, Tongji Medical College of Huazhong University of Science and Technology. Ready-to-use SABC reagent kit and rabbit anti-rat HIF-1α monoclonal antibody were purchased from Boshide Bioengineering Co.Ltd (Wuhan); Rabbit anti-rat EPO monoclonal antibody was purchased from Santa Cruz Company (USA). METHODS: This experiment was carried out in the Department of Anatomy, Medical College, Qingdao University during March 2005 to March 2006. ① The 84 rats were divided into 3 groups by a lot: IPC group (n=40), sham-operation group (n=40) and control group (n=4). In the IPC group, middle cerebral artery was occluded for 2 hours respectively on the 1st, 3rd, 7th, 14th and 21st days of the reperfusion following 10-minute preischemia was made using a modified middle cerebral artery second suture method from Zea-Longa. The rats were sacrificed 22 hours after reperfusion in the end of middle cerebral artery occlusion (MCAO). That was to say, after 10-minute preischemia, suture was exited to the external carotid artery and embedded subcutaneously. Middle cerebral artery was occluded again to form the second reperfusion at the set time point after reperfusion. Twenty-two hours later, rats were sacrificed; In the sham-operation group,the preischemia was substituted by sham-operation(only common carotid artery and crotch were exposed, and MCAO by suture was omitted), and the other procedures were the same as those in the IPC group. In the control group, rats were given sham-operation twice at an interval of one day, and they were sacrificed 24 hours after the second sham-operation. ② Brain tissue was taken from the rats in each group. Cerebral infarction area of each layer was measured with TTC staining, and total cerebral infarction volume (The total cerebral infarction area of each layer×interspace ) was calculated. After brain tissue was stained by haematoxylin-esoin (HE), the form of nerve cells was observed under an optical microscope, and the expressions of HIF-1αand EPO protein in the brain tissue were detected with immunohistochemical method. MAIN OUTCOME MEASURES: ①Cerebral infarction volume;②form of nerve cell; ③ the expression of HIF-1α and EPO protein in the brain tissue. RESULTS:Totally 84 rats were enrolled in the experiment. The dead rats were randomly supplied during the experiment, and finally 84 rats entered the stage of result analysis. ① Detection of cerebral infarction volume of rats in each group: Cerebral infarction volume in the IPC group was significantly smaller than that in the sham-operation group on the 1st, 3rd and 7th days after reperfusion respectively [(161.2±6.9) mm3 vs (219.9±11.2) mm3, (134.9±9.0) mm3 vs (218.6±13.0) mm3, (142.9±13.7) mm3 vs (221.3±14.2) mm3, t=8.924,10.587,7.947, P < 0.01]. ② Observation of nerve cell form of brain tissue: HE staining showed that the ischemic degree, range and cerebral edema degree of IPC group were significantly milder than those of sham-operation group. ③ The expressions of HIF-1α and EPO protein in cerebral cortex and hippocampus : The expression of HIF-1αof IPC group was significantly higher than that of sham-operation group on the 1st, 3rd and 7th days after reperfusion respectively (125.93±3.79 vs 117.65±5.60, 140.63±4.64 vs 119.33±4.26, 131.15±2.74 vs 107.60±3.89, t=2.449, 6.763,9.899,P < 0.05-0.01). The expression of EPO of IPC group was significantly higher than that of sham-operation group on the 3rd and 7th days after perfusion respectively (141.68±3.29 vs 126.33±4.51, 138.88±2.59 vs 125.58±6.18,t=5.499,3.970, P < 0.05). | 8 | Commercial Production of Tiger Puffer(Takifugu rubripes)in Winter Using a Recirculating Aquaculture System显示文摘Tiger puffer(Takifugu rubripes) is a promising species for aquaculture production because of its high value and limited supply. However, in the north of China, using sea cages to culture this species in winter is hampered by the fact that the seawater temperature is extremely low. Here, a large scale commercial production of tiger puffer has been successfully realized using a recirculating aquaculture system(RAS) from 3 October 2012 to 31 May 2013. The RAS was comprised of nine culture tanks(total water volume 200 m3) and stocked with approximately a total of 14400 fish(initial mean weight 160 g). The tiger puffer was hand-fed at a rate of 0.7% of total body weight per day, and the feed conversion rate was(1.21 ± 0.3) kg kg-1. The recycle water in RAS was treated by a sieve bend screen, a foam fractionator, a submerged biofilter, an UV sterilizer and a submersible aerator. During the whole culture period, an excellent water quality control was achieved in RAS. At the end of this experiment, the survival rate of tiger puffer was more than 98%. | 16 | Influence of the low-frequency source parameters on the plasma characteristics in a dual frequency capacitively coupled plasma reactor:Two dimensional simulations显示文摘A two-dimensional (2D) fluid model is presented to study the discharge of argon in a dual frequency capacitively coupled plasma (CCP) reactor. We are interested in the influence of low-frequency (LF) source parameters such as applied voltage amplitudes and low frequencies on the plasma characteristics. In this paper, the high frequency is set to 60 MHz with voltage 50 V. The simulations were carried out for low frequencies of 1, 2 and 6 MHz with LF voltage 100 V, and for LF voltages of 60, 90 and 120 V with low frequency 2 MHz. The results of 2D distributions of electric field and ion density, the ion flux impinging on the substrate and the ion energy on the powered electrode are shown. As the low frequency increases, two sources become from uncoupling to coupling. When two sources are uncoupling, the increase in LF has little impact on the plasma characteristics, but when two sources are coupling, the increase in LF decreases the uniformities of ion density and ion flux noticeably.
