| 2 | Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. | 7 | Linear Acceleration of an Undulatory Robotic Fish with Dynamic Morphing Median Fin under the Instantaneous Self-propelled Condition显示文摘Fish commonly execute rapid linear accelerations initiated during steady swimming,yet the function of the median fins during this process is less understood.We find that the erection/folding time(from the starting time of the linear acceleration(0 s)to the starting time of the folding movement of the fin),as well as the spreading area of the median fins,actively change during the linearacceleration of the live largemouth bass(Micropterus salmoides).To better understand the influence of the folding time and the area change of the median fins on the linear acceleration,we implemented an undulatory biomimetic robotic fish with soft median fins that can be programmed to erect and fold,just like a live fish.To characterize the acceleration performance of the robotic fish,we developed a'self-propelled'experiment technique based on the Kalman filter and Proportional-Integral-Derivative(PID)control algorithm.The experiments on the robotic fish show the acceleration induced by fully-erected median fins increases by 46.3%.Fully-erected median fins positively contribute to propulsion primarily at the onset stage of the linear acceleration while result in a significant decrease in steady swimming speed by 25%,which suggests a large drag force is induced at the steady swimming stage due to the enlarged wetted area.Parametric sweeping experiments on erection/folding time and spreading area demonstrate a proper combination of the erection/folding time and the spreading area enhances the mean linear acceleration by up to 85%.Particle Image Velocimetry(PIV)results reveal that the vortexes shed by the erected dorsal fin are stronger than those shed by the folded fin.As the acceleration process progresses,the thrust generated by the dorsal fins gradually is weakened until only resistance is generated in the end.Our findings may shed light on the realization of controllable surfaces on high performance fish-inspired robotic systems in the future. | Wenguang Sun Zemin Liu Ziyu Ren Gang Wang Tao Yuan Li Wen | 2020 | Journal of Bionic Engineering2020,17,2: | 1 |
| 8 | Candidate therapeutic agents in a newly established triple wild-typemucosalmelanoma cell line显示文摘Background:Mucosalmelanoma has characteristically distinct genetic features and typically poor prognosis.The lack of representativemucosal melanoma models,especially cell lines,has hindered translational research on this melanoma subtype.In this study,we aimed to establish and provide the biological properties,genomic features and the pharmacological profiles of a mucosal melanoma cell line that would contribute to the understanding and treatment optimization of molecularly-defined mucosal melanoma subtype.Methods:The sample was collected from a 67-year-old mucosal melanoma patient and processed into pieces for the establishment of cell line and patientderived xenograft(PDX)model.The proliferation and tumorigenic property of cancer cells from different passageswere evaluated,andwhole-genome sequencing(WGS)was performed on the original tumor,PDX,established cell line,and the matched blood to confirm the establishment and define the genomic features of this cell line.AmpliconArchitect was conducted to depict the architecture of amplified regions detected by WGS.High-throughput drug screening(HTDS)assay including a total of 103 therapeutic agents was implemented on the established cell line,and selected candidate agents were validated in the corresponding PDX model.Results:A mucosal melanoma cell line,MM9H-1,was established which exhibited robust proliferation and tumorigenicity after more than 100 serial passages.Genomic analysis of MM9H-1,corresponding PDX,and the original tumor showed genetic fidelity across genomes,and MM9H-1 was defined as a triple wild-type(TWT)melanoma subtype lacking well-characterized“driver mutations”.Instead,the amplification of several oncogenes,telomerase reverse transcriptase(TERT),v-Rafmurine sarcoma viral oncogene homolog B1(BRAF),melanocyte Inducing transcription factor(MITF)and INO80 complex ATPase subunit(INO80),via large-scale genomic rearrangement potentially contributed to oncogenesis of MM9H-1.Moreover,HTDS identified proteasome inhibitors,especially bortezomib,as promising therapeutic candidates for MM9H-1,which was verified in the corresponding PDX model in vivo.Conclusions:We established and characterized a new mucosal melanoma cell line,MM9H-1,and defined this cell line as a TWT melanoma subtype lacking well-characterized“driver mutations”.The MM9H-1 cell line could be adopted as a unique model for the preclinical investigation of mucosal melanoma. | Chaoji Shi Ziyue Gu Shengming Xu Houyu Ju Yunteng Wu Yong Han Jiayi Li Chuwen Li Jing Wu Lizhen Wang Jiang Li Guoyu Zhou Weimin Ye Guoxin Ren Zhiyuan Zhang Rong Zhou | 2022 | Cancer Communications2022,42,7: | 1 |
| 9 | Regulation of serotonin production by specific microbes from piglet gut显示文摘Background Serotonin is an important signaling molecule that regulates secretory and sensory functions in the gut.Gut microbiota has been demonstrated to affect serotonin synthesis in rodent models.However,how gut microbes regulate intestinal serotonin production in piglets remains vague.To investigate the relationship between microbiota and serotonin specifically in the colon,microbial composition and serotonin concentration were analyzed in ileum-cannulated piglets subjected to antibiotic infusion from the ileum when comparing with saline infusion.Microbes that correlated positively with serotonin production were isolated from piglet colon and were further used to investi-gate the regulation mechanisms on serotonin production in IPEC-J2 and a putative enterochromaffin cell line RIN-14B cells.Results Antibiotic infusion increased quantities of Lactobacillus amylovorus(LA)that positively correlated with increased serotonin concentrations in the colon,while no effects observed for Limosilactobacillus reuteri(LR).To understand how microbes regulate serotonin,representative strains of LA,LR,and Streptococcus alactolyticus(SA,enriched in feces from prior observation)were selected for cell culture studies.Compared to the control group,LA,LR and SA supernatants significantly up-regulated tryptophan hydroxylase 1(TPH1)expression and promoted serotonin production in IPEC-J2 cells,while in RIN-14B cells only LA exerted similar action.To investigate potential mechanisms mediated by microbe-derived molecules,microbial metabolites including lactate,acetate,glutamine,andγ-aminobutyric acid were selected for cell treatment based on computational and metabolite profiling in bacte-rial supernatant.Among these metabolites,acetate upregulated the expression of free fatty acid receptor 3 and TPH1 while downregulated indoleamine 2,3-dioxygenase 1.Similar effects were also recapitulated when treating the cells with AR420626,an agonist targeting free fatty acid receptor 3.Conclusions Overall,these results suggest that Lactobacillus amylovorus showed a positive correlation with sero-tonin production in the pig gut and exhibited a remarkable ability to regulate serotonin production in cell cultures.These findings provide evidence that microbial metabolites mediate the dialogue between microbes and host,which reveals a potential approach using microbial manipulation to regulate intestinal serotonin biosynthesis. | Ziyu Liu Yidan Ling Yu Peng Shuibing Han Yuting Ren Yujia Jing Wenlu Fan Yong Su Chunlong Mu Weiyun Zhu | 2023 | Journal of Animal Science and Biotechnology2023,14,6: | 0 |
