1A bHLH transcription activator regulates defense signaling by nucleo-cytosolic trafficking in rice显示文摘Crosstalk between plant hormone signaling pathways is vital for controlling the immune response during pathogen invasion.Salicylic acid(SA)and jas-monic acid(JA)often play important but antagonistic roles in the immune responses of higher plants.Here,we identify a basic helix-loop-helix transcription acti-vator,OsbHLH6,which confers disease resistance in rice by regulating SA and JA signaling via nucleo-cytosolic trafficking in rice(Oryza sativa).OsbHLH6 expression was upregulated during Magnaporthe oryzae infection.Transgenic rice plants overexpressing OsbHLH6 display increased JA responsive gene expression and enhanced disease susceptibility to the pathogen.Nucleus-localized OsbHLH6 activates JA signaling and suppresses SA signaling;however,the SA regulator OsNPR1(Nonexpressor of PR genes 1)sequesters OsbHLH6 in the cytosol to alleviate its effect.Our data suggest that OsbHLH6 controls disease re-sistance by dynamically regulating SA and JA signaling.Fanwei Meng Chao Yang Jidong Cao Huan Chen Jinhuan Pang Qiqi Zhao Zongyi Wang Zheng Qing Fu Jun Liu 2020Journal of Integrative Plant Biology2020,62,10:10
2PI3K/Akt/mTOR signaling orchestrates the phenotypic transition and chemo-resistance of small cell lung cancer显示文摘Small cell lung cancer(SCLC)is a phenotypically heterogeneous disease with an extremely poor prognosis,which is mainly attributed to the rapid development of resistance to chemotherapy.However,the relation between the growth phenotypes and chemo-resistance of SCLC remains largely unclear.Through comprehensive bioinformatic analyses,we found that the heterogeneity of SCLC phenotype was significantly associated with different sensitivity to chemotherapy.Adherent or semiadherent SCLC cells were enriched with activation of the PI3K/Akt/mTOR pathway and were highly chemoresistant.Mechanistically,activation of the PI3K/Akt/mTOR pathway promotes the phenotypic transition from suspension to adhesion growth pattern and confers SCLC cells with chemo-resistance.Such chemo-resistance could be largely overcome by combining chemotherapy with PI3K/Akt/mTOR pathway inhibitors.Our findings support that the PI3K/Akt/mTOR pathway plays an important role in SCLC phenotype transition and chemo-resistance,which holds important clinical implications for improving SCLC treatment.Xuefeng Li Cheng Li Chenchen Guo Qiqi Zhao Jiayu Cao Hsin-Yi Huang Meiting Yue Yun Xue Yujuan Jin Liang Hu Hongbin Ji 2021Journal of Genetics and Genomics2021,48,7:5
3Spatio-temporal variations of soil water content and salinity around individual Tamarix ramosissima in a semi-arid saline region of the upper Yellow River, Northwest China显示文摘Ecological restoration by Tamarix plants on semi-arid saline lands affects the accumulation, distribution patterns and related mechanisms of soil water content and salinity. In this study, spatio-temporal variations of soil water content and salinity around natural individual Tamarix ramosissima Ledeb. were invetigated in a semi-arid saline region of the upper Yellow River, Northwest China. Specifically, soil water content, electrical conductivity(EC_e), sodium adsorption ratio(SAR_e), and salt ions(including Na^+, K^+, Ca^(2+), Mg^(2+) and SO_4^(2–)) were measured at different soil depths and at different distances from the trunk of T. ramosissima in May, July, and September 2016. The soil water content at the 20–80 cm depth was significantly lower in July and September than in May, indicating that T. ramosissima plants absorb a large amount of water through the roots during the growing period, leading to the decreasing of soil water content in the deep soil layer. At the 0–20 cm depth, there was a salt island effect around individual T. ramosissima, and the EC_e differed significantly inside and outside the canopy of T. ramosissima in May and July. Salt bioaccumulation and stemflow were two major contributing factors to this difference. The SAR_e at the 0–20 cm depth was significantly different inside and outside the canopy of T. ramosissima in the three sampling months. The values of SAR_e at the 60–80 cm depth in May and July were significantly higher than those at the 0–60 cm depth and higher than that at the corresponding depth in September. The distribution of Na^+ in the soil was similar to that of the SAR_e, while the concentrations of K^+, Ca^(2+), and Mg^(2+) showed significant differences among the sampling months and soil depths. Both season and soil depth had highly significant effects on soil water content, EC_e and SAR_e, whereas distance from the trunk of T. ramosissima only significantly affected EC_e. Based on these results, we recommend co-planting of shallow-rooted salt-tolerant species near the Tamarix plants and avoiding planting herbaceous plants inside the canopy of T. ramosissima for afforestation in this semi-arid saline region. 