| 1 | Inflammatory processes in the liver: divergent roles in homeostasis and pathology显示文摘The hepatic immune system is designed to tolerate diverse harmless foreign moieties to maintain homeostasis in the healthy liver.Constant priming and regulation ensure that appropriate immune activation occurs when challenged by pathogens and tissue damage.Failure to accurately discriminate,regulate,or effectively resolve inflammation offsets this balance,jeopardizing overall tissue health resulting from an either overly tolerant or an overactive inflammatory response.Compelling scientific and clinical evidence links dysregulated hepatic immune and inflammatory responses upon sterile injury to several pathological conditions in the liver,particularly nonalcoholic steatohepatitis and ischemia-reperfusion injury.Murine and human studies have described interactions between diverse immune repertoires and nonhematopoietic cell populations in both physiological and pathological activities in the liver,although the molecular mechanisms driving these associations are not clearly understood.Here,we review the dynamic roles of inflammatory mediators in responses to sterile injury in the context of homeostasis and disease,the clinical implications of dysregulated hepatic immune activity and therapeutic developments to regulate liver-specific immunity. | Ola Ahmed Mark W.Robinson Cliona O’Farrelly | 2021 | Cellular & Molecular Immunology2021,18,6: | 5 |
| 2 | Prognostic value of pre-treatment F-18-FDG PET-CT in patients with hepatocellular carcinoma undergoing radioembolization显示文摘AIM To evaluate the value of pre-treatment 18F-FDG PET/CT in patients with HCC following liver radioembolization.METHODS We identified 34 patients with HCC who underwent an FDG PET/CT scan prior to hepatic radioembolization at our institution between 2009 and 2013. Patients wereseen in clinic one month after radioembolization and then at 2-3 mo intervals. We assessed the influence of FDG tumor uptake on outcomes including local liver control(LLC), distant liver control(DLC), time to distant metastases(DM), progression free survival(PFS) and overall survival(OS).RESULTS The majority of patients were males(n = 25, 74%), and had Child Pugh Class A(n = 31, 91%), with a median age of 68 years(46-84 years). FDG-avid disease was found in 19(56%) patients with SUVmax ranging from 3 to 20. Female patients were more likely to have an FDG-avid HCC(P = 0.02). Median follow up of patients following radioembolization was 12 months(1.2-62.8 mo). FDG-avid disease was associated with a decreased 1 year LLC, DLC, DM and PFS(P < 0.05). Using multivariate analysis, FDG avidity predicted for LLC, DLC, and PFS(all P < 0.05).CONCLUSION In this retrospective study, pre-treatment HCC FDGavidity was found to be associated with worse LLC, DLC, and PFS following radioembolization. | 4 | Suppression of monosodium urate crystal-induced inflammation by inhibiting TGF-β-activated kinase 1-dependent signaling:role of the ubiquitin proteasome system显示文摘Monosodium urate(MSU)crystals activate inflammatory pathways that overlap with interleukin-1β(IL-1β)signaling.However,the post-translational mechanisms involved and the role of signaling proteins in this activation are unknown.In the present study,we investigated the intracellular signaling mechanisms involved in MSU-induced activation of THP-1 macrophages and human nondiseased synovial fibroblasts(NLSFs)and the in vivo efficacy of an inhibitor of tumor grovArth factor-β(TGF-β)-activated kinase 1(TAK1),5Z-7-oxozeaenoI,in MSU-induced paw inflammation in C57BL76 mice.THP-1 macrophage activation with MSU crystals(25-200 ng/ml)resulted in the rapid and sustained phosphorylation of interleukin-1 receptor-activated kinase 1(IRAK1 Thr^(209))and TAK1(Thr^(184/187))and their association with the E3 ubiquitin ligase TRAF6.At the cellular level,MSU inhibited the deubiquitinases A20 and UCHL2 and increased 20s proteasomal activity,leading to a global decrease in K^(63)-linked ubiquitination and increase in K^(48)-linked ubiquitination in THP-1 macrophages.While MSU did not stimulate cytokine produaion in NLSFs,it significantly amplified IL-1β-induced IL-6,IL-8,and ENA-78/CXCL5 production.Docking studies and MD simulations followed by TAK1 in vitro kinase assays revealed that uric acid molecules are capable of arresting TAK1 in an active-state conformation,resulting in sustained TAK1 kinase activation.Importantly,MSU-induced proinflammatory cytokine production was completely inhibited by 5Z-7-oxozeaenol but not IRAK1/4 or TRAF6 inhibitors.Administration of 5Z-7-oxozeaenol(5 or 15 mg/kg;orally)significantly inhibited MSU-induced paw inflammation in C57BL/6 mice.Our study identifies a novel post-translational mechanism of TAK1 activation by MSU and suggests the therapeutic potential of TAK1 in regulating MSU-induced Inflammation. | Anil K.Singh Mahamudul Haque Kayla O'Sullivan Mukesh Chourasia Madhu M.Ouseph Salahuddin Ahmed | 2021 | Cellular & Molecular Immunology2021,18,1: | 2 |
