| 1 | Overexpression of Sirt6 ameliorates sleep deprivation induced-cognitive impairment by modulating glutamatergic neuron function显示文摘Sleep benefits the restoration of energy metabolism and thereby suppo rts neuronal plasticity and cognitive behaviors.Sirt6 is a NAD+-dependent protein deacetylase that has been recognized as an essential regulator of energy metabolism because it modulates various transcriptional regulators and metabolic enzymes.The aim of this study was to investigate the influence of Sirt6 on cerebral function after chronic sleep deprivation(CSD).We assigned C57BL/6J mice to control or two CSD groups and subjected them to AAV2/9-CMV-EGFP or AAV2/9-CMV-Sirt6-EGFP infection in the prelimbic cortex(PrL).We then assessed cerebral functional connectivity(FC) using resting-state functional MRI,neuron/astrocyte metabolism using a metabolic kinetics analysis;dendritic spine densities using sparse-labeling;and miniature excitato ry postsynaptic currents(mEPSCs) and action potential(AP) firing rates using whole-cell patchclamp recordings.In addition,we evaluated cognition via a comprehensive set of behavioral tests.Compared with controls,Sirt6 was significantly decreased(P<0.05) in the PrL after CSD,accompanied by cognitive deficits and decreased FC between the PrL and accumbens nucleus,piriform cortex,motor co rtex,somatosensory co rtex,olfactory tubercle,insular cortex,and cerebellum.Sirt6 ove rexpression reve rsed CSD-induced cognitive impairment and reduced FC.Our analysis of metabolic kinetics using [1-13C] glucose and [2-13C] acetate showed that CSD reduced neuronal Glu4and GABA2synthesis,which could be fully restored via forced Sirt6 expression.Furthermore,Sirt6 ove rexpression reversed CSD-induced decreases in AP firing rates as well as the frequency and amplitude of mEPSCs in PrL pyramidal neurons.These data indicate that Sirt6 can improve cognitive impairment after CSD by regulating the PrL-associated FC network,neuronal glucose metabolism,and glutamatergic neurotransmission.Thus,Sirt6 activation may have potential as a novel strategy for treating sleep disorder-related diseases. | Jinpiao Zhu Chang Chen Zhen Li Xiaodong Liu Jingang He Ziyue Zhao Mengying He Binbin Nie Zili Liu Yingying Chen Kuanpin Su Xiang Li Juxiang Chen Hongbing Xiang Fuqiang Xu Kangguang Lin Zongze Zhang Jie Wang | 2023 | Neural Regeneration Research2023,18,11: | 1 |
| 2 | A critical review on the technique and mechanism of microwave-based denitrification in flue gas显示文摘Microwave radiation has received extensive attention due to its significant thermal and nonthermal effects, and the development of MW-based denitrification in flue gas has become one of the most promising methods to avoid the defects of ammonia escape, high temperature and cost in traditional SCR. This review introduces the thermal and non-thermal effects of microwaves and divides MW-based denitrification methods into MW reduction and oxidation denitrification, systematically summarizes these denitrification methods, including MW discharge reduction, MW-induced catalytic reduction using active carbon, molecular sieves, metal oxides(transition metals, perovskites, etc.), MW-induced oxidation denitrification with and without additional oxidant, and discusses their removal pathway and mechanism. | 4 | Microcarriers promote the through interface movement of mouse trophoblast stem cells by regulating stiffness显示文摘Mechanical force is crucial in the whole process of embryonic development.However,the role of trophoblast mechanics during embryo implantation has rarely been studied.In this study,we constructed a model to explore the effect of stiffness changes in mouse trophoblast stem cells(mTSCs)on implantation:microcarrier was prepared by sodium alginate using a droplet microfluidics system,and mTSCs were attached to the microcarrier surface with laminin modifications,called T(micro).Compared with the spheroid,formed by the self-assembly of mTSCs(T(sph)),we could regulate the stiffness of the microcarrier,making the Young’s modulus of mTSCs(367.70±79.81 Pa)similar to that of the blastocyst trophoblast ectoderm(432.49±151.90 Pa).Moreover,T(micro)contributes to improve the adhesion rate,expansion area and invasion depth of mTSCs.Further,T(micro)was highly expressed in tissue migration-related genes due to the activation of the Rho-associated coiled-coil containing protein kinase(ROCK)pathway at relatively similar modulus of trophoblast.Overall,our study explores the embryo implantation process with a new perspective,and provides theoretical support for understanding the effect of mechanics on embryo implantation. | Zili Gao Jia Guo Bo Gou Zhen Gu Tan Jia Sinan Ma Liyuan Jiang Wenli Liu Lixun Zhou Qi Gu | 2023 | Bioactive Materials2023,,10: | 0 |
| 5 | An experimental study on the effects of friction modifiers on wheel-rail dynamic interactions with various angles of attack显示文摘By modifying friction to the desired level,the application of friction modifiers(FMs)has been considered as a promising emerging tool in the railway engineering for increasing braking/traction force in poor adhesion conditions and mitigating wheel/rail interface deterioration,energy consumption,vibration and noise.Understanding the effectiveness of FMs in wheel–rail dynamic interactions is crucial to their proper applications in practice,which has,however,not been well explained.This study experimentally investigates the effects of two types of top-of-rail FM,i.e.FM-A and FM-B,and their application dosages on wheel–rail dynamic interactions with a range of angles of attack(AoAs)using an innovative well-controlled V-track test rig.The tested FMs have been used to provide intermediate friction for wear and noise reduction.The effectiveness of the FMs is assessed in terms of the wheel–rail adhesion characteristics and friction rolling induced axle box acceleration(ABA).This study provides the following new insights into the study of FM:the applications of the tested FMs can both reduce the wheel–rail adhesion level and change the negative friction characteristic to positive;stick–slip can be generated in the V-Track and eliminated by FM-A but intensified by FM-B,depending on the dosage of the FMs applied;the negative friction characteristic is not a must for stick–slip;the increase in ABA with AoA is insignificant until stick–slip occurs and the ABA can thus be influenced by the applications of FM. | Zhen Yang Pan Zhang Jan Moraal Zili Li | 2022 | Railway Engineering Science2022,30,3: | 0 |
| 6 | Toxicity and bio-distribution of carbon dots after single inhalation exposure in vivo显示文摘Because of the advantages of excellent light stability, carbon dots(CDs) are considered to be a promising agent in the bio-marker application. Nevertheless, there are many unresolved issues with the toxicity of CDs in vitro and in vivo. In the study, CDs were synthesized by citric acid and ethylenediamine into deionized water, then the inhalation toxicity and bio-distribution of CDs in vivo were systematically assessed. The results showed that CDs caused animals death at higher dosages and induced injury in the lung and liver including inflammation and necrosis after single inhalation exposure at 5, 2 and 1 mg/kg dosages of the CDs. We also found that the injury increase with a dose-dependent and time-dependent manner. Fluorescent examination and TEM results showed that CDs mainly located at the lung and liver.And the fluorescent intensity increase with a time-dependent manner.