5Reductive damage induced autophagy inhibition for tumor therapy显示文摘Numerous therapeutic anti-tumor strategies have been developed in recent decades.However,their therapeutic efficacy is reduced by the intrinsic protective autophagy of tumors.Autophagy plays a key role in tumorigenesis and tumor treatment,in which the overproduction of reactive oxygen species(ROS)is recognized as the direct cause of protective autophagy.Only a few molecules have been employed as autophagy inhibitors in tumor therapy to reduce protective autophagy.Among them,hydroxychloroquine is the most commonly used autophagy inhibitor in clinics,but it is severely limited by its high therapeutic dose,significant toxicity,poor reversal efficacy,and nonspecific action.Herein,we demonstrate a reductive-damage strategy to enable tumor therapy by the inhibition of protective autophagy via the catalytic scavenging of ROS using porous nanorods of ceria(PN-CeO_(2))nanozymes as autophagy inhibitor.The antineoplastic effects of PN-CeO_(2)were mediated by its high reductive activity for intratumoral ROS degradation,thereby inhibiting protective autophagy and activating apoptosis by suppressing the activities of phosphatidylinositide 3-kinase/protein kinase B and p38 mitogen-activated protein kinase pathways in human cutaneous squamous cell carcinoma.Further investigation highlighted PN-CeO_(2)as a safe and efficient anti-tumor autophagy inhibitor.Overall,this study presents a reductive-damage strategy as a promising anti-tumor approach that catalytically inhibits autophagy and activates the intrinsic antioxidant pathways of tumor cells and also shows its potential for the therapy of other autophagy-related diseases.Yuqian Wang Yingjian Huang Yu Fu Zhixiong Guo Da Chen Fangxian Cao Qi Ye Qiqi Duan Meng Liu Ning Wang Dan Han Chaoyi Qu Zhimin Tian Yongquan Qu Yan Zheng 2023Nano Research2023,16,4:1
6Aristolochic acids exposure was not the main cause of liver tumorigenesis in adulthood显示文摘Aristolochic acids(AAs) have long been considered as a potent carcinogen due to its nephrotoxicity. Aristolochic acid I(AAI) reacts with DNA to form covalent aristolactam(AL)-DNA adducts,leading to subsequent A to T transversion mutation, commonly referred as AA mutational signature. Previous research inferred that AAs were widely implicated in liver cancer throughout Asia. In this study, we explored whether AAs exposure was the main cause of liver cancer in the context of HBV infection in China's Mainland. Totally 1256 liver cancer samples were randomly retrieved from 3 medical centers and a refined bioanalytical method was used to detect AAI-DNA adducts. 5.10% of these samples could be identified as AAI positive exposure. Whole genome sequencing suggested 8.41% of 107 liver cancer patients exhibited the dominant AA mutational signature, indicating a relatively low overall AAI exposure rate. In animal models, long-term administration of AAI barely increased liver tumorigenesis in adult mice, opposite from its tumor-inducing role when subjected to infant mice. Furthermore, AAI induced dose-dependent accumulation of AA-DNA adduct in target organs in adult mice, with the most detected in kidney instead of liver. 7The formation kinetics and control of biofilms by three dominant fungi species isolated from groundwater显示文摘Filamentous fungi can enter drinking water supply systems in various ways,and exist in suspended or sessile states which threatens the health of individuals by posing a high risk of invasive infections.In this study,the biofilms formation kinetics of the three genera of fungal spores,Aspergillus niger(A.niger),Penicillium polonicum(P.polonicum)and Trichoderma harzianum(T.harzianum)isolated fromthe groundwater were reported,as well as the effects of water quality parameters were evaluated.In addition,the efficiency of low-concentrations of chlorine-based disinfectants(chlorine,chlorine dioxide and chloramine)on controlling the formation of fungal biofilms was assessed.The results showed that the biofilms formation of the three genera of fungi could be divided into the following four phases:induction,exponential,stationary and sloughing off.The optimum conditions for fungal biofilms formation were found to be neutral or weakly acidic at 28°C with rich nutrition.In fact,A.niger,P.polonicum,and T.harzianum were not observed to form mature biofilms in actual groundwater within 120 hr.Carbon was found to have the maximum effect on the fungal biofilms formation in actual groundwater,followed by nitrogen and phosphorus.The resistance of fungal species to disinfectants during the formation of biofilms decreased in the order:A.niger>T.harzianum>P.polonicum.Chlorine dioxide was observed to control the biofilms formation with maximum efficiency,followed by chlorine and chloramine.Consequently,the results of this study will provide a beneficial understanding for the formation and control of fungal biofilms.Xinyu Luo Xiangqian Xu Ruihua Cao Qiqi Wan Jingyi Wang Huining Xu Yingzi Lin Gang Wen Tinglin Huang 2021Journal of Environmental Sciences2021,33,11:1