| 5 | Frequency of diabetic retinopathy and associated risk factors in Khartoum,Sudan:population based study显示文摘AIM: To assess the frequency and associated risk factors of diabetic retinopathy among Sudanese individuals with diabetes attending Makka Eye complex in Khartoum, Sudan. METHODS: The cross sectional hospital based study recruited 316 individuals with diabetes from Makkah Eye Complex Retina Clinic. Standard questionnaire was used to collect demographic data, medical history and life style characteristics. Blood samples were taken to measure Hb A1 c and lipid profile. Fundus and slit lamp examination were performed for screening of diabetic retinopathy.RESULTS: Among 316 participants, 187(59.2%) were males and 129(40.8%) were females. The mean age of participants was 58.7±10.5y. The overall frequency of retinopathy was 261(82.6%). The percentages of the total participants with proliferative diabetic retinopathy(PDR) were 126(39.9%) and non-proliferative diabetic retinopathy(NPDR) were 135(42.7%). Importantly, duration of diabetes mellitus(DM)(72.2% of more than 10y), being on oral hypoglycaemic drugs(versus insulin), and hypertension were all significant risk factors for diabetic retinopathy(P=0.00, 0.01 and 0.00 respectively). Complications ofdiabetes like diabetic foot(17.7%), history of amputation(6.7%) and clinically significant macular edema(CSME)(47.4%) of the eyes were all significant risk factors(P<0.05). Logistic regression analysis showed that duration of diabetes, hypertension and CSME were found to be absolute risk factors(P=0.007, 0.003 and 0.000 respectively). Duration of DM of more than 10 y have more than double risk(OR=2.8), while having hypertension triples the risk of retinopathy(OR=3.1).CONCLUSION: High rates of diabetic retinopathy are noted among individuals with diabetes attending Makkah Eye hospital in capital Khartoum. Urgent strategies are needed to monitor and treat hypertension and optimize diabetes control in individuals with diabetes. | 6 | Mycogenic Silver Nanoparticles From Endophytic Trichoderma atroviride with Antimicrobial Activity显示文摘There is an increasing interest in developing nanoparticles with diverse biologic activities.To this end,we prepared 10 to 15 nm silver nanoparticles(AgNP)from native isolates of Trichoderma atroviride.Within this study,endophytic fungi hosted four medicinal plants in Saint Katherine Protectorate,South Sinai,Egypt have been isolated by surface sterilization technique on four isolation media.Ten species,based on their frequency of occurrence,out of twenty recovered taxa were tested for their capability to synthesize extracellular AgNPs.Trichoderma atroviride hosted Chiliadenus montanus was found to be the best candidate for the production of mycogenic AgNPs among all examined species.The mycosynthesized AgNPs were compared with chemically synthesized and characterized using Ultraviolet-visible(UV-vis)spectroscopy,Raman spectroscopy,X-ray diffraction(XRD)and high-resolution transmission electron microscopy(HRTEM)techniques.The HRTEM result showed the distribution of spherical AgNPs ranging from 10 to 15 nm.Trichoderma atroviride isolate was subjected to sequencing for confirmation of phenotypic identification.The internal transcribed spacer(ITS)1-5.8 s-ITS2 rDNA sequences obtained were compared with those deposited in the GenBank Database and registered with accession number MH283876 in the NCBI Database.Antibacterial,anticandidal and antifungal effects of chemically and mycosynthesized AgNPs were examined at various concentrations in vitro against six pathogenic bacteria and 4 pathogenic fungi by agar well diffusion technique.Standard antibiotics;Gentamicin,Amoxicillin,Clotrimazole,and Nystatin at 5μg/disk were taken as positive controls,while 5%DMSO was used as the negative control.Our data revealed that the application of mycogenic AgNPs at a concentration of 100 ppm resulted in maximum inhibition of pathogenic bacteria and fungi.These data suggest that AgNPs from native isolates of Trichoderma atroviride(MH283876)offer a source of rapid synthesis of eco-friendly,economical biomaterials that show antimicrobial activities. | Ahmed Abdel-Azeem Amr A.Nada Anthonia O’Donovan Vijay Kumar Thakur Amr Elkelish | 2020 | Journal of Renewable Materials2020,8,2: | 2 |
