| 1 | A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. | 2 | Intracoronary arterial retrograde thrombolysis with percutaneous coronary intervention: a novel use of thrombolytic to treat acute ST-segment elevation myocardial infarction显示文摘Background Clearance of coronary arterial thrombosis is necessary in patients with acute ST-segment elevation myocardial infarction (STEMI) undergoing urgent percutaneous coronary intervention (PCI). There is currently no highly-recommended method of thrombus removal during interventional procedures. We describe a new method for opening culprit vessels to treat STEMI: intracoronary arterial retrograde thrombolysis (ICART) with PCI. Methods & Results Eight patients underwent ICART. The guidewire was advanced to the distal coronary artery through the occlusion lesion. Then, we inserted a microcatheter into the distal end of the occluded coronary artery over the guidewire. Urokinase (5–10 wu) mixed with contrast agents was slowly injected into the occluded section of the coronary artery through the microcatheter. The intracoronary thrombus gradually dissolved in 3–17 min, and the effect of thrombolysis was visible in real time. Stents were then implanted according to the characteristics of the recanalized culprit lesion to achieve full revascularization. One patient experienced premature ventricular contraction during vascular revascularization, and no malignant arrhythmias were seen in any patient. No reflow or slow flow was not observed post PCI. Thrombolysis in myocardial infarction flow grade and myocardial blush grade post-primary PCI was 3 in all eight patients. No patients experienced bleeding or stroke. Conclusions ICART was accurate and effective for treating intracoronary thrombi in patients with STEMI in this preliminary study. ICART was an effective, feasible, and simple approach to the management of STEMI, and no intraprocedural complications occurred in any of the patients. | 4 | Expression and role of specificity protein 1 in the sclera remodeling of experimental myopia in guinea pigs显示文摘AIM:To study the expression of collagen I and transcription factor specificity protein 1(Sp1),a transforming growth factor-β1(TGF-β1) downstream target,and reveal the impact of the TGF-β1-Sp1 signaling pathway on collagen remodeling in myopic sclera.METHODS:Seventy-five 1-week-old guinea pigs were randomly divided into normal control,form deprivation myopia(FDM),and self-control groups.FDM was induced for different times using coverage with translucent latex balloons and FDM recovery was performed for 1wk after 4wk treatment;then,changes in refractive power and axial length were measured.Immunohistochemistry and reverse transcription-polymerase chain reaction were used to evaluate dynamic changes in collagen I and Sp1 expression in the sclera of guinea pigs with emmetropia and experimental myopia,and the relationship between collagen I and Sp1 levels was analyzed.RESULTS:In the FDM group,the refractive power was gradually changed(from 2.09±0.30 D at week 0 to-1.23±0.69 D,-4.17±0.59 D,-7.07±0.56 D,and-4.30±0.58 D at weeks 2,4,6,and 1wk after 4wk,respectively;P<0.05),indicating deepening of myopia.The axial length was increased(from 5.92±0.39 mm at week 0 to 6.62±0.36 mm,7.30±0.34 mm,7.99±0.32 mm,and 7.41±0.36 mm at weeks 2,4,6,and 1wk after 4wk;P<0.05).The m RNA and protein expression of Sp1 and collagen I in the sclera of the FDM group was lower than that of the control groups(P<0.05),and the reduction was eye-coverage time-dependent.Furthermore,correlation between Sp1 and collagen I down-regulation in the myopic sclera was observed.CONCLUSION:Our data indicate that transcription factor Sp1 may be involved in the regulation of type I collagensynthesis/degradation during myopic sclera remodeling,suggesting that TGF-β1 signaling plays a role in the development and progression of myopia. | Bo Jiang Zhang-You Wu Zi-Cheng Zhu Gen-Jie Ke Yue-Chun Wen Si-Qin Sun | 2017 | International Journal of Ophthalmology(English edition)2017,10,4: | 15 |

| 7 | Rapid and Accurate Sequencing of Enterovirus Genomes Using MinION Nanopore Sequencer显示文摘Objective Knowledge of an enterovirus genome sequence is very important in epidemiological investigation to identify transmission patterns and ascertain the extent of an outbreak.The MinION sequencer is increasingly used to sequence various viral pathogens in many clinical situations because of its long reads,portability,real-time accessibility of sequenced data,and very low initial costs.However,information is lacking on MinION sequencing of enterovirus genomes.Methods In this proof-of-concept study using Enterovirus 71(EV71) and Coxsackievirus A16(CA16) strains as examples,we established an amplicon-based whole genome sequencing method using MinION.We explored the accuracy,minimum sequencing time,discrimination and high-throughput sequencing ability of MinION,and compared its performance with Sanger sequencing.Results Within the first minute(min) of sequencing,the accuracy of MinION was 98.5% for the single EV71 strain and 94.12%-97.33% for 10 genetically-related CA16 strains.In as little as 14 min,99% identity was reached for the single EV71 strain,and in 17 min(on average),99% identity was achieved for 10 CA16 strains in a single run.Conclusion MinION is suitable for whole genome sequencing of enteroviruses with sufficient accuracy and fine discrimination and has the potential as a fast,reliable and convenient method for routine use. | WANG Ji KE Yue Hua ZHANG Yong HUANG Ke Qiang WANG Lei SHEN Xin Xin DONG Xiao Ping XU Wen Bo MA Xue Jun | 2017 | Biomedical and Environmental Sciences2017,30,10: | 11 |