11A Catalog of Quasar Candidates Identified by Astrometric and Mid-infrared Methods in Gaia EDR3显示文摘Quasars are very important in materializing the reference frame.The excess emission of active galactic nuclei(AGNs) in the mid-infrared band can be used to identify quasar candidates.As extremely distant and point-like objects,quasars also could be further selected by an astrometry method.Increasing the number of reliable quasar candidates is necessary in characterizing the properties of Gaia astrometric solution and evaluating the reliability of Gaia’s own quasars classification.We identify quasars by using appropriate All WISE [W1-W2] color and different combinations of astrometric criteria.Together with the contamination and completeness,the magnitude,astrometric properties,density distribution,and the morphological indexes of these selected quasars are evaluated.We obtain a quasar candidate catalog of 1,503,373 sources,which contains 1,186,690 candidates(78.9%) in common with the Gaia EDR3_AGN catalog and 316,683 newly identified quasar candidates.The completeness of this catalog is around 80% compared to LQAC5,and the purity of the overall catalog is about 90%.We also found that the purity of quasar candidates selected by this method will decrease in the crowded sky area and the region with less WISE observations.Qiqi Wu Shilong Liao Zhaoxiang Qi Hao Luo Zhenghong Tang Zihuang Cao 2023Research in Astronomy and Astrophysics2023,23,2:0
12Characteristics and Clinical Implication of UGT1A1 Heterozygous Mutation in Tumor显示文摘Background:The literature recommends that reduced dosage of CPT-11 should be applied in patients with UGT1 A1 homozygous mutations,but the impact of UGT1 A1 heterozygous mutations on the adverse reactions of CPT-11 is still not fully clear.Methods:A total of 107 patients with UGT1 A1 heterozygous mutation or wild-type,who were treated with CPT-11 from January 2018 to September 2021 in Peking University Third Hospital,were retrospectively enrolled.The adverse reaction spectra of patients with UGT1 A1*6 and UGT1 A1*28 mutations were analyzed.Adverse reactions were evaluated according to National Cancer Institute Common Terminology Criteria for Adverse Events(NCI-CTCAE) 5.0.The efficacy was evaluated according to Response Evaluation Criteria in Solid Tumors(RECIST) 1.1.The genotypes of UGT1 A1*6 and UGT1 A1*28 were detected by digital fluorescence molecular hybridization.Results:There were 43 patients with UGT1 A1*6 heterozygous mutation,26 patients with UGT1 A1*28 heterozygous mutation,8 patients with UGT1 A1*6 and UGT1 A1*28 double heterozygous mutations,61 patients with heterozygous mutation at any gene locus of UGT1 A1*6 and UGT1 A1*28.Logistic regression analysis showed that the presence or absence of vomiting(P=0.013) and mucositis(P=0.005) was significantly correlated with heterozygous mutation of UGT1 A1*28,and the severity of vomiting(P<0.001) and neutropenia(P=0.021) were significantly correlated with heterozygous mutation of UGT1 A1*6.In colorectal cancer,UGT1 A1*6 was significantly correlated to diarrhea(P=0.005),and the other adverse reactions spectrum was similar to that of the whole patient cohort,and efficacy and prognosis were similar between patients with different genotypes and patients treated with reduced CPT-11 dosage or not.Conclusion:In clinical use,heterozygous mutations of UGT1 A1*6 and UGT1 A1*28 are related to the risk and severity of vomiting,diarrhea,neutropenia and mucositis in patients with Pan-tumor and colorectal cancer post CPT-11 therpy.In colorectal cancer,UGT1 A1*6 is significantly related to diarrhea post CPT-11 use,efficacy and prognosis is not affected by various genotypes or CPT-11 dosage reduction.Qian LI Tao SUN Hua ZHANG Wei LIU Yu XIAO Hongqi SUN Wencheng YIN Yanhong YAO Yangchun GU Yan'e LIU Fumei YI Qiqi WANG Jinyu YU Baoshan CAO Li LIANG 2022中国肺癌杂志2022,25,3:0
13Inactivation and subsequent reactivation of Aspergillus species by the combination of UV and monochloramine: Comparisons with UV/chlorine显示文摘Ultraviolet(UV)/monochloramine(NHCl) as an advanced oxidation process was firstly applied for Aspergillus spores inactivation. This study aims to: i) clarify the inactivation and photoreactivation characteristics of UV/NHCl process, ii) compared with UV/Clin inactivation efficiency, photoreactivation and energy consumption. The results illustrated that UV/NHCl showed better inactivation efficiency than that of UV alone and UV/Cl, and could effectively control the photoreactivation. For instance, the inactivation rates for Aspergillus flavus, Aspergillus niger and Aspergillus fumigatus in the processes of UV/NHCl(2.0 mg/L) was 0.034, 0.030 and 0.061 cm^(2)/m J), respectively, which were higher than that of UV alone(0.027, 0.026 and 0.024 cm^(2)/m J) and UV/Cl(0.023, 0.026 and 0.031 cm^(2)/m J). However, there was no synergistic effect for Aspergillus flavus and Aspergillus fumigatus. As for Aspergillus niger, the best synergistic effect can reach 1.86-log 10. This may be due to their different resistance to disinfectants, which were related to the size, an outer layer of rodlets(hydrophobins) and pigments. After UV/NHCl inactivation, the degree of cell membrane damage and intracellular reactive oxygen species were higher than that of UV alone.
