1Global Dynamic Molecular Profiling of Stomatal Lineage Cell Development by Single-Cell RNA Sequencing显示文摘The regulation of stomatal lineage cell development has been extensively investigated.However,a comprehensive characterization of this biological process based on single-cell transcriptome analysis has not yet been reported.In this study,we performed RNA sequencing on 12844 individual cells from the cotyledons of 5-day-old Arabidopsis seedlings.We identified 11 cell clusters corresponding mostly to cells at specific stomatal developmental stages using a series of marker genes.Comparative analysis of genes with the highest variable expression among these cell clusters revealed transcriptional networks that regulate development from meristemoid mother cells to guard mother cells.Examination of the developmental dynamics of marker genes via pseudo-time analysis revealed potential interactions between these genes.Collectively,our study opens the door for understanding how the identified novel marker genes participate in the regulation of stomatal lineage cell development.Zhixin Liu Yaping Zhou Jinggong Guo Jiaoai Li Zixia Tian Zhinan Zhu Jiajing Wang Rui Wu Bo Zhang Yongjian Hu Yijing Sun Yan Shangguan Weiqiang Li Tao Li Yunhe Hu Chenxi Guo Jean-David Rochaix Yuchen Miao Xuwu Sun 2020Molecular Plant2020,13,8:11

3Comparison of Cloud Properties between Cloud Sat Retrievals and Airplane Measurements in Mixed-Phase Cloud Layers of Weak Convective and Stratus Clouds显示文摘Cloud microphysical properties including liquid and ice particle number concentration(NC),liquid water content(LWC),ice water content(IWC) and effective radius(RE) were retrieved from Cloud Sat data for a weakly convective and a widespread stratus cloud. Within the mixed-phase cloud layers,liquid-phase fractions needed to be assumed in the data retrieval process,and one existing linear(p1) and two exponential(p2and p3) functions,which estimate the liquid-phase fraction as a function of subfreezing temperature(from-20?C to 0?C),were tested. The retrieved NC,LWC,IWC and RE using p1 were on average larger than airplane measurements in the same cloud layer. Function p2 performed better than p1 or p3in retrieving the NCs of cloud droplets in the convective cloud,while function p1 performed better in the stratus cloud. Function p3 performed better in LWC estimation in both convective and stratus clouds. The REs of cloud droplets calculated using the retrieved cloud droplet NC and LWC were closer to the values of in situ observations than those retrieved directly using the p1 function. 4PARP-1 inhibitors enhance the chemosensitivity of leukemia cells by attenuating NF-κB pathway activity and DNA damage response induced by Idarubicin显示文摘Idarubicin(IDA),an anthracycline antineoplastic drug,is commonly used in the treatment of acute myeloid leukemia(AML)with reasonable response rates and clinical benefits.However,some patients still relapse,or do not respond,and suffer high fatality rates.Recent studies have shown that overexpression of PARP-1 may represent an important risk factor in AML patients.The aim of the present study was to determine the underlying molecular mechanisms by which the PARP-1 inhibitor Olaparib enhances the chemosensitivity of the leukemia cell line K562 and THP1 to IDA.Our data demonstrated that PARP-1 is upregulated in AML patients as well as in K562 and THP1 cells,and that the suppression of PARP-1 activity by Olaparib enhances the inhibitory effect of IDA.A mechanistic study revealed that Olaparib decreases the expressions of p-ATM,p-IκBα,XIAP and p65,and upregulates Bax,cleaved-Caspase-3 andγ-H2AX.Olaparib can enhance the induction of DNA damage by IDA,probably mediated by the inhibition of the ATM-related DNA damage response.Moreover,we also found that the nuclear translocation of p65 and the nuclear export of NEMO are inhibited when IDA and Olaparib are combined.Our results suggest that Olaparib attenuates the activity of the NF-κB pathway and decreases the DNA damage response induced by IDA.Therefore,we conclude that Olaparib is a potentially valuable chemosensitizer for leukemia patients.Bo Ke Anna Li Huan Fu Chunfang Kong Tingting Liu Qingqing Zhu Yue Zhang Zixia Zhang Chen Chen Chenghao Jin 2022Acta Biochimica et Biophysica Sinica2022,54,1:1