| 9 | Waste oyster shell as a kind of active filler to treat the combined wastewater at an estuary显示文摘Estuaries have been described as one of the most difcult environments on Earth. It is difcult to know how to treat the combined wastewater in tidal rivers at the estuary, where the situation is very diferent from ordinary fresh water rivers. Waste oyster shell was used as the active filler in this study in a bio-contact oxidation tank to treat the combined wastewater at the Fengtang Tidal River. With a middle-experimental scale of 360 m3/day, the average removal efciency of COD, BOD, NH3-N, TP and TSS was 80.05%, 85.02%,86.59%, 50.58% and 85.32%, respectively, in this bio-contact oxidation process. The living microbes in the biofilms on the waste oyster shell in this bio-contact oxidation tank, which were mainly composed of zoogloea, protozoa and micro-metazoa species, revealed that waste oyster shell as the filler was suitable material for combined wastewater degradation. This treatment method using waste oyster shell as active filler was then applied in a mangrove demonstration area for water quality improvement near the experiment area, with a treatment volume of 5 × 103m3/day. Another project was also successfully applied in a constructed wetland, with a wastewater treatment volume of 1 × 103m3/day. | 14 | A method of determining the permeability coefficient of coal seam based on the permeability of loaded coal显示文摘This study developed the equipment for thermo-fluid–solid coupling of methane-containing coal, and investigated the seepage character of loaded coal under different working conditions. Regarding the effective pressure as a variable, the variation characteristics of the gas permeability of loaded methane-containing coal has been studied under the conditions of different confining pressures and pore pressures. The qualitative and quantitative relationship between effective stress and permeability of loaded methane-containing coal has been established, considering the adsorption of deformation, amount of pore gas compression and temperature variation. The results show that the permeability of coal samples decreases along with the increasing effective stress. Based on the Darcy law, the correlation equation between the effective stress and permeability coefficient of coal seam has been established by combining the permeability coefficient of loaded coal and effective stress. On the basis of experimental data, this equation is used for calculation, and the results are in accordance with the measured gas permeability coefficient of coal seam. | 15 | Effect of tin addition on corrosion behavior of a low-alloy steel in simulated costal-industrial atmosphere显示文摘The effect of tin addition on the atmospheric corrosion behavior of a low-alloy steel in sim ulated coastalindustrial atm osphere has been investigated by indoor wet/dry cyclic corrosion test (CCT). The results indicate that tin addition can obviously make the steel substrate more resistant to atmospheric corrosion by suppressing the cathodic H^+ reduction reaction, and but tin addition is not of obvious beneficial effect when the steel is covered with a thicker rust layer during long-term corrosion process. The reason lies in the fact that the presence of un-reduced H^+ can lower the electrolyte pH value and lead to a loose and porous rust layer on tin-containing steel sample than that on tin-free steel sample. In addition, the 120 CCT cycles corrosion process of the two steels can be divided into three stages. Both the tin-free and tin-containing steels show an increasing corrosion rate during the initial corrosion stage and then exhibit a decreasing corrosion rate during the second and third corrosion stages. Growing demand from the user community for a small beam and related experimental methods have motivated upgrades of the devices in the endstation.Minibeam modes have already been developed for operation. A self-integrated diffractometer reduces the sphere of confusion of the rotatory axis to 1μm. The new diffractometer is equipped with an upgraded on-axis viewing system that can improve the resolving power. Additionally,the area detector was also upgraded to the newest generation of detectors, the EIGER X 16 M, which can collect data at 133 Hz. After these upgrades, the endstation became virtually new. | 18 | High levels of circulating GM-CSF^(+) CD4^(+) T cells are predictive of poor outcomes in sepsis patients: a prospective cohort study显示文摘Granulocyte colony-stimulating factor(GM-CSF),produced by CD4^(+)T cells,has recently been implicated in the pathogenesis of inflammatory diseases,such as multiple sclerosis and juvenile arthritis.However,the role of GM-CSF-producing CD4^(+)T cells in sepsis remains unknown.This study reports peripheral changes in GM-CSF-producing CD4^(+)T cells in septic patients and the possible underlying mechanism by which GM-CSF influences the outcome of sepsis.Forty-three septic patients,20 SIRS patients,and 20 healthy controls were enrolled in this study and followed for 28 days to assess mortality.We measured the peripheral frequency of GM-CSF^(+)CD4^(+)T cells and recorded their associated relationship with disease progression.Our data demonstrated that peripheral GM-CSF-producing CD4^(+)T cells were significantly higher in septic patients than in both SIRS patients and healthy controls.These cells exhibit a memory phenotype and impaired IFN-γ-secreting capacity in sepsis patients.Using a receiver operating curve analysis with 8.01%as a cut-off point,the percentage of GM-CSF^(+)CD4^(+)T cells could predict the outcome of septic patients.Combined with the increase in GM-CSF-producing CD4^(+)T cells,inflammatory cytokines IL-1βand IL-6 were also upregulated.Using an in vitro neutrophil model,we found that GM-CSF inhibited C3aR expression,while inducing IL-8 production.Furthermore,this effect was transferrable in plasma from sepsis patients and was attenuated by inhibition of GM-CSF using an anti-GM-CSF antibody.These results indicate that GM-CSF-producing CD4^(+)T cells may serve as a marker of sepsis severity.Thus,targeting GM-CSF overproduction may benefit sepsis patients. | Huihuang Huang Siyu Wang Tianjun Jiang Rong Fan Zheng Zhang Jinsong Mu Ke Li Yonggang Wang Lei Jin Fang Lin Jie Xia Lijian Sun Biao Xu Chengcheng Ji Jing Chen Juan Chang Bo Tu Bing Song Chao Zhang Fu-Sheng Wang Ruonan Xu | 2019 | Cellular & Molecular Immunology2019,16,6: | 6 |
